        We evaluated the antioxidative activity of extracts of P. lobata root depending on roasting conditions. P. lobata roots were roasted at three different temperature at 150℃, 200℃, and 250℃ and three different time at 10 min, 20 min, and 30 min respectively. Roasted P. lobata root was extracted using water at 85℃ for 6 h and filtered using filter paper, followed by then evaporated (12±0.3 °Brix) and freeze-dried. The concentration of maker compound puerarin was determined using a high performance liquid chromatography system. 2 phenolic compounds, flavonoid contents, and antioxidant activities of the extract powder were evaluated. Puerarin contents, Phenolic compounds, and flavonoid contents of roasted P. lobata root were higher than those of unroasted P. lobata root. The results of DPPH and ABTS showed that roasted P. lobata root possessed higher antioxidant activity than unroasted P. lobata root. This study suggested that roasting process could be applied to P. lobata root in order to achieve its high quality and functionality.

        This paper presents the development of a 3D printer based piezo-driven vertical micro-positioning stage. The stage consists of two flexure bridge structures which amplify and transfer the horizontal motion of the piezo-element into vertical motion of the end-effector. The stage is fabricated with ABS material using a precision 3D printer. This enables a one-body design eliminating the need for assembly, and significantly increases the freedom in design while shortening fabrication time. The design of the stage was optimized using response surface analysis method. Experimental results are presented which demonstrate 100nm stepping in the vertical out-of-plane direction. The results demonstrate the future possibilities of applying 3D printers and ABS material in fabricating linear driven motion stages.

        Floral pattern plays a vital role in flower structure. The most basic factors that make up the floral pattern are spots and lines in Sedirea japonica. These factors were expressed in a various pattern according to their location and the type of the flower. Frequent expression pattern is higher in the left side of lateral sepal and labellum than petal and dorsal sepal. The expression type in one plant is usually very different according to right or left of sepals, flowering orders, and flowering times and in some individuals, expression pattern remained constant in all flowers. Two hybrids from the cross between Hygrochilus parishii and H. pasishii var. purpureus hybrid (Hongjadan) have shown similar flower characteristics (flower size, flower background color, and pattern distribution) to those of H. pasishii with the exception of the pattern, size, and expression of the number of spots, and pattern shape of labellum including morphology. The flower of an offspring from Hongjadan x S. japonicum (K10) showed petals and sepals without pattern, red flecks, red spot, and red sectors. The shape and back ground color of labellum were similar to those of Hongjadan and S. japonica. The results of this study will be helpful for breeding new cultivars of Sedirea japonica.

        Background : Panax ginseng C. A. Meyer is a slow-growing perennial herb that is cultivated in shading condition. Climate change occur around the world that make a lot of problem such as damage of high temperature, drought, salinity and disease. The problems lower the ginseng productivity that cause income reduction of farmers. To achieve stable ginseng production, development of elite varieities resistant to abiotic and biotic stresses is consistently required. It is very time consuming process in order to develop new ginseng varieties because ginseng flowers after 3 years of growth. So, early selection system of elite line must be established. This study was conducted to develope efficient ginseng breeding techniques for early identification of heat or salinity resistance. Methods and Results : Ginseng petioles was soaked in mixed salts solution consisting of KNO3, KH2PO4, MgSO4․H2O in order to test resistant or susceptible salinity. The degree of resistance was quantified according to damage size. Also, ginseng lines transplanted in pot were treated 46℃ for 1 hour and then chlorophyll fluorescence reaction were measured in order to test resistant or susceptible high-temperature. The measured values such as Fm/Fo, Fv/Fm, Rfd were differentiated between resistant and susceptible line. Conclusion : Several lines showed that they are resistance to high temperature or salinity. The selected lines will be utilized for parents to develop new varieties.

        Background : When ginseng seeds were gathered, the seeds were unripe. To grow immature embryo definitely, special treatment called dehiscence must be performed. Even though dehiscence is completed, most ginseng seeds are on enforced dormancy. The breaking seed dormancy is generally achieved using cold treatment. Also it is reported that gibberellin treatment can replace the treatment. It is very time consuming process in order to develop new ginseng cultivar because ginseng flowers after 3 years of growth. To shorten the ginseng breeding period, it is necessary to establish fast generation progress. Therefore, this study examined the possibility of breaking seed dormancy of ginseng using GA3 treatment and alternating temperature. Methods and Results : Seeds were obtained from local variety fruit which is not inbred. Gibberellin of 100 ppm was treated at seeds for 24 hours. Fixed cold condition was treated on both –2℃ and 2℃. Alternating cold condition was treated on 2℃ and then –2℃, finally 2℃. Fixed and alternating temperature was continued for 15, 30, 45, 60, 90 days that 15 days of alternating temperature is first 2℃ for 5days and then -2℃ for 5days, finally 2℃ for 5days. The other treatment periods such as 30, 45, 60, 90 days mean 10, 15, 20, 30 days respectively. Each of 48 seeds were sowed on tray in greenhouse at 3 replication. Experimental plot was completely randomized. Conclusion : Seeds untreated with GA3 were germinated little and there is no difference between 2℃ and –2℃. Alternating temperature until 60days made no difference with fixed temperature but germination rate increased up to 70.8% when seeds were treated for 90days. Germination of seeds treated with GA3 is much higher than untreated seeds especially combined with alternating temperature.

        본 연구에서는 시중에서 유통 중인 어린이용 화장품 총 125건(n = 125)을 대상으로 하여 파라벤 6종과 벤질알콜, 페녹시에탄올, 소르빈산, 벤조산 등 10종의 보존제에 대한 혼합 사용실태와 함량을 HPLC를 이용하여 조사하였다. 유형별 보존제 검출률은 세정제 63%, 크림류 48%, 자외선차단제 46%, 로션류 38%, 오일이 13%의 순으로 검출되어 총 125건 중 63건(50%)의 제품에서 1종류 이상의 보존제를 사용하고 있는 것으로 조사되었다. 보존제별 검출범위는 페녹시에탄올 0.01 ~ 0.91% (n = 35), 벤조산 0.01 ~ 0.48% (n = 28), 벤질알콜 0.01 ~ 0.78% (n = 9), 소르빈산 0.01 ~ 0.11% (n = 3)였으며, 파라벤류 중 메틸 파라벤과 프로필 파라벤은 0.04 ~ 0.21% (n = 8)와 0.02 ~ 0.09% (n = 8)의 검출범위를 보였고, 에틸 파라벤은 1개 제품에서 0.04%로 모두 배합한도 이내로 검출되었다. 조사결과 어린이용 화장품에서는 파라벤류 보다 페녹시에탄올과 벤조산의 사용빈도가 높았으며 검출된 파라벤류 중에서는 메틸 파라벤과 프로필 파라벤이 주로 혼합 사용되는 것으로 나타났다.

        Tomato fruit color, which is the most visible characteristic among the other fruit traits, is considered to have a substantial influence on consumers. The pink-colored tomatoes with high soluble solids content are considerably preferred especially in Asia compared to the other colors. Generally the pink fruit trait of tomatoes is easily determined by visual examination of intact fruit, however, it is technically determined by the characteristic of the fruit peel. The pink trait is regulated by variations of the SlMYB12(y) gene located on chromosome 1, which controls the accumulation of the naringenin chalcone, which comprises a large proportion of flavonoids. In this study, we developed a derived Cleaved Amplified Polymorphic Sequences (dCAPS) marker and a sequence characterized amplified regions (SCAR) marker in order to discriminate of pink/non-pinktomatoes in the domestic breeding lines. Quantitative RT-PCR analysis indicated that the SlMYB gene is highly expressed in non-pink fruit peel, whereas the expression is significantly lowered in the pink fruit peel. These gene based markers are expected to enhance the efficiency and accuracy of selection pink-tomatoes in tomato breeding programs.

        Polyethyleneglycol-adsorbed–superoxide dismutase (PEG-SOD), has been proposed as an effective agent for reducing free radical-mediated injury. The objective of this study was to investigate a protective effect of PEG-SOD supplementation on ovarian tissue during transplantation. Ovaries from F-1 mice were collected and vitrified. After warming, ovaries were autotransplanted under kidney capsule. Mice were randomly divided into four groups according to dose of PEG-SOD, (0 U/ml, 100 U/ml, 1,000 U/ml and 10,000 U/ml respectively). Grafted ovaries were retrieved 2, 7 and 21 days later. PEG-SOD was treated by intraperitoneal injection once every 48 hours and especially for 21 days group, after first week treatment, PEG-SOD was treated once every 4 days. Morphology of ovaries was assessed histological analysis and ELISA for FSH was performed to evaluate restoration of ovarian function. In 2 days groups, morphologically intact follicle ratio of 10,000 U/ml group was significantly higher than other groups. In 7 days groups, morphologically intact follicle ratio was significantly higher in all treatment groups. In 21 days groups, there was no significant difference of intact follicle ratio in total follicles in all groups but intact primordial, primary and secondary follicles ratio was higher in 10,000 U/ml group. FSH levels in blood serum were decrease as time goes on, but there is no statistical difference in each groups. In conclusion, the data of the present study show that PEG-SOD has a beneficial effect on preservation of the morphologically intact follicle.

        Ovarian tissue cryopreservation and transplantation causes follicle depletion. To overcome this problem, we investigate the effect of Anti-Müllerian hormone (AMH), a follicle recruitment control hormone, supplementation before and/or after mouse ovarian transplantation. A total of 120 5-week-aged BD F-1 female mice were used. The mice were randomly divided into four groups according to AMH doses (0, 5, 25, 125 μg/mL, respectively). AMH was injected intraperitoneally on every other day for a week before, after, or before and after transplantation of ovaries under kidney capsules was performed. One week after transplantation, follicular normality was evaluated by histological analysis and TUNEL assay. In Group A and C, morphologically intact follicle (G1) ratios of AMH treated groups showed no statistically significant difference. In Group B, G1 ratios of 25 and 125 μg/mL of AMH treated groups were higher than those of 5 μg/mL treated group, but there was no improvement in G1 ratio after AMH treatment. In every group, apoptotic follicle ratios did not show any trend according to AMH treatment. Proportions of primordial follicle were not significantly different according to AMH treatment in all groups. The result of the present study demonstrated that AMH treatment during on transplantation of cryopreserved ovaries has no significant effect on follicle survival and prevention of follicle depletion.

        Objective : To investigate the effects of Simvastatin and Methylprednisolone on ovarian tissue cryopreservation and transplantation using mouse models. Methods : The mice were randomly distributed into 1 control and 3 experimental groups. The B6D2F1 mice were given oral Simvastatin (5 mg/kg), intravenous Methylprednisolone (15 mg/kg), or a combination of both at 2 hours before ovariectomy. Same volume of normal saline was given perorally in the control group at 2 hours before ovariectomy. The ovarian tissues were vitrified accrording to our protocols. The vitrified ovaries were warmed 1 week later and auto-transplanted under bilateral kidney capsules. The ovaries and blood sera were collected at 2, 7 or 21 days after transplantation. Histological analysis, TUNEL assay, immuno-histochemistry for CD31, serum AMH level and embryonic development after in vitro fertilization were assessed for evaluation. Results : With regard to the total grade 1 follicle rate, both Simvastatin or Methylprednisolone treated groups were significantly increased at 2, 7 or 21 days after transplantation (except Simvastatin treated group at 7 days). A combination of Simvastatin and Methylprednisolone group was significantly improved in terms of the total G1 follicle rate, apoptotic follicle rate, CD31 positive area and serum AMH after ovarian tissue transplantation. However, there were no statistically difference with respect to the oocyte maturation rate, blastulation rate, and the other embryonic development parameters after in vitro fertilization procedure among the four groups. Conclusion : Our results suggest that combined donor Simvastatin and Methylprednisolone have beneficial effects on the quality and function of transplanted ovarian tissues.

        Platycodon grandiflorum, which is the only species in the genus Platycodon of the family Campanulaceae, is an herbaceous flowering perennial. P. grandiflorum is generally known as bellflower or balloon flower indicating its ornamental uses. It has also been traditionally used as a medicinal crop in East Asia, which is widely employed as an antiphlogistic, antitussive, and expectorant. However, marker-assisted selection and molecular breeding in P. grandiflorum has lagged behind other plants such as pepper and tomato because of the lack of genetic information and effective molecular markers. Transcriptome sequencing provides an effective way to obtain large amount of sequence data when there is no available genome sequence. In this study, we performed a transcriptome analysis in platycodons, which has not been attempted previously. We analyzed simple sequence repeats (SSRs) using RNA-seq data. Di-nucleotide motifs were the most abundant repeats (39% ~ 40%) followed by mono- (26% ~ 32%), tri- (25% ~ 31%), tetra- (1.5% ~ 1.9 %), penta- (0.3% ~ 1%) in three platycodon accessions. Based on the SNP information obtained from RNA-seq analysis, we developed 12 PCR-based markers in Platycodon. The number of alleles ranged from two to seven with the average PIC value of 0.373. These 12 markers were applied to 21 platycodon accessions and a phylogenetic tree was constructed. The markers developed in this study could be introduced in molecular breeding program of platycodons. The SSR information obtained from RNA-seq analysis could be further utilized for developing genic-SSR markers in platycodons. Since platycodon is considered as an orphan crop, which has not been actively deployed for genetic study, the sequence information obtained from this study will contribute to further genetic improvements, genomic information and gene discovery in platycodon

        Genome sequencing researches for considerable numbers of crops and wild plants are being developed. Cytogenetic researches according to chromosome number and size are essential to confirm and comprehend ploidy level and genome size before genome sequencing project is actually conducted. Cytogenetic researches on six food crop plants were carried out by DAPI staining and fluorescence in situ hybridization (FISH) method. Fagopyrum esculentum Moench showed 2n=2x=16, each chromosome length of 1.42㎛ to 1.77㎛, total chromosome length of 13.31㎛, and karyotypic formula of 2n=8m; Phaseolus angularis W.F. Wight, 2n=2x=22, 2.01㎛ to 3.84㎛, total 28.03㎛, 2n=9m+2sm, Perilla frutescens var. japonica Hara, 2n=2x=40, 1.73㎛ to 2.76㎛, total 44.36㎛, 2n=5m+13sm+2st. Chromosome sizes of the other three species such as, Panicum miliaceum L., 2n=2x=36, total chromosome length of 30.83㎛, Sesamum indicum L., 2n=2x=26, 27.39㎛, lpomoea batatas L., 2n=2x=30, total 33.51㎛ were too small for each chromosome type to be identified and analyzed. The result of FISH analysis using 5S and 45S rDNA probe showed species-specific chromosome locations in the genome. These preliminary analyses were carried out to decide which food crop to prioritize for genome sequencing.         The fruit shape is an important character in tomato. OVATE is one of genes controlling fruit elongation in tomato. Two loci suppress the ovate mutation, sov1 and sov2, on chromosome 10 and chromosome 11 respectively. sov1 appears to control neck constriction in the fruits (Rodriguez et al, 2013). We sequenced the genomes of Gold Ball Livingston and Yellow Pear using the Illumina Hiseq 2000 generating 101 PE reads and developed molecular markers tightly linked to sov1. The locus was confirmed by fruit shape index analysis, marker genotyping and progeny testing of recombinants. We find mapped sov1 to a 145 kb interval corresponding to a region comprising two candidate genes. One of the candidate genes for sov1 is SlOFP20 another member of the Ovate Family Protein class. Although there is no difference expression of SlOFP20 in the parents at anthesis, when the gene is expressed very high, the mutation appears to be a 34 kb promoter deletion of SlOFP20 in Yellow Pear, conferring a pear shaped and neck-constricted fruit.
