During the last couple of decades, molecular techniques are widespread tools for the species diversity. However, their application to taxonomy provoked intense debates between traditional and molecular taxonomists. To prevent every kind of disagreement, it should be required to a threshold for standarizing the species delimitation. Here, we tested three species delimitation methods (ABGD, PTP and bPTP) and compared their results with morphospecies on the widest DNA barcoding dataset. Moreover, a possible threshold for species determination within the superfamily has been suggested. It is believed that it could be an important criterion in detection for hidden or cryptic species, tracing of variation, and can help in identification on immature stages of pests.

Insecticide Resistant Management (IRM) and Positive List System (PLS) is the main keyword in current insect pest management. So we are trying to develop a integrated insect pests control prescription issuing system based on big data which linked National Crop Pest Management System (NCPMS). To manage the insecticide resistance, IRAC suggest some tips for effective control : choosing the effective insecticide and alteration of these insecticides based on MOA. Then, how to distinguish the effective or non-effective insecticide. Until now, bioassay is the right answer. However that is time-consuming, labor-intensive and costly process. So we have been steadily advancing molecular based insecticide resistance diagnosis for supporting and/or substitution of classical bioassay. And it will link to PLS. Here we discuss more detailed methodology of molecular based insecticide resistance diagnosis and its application for the insect pests control prescription issuing system.

Mosquitoes are the most important medical species by transmitting some of deadly infectious diseases to human. Established vector control methods involving the elimination of larval habitats, the use of physical barriers, or those involving insecticides are not sufficient to maintain the populations of these species below the epidemic risk threshold. The sterile insect technique (SIT) is a species-specific, non-polluting and environmentally benevolent method of insect control. This technique involves the release of massive numbers of artificially-reared sterile male insects that, due to their abundance, outcompete fertile wild males for mating opportunities with wild female insects. In this study we discuss optimization of the sterilization process and present a simple model and procedure for determining the optimum dose.

It is known that the growth and development of the mosquito are greatly affected by the change of the meteorological factors. In particular, temperature and precipitation are closely related to the life cycle of the mosquito, and their effects have different characteristics for each species of mosquito. Therefore, to develop a mosquito activity index based on mosquito density, it is essential to develop a prediction model based on weather data. In this study, we developed a functional formula that can estimate the change of mosquito density according to the change of meteorological factors using the mosquito classification data of Incheon region collected from 2011 to 2017. Also, using the data of the digital mosquito monitoring system (DMS) from April to October 2018, mosquito activity index according to characteristics of space in city was developed. In order to reflect the temporal characteristics of the mosquito life-cycle, we used data of temperature and precipitation prior to 1-2 weeks, and used land cover data to reflect the spatial characteristics of mosquito density. Density of Culex pipiens collected in the Incheon area were gradually increased when the average temperature increased two weeks ago after adjusting the precipitation. However, when the average temperature reached 22°C, the density of Culex pipiens marked a peak, and above the 22°C, the density was decreased. The predicted mosquito activity index calculated by applying the machine learning method to the DMS data of the Incheon area is designed to calculate from 1 to 10 grades. The accuracy of the mosquito activity index was 87% when the one grade error was allowed.

Malaria is notorious disease to transmitted by Anopheline mosquitoes. In Korea, Plasmodium vivax malaria was re-introduce in 1993 and exceed to more than 4,000 cases in early 2000s. Based upon our control efforts, it was sharply decreased and diagnosis 576 cases in last year. We made Action Plan in this year for certification of malaria elimination by WHO until year 2024. In this plan, we need to strengthen our capacity of vector surveillance and control. For vector surveillance, we will introduce daily mosquito population monitoring system. In 2020, every malaria high risk area install two mosquito counters and send information to main server. For vector control, we will try building capacity of malaria mosquito control people in the Public Health center and military section. To decrease no. of mosquitoes, we will concentrate our resource to cattle shed, which is main blood source of Anopheline mosquitoes. Based on the ecological characteristic of Anopheline, we will adopt Integrate Vector management (IVM) including physical, chemical and biological method. Based on the reconciliation with North Korea, we will support technical and material support for malaria elimination.

모기는 말라리아, 황열, 뎅기열, 웨스트나일열, 사상충, 치쿤구냐, 일본뇌염, 지카 등의 여러 질병을 매개하고 있고 자연적 요인과 인위적 요인에 의한 지구의 기후변화는 이들 병원성 매개모기들이 점점 더 번성하게 만드는 요인이 되고 있다. 현재 우리나라에서 주목해야 될 감염성 매개질병으로 말라리아와 뎅기열은 기후변화에 의한 기온 상승 요인으로 매개모기의 분포지역 확대와 함께 교통수단의 발달 및 국가 간 빈번한 교류에 의해서 유럽을 비롯한 오세아니아, 동아시아의 온대 지역에서도 감염자의 발생빈도 수가 최근 지속적으로 증가 추세를 보이고 있다. 따라서 기후변화 요인과 인간 활동 등에 의한 요인으로 감염 지역과 매개모기 분포가 미래 예측 불확실성과 복잡성을 가지고 있지만 국내 침입을 막기 위한 해외 유입 방지에 대한 지속적인 모니터링과 방제뿐만 아니라 국가 간 방제 프로그램 공조를 통한 모기에 의해 매개되는 질병 확산에 대비하는 것이 중요할 것으로 생각된다.

Ticks are obligatory hematophagous ectoparasites, which can transmit various pathogens including bacteria, protozoa, and viruses via salivary secretion during feeding. Dopamine is known to be the most potent molecule for tick salivary secretion during blood feeding. Two dopamine receptors were recently characterized in tick salivary glands: dopamine receptor (D1) and invertebrate specific D1-like dopamine receptor (InvD1L). Dopamine’s action controlls the salivary glands by two distinct physiological actions: 1) inward fluid transport into the lumen of salivary glands acini via D1 receptor, 2) emptying lumen solute into salivary duct by pumping and gating via InvD1L. Between blood feeding, ticks can survive long period of time without blood feeding because of their highly developed osmoregulatory mechanisms. Off host ticks can maintain water balance by obtaining atmospheric water vapor via salivary glands. Understanding osmoregulation in ticks is crucial for the development of novel methods to prevent tick-borne disease transmission. S3-5

Among the various expression systems used for foreign protein expression, baculovirus expression system (BES) has the high level of post-translational modification ability such as glycosylation, folding and disulfide bonding. BES is widely used now in the production of VLPs because it is possible for the efficient multi-gene expression. However, there are not many cases of VLPs being manufactured through BES. Therefore, in this study, three improvements were made to increase the productivity of VLP through BES. A new heper enhanced expression vector was constructed to increase the expression of structural proteins of virus-like particles, and baculovirus bacmid was modified to increase production time. In addition, an easy purification system was constructed to efficiently purify VLP, and finally the construction of BES optimized for VLP production was completed.

As world become closer and many people travel around the world, infectious diseases are likely to be prevalent, Dengue, chikungunya, Zika virus would become endemic diseases in many places in the world as climate is more likely to be fit to mosquito vectors than before. As a matter of fact, Aedes albopictus mediating these diseases became endemic species in Japan and France recently. One of the things that we employ is that we need to apply the Internet of Things concept in disease vector control soon to increase the accuracy of vector species in time as they are present unexpectedly. Here in this presentation, many lines of current state-of-the-art technologies to control the disease vectors using big data informatics application and machine learning strategies. With this trend, this presentation will add future directions of integrated vector management with new research endeavors to monitor the population of disease vectors concisely and precisely, Application of internet of things (IoT) in disease vector management will be addressed in this presentation. This will change future research and development of apparatus to monitor and control disease vectors.

The oriental fruit flies, Bactrocera doralis (Hendel) (Diptera: Tephritidae), a tropical pest, have recently been expanded to temperate region neighboring with Korea, with increasing international trade and global climate change. The biological data of several B. dorsalis complex was collected, and the establishment potential in Korea was examined using ecoclimatic index and population model results obtained from CLIMEX and Popmodel, respectively. In the CLIMEX, B. dorsalis could be established in the small part of Jeju Island in the 2010s, and the range gradually spread until the 2090s, and many parts of the lowland of Jeju Island were assigned in the marginal to optimal range. Using the population model and the temporal distribution of the host fruits, it was found that B. dorsalis can use a variety of host plants without time lag, allowing them to increase its population and to damage citrus, the main crop of Jeju.

Jumping plant-lice (Hemiptera: Psylloidea) are known for a few deleterious pest species worldwide, yet the phylogeny of the group has been poorly understood. The goal of researching the Korean Psylloidea is to achieve a total overview of the group. The systematic study relies on three main pillars which are i) the taxonomic review of superfamily Psylloidea in the Korean Peninsula, ii) the correction of taxonomic confusions caused by phenetic plasticity and iii) the molecular phylogeny of Psylloidea to elucidate the higher-level phylogenetic relationships among the families and subfamilies of the superfamily Psylloidea. Discussions of the early diversification and evolution of Psylloidea will be presented together with comments on biogeography and host plant association.

최근 파밤나방 등 다양한 나비목 해충에서 diamide 계통의 살충제 (IRAC group 28)의 저항성이 많이 보고가 되고 있다. 국내에서도 이미 배추좀나방과 파밤나방의 저항성이 보고되었는데, 본 연구에서 사용된 강릉에서 2018년 채집한 파밤나방의 경우 Chlorantraniliprole에 대하여 감수성 대비 2,500배 이상의 높은 저항성을 나타냈다. 추천 농도 처리 시 30% 이하의 방제가를 나타냈으며 Cyantraniliprole, Cyclaniliprole, Flubendiamide에서 높은 교차저항성을 나타냈다. 이 저항성 집단에서 diamide 계통 살충제의 작용점인 ryanodine receptor에 이미 보고된 G4946E 돌연변이는 발견되지 않았으나 새로운 I4760M 돌연변이를 발견하였는데, 2014년 배추좀나방에서 보고된 돌연변이와 위치가 동일하였다. 따라서 I4760M 돌연변이는 파밤나방에서 diamide 저항성에 관여 할 것으로 판단한다.

A residual contact vial plus water (RCVpW) bioassay method was established to monitor insectiside resistance in field populations of the melon thrips, Thrips palmi. Resistance level against six major insecticides were evalutated in five regions to test applicability of RCVpW as an on-site resistance monitoring tool. Reduced mortality in response to six test insecticides were exhibited compared to the RDA susceptable strain showing 100 % mortality, indicating different degree of resistance. An apparently reduced mortality to emamectin benzoate and chlofenapyr was observed in some field populations, suggesting uneven distribution of resistance to these insecticides in field populations. In addition, spinosad resistance was high and widely distributed in the test regions. Synergistic bioassay revealed that cytochrome P450-mediated metabolic factor is involved in spinosad resistance in the Korean population.

A specific serotonin receptor (Se-5HTR) has been identified in the beet armyworm, Spodoptera exigua and classified into 5-HT7 type. Se-5HTR expression was up-regulated in hemocytes and fat body in response to immune challenge. As being a GPCR, this receptor is presumably coupled with intracellular trimeric Gαs protein activating cAMP-dependent protein kinase (PKA) pathway to regulate several cellular functions. RNA interference (RNAi) of Se-5HTR as well as its downstream signal proteins exhibited significant suppression in cellular immune responses including nodulation and phagocytosis. Application of inhibitors to the signaling cascade suppressed the immune responses as well. To validate the Se-5HTR involvement in mediating cellular immunity, 5-HTR knock-out mutants were developed using CRISPR-Cas9 technique and suffered significant developmental anomalies.

The two-spotted spider mite, Tetranychus urticae Koch (Acari: Tetranychidae), is one of the most important pest species, because it devastates many horticultural and ornamental crops and fruit trees. The resistance ratios calculated for the LC50 value in acequinocyl- and pyridaben-resistant strain was 4,237- and 5,555-fold higher than that of the susceptible strain, respectively. The objective of this study was to evaluate the cross-resistance to several acaricides and to identify the mechanisms associated with acequinocyl- and pyridaben-resistant strain of T. urticae.

Aerosol-type insecticides are one of the most widely used household insecticides to control mosquitoes. Not only the structural difference such as the size and shape of orifice, but also the difference in formulation including solvent system and proportion of propellent can affect the efficacy of those aerosol products. Orifice structure can be represented by nozzle diameter while solvent systems are divided into water-based and oil-based types in wide view. In the present study using Asian Tiger Mosquito, Aedes albopictus, we found that the orifice nozzle size made a significant difference in shoot-out range, followed by heterogeneous mortality by distance. Additionally, oil-based spray affected farther space than water-based one. Oil-based solvent system was also responsible for lowering the LT50 and KT50(knock-down time 50) values. Therefore, they are one of the definitive parts in insecticides formulation that determine the way of insecticide delivery. In terms of insecticide resistance management, we define efficiency of insecticide as the proportion of insecticide treated to environment that is solely acted as toxin to target pest. The higher efficiency of insecticide, the less overused chemicals we face, which may imply lower risk of resistance development. Thus, we argue that formulations, the specific route of insecticide discharge to environment, should be deliberately considered not only for consumer satisfaction, but also for insecticide resistance management.

Eurytomidae Walker, 1832 are relatively medium-sized group in the superfamily Chalcidoidea with approximately 1600 species of three subfamilies and 73 valid genera. In Palearctic region, more than 600 species have been recorded, and only 11 species are recorded in South Korea. In this presentation, we report 21 eurytomid species including one subfamily, three genera and 10 species new to South Korea.

The subfamily Lachninae Herrich-Schaeffer, 1854 consist of about 400 species belonging to 19 genera world-wide. Lachninae are distributed mostly in the Nearctic, Oriental and Palaearctic Regions. Their host plants are known as grasses, deciduous tree and conifers. Taxonomic study of Lachninae has been dealt with some authors in several countries, whereas it has rarely performed in the Korean Peninsula after Lee et al. (1994). Here, we review the subfamily Lachninae from the Korean Peninsula, focusing on its taxonomic history and diversity.

The genus Placosternum Amyot & Serville (Heteroptera: Pentatomidae) currently belongs to the tribe Pentatomini Leach with 13 described species. Species are medium to big size and are defined by having characteristic thoracic sternal structures and the pronotal lateral angles somewhat protruded and/or serrate. The genus follows a typical Indo-Malayan distribution limited by the Wallace line south-eastwards and extending to some regions of the eastern Palaearctic. The species in the genus can be considered somehow cryptic as morphologically they are similar and most of them have been classified as a single species. Additionally, as most of the species descriptions are extremely poor, after the revision of the type materials and material deposited in many of the worldwide institutions, we present the systematic knowledge on the genus.

The leafhopper subfamily Deltocephalinae are the biggest group belonging to the Cicadellidae. Deltocephalinae consists of 38 tribes, 923 genera, and 6,683 valid species in worldwide. The genus Scaphoideus Uhler of the subfamily is the largest genus of the tribe Scaphoideini with two subgenera and comprised about 160 species found in the world. Some of Scaphoideus species are serious pests and vectors of phytoplasma such as Scaphoideus titanus, S. luteolus. In this talk, the leafhopper genus Scaphoideus from Korea peninsula is reviewed with one new species and one new recorded species. Additionally, morphological information and descriptions of genitalia, and a key to the species of Scaphoideus are provided.
