        Liquid-state fermentation (LSF) and solid-state fermentation (SSF) were carried out to produce enzymes and β-glucan. The modified synthetic medium composition in liquid-state fermentation (LSF) was determined to be 5% (w/v) whole oat flour, 2% (w/v) tryptone, and 2% (w/v) rice bran. The different humidity conditions of oat by solid-state fermentation were evaluated in terms of maximum production of fungal biomass, amylase, protease, and β-glucan, which were 0.23 mg/g, 7,157.38 U/g, 413.67 U/g, and 8.26% (w/w), respectively, at 60% of humidity condition. Moreover, α-glucosidase inhibition activities of fermented oat with 60% humidity at 48 h were shown to be high 49.08% (w/w). Therefore, liquid- and solid-state fermentation processes were found to enhance the overall fermentation yields of oat to produce enzymes and β-glucan.

        The structure of isoflavone moiety in soybean could be altered by thermal treatment. Thus, this study analyzed the isoflavone profiles of raw soybean extract (RSe, no pretreatment), blanched soybean extract (BSe, at 85oC for 3 min), and cooked soybean extract (CSe, at 95oC for 30 min) and measured their α-glucosidase inhibitory activities. The content of malonylglucosides in RSe decreased considerably during their transformation into BSe and CSe. The total content of isoflavones in RSe and BSe was fairly similar, although there was a significant difference in the respective values for RSe and CSe. It was concluded that the cooking treatment significantly impacted soybean isoflavone content and, eventually, one-quarter of isoflavones were lost in CSe. According to authentic isoflavone tests, the inhibition of α-glucosidase by isoflavone aglycones was more effective than that of the corresponding isoflavone glucosides. The α-glucosidase inhibitions were observed in the order of BSe (70.0%), CSe (53.3%), and RSe (32.3%). However, it was challenging to identify which isoflavone derivative had contributed to the phenomenon mentioned above. Hence, the blanching of soybean seemed to be more appropriate than cooking for preparing soybean extract to inhibit α-glucosidase due to the higher loss of isoflavone during cooking.

        This study investigated optimal extraction conditions for applying Capsicum annuum L. leaf as a functional food resource. Capsicum annuum L. leaf was extracted at different extraction solvents (water and 95% ethanol), extraction temperatures (80oC and 100oC), and e xtraction times ( 30, 60, and 9 0 min), a nd t he extracts were e valuated for extraction yield, luteolin content as a major flavonoid component, antioxidant activity, and α-glucosidase inhibitory activity. The extraction yield, luteolin content, DPPH and ABTS radical scavenging activities, and α-glucosidase inhibitory activity of the hot water extract were higher than those of the ethanol extract. In evaluating the extraction temperature of Capsicum annuum L. leaf, the antioxidant activities were similar, but the extraction yields, luteolin contents, and α-glucosidase inhibitory activities were higher at 100oC extraction. In evaluating the extraction time of Capsicum annuum L. leaf, extraction yield increased as the extraction time increased, but antioxidant activity and α-glucosidase inhibitory activity were the highest at 60 min of extraction. These results suggest that the optimum extraction conditions of Capsicum annuum L. leaf are hot water for 60 min at 100oC, and the extracts can be used as a functional food resource.

        The antioxidant activity and α-glucosidase inhibitory activity of the solvent fraction fractionated from the methanol extract of Saururus chinensis Baill were examined. As a result of measuring the yields of methanol, hexane, chloroform, ethylacetate, butanol, and water fractions, the extraction yield of fraction was 18.60, 3.38, 24.03, 7.75, 8.11 and 62.57%, respectively. The total polyphenol content of the methanol extract of Saururus chinensis Baill was 13.40, 4.62, 7.39, 31.24, 25.76 and 5.64 mg GAE/g, respectively. DPPH radical scavenging activity (IC50%) results were 20.81, 5.47, 10.15, 22.63, 19.68 and 21.06 ug/mL, respectively, and hydroxyl radical scavenging activity (IC50%) results were 15.81, 2.69, 8.84, 12.80, 3.70 and 3.39 ug/mL. Hydrogen peroxide scavenging activity scavenging activity measurement (IC50%) showed 33.63, 8.88, 16.93, 32.84, 33.79, and 33.71 ug/mL in methanol, hexane, chloroform, ethyl acetate butanol, and water fractions, respectively. The α-glucosidase inhibitory activity of the solvent fraction fractionated with the methanol extract of 300 sec was measured for the α-glucosidase inhibitory activity of methanol, hexane, chloroform, ethyl acetate butanol, and water fraction, respectively, 15.85, 10.84, 15.74, 24.90, 2.58 and 35.70%.

        본 연구에서는 α-glucosidase 저해활성이 우수한 천연 자원을 개발하기 위하여 개똥쑥으로부터 천연 물질을 분리·동정하고 분리된 물질에 대하여 α-glucosidase 저해활성을 측정하였다. 개똥쑥 잎과 줄 기의 chloroform층과 ethyl acetate층으로부터 각각 6개와 5개의 화합물을 분리하였다. 분리한 화합물 중 chloroform층으로부터 분리된 ArteCA(M.W. 316g/mol, C16H12O7)는 isorhamnetin, ArteCB(M.W. 360g/mol, C18H16O8)는 chrysosplenol D, ArteCC(M.W. 316 g/mol, C16H12O7)는 rhamnetin, ArteCD (M.W. 374g/mol, C18H16O8)는 chrysosplenetin, ArteCE(M.W. 284g/mol, C21H20O12)는 acacetin 그리 고 ArteCF(M.W. 270g/mol, C16H14O4)는 imperatorin으로 확인되었다. 한편 EtOAc층으로부터 분리된 ArteEAA(M.W. 178g/mol, C9H6O4)는 6,7-dihydroxy coumarin, ArteEAB (M.W. 192g/mol, C10H8O4) 는 scopoletin, ArteEAC (M.W. 448g/mol, C21H20O11)는 kaempferol-3-O-b-D-glucoside, ArteEAD (M.W. 464 g/mol, C21H20O12)는 quercetin-3-O-b-D-glucoside 및 ArteEAE(M.W. 318g/mol, C15H10O8)는 myricetin으로 확인되었다. 이 중에서 ArteEAE(myricetin)는 α-glucosidase에 대하여 97.3%의 높은 저해활성을 가지고 있었으므로, 혈당조절용 건강식품 또는 치료제 개발을 위한 물질로 활용될 수 있을 것으로 판단되었다.

        Cr3+ 및 Se을 첨가하여 수경 재배한 치커리 3종의 에탄 올 추출물을 각각 제조하여 추출물별 항산화 활성을 비교 검토하였다. 그 결과 양액만으로 재배한 치커리보다 양액에 Se이나 Cr3+을 첨가한 치커리 추출물에서 총 폴리페놀 함량, 총 플라보노이드 함량 및 FRAP 환원능이 증가하였으며, DPPH 라디칼 소거능 및 ABTS 라디칼 소거능도 증가하였다. 특히 Se을 첨가한 치커리보다 Cr3+을 첨가한 치커리에서 더 우수한 항산화 활성을 나타내었다. 또한 Cr3+ 을 첨가한 CLER 및 CLE의 경우 α-glucosidase 저해활성이 있음을 알 수 있었다. 따라서 치커리 추출물의 수경재배 과정에서 양액에 Cr3+, Se의 첨가는 치커리의 항산화 활성 및 α-glucosidase 저해활성 증진에 영향을 주는 것으로 확인되었다.

        Russula compacta, a wild mushroom, belongs to Russulaceae, Russulales of Basidiomycota. This study was conducted to evaluate the free radical scavenging, anti-inflammatory, anticholinesterase and anti-α-glucosidase effects from fruiting bodies of R. compacta extracted with methanol and hot water. In 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging effects, the methanol and hot water extracts showed good scavenging effects comparable with positive control, BHT. The chelating effect of methanol and hot water extracts of the mushroom were significantly higher than the positive control, BHT. The reducing power of the methanol and hot water extracts of the mushroom were lower than the positive control at the concentrations tested. In the HPLC anaysis of phenolic acids profile of the mushroom extract, 7 phenolic acids such as gallic acid, vanillin, rutin hydrate, resveratol, quercetin formononetin, and biochanin-A were detected. Nitric oxide (NO) production in lipopolysaccahride (LPS) activated RAW 264.7 cells was inhibited by 1.5-fold with the treatment of methanol extract when compared with the control. In the anti-cholinesterase activity assay, the methanol extract inhibited the acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) effects by 73.9% and 81.05% at the 1.0 mg/mL concentration, whereas galanthamine, the standard drug, inhibited the AChE and BChE activities by 97.80% and 81.12%, respectively at the same concentration. The methanol and hot water extracts of the mushroom inhibited the α-glucosidase activity by 55.44% and 62.00%, respectively at the 2.0 mg/mL concentration, while acarbose, the positive control inhibited the α-glucosidase activity by 81.81% at the 2.0 mg/mL concentration. From the experimental results, the fruiting bodies of R. compacta contained natural antioxidant, anti-inflammatory, anti-cholinesterase, and anti-diabetic substances, which might be used for health foods.

        Coprinellus miaceus, belongs to Coprinaceae of Agaricales, Basidiomycota, has been used for an edible purposes in asian countries. This experiment was initiated to evaluate the free radical scavenging, free radical scavenging, anti-acetylcholinesterase, anti-inflammatory and α-glucosidase inhibitory activities of C. micaceus fruiting bodies extracted with methanol and hot water. In 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay, the scavenging activities of methanol and hot water extracts were lower than that of positive control, BHT. The chelating effects of methanol and hot water extracts were significantly higher than positive control, BHT at the concentrations of 0.125-2.0 mg/mL. In the reducing power assay, methanol and hot water extracts exhibited the lower activities than the positive control, BHT at the 0.125-0.2 mg/mL concentration. In the HPLC analysis of phenolic acids profile of the mushroom fruiting bodies, 4 phenolic compounds including procatechuic acid, chlorogenic acid, (-)-epicatechin, and naringin were detected. The tyrosinase inhibitory activities of methanol and hot water extracts were 91.33% and 91.99% at 2.0 mg/mL concentration, while the inhibitory activity of kojic acid, the positive control, was 99.61%. Nitric oxide (NO) production in lipopolysaccahride (LPS) activated RAW 264.7 cells were inhibited by the methanol extract in a concentration dependent manner. In the acetylcholinesterase (AChE) inhibitory activity assay, methanol and hot water extracts of the mushroom inhibited the AChE by 94.64% and 74.19%, respectively at the 1.0 mg/mL concentration, whereas galanthamine, the standard drug inhibited the AChE activity by 97.80% at the same concentration. The methanol and hot water extracts of the mushroom inhibited the α-glucosidase activity by 62.26% and 67.59%, respectively at the 2.0 mg/mL concentration, while acarbose, the positive control inhibited the α-glucosidase activity by 81.81% at the same concentration. Therefore, it is concluded that fruiting bodies of C. micaceus contained natural antioxidant, anti-inflammatory, anti-acetylcholinesterase and α-glucosidase inhibitory substances which might be used for promoting human health.

        This study was carried out to investigate the effect on the growth and antioxidant activities of Cichorium intybus L.(CLE), Cichorium intybus L. var. folisum ‘treviso’ (CLET), Cichorium intybus L. var. folisum ‘rosaitaliana’ (CLER) in hydroponics added with Cr3+ or Selenium (Se) for 4 weeks. Total polyphenol, total flavonoids contents and FRAP values of three species of chicory were grown hydroponically with Cr3+ or Se were increased. These extracts were also showed stronger DPPH and ABTS scavenging activity than chicory extracts. In particular, chicories added with Cr3+ had higher antioxidant activities than chicories added with Se. CLER and CLE extracts added with Cr3+ were also showed α-glucosidase inhibition activities. These results indicate that chicories were cultivated in culture fluid added with Cr3+ or Se could be used as high functional vegetables.

        본 연구는 짚신나물 열수 추출물의 α-glucosidase 저해 활 성을 측정하고, 분화된 근육세포에서 glucose 이용과 인슐린 신호전달에 미치는 영향을 분석하였다. 짚신나물 열수 추출 물(10 ㎎/㎖)은 α-glucosidase 활성을 67% 저해하였으며, 같은 농도의 양성대조구인 acarbose(63%)와 유사한 저해 효과를 보였다. 짚신나물 열수 추출물이 α-glucosidase에 의한 단당 류 생성을 저해함으로 식사 후 혈당이 급격히 상승하는 것을 억제하는데 효과적인 소재로 이용 가능성을 확인하였다. 또한 근육세포에서 인슐린 저항성을 유발하기 위해 지방산(1 mM, palmitic acid)를 처리하였고, glucose의 세포내 유입이 감소 되는 것을 확인하였다. 지방산 처리 세포 모델에서 짚신나 물 열수 추출물(10 ㎍/㎖)은 glucose 이용을 유의적으로 회복 시켜 주었다. Normal 상태의 배양조건에서 근육세포의 포도 당 이용능은 짚신나물 열수 추출물(100 ㎍/㎖) 처리에 의해 유의적으로 증가하였다. 근육세포 내로 glucose 유입은 운반 단백질인 Glut4를 통해 이루어지며, 이것은 인슐린이 신호전 달을 통해 조절한다. 짚신나물 열수 추출물의 세포 내 glucose 이용 증가 효과는 인슐린 신호전달 관련 분자인 Akt 유전자 와 단백질 발현을 증가시킨 것과 관련되는 것으로 추정된다. 결론적으로, 짚신나물 열수 추출물은 소화기관에서의 탄수화 물 흡수 저해와 근육세포 내 glucose 이용 증가를 통해 혈당 조절 및 당 대사 개선에 긍정적인 영향을 미치고 있음을 확인 하였다.

        We studied antioxidant activity and inhibitory effect of α-glucosidase from aqueous, ethanolic and methanolic fractions of Galla rhois. In FRAP and ORAC assay for measuring antioxidant activity, we confirmed that Galla rhois extracts had strong antioxidant activity and ethanolic and methanolic extracts were relatively stronger than aqueous extract. We used trolox as a positive control. In order to measure the inhibitory effect of α-glucosidase, we compared acarbose and Galla rhois extracts. As a result of α-glucosidase inhibitory assay, aqueous, ethanolic and methanolic extracts of Galla rhois showed high inhibitory activitity and ethanolic and methanolic extracts were relatively stronger than aqueous extract. The 50% inhibitory concentrations (IC50s) of acarbose, aqueous, ethanolic and methanolic fractions were 0.45 mM, 0.53 μg/mL, 0.415 μg/mL and 0.37 μg/mL, respectively. These results suggest that Galla rhois extracts can be a clinically useful anti-diabetic ingredient, indicating that it needs to be fractionated and isolated and should be further investigated.

        After a mixed carbohydrate diet, inhibition of α-amylase and α-glucosidase involved in the digestion and absorption of carbohydrates can significantly decrease the postprandial increase of blood glucose level. In the course of screening these useful enzyme inhibitors, we selected five kinds of bean, using an in-vitro enzyme inhibition assay method. To evaluate the effect of germination process on the functionality of the bean, we investigated the inhibitory activities of the water extracts of non-germinated bean and germinated bean against α-amylase and α-glucosidase, relevant to postprandial hyperglycemia. We also investigated the oxygen radical absorbance capacity(ORAC), total phenolics content, and postprandial blood glucose lowering effect in rats(Sprague-Dawley rat model). Most germinated beans showed significantly higher α-glucosidase inhibitory activity, compared with non-germinated beans. Among germinated beans, Glycine max had the highest α-glucosidase inhibitory activity(53.3%). The water extract of germinated Phaseolus vulgaris L. had the highest α-amylase inhibitory activity(95.1%), followed by Glycine max(58.7%), and Glycine max L. Merr(54.1%). Furthermore, the five germinated beans also showed high antioxidant activities in ORAC assay. Results suggested that the germination process may improve and enhance the anti-hyperglycemia potential and antioxidant activity of the bean.
