        Insulin, transferrin and selenium (ITS) complex is reported to improve in vitro development of oocytes and embryos. This study was carried out to investigate the effects of ITS during in vitro culture (IVC) of porcine parthenogenetic and nuclear transfer (NT) embryos on subsequent developmental capacity in vitro. The electrically activated oocytes were cultured in Porcine Zygote Medium (PZM-3) with various concentrations (0, 0.1, 0.5, and 1.0%) of ITS for 7 days. Also, the electrically activated reconstructed embryos were cultured in PZM-3 with various concentrations (0, 0.1, 0.5, and 1.0%) of ITS for 6 days. Addition of ITS to culture medium did not affect development of porcine parthenogenetic embryos in vitro. To test the effect of ITS on the in vitro development of porcine NT embryos, factorial experiments were also performed for in vitro maturation (IVM) medium (TCM-199) with or without 1% ITS and culture medium (PZM-3) with or without 0.5% ITS. Addition of 0.5% ITS to culture medium increased (p<0.05) the proportion of NT blastocysts compared with non-treated group. In contrast, addition of 1% ITS to culture medium was ineffective or had a detrimental effect. Also, addition of ITS only to maturation medium increased (p<0.05) the percentage of NT blastocysts formation compared with the control group. In conclusion, addition of ITS to IVM or IVC medium could improve subsequent blastocyst development of porcine NT embryos.

        To examine the differential protein expression pattern in the 11.5 day post-coitus (dpc) and 18.5 dpc placenta of mouse, we have used the global proteomics approach by 2-D gel electrophoresis (2-DE) and MALDI-TOF-MS. The differential protein patterns of 3 placentae at the 11.5 dpc and 18.5 dpc from nature mating mice were analyzed. Proteins within isoelectric point range of 3.0~10.0, separately were analyzed in 2DE with 3 replications of each sample. A total of approximately 1,600 spots were detected in placental 2-D gel stained with Coomassie-blue. In the comparison of 11.5 dpc and 18.5 dpc placentae, a total of 108 spots were identified as differentially expressed proteins, of which 51 spots were up-regulated proteins such as alpha-fetoprotein, mKIAA0635 protein and transferrin, annexin A5, while 48 spots were down-regulated proteins such as Pre-B-cell colony-enhancing factor 1(PBEF), aldolase 1, A isoform, while 4 spots were 11.5 dpc specific proteins such as chaperonin and Acidic ribosomal phosphoprotein P0, while 3 spots were 18.5 dpc specific proteins such as aldo-keto reductase family 1, member B7 and CAST1/ERC2 splicing variant-1. Most identified proteins in this analysis appeared to be related with catabolism, cell growth, metabolism and regulation. Our results revealed composite profiles of key proteins involved in mouse placenta during pregnancy.

        To study the signaling effect of insulin-like growth factor-Ⅰ(IGF-1), transgenic mice containing IGF-1 Receptor (IGF-1R) cDNA fused to metallothionein promoter were produced by DNA microinjection into the pronucleus of mouse zygote. Three founders were produced with transgenic mice containing IGF-1R gene. Transgenic mice lines contained approximately 4~20 copies of transgenes per cell and transmission of this gene into the progeny with Mendelian manner were determined. The founder mice were mated with normal mice to produce F1 mice and then F2 mice. Transmission rates of IGF-1R transgene in the progeny mice were 25~60% in F1 generation and 40~50% in F2 generation. The mRNA expression of IGF-1R transgene in liver was analyzed using RT-PCR for IGF-1R gene in liver. When body weights of transgenic pups were measured during 4, 10 and 14 weeks after birth, IGF-1R transgenic mice grew faster than non transgenic littermates. This study indicated that growth regulation by IGF-1 signaling through IGF-1R can be elucidated using IGF-1R transgenic mice.

        원형질체 융합에 의한 화합성 및 불화합성 종간 체세포잡종을 얻었다. 화합성 종간인 Pleurotus ostreatus 와 P. florida 의 융합체는 이질핵체 (heterokaryon) 를 형성하였고, 불화합성 종간인 P. cornucopiae + P. florida , P. ostreatus + Ganoderma applanatum, P. florida + Ganoderma lucidum, 그리고 P. ostreatus + Flammulina velutipes 는 합핵체(synkaryon) 를 형성하였다. 이질이핵체는 동일한 양상의 자실체를 형성하는데 비해 합핵체는 유사분열상의 꺽쇠연결체 형성, 한쪽 친과 유사한 자실체 형성, 비정상적 유전형질 분리 및 유전자재조합 현상을 나타내었다. 화합성 및 불화합성 계통간 융합체의 RAPD 분석결과 화합성 종간 융합체는 동일한 DNA 패턴을 나타내었고, 불화합성 종간 융합체는 한쪽 친과 유사한 DNA 양상이면서 비양친 DNA 밴드도 형성하였다. 합핵체의 패턴은 microgenome insertion type 과 macrogenome insertion type 으로 구분되었다. 합핵체의 자실체 발생은 융합 모균주 양친의 자가임성에 의존하는데 이는 느타리의 동형핵체 자가임성과 유사한 양상이었고, 교배형 전환과 관련이 있는 것으로 사료된다. 여기서는 이러한 관점에서 논할 것이다.

        진흥(振興)과 유신(維新)을 공시(供試)하여 이앙시기(移秧時期), 재식밀도(栽植密度), 질소시비(窒素施肥) 수준(水準)과 벼잎집무늬마름병(病) 발생(發生)과의 관계(關係)를 검토(檢討) 하였다. 잎집무늬마름병(病) 발생(發生)은 5월(月) 26일(日) 이앙구(移秧區)보다 5월(月) 16일(日) 이앙구(移秧區)에서, 당 100주(株) 재식구(栽植區)가 80주(株) 재식구(栽植區)보다 , 질소질배비구(室素質倍肥區)(진흥(振興) 220kg/ha, 유신(維新) 300kg/ha) 가 보비구(普肥區)에서보다 발병(發病)이 심(甚)하였다. 또한 생육기간중(生育期間中) 발병(發病)이 심(甚)했던 재배조건(栽培條件)에서 성숙기(成熟期)의 피해도(被害度)가 높았다

        Background : Insect pests damages are increasing on the field of Lycium chinense under organic farming management and especially, pest damage of Gelechiidae (Ilseopsis parki Povolny) is serious. Currently, various organic farming materials are registered, but insecticidal activity against Gelechiidae was not verified. Therefore, this experiment was carried out to screening for effective organic farming materials showing insecticidal activity in Gelechiidae. Methods and Results : Screening for effective organic farming materials (OFM) showing insecticidal activity in Gelechiidae, we screening OFMs through in vitro testing Among them, OFMs showing insecticidal activity were first selected through field test. In the field test method, OFMs was sprayed at the time of moth occurrence, after 5 days of treatment, insecticidal activity was investigated as a result, a total of five OFMs were selected through the in vitro test and the Sophora japonica and three plant extracts showed the best control effect at 81.7%. in 2017. In 2018, the 2017 selected OFM and 4 kinds of new OFMs were field tested. The field test was performed in the same method as before. As a result, 2 kinds of OFMs made of Sophora japonica and three plant extracts extract and Sophora flavescens extract showed high control value of 87.3% and 88.3%, respectively. In contrast one organic agricultural material showed a chemical injury. To determine the timing of control, the extract was treated before and after topping and pretreatment of topping showed better control value by 80%. Conclusion : Insecticidal activity against Gelechiidae was not effective in all tested OFMs, but the 2 OFMs showed a high control value of over 80%. In this study, we selected 2 materials out of 9 OFMs against Gelechiidae, and It was effective to treat OFM before topping.
