본 연구에서는 수확 후 저장 기간에 따른 절화 장미의 수명 변화를 알아보았다. 절화장미 ‘Antique Curl’을 수확 직후 4℃ 암조건의 저장고에서 0일(무처리), 1일, 3일, 5일, 7일간 보존 용액 처리 후 습식 저장하였다. 저장이 끝난 절화는 24℃ 실 내 광환경으로 옮겨 절화 보존제 유무 조건에서 절화수명을 조사하였다. 그 결과, 절화수명은 저장기간 3일차까지는 20일 전후로 높았으나, 5일차 이후 유의적으로 감소했다. 절화 보 존제는 절화수명 연장에 효과적이지는 않았으나 절화의 꽃목 굽음 현상을 완화시켰다. 엽록소 형광(Fv/Fm)은 저장 기간과 관계없이 0.78~0.82 수준으로 나타났으며 저장기간에 의한 광 생리 기능저하는 확인되지 않았다. 결과적으로 절화장미 ‘Antique Curl’의 상업적 저장 기한은 최대 5일로 확인되었다.
Functional dyspepsia (FD) is a gastrointestinal disorder with diverse symptoms but no structural or organic manifestations. Benachio-F® (herein named ‘BF-1’) is an over-the-counter liquid digestive formulated with multiple herbal extracts, which has been reported to improve symptoms of FD. A total two experiments were conducted. First, we examined whether BF-1 can modulate the progression of FD through two experimental rat models. A total of three doses (0.3x, 1x, 3x of the human equivalent dose) were used. In the gastric emptying model, both 1x (standard) or 3x (3-fold-concentrated) BF-1 enhanced gastric emptying was compared with that of vehicle-treated animals. In a feeding inhibition model induced by acute restraint stress, treatment with 1x or 3x BF-1 led to a similar degree of restoration in food intake that was comparable to that of acotiamide-treated animals. Among the constituents of BF, fennel is known for its choleretic effect. Thus, we next investigated whether a novel BF-based formula (named ‘BF-2’) that contains an increased amount of fennel extract (3.5-fold over BF-1), has greater potency in increasing bile flow. BF-2 showed a superior choleretic effect compared to BF-1. Furthermore, the postprandial concentration of serum secretin was higher in animals pretreated with BF-2 than in those pretreated with BF-1, suggesting that the increased choleretic effect of BF-2 is related to secretin production. Our results demonstrate that BF-1 can modulate the pathophysiological mechanisms of FD by exerting prokinetic and stress-relieving effects, and that BF-2 has a better choleretic effect than BF-1.
Mesenchymal stem cells (MSCs) have been recognized as a therapeutic tool for various diseases due to its unique ability for tissue regeneration and immune regulation. However, poor survival during in vitro expansion and after being administrated in vivo limits its clinical uses. Accordingly, protocols for enhancing cell survivability is critical for establishing an efficient cell therapy is needed. CDDOMe is a synthetic C-28 methyl ester of 2-cyano-3,12-dioxoolean-1,9-dien-28-oic acid, which is known to stimulate nuclear factor erythroid 2-related factor 2 (Nrf2)- antioxidant response element (ARE) pathway. Herein, report that CDDO-Me promoted the proliferation of MSCs and increased colony forming units (CFU) numbers. No alteration in differentiation into tri-lineage mesodermal cells was found after CDDO-Me treatment. We observed that CDDO-Me treatment reduced the cell death induced by oxidative stress, demonstrated by the augment in the expression of Nrf2-downstream genes. Lastly, CDDO-Me led to the nuclear translocation of NRF2. Our data indicate that CDDO-Me can enhance the functionality of MSCs by stimulating cell survival and increasing viability under oxidative stress.
부추속 식물은 수선화과의 다년생 초본으로 약 1000여종에 이른다. 한국에서는 24종의 자생종이 분포하며 유전자원으로서 큰 가치를 지닌 분류군이다. 그러나 자생부추속 식물에 대한 연구는 일부 종에 대하여 한정적으로 이루어지고 있으며, 갯부추, 두메부추, 울릉산마늘과 같이 최근 분류되거나 잘 알려지지 않은 종에 대한 연구는 매우 부족하였다. 따라서 본 연구는 자생부추속 4종(강부추, 한라부추, 갯부추, 산부추)에 대한 온도 및 명암처리에 대한 발아특성을 조사하였다. 발아 특성은 발아율, T50, 평균발아일수, 발아균일도, 발아속도로 평 가하였다. 강부추와 갯부추는 5℃에서 50%이상의 발아율을 보였으나 한라부추와 산부추는 전혀 발아하지 않았다. 갯부추 는 남부지방 해변가, 강부추는 강이나 하천주변에 서식하며 한라부추와 산부추는 주로 산지에 서식하는 것으로 알려져있다. 따라서 이러한 차이는 종별 생육지의 환경조건에 따른 차 이로 추정된다. 5℃에서 강부추의 발아율은 명조건(70.0%)보다 암조건(93.3%)에서 향상되었으며, T50과 평균발아일수도 단축되었다. 한라부추는 15℃에서 암조건이 명조건보다 발아율이 증가하고 15℃와 20℃에서는 T50과 평균발아일수가 단축 되었다. 그에 따라 발아속도도 증가하였으며 발아균일도도 상 승하였다. 갯부추는 5℃에서 발아율은 명조건(70.0%)보다 암 조건(83.3%)에서 상승했고, 15℃에서 T50과 평균발아일수가 단축되었으며 발아속도가 증가하였다. 산부추는 15℃와 20℃ 에서 T50과 평균발아일수가 단축되었으며 발아속도가 증가하 였다. 따라서 상대적으로 낮은 온도 처리구에서 명조건보다 암조건이 일부 발아특성을 향상시켰다. 실험에 사용된 4종의 종자는 대부분 15~25℃에서 90%가 넘는 발아율을 보였으나, 다른 발아특성은 온도와 광에 따른 차이를 보였다. 강부추와 산부추의 T50, 평균발아일수, 발아속도는 20℃와 25℃에서 가 장 향상되었고, 한라부추와 갯부추는 25℃에서 T50과 평균발 아일수가 가장 단축되었으며, 발아속도도 가장 빨랐다. 결론적으로 발아특성을 종합적으로 고려했을 때, 강부추는 광과 관계없이 20~25℃, 한라부추는 광과 관계없이 25℃, 갯부추는 암조건 25℃, 산부추는 명조건 25℃와 암조건 20~25℃를 발아 에 적합한 조건으로 제시하였다.
본 연구는 다양한 보존용액이 절화 수명에 미치는 영향과, 절단된 두메부추(Allium dumebuchum H.J.Choi)의 줄기의 수분흡수율 및 상대 생체중을 평가하여 두메부추를 절화로 개발 하기 위해 수행되었다. 두메부추 절화가 수확된 후, gibberellic acid(GA3) 50, 75, 100mg・L-1, 8-hydroxyquinoline sulfate(8-HQS) 25, 50, 100mg・L-1, silver thiosulfate(STS) 0.1, 0.3, 0.5mM, 그리고 Chrysal 8mL・L-1, Floralife 10mL・L-1의 보존용액에 처리되었다. 실험기간 동안 환경조건으로 온도는 24℃로 유지되었으며 상대 습도 43±2.1%, 평균 광도는 22.81±1.2μmol・m- 2・s-1, 일장은 9/15h이었다. 두메부추의 절화수명은 대조구인 절화에서 9.0일로 조사된 것에 비해 GA3 100mg・L-1 보존용액 처리된 절화에서 14.0일로 가장 높게 측정되었다. 수분흡 수율 또한 GA3 100mg・L-1 처리에서 실험이 종료되는 시점에 0.07mL・g-1・d-1로 조사되어 다른 처리보다 높았다. 상대 생체 중은 시판중인 절화수명 연장제 Floralife와 Chrysal 보존용액 처리가 GA3 처리보다 유의하게 높게 조사되었다. GA3를 절화 보존용액으로 사용한 결과, 두메부추는 다른 처리들에 비해 높은 수분흡수율로 절화수명이 대조구보다 평균 5일이상 연장 되었다. 그러나, 8-HQS와 STS 보존용액 처리는 대조구에 비하여 두메부추 절화수명에 큰 차이가 없었다.
Hepatic stellate cells (HSCs) play essential roles in normal and pathophysiological function in liver. In steady state, HSCs contribute to retinoid storage, immune tolerance, and extracellular matrix (ECM) homeostasis. Upon liver injury, they become activated and lead to morphological and functional changes. Studies have demonstrated that activation of HSCs by various stimuli such as toxins, microbial infection, or metabolic overload can promote the fibrotic changes in liver by production of ECM. Herein, we provide current knowledge about the basic characteristics of HSCs and the mechanism by which they are activated.
This study was conducted to investigate effect of feeding beet pulp on reproductive performance, colostrum composition and microbiological characteristics in sows. A total of 12 sows(Landrace×Yorkshire) were randomly allotted three dietary treatments at gestating 90 days. The diets were prepared by adding 0, 5 and 10% of the beet pulp in basal diets. The pigs fed 5% beet pulp showed higher litter size than the 0 and 10% beet pulp treatments, although there was not significant among the treatments. Similarly, birth and 21days weight in piglet were not different among the treatments. In colostrum fat, protein, lactose, citric acid, solid, ffa, true protein were ranged to reference values, but not difference among the treatments. E.coli of feces was lower(P<0.01) in the 5% beet pulp than the 0 and 10% beet pulp treatments. But no effect was observed on Lactobacilli of feces among dietary treatments. These our results indicate the beet pulp has no significant effect of reproductive performance and colostrum composition in sows. But, 5% level beet pulp is effective for reduction of intestinal pathogenic microorganisms.
Royal jelly (RJ) is a well-known functional and medicinal food for human health promotion. Major royal jelly proteins (MRJPs), which are the major protein components in RJ, exhibit antimicrobial activities. However, the identities of the MRJPs of RJ responsible for its antioxidant effects have remained unclear. Here, we report that honeybee (Apis cerana) MRJP 2 (AcMRJP2) acts as an antimicrobial and antioxidant agent in RJ. Using recombinant AcMRJP2, which was produced in baculovirus-infected insect cells, we established the antimicrobial and antioxidant roles of MRJP 2. AcMRJP2 bound to the surfaces of bacteria, fungi, and yeast, which then induced structural damage in the microbial cell walls and led to a broad spectrum of antimicrobial activities. AcMRJP2 protected mammalian and insect cells via the direct shielding of the cell against oxidative stress, which led to reduced levels of caspase-3 activity and oxidative stress-induced cell apoptosis, followed by increased cell viability. Moreover, AcMRJP2 exhibited DNA protection activity against reactive oxygen species (ROS). Our data indicate that AcMRJP2 could play a crucial role as an antimicrobial agent and antioxidant in RJ, suggesting that MRJP 2 is a component responsible for the antimicrobial and antioxidant activities of RJ.
Major royal jelly proteins (MRJPs), important protein components of bee royal jelly (RJ) and exclusive nourishments for queen, exhibit various biological and pharmacological activities. RJ is one of the most studied bee products, but the crucial roles for MRJP2 as an antimicrobial and antioxidant agents remain largely unknown. Here we demonstrated the antimicrobial and antioxidant functions of the Asiatic honeybee (Apis cerna) MRJP2 (AcMRJP2). Recombinant AcMRJP2 of approximately 53 kDa was expressed in baculovirus-infected insect cells, and it exhibited antimicrobial activity against bacteria, fungi, and yeast via binding to microbial surfaces and inducing structural damage in microbial cell walls. AcMRJP2 protected mammalian and insect cells against oxidative damage through shielding of cell membranes. Interestingly, AcMRJP2 exhibited DNA protection activity and DPPH radical-scavenging activity. Altogether, our data demonstrated that AcMRJP2 functions as antimicrobial and antioxidant agents.
