        본 연구는 농․식품부산물 중 비지박, 사자발약쑥 그리고 커피박의 유효미생물(L. acidophilus ATCC 496, L. fermentum ATCC 1493, L. plantarum KCTC 1048, L. casei IFO 3533)발효 사료원이 젖소 급여용 TMR(대조구) 중 대두박을 주로 대체한 각 비지박(SC), 발효 비지박(FSC), 발효 비지박+발효 사자발약쑥 부산물(1:1, DM basis, FSCS) 3, 5, 10% 그리고 발효 비지박+발효 커피박(1:1, DM basis, FSCC) 3, 5, 10%의 반추위 내 미생물 발효특성을 알아보기 위해 9처리구를 이용하여 3반복으로 in vitro 시험이 실시되었다. 배양 6~8시간대에 FSCS와 FSCC 처리구는 SC와 FSC 처리구에 비해 높은 pH 수준을 유지하였고, 대조구에 비해 낮은 pH 수준을 유지하였다(p<0.05). 또한 배양 24시간대 FSC와 FSCC 3% 처리구는 가장 높은 pH를 유지하였다(p<0.05). Gas 생성량은 배양 4~10시간대까지 FSCS와 FSCC 처리구가 대조구 보다 유의적으로 높았으나 SC와 FSC 처리구 보다 낮은 결과를 나타내었다(p<0.05). 건물소화율은 배양 12시간대까지 FSC 처리구에서 유의적으로 높았고(p<0.05), 24시간대에는 FSCS 10% 처리구가 가장 높은 결과를 나타내었다(p<0.05). NH3-N 함량은 배양 4시간대까지 대조구에서 가장 낮았고(p<0.05), 24시간에서는 FSC 처리구가 가장 높은 결과를 나타내었다(p<0.05). 미생물단백질 합성량은 배양 시작 시 FSC, FSCS 그리고 FSCC처리구가 4종의 Lactobacillus spp.에 의한 발효 때문에 대조구와 SC 처리구에 비해 높았으며(p<0.05), 배양 10시간대 FSC 처리구 보다 SC, FSCS 그리고 FSCC 처리구에서 유의하게 높은 결과를 나타내었다(p<0.05). Total VFA 농도는 배양 6~12시간대에는 FSC 처리구에 유의하게 높았고(p<0.05), 배양 24시간대에는 FSCS 처리구에서 가장 높았다(p<0.05). Acetate 농도는 배양 0~12시간대에 FSC 처리구에서 가장 높았으며(p<0.05), 24시간대에는 FSCS 5, 10% 처리구에서 가장 높은 결과를 나타내었다(p<0.05). Propionate 농도는 FSC 처리구가 배양 0~10시간대까지 가장 높았으며(p<0.05), 전체 배양기간 동안 FSCS 5, 10% 처리구와 FSCC 처리구에 비해 높은 경향을 나타내었다. Acetate/Propionate 비율은 배양 12시간대에 SC 처리구에서 가장 높았으며, 배양 24시간대에는 FSCS 3% 처리구가 가장 낮은 결과를 나타내었다(p<0.05). CH4 가스 생성량은 FSC 처리구에서 배양 0~10시간대까지 가장 높았으며(p<0.05), 24시간대에는 가장 낮은 결과를 나타내었다(p<0.05). 배양 4~10시간대까지 FSCS와 FSCC 처리구는 SC와 FSC 처리구에 비해 유의하게 낮은 CH4 가스 생성량을 나타내었고(p<0.05), 사자발약쑥과 커피박 함량이 증가할수록 CH4 가스 생성량은 배양 종료 시까지 낮은 경향을 나타내었다.따라서, 본 in vitro 시험에서 젖소 급여용 TMR에 SC, FSC, FSCS 그리고 FSCC의 대체효과는 FSC 처리구에서 배양 초기부터 12시간대까지 반추위 내 미생물 발효 특성이 가장 높았다. FSCS와 FSCC 처리구는 배양 6~8시간대부터 반추위 내 미생물 작용이 높아지는 특징을 나타내었으나, 배양 시작 후 6~8시간대까지 gas 생성량 및 CH4 가스 발생량은 낮아지는 특징을 나타내었다. 이와 같은 결론은 FSC는 젖소용 TMR 사료원 대체 효과가 높으며, TMR사료원 중 FSCS와 FSCC 대체효과는 배양 초기 다소 낮은 반추위 내 미생물 발효 특성을 나타내었으나 CH4 저감효과를 나타내었다. 또한 배양 후기에는 SC와 FSC의 반추위 내 미생물 발효 특성과 비슷한 경향을 나타내어 TMR 사료원으로써 이용 가치가 높을 것으로 사료된다.

        한약추출 부산물의 영양가치 및 사료 가치를 평가하기 위해 in vitro 반추위 분해 시험을 하였다. 시험에 사용한 10가지 한약추출 부산물은 인삼(Ginseng radix alba), 백복령(Hoelen), 감초(Glycyrrhizaeradix), 당귀(Angelicae gigantis radix), 천궁(Cnidii rhizoma), 작약 (Paeoniae radix), 황기 (Astragali radix), 생강(Zingiberis rhizoma crudus), 대추(Ziziphi fructus), 진피(Aurantii nobilis pericarpium)로 한약재 중 빈번하게 사용하는 약재이다. In vitro 시험을 위해 50 mL serum bottle에 혐기상태로 McDougall's buffer와 반추위액을 2:1비율로 15 mL을 넣었다. 한약추출 부산물은 timothy(300 mg)를 기질로 3%, 5% 수준으로 첨가하였다. Serum bottle은 배양 시간대별(6, 12, 24, 48, 및 72 h)로 39℃에서 120 rpm으로 진탕 배양하였다. 총 가스 발생량은 한약추출 부산물 3% 첨가구 에서는 발효 24시간대에서 대조구가 생강(Zingiberis rhizoma crudus)구에 비해 많았으며, 발효 72시간 대에서도 대조구가 가장 많았고, 진피(Aurantii nobilis pericarpium)구에서 적었다(P<0.05). 한약추출부산물 5% 첨가구에서는 48시간대 가스 발생량이 진피(Aurantii nobilis pericarpium)구보다 대조구에서 많았으며, 72시간 발효구에서는 황기(Astragali radix)구에서 가장 적었다(P<0.05). 미생물 성장률은 한약추출 부산물 5% 첨가구의 48시간대를 제외한 모든 배양에서 유의적으로 증가하였다(P<0.05). VFA 농도는 배양시간의 따라 증가되는 경향을 보였으며, 배양 초기(6 h)에서 한약추출 부산물의 총 VFA 농도는 일부 한약추출 부산물 처리(작약, 황기, 진피)를 제외하고 유의적으로 증가하였다(P<0.05). 위의 결과로 한약추출 부산물은 미생물 성장률 및 발효성상 개선을 증진시킬 수 있으며, 결론적으로 조사료에 대한 한약추출 부산물 5% 대체가 가능할 것으로 예상된다.

        본 연구는 반추동물의 조사료 자원으로서 거대억새를 개발하기 위한 목적으로 수행되었다. 우리나라에서 새롭게 개발된 품종인 거대억새 1호를 완숙기 이후에 채취하여 in vitro 반추위 발효를 이용해반추위내 pH, 암모니아태 질소, 가스발생량, 휘발성 지방산 생성량 및 건물소화율을 조사하였으며,볏짚과 비교하여 평가하였다. 거대억새는 볏짚에 비하여 유의적으로 높은 반추위내 pH를 나타내었다(p<0.01). 암모니아태 질소의 경우 배양 12시간 이후에는 두 처리구간의 유의적인 차이를 나타내지 않았다 (p>0.05). 배양 6시간 이후 부터는 거대억새의 가스발생량이 볏짚에 비하여 유의적으로 낮게 나타났다 (p<0.05). 휘발성 지방산 생성량에 있어 acetate, propionate, butyrate, valerate 및 총생상량에서 볏짚이 거대억새보다 높게 나타났다. 그러나 iso-butyrate와 iso-valerate에서는 두 조사료원별 차이는 발견되지 않았다. 건물소화율에 있어 배양 12~24시간 사이의 거대억새 소화율이 볏짚에 비하여 유의적으로 나타났다. 결론적으로 거대억새의 이용성은 볏짚의 약 80% 수준인 것으로 나타났다.

        Substantial efforts have been made to manipulate ruminal environment in a hope to enhance ruminal fermentation efficiency for better ruminant productivity. Some of examples are methane inhibitors, antibiotics, microbial enzymes, fatty acids and/or lipid feeding, buffering agents, ionophores and probiotics. Of these efforts, the non-ionic surfactant (NIS) has been known for its stimulation to release enzymes from a range of anaerobic microbes. A series of studies were conducted 1) to evaluate NIS diluted with water and ethanol on in vitro ruminal fermentation and 2) to determine the influence of diluted NIS on digestibility of feedstuffs. In first experiment (Exp. 1), NIS was diluted with water or ethanol to measure its effects on in vitro microbial growth, ruminal enzyme activities and gas production by mixed ruminal microbial culture. The NIS was diluted with water or ethanol separately in a 1:5 ratio (w/v). Water and ethanol-diluted NIS with wheat flour were added with rice straw-based mixed ruminal microbial cultures at the rate of 2 ㎎ NIS/16 ㎖ McDougall buffer plus 4 ml ruminal fluid solution. The mixed ruminal microbial culture was run without any NIS addition as control. Addition of NIS either diluted with water or ethanol has significantly reduced the gas production in mixed ruminal microbial culture at 12 and 24 h of incubation. At 48 h post incubation, gas production was the highest with the addition of ethanol diluted NIS followed by water-diluted NIS and control. Carboxy methyl cellulase activity in rice straw-based mixed ruminal bacterial culture was significantly higher with the addition of ethanol-diluted NIS compared with the addition of water-diluted NIS and control at 24 and 72 h post incubation. In second in vitro experiment (Exp. 2), effects of addition of ethanol diluted NIS on dry matter (DM) digestibility of alfalfa hay, gas production, pH and cellular growth in mixed ruminal microbial culture were examined. Alfalfa hay based mixed ruminal microbial culture without any NIS addition was run as a control. The pH of mixed ruminal microbial culture was significantly lower than control at all post incubation sampling hours. In vitro DM digestibility of alfalfa hay was significantly higher with the addition of NIS compared with control. Gas production was significantly less with NIS addition compared with control at all post incubation sampling hours. Microbial growth in mixed ruminal microbial culture was significantly increased with the addition of NIS compared to control.

        The present study investigated the effect of forage sources and their inclusion levels in diet on the rumen fermentation of Korea traditional goat. Timothy and alfalfa were used as forage sources. Forages were mixed with concentrate diet in different ratios. The ratios of forage to concentrate diets were varied to 1:9, 5:5 and 9:1. The rumen fluid of goat was gathered from slaughter house. Dry matter digestibility was decreased and methane production was increased as forage levels in diet was increased. When forage sources, timothy and alfalfa, were compared, groups with timothy showed greater methane production than the groups of alfalfa. Molar ratio of produced acetate and valerate were increased when forage level in diet was increased. In the case of propionate and butyrate, they were decreased as elevated forage levels in diet. The result of this study provided a basic information for rumen fermentation of Korean traditional goat and these information could be applied in the development of nutritional and feeding strategy.

        The aim of present study was to investigate the effect of three types of Chestnut Meals (CM) on chemical composition and rumen fermentation characteristics of the fermented diet. The inoculants consisted of Lactobacillus acidophilus, Bacillus subtilis, and Sacaromyces cerevisiae and were applied to three different types of CM; Whole Chestnut (WC), endodermis (EN), and kernel (KE). All types of CMs were ensiled at 39°C for 0, 1, 2, 4, or 6 days. After ensiling, the fermented CMs were sub-sampled for laboratory assays. On day six of fermentation, counts of the lactic acid-producing Bacillus subtilis, and yeast were higher (P<0.05) in WC than in the other CM types. On day four, KE had higher (P<0.05) crude protein content but lower (P<0.05) neutral detergent fiber and acid detergent fiber contents than the other treatments. In terms of rumen digestibility, KE had the highest (P<0.05) in vitro digestibility of dry matter (IVDMD), neutral detergent fiber digestibility (IVNDFD), total volatile fatty acid (VFA), propionate, butyrate concentrations, and total gas volume, as well as the lowest (P<0.05) acetate concentration. On the other hand, EN had the highest (P<0.05) pH and ammonia-N concentration in the rumen. In the rumen, even though WC application produced the highest microbial count and fermentation characteristics, it did not have a beneficial effect on rumen digestibility. Therefore, this study concluded that application of KE could be recommended due to the observed improvements in IVDMD and IVNDFD.

        This study was conducted to investigate for the natural methane emission inhibitor as a feed additive no adversely effect on rumen fermentation. Five different Control (Wheat barn (0.05 g), MRA(Methane Reduction Additive)-1 (Allium fistulosum L. (0.05 g)), MRA-2 (Sodium Lauryl Sulfate (0.025 g) +Wheat barn (0.025 g) mixed), MRA-3 (Sodium Dodecyl Sulfate (0.025 g) + Wheat barn (0.025 g) mixed), and MRA-4 (Allium fistulosum L. (0.02 g) + Tannic acid (0.02 g) + Wheat barn (0.01 g) mixed) contents were used to perform 3, 6, 9, 12, 24 and 48 h incubation for in vitro fermentation. Ruminal pH values were ranged within normal ruminal microbial fermentation. Dry matter digestibility was not significantly different across the treatments during the whole fermentation time. Also, the result of microbial growth had no adversely effect on during the whole fermentation time. At 24 h, methane emission was significantly lower (P<0.05) than all treatments except to MRA-1. Especially, MRA-4 carbon dioxide emission was significantly lower (P<0.05) than control at 9, 24 and 48 h incubation. In addition MRA-4 propionate concentration was significantly higher (P<0.05) than control at 24 h incubation. The result of RT-PCR Ciliate-associated methanogens were significantly lower (P<0.05) at MRA-1, MRA-3 and MRA-4 than control at 24 h incubation. Based on the present results, MRA-4 could be suggestible methane emission inhibitor as a natural feed additive.

        This study aimed to estimate the effect of inoculant application level on chemical composition and bacterial count of fermented chestnut meal (FCM), and its rumen fermentation characteristics. The inoculant contained Lactobacillus acidophilus (1.2 x 1010 cfu/g), Bacillus subtilis (2.1 x 1010 cfu/g), and Saccharomyces cerevisiae (2.3 x 1010 cfu/g). The chestnut meal mixed with molasses, double distilled water, and inoculant at 1 kg, 3 g, 480 mL, and 20 mL ratio for the basal chestnut meal diet. The double distilled water from basal chestnut meal diet was substituted with bacterial inoculant at a level of 0 (Control), 20 (Medium), and 40 mL (High) in the experimental diets. The mixed experimental diets were incubated at 39°C for 7, 14, and 21 days, respectively. On 7 days of FCM incubation, the contents of crude protein (CP) (quadratic, P=0.043) and neutral detergent fiber (quadratic, P=0.071) decreased by increases of inoculant application levels, whereas bacterial count (quadratic, P=0.065) and rumen NH3-N (linear, P=0.063) increased. By increases of inoculant application levels on 14 days of FCM incubation, the increases were found on dry matter (DM) (quadratic, P=0.085), CP (quadratic, P=0.059), acid detergent fiber (quadratic, P=0.056), in vitro DM digestibility (linear, P=0.002), rumen total volatile fatty acid (VFA) (linear, P=0.057), and rumen iso-butyrate (linear, P=0.054). However, the decreases were found on bacterial count (linear, P=0.002), propionate (linear, P=0.099), and butyrate (quadratic, P=0.082). On 21 days of FCM incubation, in vitro DM digestibility (linear, P=0.002) and total VFA (linear, P=0.001) increased by increases of inoculant application levels, whereas the contents of CP (quadratic, P=0.034) and neutral detergent fiber (quadratic, P=0.047) decreased. These results indicate that the FCM with a medium level of inoculant application and 14 of fermentation had beneficial effects by increasing DM digestibility and rumen total VFA content, without altering bacterial count.

        We inoculated a spent mushroom substrate from Flammulina velutipes (SMSF) with a microbial additive and assessed the effects on chemical composition, ruminal fermentation parameters, and total-tract nutrient digestibility. In Exp. 1, three cannulated Hanwoo steers were used in an in situ trial to determine the degradation kinetics of dry matter (DM) and crude protein (CP). In Exp. 2, three Hanwoo steers were randomly assigned to experimental diets according to a 3 × 3 Latin square for a 3-week period (2 weeks for adaptation and 1 week for sample collection). Experimental diets included the control diet (3.75 kg/d formulated concentrate mixture + 1.25 kg/d rice straw), SMSF diet (3.19 kg/d formulated concentrate mixture + 1.25 kg/d rice straw + 0.56 kg/d SMSF), and inoculated SMSF (ISMSF) diet (3.19 kg/d formulated concentrate mixture + 1.25 kg/d rice straw + 0.56 kg/d ISMSF). The chemical composition of ISMSF did not differ from that of SMSF. Microbial additive inoculation decreased pH (P<0.05) and improved preservation for SMSF. The percentages of DM, neutral detergent fiber (NDF), and acid detergent fiber (ADF) in ISMSF were slightly lesser than those in SMSF. Ruminal fermentation characteristics and total-tract nutrient digestibility were not affected by diet. Overall, microbial additive inoculation improved preservation for SMSF and may allow improved digestion in the rumen; however, the total digestible nutrients (TDN) of SMSF and ISMSF diets were slightly lesser than the control diet. The ISMSF can be used as an alternative feedstuff to partially replace formulated concentrate feed.

        Total mixed ration (TMR) including concentrate diet and roughage together have been used for the ruminant animal. Relatively high concentrations of moisture and water soluble carbohydrate are representative feature of TMR. Those moisture and water can also provide a niche for bacterial growth. Therefore, a possible fermentation of TMR induced by micro-organism is generally accepted. The present study hypothesized that different lactic acid bacteria could alter fermentation of TMR and subsequently rumen fermentation. Three lactic acid bacteria, Lactobacillus paracasei (A), L. plantarum (B) and L. parabuchneri (C), were employed and 7 treatments under full factorial design were compared with control without inoculation. TMR for dairy cow was used. Significant alterations by treatments were detected at lactic acid and butyric acid contents in TMR (p<0.05). Treatment AC (mixture of A and C) and BC (mixture of B and C) showed great lactate production. Great butyrate production was found at treatment C. At in vitro rumen fermentation, treatments B, C and AB (mixture of A and B) showed significantly great total gas production (p<0.05). All treatments except treatments B and AB, showed less dry matter digestibility, significantly (p<0.05). Total volatile fatty acid production at treatment AC was significantly greater than others (p<0.05). In individual volatile fatty acid production, treatment AB and AC showed great acetate and propionate productions, significantly (p<0.05). This study investigated correlation between organic acid production in TMR and rumen volatile fatty acid production. And it was found that butyric acid in TMR had significant negative correlation with acetate, propionate, total volatile fatty acid, AP ratio and dry matter digestibility.

        This study aimed to estimate the effects of replacing Mushroom By-Product (MBP) with Tofu By-Product (TBP) on the chemical composition, microbes, and rumen fermentation indices of Fermented Diets (FDs). The basal diet was formulated using MBP, TBP, rice bran, molasses, and inoculants. The MBP in the basal diet was replaced with TBP at 0, 5, and 10% on Dry Matter (DM) basis for the experimental diets. The experimental diets were fermented at 39°C for 144 h. Chemical composition, pH, microbes, and rumen fermentation indices of the FDs were analyzed. With increasing TBP replacement, crude protein content of FDs increased (L, P < 0.001), whereas crude ash content decreased (L, P = 0.002). Lactic acid bacteria and Bacillus subtilis contents in the TBP-replaced FDs were higher than those in the control (P < 0.05), whereas pH level and mold count were lower (P < 0.05). With increasing TBP replacement, in vitro rumen digestibility of DM (L, P = 0.053) and neutral detergent fiber (L, P = 0.024) increased, wheres rumen pH changed (P = 0.026) quadratically. Rumen total volatile fatty acid (L, P = 0.001) and iso-butyrate contents (Q, P = 0.003) increased with increasing TBP replacement. In conclusion, this study indicates that the replacement of MBP with TBP could improve the quality of FD.
