        This study was designed to observe the effects of hot water soluble polysaccharides extract(PS) from Lentinus edodes on the enzyme activities related with hepatic function and peroxidation in the rats fed butter yellow. The four groups of male SD rats were fed with the diets contained 15% casein(basal diet ; NO group), added butter yellow(BO group) or/and PS(NP, BP group) for 6 weeks. The activities of γ-GTP and GPT in BP were significantly lower compared with BO. The activities of glutathione peroxidase, catalase and lactate dehydrogenase were not significantly different between NP and NO, while those activities were significantly lower value in BP than BO. The activities of glutathione S-transferase of the microsomal and cytosol fractions were significantly low in BP than in BO. The contents of glutathione and malondialdehyde in the liver were considerably low value in BP. In a view of these results the PS of Lentinus edodes prevents the lipid peroxidation and diminishes the liver toxicity caused with butter yellow. The superoxide dismutase activity in cytosolic fraction of liver was not found any effect in all groups. But hepatic function enzyme activities such as catalase and glutathione peroxidase, LDH activities were remarkably decreased in the groups 2(basal diet + PS) and the γ-GTP, GOT and GPT activities, too. In liver, the contents of glutathione decreased by PS supplementation but HDL-cholesterol and total cholesterol ratio in plasma decreased at the groups 3, 4. The γ-GTP, GOT and GPT in plasma were remarkably higher in the rats fed the p-DAB than the control group, too. But above enzyme activities significantly decreased in the groups fed PS.

        국내산 재배 생약류 28종 중 열수 추출물에서 항산화력을 나타낸 것으로 조사된 작약, 목단, 황금, 두충, 시호 그리고 산수유의 열수 추출물 6종을 냉동건조한 후 70% ethanol로 용해하여 ethanol 가용성 획분(ESF)과 ethanol 불용성 획분(EIF)으로 분획하였다. 이 분획물들을 60% linoleic acid에 3, 000ppm씩 가한 후 35℃의 항온기에서 저장하면서 BHA 첨가구와의 산화안정성을 비교하였다. 이 결과 작약과 황금의 ESF는 유도기간이 각각 12일과 9일인데 반해 BHA 첨가구는 9일로 나타나 산화 안정성이 더 높거나 같은 것으로 나타났다. 한편 6종의 생약류의 ESF는 EIF 보다 linoleic acid에 대한 항산화 효과가 모두 높은 것으로 나타났다.

        고사리 열수 추출물로부터 보체계 활성화 산성 다당을 분리하였다. 열수 추출물의 메탄올 환류, 에탄올 침전, 투석 및 동결건조를 통하여 조다당획분인 HPA-1을 분리하였다. HPA-1은 총당 81.80%, 산성당 30.40%, 단백질 15.60%를 함유하였으며, 주요 구성당으로서 rhamnose, galactose, galacturonic acid, glucuronic acid등이 포함되어 있었다. HPA-1을 cetavlon(cethyltrimethyl ammonium bromide)처리에 의해 HPA-2, 3, 4, 5의 획분들로 분리하였으며, 활성과 수율이 가장 높았던 HPA-2로부터 DEAE-Sepharose CL-6B 이온교환 및 Sephadex G-100 and Sepharose CL-6B, Sepharose CL-4B 겔여과 크로마토그라피를 통하여 두 개의 주요 활성 다당체인 HPA-2-Ⅳa와 HPA-2-Ⅴa-2를 정제하였다. 이 다당들은 HPLC에서 단일 획분으로 확인되었으며 분자량은 각각 500, 000과 560, 000으로 측정되었다. HPA-2-Ⅳa는 총당 81.63%, 산성당 31.61%로 구성되었으며 fucose, galacturonic acid, glucuronic acid를 1.40:0.97:1.88의 몰비율로 함유하였다. HPA-2-Ⅴa-2는 주요 구성당으로서 rhamnose, galactose, galacturonic acid를 1.00: 1.38: 139의 몰비율로 함유하였다. 이 다당들의 Ca^2+존재 유무시의 황보체 활성 측정과 교차면역 전기영동 결과, 고전경로(classical pathway)와 부경로(alternative pathway)를 통한 보체계 활성화함에 관여함을 알 수 있었다.

        Background : Dry eyes are caused by highly increased osmolarity of tear film, inflammation, and apoptosis of the ocular surface. Polygonum cuspidatum is a herbaceous perennial plant of the genus Polygonum found in Asia and North America. However, the effects of P. cuspidatum aqueous extract (PCE) on hyperosmolarity-induced inflammation and apoptosis in human corneal epithelial cells have not been examined. Methods and Results : Hyperosmotic media induced human corneal epithelial cell (HCEC) cytotoxicity though increased inflammation, apoptosis, and oxidative stress. PCE treatment significantly inhibited expression of cyclooxygenase-2 and inflammatory cytokines (interleukin-6 and tumor necrosis factor-α), and activation of NF-κB p65 in hyperosmolar stress-induced HCECs. In addition, Hyperosmolarity-induced increase in BAX expression and activation of cleaved poly (ADP-ribose) polymerase and caspase 3 were attenuated in a concentration-dependent manner by PCE. PCE treatment restored anti-oxidative proteins such as Heme oxygenase-1 (HO-1), Superoxide dismutase-1 (SOD-1), and Glutathione peroxidase (GPx) in hyperosmolar stress-induced HCECs. Conclusion : PCE protected against hyperosmolar stress-induced inflammation, apoptosis, and oxidation by inhibiting expression of COX-2, BAX, MMP9; activation of NF-κB, caspase 3, and PARP; and increasing expression of MUC4 and anti-oxidative proteins. Overall, our data provide insight into the protective effects of PCE as a candidate for eye health.

        In this study, the DPPH free radical scavenging activity, antimicrobial effects, growth inhibition and cytotoxicity of herb extracts were determined to screen alternative antibiotics. Hot water extracts of 10 species herbs (Origanum vulgare, Monarda didyma, Echinacea purpurea, Ocimum basilicum, Mentha piperita, Melissa officinalis, Thymus vulgaris, Stevia rebaudiana, Rosmarinus officinalis, Matricaria recutica) were used. DPPH free radical scavenging activity of all herb extracts was ranged from 31.4-49.9%, and significantly great activties were found at Rosmarinus officinalis, Origanum vulgare and Matricaria recutica (P<0.05). Hot water extracts of monarda didyma, origanum vulgare, thymus vulgaris and rosmarinus officinalis showed greater antimicrobial activities compared to others. Additionally, those four extracts represented relatively low cytotoxicity compared to others. As a result, it was found that Origanum vulgare and Rosmarinus officinalis which possessed great antioxidant and antimicrobial activity with less cytotoxicity. So these two herb extracts can be used as an alternative of antibiotics for organic farming.

        This study was carried out to investigate the physicochemical and antioxidant properties of fermented milk with addition of hot water extract of Cordyceps militaris grown upon Tenebrio molitor. The general components of Cordyceps militaris grown upon Tenebrio molitor are moisture 10.85%, crude protein 18.44%, crude fat 2.07%, crude ash 5.46%. The DPPH radical scavenging activities of different solvents were the highest (74.81 EDA%) with hot water extract sample. The acidity of fermented milk was high with increasing amount of extracts. The pH of fermented milk reached 4.60-4.66 after 4 h of fermentation, and the number of lactic acid bacteria was highest (11.70 log CFU/mL) with 1% fermented milk. The moisture content of fermented milk showed no significant difference. In addition, contents of crude protein were not significantly different according to addition amount. Regarding Hunter’s color values, L value decreased as the amount of extract increased, whereas a value and b value increased. The content of free amino acid increased with increasing amount of extract. The DPPH free radical scavenging ability and ABTS free radical scavenging ability of fermented milk were significantly different as the addition amount of extract increased. In the sensory evaluation, fermented milk containing 1% of extract showed the highest preference.

        This study was carried out to investigate the anti-oxidative and anti-inflammatory effects of hot water extracts of Ligularia fischeri cultivated in Youngyanggun. We obtained hot water extract (HWE) and cold water extract (CWE) from L. fischeri. The anti-oxidative activities of L. fischeri extracts were measured by 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity. The anti-inflammatory effects of L. fischeri were evaluated in human mast cell line-1 (HMC-1) cells stimulated with phorbol-12-myristate-13-acetate plus A23187 (PMACI). The solid yields of HWE was 150% higher than CWE solid yield. Total polyphenol contents of HWE were 198.07±0.24 mg/g. The value of anti-oxidative activities of HWE were shown IC50 28.2±0.04 ug/mL. We showed that HWE significantly reduced the PMACI-induced the production of IL-6 (0.01-1 mg/mL), IL-8 (0.1-1 mg/mL), and TNF-α (0.01-1 mg/mL). These results indicate that the HWE of L. fischeri can be used as a functional material due to its antioxidant and anti-inflammatory activities.

        Spiraea prunifolia Sieb. et Zucc. var. simpliciflora Nakai (SSN) has been used for the anti-inflammation in traditional folk medicine. To compare water and methanol extracts of SSN, we analyzed major components using LC IT TOF MS. The major components of hot water extract were identified as caffeic acid and p-coumaric acid, but methanol extract was not well established. However, methanol extract was detected with less polarity compounds compared to hot water extract. Next, we investigated the inhibitory effects of SSN water extract on the lipopolysaccharide (LPS)-induced inflammatory response or H2O2-induced oxidative stress in Raw 264.7 macrophage cells. SSN strongly suppressed the production of nitric oxide in LPS-induced inflammatory response without cytotoxcity. The SSN possessed free radical scavenging activities such as DPPH (IC50=320.2 ㎍/㎖), ABTS (IC50=124.0 ㎍/㎖), and superoxide anion radical (IC50=122.6 ㎍/㎖). The total phenol and flavonoid content of SSN was 56.7 ㎎/g, and 15.1 ㎎/g, respectively. Furthermore, SSN decreased the H2O2-induced cytotoxicity by enhancing the cell viability, and SSN significantly reduced the intracellular reactive oxygen species (ROS) level. Therefore, SSN may be recommended as an effective strategy to prevent and/or treat various inflammation and ROS-induced diseases.

        Menopause is characterized by a decrease in estrogen production by the ovaries. Furthermore, the risk of developing chronic diseases including cardiovascular disease, obesity, and diabetes increases in menopausal women. In this study, we evaluated the ability of Caragana sinica (CS) to affect lipid and glucose metabolism in an ovariectomized Sprague-Dawley rat model of induced menopause. The animals were divwided randomly into three groups: sham-operated rats (SHAM), ovariectomized rats (OVX), and ovariectomized rats treated with 1% water extract of CS (OVX-CS). The OVX-CS treated mice showed a significant decrease in body weight, adipose tissue, triglyceride, and total cholesterol. Improved lipid and glucose profiles were also observed, and were attributed to suppression of hepatic lipogenesis and adipokine production and regulation of glucose-regulating enzyme activity. Therefore, these findings indicate a potential use for CS as a functional food for menopause-induced hyperlipidemia and hyperglycemia.

        We investigated the effects of Acorus gramineus water extract on the blood lipid levels in high fat diet-fed obese male mice. We divided thirty-five C57BL/6 mice into 5 groups: normal group, control group, and groups treated with Acorus gramineus water extract at concentrations of 20, 100, and 500 ㎎/㎏. We inoculated Acorus gramineus water extract per orally once a day for 6 weeks respectively. The results revealed that Acorus gramineus water extracts had positive effects on the total cholesterol, triglyceride, high-density lipoprotein cholesterol, and low-density lipoprotein cholesterol levels.

        본 연구에서는 다양한 생리활성을 가진다고 알려져 있는 약용열매인 산수유, 오미자, 복분자 및 구기자 4종의 기능 성식품 소재로 활용하고자 혼합비율별로 추출한 열수 추출 물의 이화학적 특성 및 항산화 활성을 조사하였다. 약용열 매 혼합비율에 따른 열수 추출물의 pH는 3.22~3.52로 산성 을 나타내었으며, 당도는 M3이 3.20 °Brix로 다른 시료에 비해 낮았고 산도는 3.60~5.85%으로 오미자 혼합비율이 높은 구간에서 5.85%로 가장 높게 나타났다. 추출수율은 M2, M1, MS, M4 및 M3 순으로 나타났으며, 총 폴리페놀 및 플라보노이드 함량은 각각 14.54 및 5.65 g/100 g으로 복분자 혼합비율이 높은 추출물인 M3에서 가장 높은 값을 나타내었다. DPPH 및 ABTS radical 소거활성은 M3이 250~1,000 μg/mL 농도에서 25.80~86.09% 및 33.43~90.49% 로 가장 높게 나타났다. FRAP 및 환원력은 M3가 250~1,000 μg/mL 농도에서 각각 0.36~0.86 μM 및 0.21~0.96으로 가장 우수하였으며, 모든 혼합 열수 추출물에서 농도 의존적으 로 증가하였다.

        본 연구에서는 연화 열수 및 에탄올 추출물의 3T3-L1 지방세포 분화 및 ROS생성 저감효과를 구명하기 위하여 3T3-L1 전지방세포 분화과정 중 연화 열수 및 에탄올 추출 물에 의한 지방축적과 ROS 생성을 관찰하였다. 연화 열수 및 에탄올 추출물은 XTT assay에서 100, 200 및 400 μg/mL 농도에서 세포 독성을 보이지 않았다. 지방세포 분화 중 세포 내 지방축적 및 ROS 생성량을 비교한 결과, 연화 열수 및 에탄올 추출물을 처리한 지방세포의 경우 지방축적량과 ROS 생성량 모두 유의적으로 억제되는 것으로 나타났다. 특히 연화 열수 및 에탄올 추출물을 처리함으로써 지방세포 분화와 관련된 전사인자인 PPARγ, C/EBPα 및 aP mRNA의 발현을 유의적으로 감소시켰으며, ROS의 생성과 관련이 있는 주요 유전자인 NOX4 및 catalase의 유전자발현 또한 유의적으로 감소하였다. 이 결과를 통해 연화 열수 및 에탄 올 추출물이 3T3-L1 지방세포 내 중성지방의 축적 억제 효과와 더불어 ROS 생성 억제에 효과적으로 작용함을 확인 하였다. 따라서 연화 열수 및 에탄올 추출물은 비만과 같은 대사증후군 관련 질환의 개선을 위한 천연물 기능성 소재로 의 활용이 기대된다.

        Onion (Allium cepa L.) is one of the richest sources of flavonoids in human diet. Onion peel contains over 20times more quercetin than onion flesh. In this study, we studied the effects of onion peel water extract (OPE) on the bloodlipid profiles in mice. The onion peel extracts was extracted with hot water. The experimental groups were divided with 3groups (n=6) of ICR male mice: normal diet+distilled water (NC), high-fat diet+distilled water (HF), high-fatdiet+onion peel water extract 20㎎/㎏ (OPE-20). The oral administration was conducted daily. The experimental periodwas 7 weeks. Onion peel water extract showed higher concentration of polyphenol gallic acid and anti-oxidant trolox equiv-alent than the ethanol extract. The body weight gain and food efficiency ratio was significantly lower in the OPE-20 group ascompared with HF group (p<0.05). The epididymal fat and retroperitoneal fat showed significantly lower weights and sizesin the OPE-20 group as compared with HF group (p<0.05). The serum levels of total cholesterol, LDL cholesterol and trig-lyceride were significantly lower in the OPE-20 group as compared with HF group (p<0.05). The OPE-20 group showedhigher HDL cholesterol concentration than HF group (p<0.05). Atherogenic index was ignificantly lower in as comparedwith HF group (p<0.05). The serum levels of glucose, GOT and GPT were significantly lower in the OPE-20 group as com-pared with HF group (p<0.05). In these results, we suggests that onion peel water extracts supplementation can reduces theserum lipid components and improves the lipid metabolism in hyperlipidemic mice induced with a high-fat diet.

        본 연구는 인공 재배 산채의 생산량 및 산채의 가공제품의 개발의 증대함에 따라 야생 및 재배 참취의 품질 특성을 비교하고자 재배 및 야생 참취의 건조방법에 따른 항산화성을 비교 분석하였다. 참취의 열수 추출물의 수율은 야생참취에 비해 재배 참취에서 수율이 높았고, 블랜칭 건조추출물의 수율이 높았다. 야생 참취 추출물의 총 폴리페놀과 플라보노이드의 함량은 재배 참취 추출물보다 높았으며, 건조방법에 따른 총 폴리페놀의 함량은 블랜칭 건조추출물에서 높았고, 플라보노이드의 경우에는 생채 추출물에서 높았다. 참취의 열수 추출물의 전자공여능은 농도가 증가함에 따라 증가하였고, 블랜칭 건조의 열수 추출물에서 높은 전자공여능을 보였다. 참취 열수 추출물의 SOD 유사활성능은 추출물의 농도가 높아질수록 증가하였으며, 생채 열수 추출물에서 다른 추출물에 비해 높았다. pH 1.2에서의 아질산염 소거능은 야생 및 재배의 생채, 블랜칭건조, 생채건조 열수 추출물 순으로 높았다. Xanthine oxidase 저해활성은 열수 추출물의 농도가 증가할수록 저해효과가 높았으며, 생채건조 추출물에서 가장 높았다. Tyrosinase 저해활성과 환원력은 농도가 증가함에 따라 블랜칭 건조 열수 추출물에서 높게 나타났다. 따라서 참취의 재배 및 건조 방법에 따른 열수 추출물은 항산화성이 우수하여 천연 항산화 소재 및 기능성 식품소재로서의 활용 가능한 약용식물자원이며, 이를 활용한 가공산업 발전의 기초자료가 될 것으로 생각된다.

        본 연구는 식품, 화장품 및 의약품 소재로 활용 가능성에 대해 알아보고자 우산나물 뿌리와 지상부에 대한 항산화 활성을 측정하였다. 우산나물 뿌리 열수 추출물(RHW)은 4.58 mg/g의 플라보노이드와 59.11 mg/g의 폴리페놀 화합물을 함유하였으며, 지상부 열수 추출물(APHW)은 각각 2.79 mg/g과 48.01 mg/g을 함유하였다. SOD 유사활성능은 RHW에서 23.74%, APHW는 21.61%를 나타내었다. 아질산염 소거능은 2,000 μg/mL에서 RHW는 pH 1.2에서 63.06%였으며, pH 3.0에서는 47.16%로 아질산염을 50% 소거하는 IC50은 ascorbic acid 99.93 ug/mL, RHW 1,150.85 μg/mL, 그리고 APHW에서는 1,610.25 μg/mL이었다. DPPH free radical 소거능에 대한 IC50은 RHW 99.87 μg/mL, APHW 118.29 μg/mL를 나타내었다. Xanthine oxidase에 대한 IC50은 RHW에서 139.62 μg/mL였으며, APHW는 111.11 μg/mL으로 xanthine oxidase 저해능은 우산나물 지상부의 열수 추출물인 APHW가 뿌리 열수 추출물인 RHW보다 좀더 낮은 농도에서 50%의 활성을 나타내었으나, 플라보노이드와 폴리페놀 화합물 함량, SOD 유사활성, 아질산염 소거 및 DPPH free radical 소거능은 RHW가 APHW보다 높은 활성을 나타내었다. 이상의 결과 우산나물은 기능성 식품과 화장품 그리고 의약품 개발을 위한 항산화 소재로 활용 할 수 있을 것으로 판단된다.

        This study was conducted to investigate the extraction characteristics including total polyphenol compound content (TPC) and the antioxidative effect of Rumex crispus root extract on commercial corn oil. Extraction yield was increased with extraction temperature but decreased with extraction ratio. No significant differences in aromatics content were found among the extracts prepared with various extraction conditions; extraction ratio, extraction temperature, extraction pH and composition of extractant. Total flavonoid content of the extract was increased with extraction temperature and extraction pH, and highest value of it was found when extractant composition of ethanol in water was 75%. Total polyphenol compound content (TPC) of the extract with 10 fold extraction ratio showed the highest value, but no significant difference in TPC was found with extraction temperature. Reducing power and DPPH (α,α-diphenyl-β-picrylhydrazyl) radical scavenging ability (RSA) of the extract in spectrophotometric absorbance were decreased with extraction ratio but increased with extraction temperature and showed 63.1%~98.4% and 67.6%~86.7% of those of ascorbic acid in reducing power and DPPH RSA, respectively. The antioxidative effects of the extract on corn oil were 84.8~93.0% of that of commercial butylated hydroxytoluene, the antioxidant index value was highest when the ethanol composition to water in extractant was equal ratio.

        Recently, NADPH oxidase 4 (NOX4)-mediated generation of intracellular reactive oxygen species (ROS) was proposed to accelerate adipogenesis of 3T3-L1 cell. We have previously shown that Cheonnyuncho (Opuntia humifusa) extract significantly inhibited adipocyte differentiation via downregulation of PPARγ (peroxisome proliferator-activated receptor gamma) gene expression. In this study, we focused on the molecular mechanism(s) of NOX4, G6PDH (glucose-6-phosphate dehydrogenase) and antioxidant enzymes in anti-oxidative activities of 3T3-L1 adipocytes. Our results indicate that Cheonnyuncho extracts markedly inhibits ROS production during adipogenesis in 3T3-L1 cells. Cheonnyuncho extracts suppressed the mRNA expression of the pro-oxidant enzyme such as NOX4 and theNADPH-producing G6PDH enzyme. In addition, treatment with Cheonnyuncho extract was found to upregulate mRNA levels of antioxidant enzymes such as Mn-SOD (manganese-superoxide dismutase), Cu/Zn-SOD (copper/zinc-SOD), glutathione peroxidase (GPx), glutathion reductase (GR), and catalase, all of which are important for endogenous antioxidant responses. These data suggest that Cheonnyuncho extract may be effective in preventing the rise of oxidative stress during adipocyte differentiation through mechanism(s) that involves direct down regulation of NOX4 and G6PDH gene expression or via upregulation of endogenous antioxidant responses.
