        나노 크기인 5nm 이하로 제조한 은나노제품(파이투패치)은 주요 식물병원균인 탄저병원균(Collectotrichum gloeosporioides), 잿빛곰팡이병원균(Botrytis cinerea), 균핵병원균(Sclerotinia sclerotiorum)에 대해 포자발아 및 균사생장을 억제하는 항균력이 있었다. 파이투패치 살포에 의한 고추탄저병 방제효과를 실험하기 위해 파이투패치 희석액에 포자를 침지하여 포습시킨 후 발아율을 조사한 결과 5ppm까지 희석한 처리구에서 병원균의 포자발아억제효과를 보였으며, 균사는 10ppm에서 생장억제효과가 15일간 지속되었다. 특히 파이투패치를 10ppm으로 희석하여 배지표면에 도말한 후 탄저병원균 포자를 접종하면 3일간 발아가 억제되어 식물체 감염을 효과적으로 예방하였으며, 40% 이상 발병한 시험구에 4ppm 파이투패치를 살포한 결과 21일 후 7% 이하의 발병과율로 무처리 대비 70% 방제효과가 있었다. 장마철 탄저병 발생율이 94.6%인 시험포장에서 10ppm 농도로 파이투패치를 7일간격으로 엽면살포한 결과 발병과 발생율이 5.8%로 방제효과를 확인하였으며, 수확한 홍고추를 자연건조한 후에도 발병과율이 24.2%로 건고추 수확량도 증가하였다. 장마철 고추역병(Phytophthora capsici)은 장마가 끝난 무피복시험구에서 8월 11일 15%이었으며 고온기를 지난 9월 7일에는 발병율이 74%로 수확을 포기하였으나, 파이투패치를 코팅처리한 피복재를 씌운 시험구에서는 발병주가 2.3%로 장마철 역병발생이 효과적으로 방제되었다.

        The study indicated that antimicrobial activity about gram positive and gram negative bacteria of ginger-oleoresin(GO) extract with the condition of ethanol and supercritical fluid extractions. As the concentration of extraction increases, the clear zone of GO ethanol extract also increased dependently. This led the antimicrobial activity of gram positive bacteria to take bigger place than gram negative bacteria especially in Listeria monocytogenes. There was a high antimicrobial activity in E-III treatment where the ratio of the ginger powder extract to ethanol extraction was 1:6. It was quite effective to treat the antimicrobial activity of GO ethanol extract under 80oC and there was not big difference in the intervals which were the extraction time - 1 to 7 hours. The antimicrobial activity of supercritical fluid extract seemed to take the biggest place in Listeria monocytogenes. From the supercritical fluid extract, it was shown the strong ability of antimicrobial activity in the condition with 100 bar 35oC, 250 bar 35oC and 250 bar 65oC. Furthermore, according to the case of solvent extract, there was not any significant difference in the antimicrobial activity with condition of extraction. However, there was significant antimicrobial activity in E-III treatment of 100 bar and 500 bar of extraction pressure, and 35oC and 65oC of extraction temperature.

        미강으로부터 분리 정제된 피틴산을 식육에 적용하여 식육에서의 항균활성과 항산화능을 조사함으로써 식품보존제로서의 가능성을 확인하였다. 피틴산이 첨가된 Tryptic soy broth에 S. Typhimurium KCCM 11806을 배양하였을 경우 1%(w/v) 피틴산 첨가 시 S. Typhimurium의 생육을 완전히 저해시켰다. E. coli O157:H7에 대한 항균 효과는 1.0%(w/v) 피틴산 첨가 시 약 3.0 log 단위의 생육저해효과를 확인하였다. 신선한 닭고기, 돼지고기, 소고기에 S. Typhimurium을 접종하고 피틴산을 첨가하여 37oC에서 24시간 배양하여 피틴산의 항균효과를 조사한 결과 모든 식육에서 항균효과가 피틴산 농도에 의존적으로 나타났으며 1%(w/v) 피틴산 첨가 시 약 3.0 log 단위의 항균효과를 나타내었다. 생육인 닭고기, 돼지고기, 소고기에 피틴산을 여러 농도로 첨가한 결과 E. coli O157:H7에 대한 항균효과는 모든 생육에서 0.5% 피틴산 첨가 시 약 2.0 log 단위의 생육저해 효과를 보여 주었으며, 가열 처리된 식육에서도 첨가된 피틴산 농도에 의존적으로 유의성 있게 증식을 억제하였다. 생육에 피틴산을 첨가하여 4oC에서 3일간 저장할 경우 저장기간이 증가함에 따라 TBARS(thiobarbituric acid-reactive substances) 값이 증가하였으며 피틴산을 5 mM 첨가하였을 때 TBARS 값이 유의적으로 감소하여 식육에서의 항산화능을 확인하였다.

        본 연구는 probiotics 균주 선발을 위해 동치미로부터 식중독균에 대해 높은 항균활성을 보이는 유산균을 분리동정하여 배양조건을 확립하고, 위산, 담즙산, 췌장액에 대한 저항성 실험을 비교, 분석하여 probiotics 균주로 사용 가능성을 탐색하고자 하였다. 2% CaCO3를 첨가한 MRS agar를 이용하여 김치 무리로부터 식중독균의 저해능이 우수한 8 균주를 분리하였다. 식중독균에 대해 항균활성실험 결과 J4가 B. cereus는 19.85±0.53 mm, L. monocytogenes는 18.37±0.38 mm, S. aureus는 22.49±0.37 mm, E. coli는 17.90±0.64 mm, S. enterica는 16.28±0.53 mm, V. parahaemolyticus는 24.59±0.29 mm의 생육저해환을 형성하여 가장 높은 항균력을 나타내었다. J4 균주의 16S rDNA 염기서열을 분석한 결과 L. sakei와 99% 상동성을 나타내어 L. sakei J4라 명명하였다. L. sakei J4의 최적배양조건은 pH 8.5, 30, 정치배양시 가장 활발하게 생육이 진행됨을 확인하였다. L. sakei J4는 인공위액에서의 생존율 24.3±3.0%, 담즙산에서의 생존율 25.8±0.4%, 췌장액에서의 생존율 101.2±4.5%로 다른 균주보다 상대적으로 생존율이 높았으며, 향후 alginate를 이용한 캡슐화, chitosan을 이용한 재코팅 등의 연구방법을 정립하여 인공위액에 대한 저항성을 높인다면 프로바이오틱스 균주로서 조건을 충족시킬 수 있으리라 사료된다.

        To determine current rate of antimicrobial resistance, a total of 236 isolates from milk samples of dairy cattle with mastitis in Korea during 2010-2011 were examined against 12 antimicrobials using disc diffusion method: 67 Staphylococcus aureus, 74 coagulase-negative Staphylococcus spp. (CNS), and 95 Escherichia coli isolates. The isolates examined in this study were submitted by Local Veterinary Service Laboratories located in 13 provinces and metropolitan cities nationwide. The highest rates of resistance among S. aureus isolates were against ampicillin (56.7%) and penicillin (56.7%), followed by kanamycin (11.9%). All S. aureus isolates were sensitive to lincomycin, amikacin, and cephalothin. Only one isolate showed resistance to tetracycline and oxacillin, respectively. Less than 10% of the S. aureus isolates presented resistance to erythromycin, neomycin, and gentamicin. Among CNS isolates, the most frequently observed resistance was to lincomycin (44.5%), followed by penicillin (28.3%), ampicillin (18.9%), tetracycline (17.5%), kanamycin (13.5%), and erythromycin (9.4%). All or most of the CNS isolates were sensitive to cephalothin, amikacin, neomycin, and gentamicin. The highest rate of resistance among E. coli isolates was against tetracycline (26.3%), followed by streptomycin (21%), neomycin (15%), kanamycin (12.6%), and gentamicin (10.5%). Amikacin was the only antimicrobial to which no E. coli isolates showed resistance. Around 10% of the S. aureus isolates and 15% of the CNS isolates showed resistance against three or more antimicrobials simultaneously, while more than 30% of the E. coli isolates did.

        The present study was evaluated the antibacterial effect of the combination of Coptidis rhizoma, Glycyrrhiza uralensis Fischet, Schizandra chinensis and Corni Fructus(1:1:1) extracts(CGSC10). Furthermore, the effectiveness of CGSC10, sodium chlorate, and the combination of CGSC10 and sodium chlorate(CGSCS10) against E. coli O157:H7 infection was studied using ICR female mice. During the incubation period, the dose of 5, 10, and 20% CGSC10 was inhibited the growth of E. coli O157:H7 by 34.7, 60.2, and 76.4%, respectively. For 7 days after single challenge with E. coli O157:H7, forty female ICR mice were divided into four experimental groups which were administered in drinking water with saline, 10% CGSC10, 15 mM sodium chlorate, and CGSCS10, respectively. On the 3rd day, the number of E. coli O157:H7 in mouse feces was significantly decreased by administration of CGSC10, 15 mM sodium chlorate, and CGSCS10 (p < 0.001). On the 7th day-after administration, CGSC10, sodium chlorate, and CGSCS10 were decreased the number of E. coli O157:H7 by 27.1, 67.7, and 83.3%, respectively. According to the results of the present study, administration of CGSCS10 to mice can reduce the severity of E. coli O157:H7 infection. In addition, it is suggested that CGSCS10 represents a good candidate for the treatment of enteric infections in domestic animals.

        This study was carried out to investigate efficacy of aerosol sanitizer with natural antimicrobial and organic acids against Escherichia coli O157:H7, Salmonella Typhimurium and Listeria monocytogenes. The artificially inoculated pathogens on stainless steel coupon were treated with grapefruit seed extract (GFE), acetic acid,citric acid and lactic acid in model cabinet for 5 min. The number of three foodborne pathogens with individual treatment was reduced by 0.34-3.77 log units, treatment with GEF + organic acid was reduced by 1.72-3.89 log units and treatment with GEF + organic acid + alcohol was reduced by 1.46-5.05 log units. By treatment with GEF + lactic acid + alcohol in scale-up model system for 10 min. Populations of E. coli O157:H7, S. Typhimurium and L. monocytogenes were reduced by 3.42, 2.72 and 2.30 log units from the untreated control respectively. From the above result,aerosol sanitizer with natural antimicrobial agents and organic acid can be used as an environmental sanitation method with satisfying the consumer demand on safe food.

        Salmonellosis constitutes an important public health problem in both developing and developed countries, including Korea. The aims of present study were to investigate the serovar and antimicrobial resistance of Salmonella isolated from food animals and animal products in slaughterhouses and farms. A total of 323 Salmonella were isolated from food animals (n=277) and meats (n=46) during 2010. Of the isolates, 21 different serovars were identified. The predominant serovars were S. Rissen (35%) and S. Montevideo (24.3) in healthy pigs, while S. Enteritidis (25.5%) in healthy chicken. S. Typhimurium (88.8%) was predominant in disease pigs, while S. Gallinarum (29.2%) and S. Montevideo (26.9%) were in diseased chickens. Among meat samples, S. Typhimurium (57.1%) was the most common serovar in pork but S. Enteritidis (38.7%) and S. Montevideo(32.3%) were in chikcen meats. Analysis of antimicrobial resistance patterns revealed that 20.7% of the isolates were sensitive to all the 15 drugs tested. The isolates were frequently resistant to nalidixic acid (47.7%), tetracycline (38.4%), streptomycin (33.7%), and ampicillin (32.8%). The resistance to quinolone and 3rd generation cephalosporins was higher in chicken and chicken meat isolates. Of the 323 isolates, 174 (53.9%) were resistant to one or more CLSI subclass, and 117 (36.2%) showed multiple-resistance. Our findings showed that multiple resistant Salmonella organism are widespread in animals and animal products in Korea. To prevent the transmission or exposure for consumers of antimicrobial resistant Salmonella, policies and guidelines aiming at prudential use of critical antimicrobials for humans are needed.

        Campylobacterosis is the most common food borne bacterial disease in many countries. Food animals and animal products are considered to be the reservoir of the Campylobacter species. The aim of this study was to investigate the antimicrobial resistance of Campylobacter spp. from food animals and raw meats in slaughterhouses. A total of 90 Campylobacter jejuni (C. jejuni) and 127 Campylobacter coli (C. coli) were isolated, for which antimicrobial susceptibility was examined using broth dilution method. Resistance to macrolide antimicrobials was higher among C. coli isolates than among C. jejuni. Among both C. jejuni and C. coli isolates, the most frequently observed resistance was to nalidixic acid, ciprofloxacin, and tetracycline. No erythromycin resistance was observed among C. jejuni isolates from cattle, pig and beef. However, 28.3% (n=13) and 25% (n=3) of C. coli isolates from pigs and pork showed resistance to erythromycin, respectively. The predominant profile of multiple resistance among C. jejuni and C. coli isolates was ciprofloxacin/tetracycline/nalidixic acid resistance (46.7%) and ciprofloxacin/nalidixic acid resistance (31.5%), respectively. This finding has important implication for food safety and public health.

        Staphylococcus aureus is an important food-borne pathogen, which is present on the skin and mucosa of animals. Some of the S. aureus strains are causative agent of food poisoning syndrome. The aim of this study was to investigate the antimicrobial resistance of S. aureus isolates from raw meats in slaughterhouses during 2010. From 17,874 raw meat samples tested, a total of 190 S. aureus were isolated, for which antimicrobial susceptibility to 17 agents was examined using broth dilution method. Among isolates from beef, chicken and pork, 20 (51.3%), 20 (24.7%) and 9 (12.9%) were sensitive to all antimicrobials tested, respectively. Isolates from pork and chicken meats showed much higher resistance, compared to isolates from beef. Penicillin resistance was the most frequent among isolates from beef (35.3%) and pork (75.7%), while tetracycline resistance was among those from chicken meats (48.1%). A total of 3 methicillin resistant S. aureus (MRSA) were detected from beef (5.1%, 2/39) and pork (1.4%, 1/70). Although the prevalence of MRSA was low, the presence of antimicrobial resistant S. aureus such as MRSA suggests that further investigation and strict surveillance on MRSA and antimicrobial resistance are needed.

        Antimicrobial peptides are widely found in living organisms and are known to play a critical role in innate immunity. Numerous antimicrobial peptides from diverse species appear to be effective against pathogenic microorganisms of bacteria, fungi, protozoa, and viruses. Because antibiotic resistance is a global health issue in the fight against pathogenic microorganisms, there has been an urgent need for development of new antibiotic substances. In the current study, we performed yeast two hybrid screening using Beclin1 bait in order to find new peptide antibiotics from a random peptide library. Two candidate peptides from the screening were expressed in a yeast secretory system of Pichia pastoris and tested for any antimicrobial activity against Staphylococcus aureus, MRSA, MRSA2242, MRSA2250, Lactobacillus casei, and Lactobacillus acidophilus. Disc clear zone assay and spectrophotometric analysis revealed that the two peptides exert a decent activity against the pathogenic bacteria, in contrast to minimal effect on the commensal Lactobacillus strains. Taken together, this study presents novel peptides with antibacterial activity against the pathogenic forms of Staphylococcus aureus and suggests the possibility that these peptides, upon further characterization, may be developed as clinically useful antibiotics.

        N-(fluorodichlomethylthio) phthalimide-silver complexes were prepared and investigated the antimicrobial activity on rubber mat manufactured with waste rubber. We are exposed to harmful bacteria and fungi all the time. We manufactured antimicrobial mat to be imposed to mats that it can prevent generation of bacteria and microorganisms, and restrict their reproduction. Infection of medical devices causes significant morbidity and mortality. For aim of this study, we measured the antimicrobial mat manufactured with N-(fluorodichlomethylthio) phthalimide-Ag complex by CCD, FT-IR and NMR. The effect of mole ratio of N-(fluorodichlomethylthio) phthalimide-Ag complex on antibacterial activity to bacteria and fungi is investigated. Reduction rate is evaluated using the Quinn method. Escherichia Coli is effectively inhibited than Staphylococcus aureus by antimicrobial mat.

        The antimicrobial susceptibility of the 41 Bordetella bronchiseptica isolates was tested by using the Kirby-Bauer agar disk diffusion method. The B. bronchiseptica isolates were found to be sensitive to amoxicillin/clavulanic acid (100%), gentamicin (100%), neomycin (100%) and amikacin (97.6%), whereas they were resistant to streptomycin (100%), trimethoprim/sulfamethoxazole (100%), penicillin (100%) and ampicillin (97.6%). All the B. bronchiseptica isolates resisted to at least 4 antimicrobial agents and totally 8 different combinations of multiple antibiotic resistance patterns were noted. All of the B. bronchiseptica isolates, except one, were simultaneously resistant to streptomycin, trimethoprim/sulfamethoxazole, penicillin and ampicillin. The observed antibiotic resistance is not plasmid mediated as plasmids were absent from all the B. bronchiseptica isolates.

        Bacillus thuringiensis (Bt) is characterized by its ability to synthesize crystal toxins and also able to produce bacteriocins such as thuricin, tochicin, entomocin and bacthuricin. The present work, for the first time, describes the biological activity of bacteriocins from B. thuringiensis subsp. cameroun (Btc). Supernatant which was produced from a liquid culture of Btc had antimicrobial activity against Bacillus cereus, ending up to making a inhibition zone on an agar medium. A significant reduction in antimicrobial activity was observed when the supernatant was exposed to heat at 75~100°C for 15 min. Proteins were separated from the supernatant by a fast protein liquid chromatography (FPLC) given the thermal instability. A group of FPLC fractions had antimicrobial activity against Bt subsp. palmanyolensis, israelensis, 1-3, morrisoni, toguchini and kurstaki, and B. cereus ACTC21768, ATCC14579 and NRRLB-569. Interestingly, when the supernatant was individually incorporated into the liquid cultures of Bt subsp. israelensis (Bti) and mogi (Btm) with mosquitocidal activity, a vegetative cell growth was observed only in the Btm culture 10 h post-incubation. A possible recovery of vegetative Btm cell growth was observed, compared to a control without the supernatant. These results suggest that Btc produced proteinous antimicrobial substances, one of which may be used as a selection marker to separate Btm after possibly conjugating the two mosquitocidal strains.

        Ursolic acid is a triterpenoid compound present in many plants. This study examined the antimicrobial activity of ursolic acid against mutans streptococci (MS) isolated from the Korean population. The antimicrobial activity was evaluated by the minimum inhibitory concentration (MIC) and time kill curves of MS. The cytotoxicity of ursolic acid against KB cells was tested using an MTT assay. The MIC90 values of ursolic acid for Streptococcus mutans and Streptococcus sobrinus isolated from the Korean population were 2 μg/ml and 4 μg/ml, respectively. Ursolic acid had a bactericidal effect on S. mutans ATCC 25175 T and S. sobrinus ATCC 33478 T at > 2 × MIC (4 μg/ml) and 4 × MIC (8 μg/ml), respectively. Ursolic acid had no cytotoxic effect on KB cells at concentrations at which it exerted antimicrobial effects. The results suggest that ursolic acid can be used in the development of oral hygiene products for the prevention of dental caries.

        This study was carried out to investigate the antimicrobial resistance pattern and distribution of resistance gene determinants in fecal E. coli from chicken. Antimicrobial susceptibility test was performed on a total of 109 fecal E. coli isolates from chicken, collected in Gyeonggi, Chungbuk, Jeonnam and Jeonbuk province from March to November 2003, by the disk agar diffusion method. Eighteen commonly used antimicrobial agents approved in Korea as veterinary medicine were tested: ampicillin (AM), amoxicillin/clavulanic acid (AMC), cephalothin (CF), cefozolin (CZ), cefoxitin (FOX), cefotaxime (CTX), cefepime (FEP), imipenem (IPM), streptomycin (S), gentamicin (GM), amikacin (AN), ciprofloxacin (CIP), enrofloxacin (ENO), norfloxacin (NOR), trimethoprim/sulfamethoxazole (SXT), erythromycin (E), chloramphenicol (C) and tetracycline (TE). Higher resistance rates (≥50%) were observed against 9 antimicrobial agents including AM, CF, S, CIP, ENO, NOR, SXT, E and TE. Resistance was most frequent for TE in 105 E. coli isolates (96.3%). Twenty-two isolates (20.2%) of the isolates showed multiple antimicrobial resistance to 8, and 19 isolates (17.4%) showed to 7 antimicrobial agents. The distribution of the resistance gene determinants for S and TE was assessed by PCR in resistant isolates. Thirty isolates possessed the strA, strB, and aadA gene, 25 isolates possessed the strA and strB gene among the 66 streptomycin-resistant isolates. Fifty one isolates possessed only the tetA gene, 22 isolates possessed the tetA and tetB gene, 11 isolates possessed only the tetB gene among the tetracycline-resistant isolates.

        In this study, an enthalpy exchanger was coated by silver nano particles via spark discharge method and its antimicrobial and heat exchange efficiencies were evaluated. A method utilizing thermophoretic force was used to improve coating efficiency. Four spark discharge systems were parallel connected and generated silver nano aerosol particles (number concentration of 1.65×108 particles/cc, mode diameter of 31 nm). The coating efficiency was evaluated according to various face velocities (V=0.25~1 m/s) and temperature gradients ((Thot-Tcold)/Thot=0~0.09). The maximum coating efficiency was 90.8 % when the face velocity was 0.25 m/s and the temperature gradient was 0.09 (Thot=30℃, Tcold=2℃). Silver nano particles were coated onto the enthalpy exchange element and two different coating amounts of silver nano particles (0.11 ㎍/cm3 , 0.22 ㎍/cm3 ) were tested. For evaluation of antimicrobial efficiency, the suspension test method with E. coli was used. After the suspension test method, CFU(colony forming unit)s of each test sample were counted and colony ratio was calculated. The colony ratio was decreased more quickly when the amount of coated silver particles was increased. When the contact time between each sample and suspension was over 3 hours, antimicrobial efficiencies of coated samples were more over 99.9 % for both amount of silver nano particle(0.11 ㎍/cm3 , 0.22 ㎍/cm3 ). The coating of silver nano particles did not affect the heat exchange efficiency.

        The incidence of multidrug-resistant strains of Staphylococcus aureus continues to increase in hospitals and the community all over the world. As the importance of miner surgery in dentistry gets greater recently, the frequency in use of antibiotics has been increasing. Although several cases of acquired infection in oral cavity in dental hospital has been reported, the research on isolation of S. aureus and evaluation of its antibiotic resistance from outpatients in the dental clinic has not been found yet. The aim of this study was to evaluate the antimicrobial resistance of S. aureus isolates. The isolation rate of S. aureus were 41.7% in dental hospital personnel, 60.4% in outpatients and 62% in dental hygiene students. S. aureus which showed resistance to penicillin were 88.6%, ampicillin 88.6%, erythromycin 8.6%, tetracycline 4.3%, oxacillin 4.3%, cefoteten 4.3%. Six strains of S. aureus showed the susceptible to all antibiotics tested. Sixty-four(91.4%) strains were multiple antibiotic resistance.         Oleanolic acid is a natural triterpenoid that exists widely in foods and some medicinal herbs. The purpose of this study was to determine the antimicrobial activity of oleanolic acid against Streptococcus mutans strains isolated from a Korean population. Antimicrobial activity against these bacteria was evaluated by minimal inhibitory concentration (MIC) and time kill curves. The tolerance of human gingival fibroblasts and human periodontal ligaments to oleanolic acid was tested using a methyl thiazolyl tetrazolium (MTT) assay. The MIC90 value of oleanolic acid for both S. mutans and S. sobrinus isolated from Koreans was 8μg/mℓ. Oleanolic acid showed bactericidal effects against S. mutans ATCC 25175T and S. sobrinus ATCC 33478T at 1 × MIC(8μg/mℓ) and had no cytotoxic effects against KB cells at this dose. The results suggest that oleanolic acid could be useful in the future development of oral hygiene products for the prevention of dental caries.
