        본 연구는 항균성 코팅제의 개념과 동향파악으로 항균성 코팅제의 연구개발의 방향을 설정하는데 도움을 주기 위한 것이다. 항균제는 미생물을 제거하거나 성장을 저지하는데 사용되는 화합물이며 항균 코팅제에 함유되는 항균제용 재료는 무기물, 금속, 저분자 유기물, 천연유기물, 고분자가 있다. 항균코팅제는 생활용품, 병원용품, 산업용품, 전자제품, 의류, 건축 내장재 등의 표면의 항균성 부여에 쓰인다. 기존 항생제는 세균의 세포벽을 손상하지 않고 미생물을 침투하나 항균성 고분자는 세포막을 파괴하므로 항생제의 내성을 방지할 수 있다. 대부분의 고분자 항균제는 양이온 고분자에 초점을 맞추고 있다. 항균제의 분자설계와 코팅제 배합의 합리화로 항균제의 선택성, 내구성, 독성 문제가 개선될 것이다.

        In this study, antimicrobial and antioxidative activities were investigated using the hot water extracts of Rhus verniciflua Stokes (RVS) in order to develop RVS as a natural antimicrobial and antioxidative material. The antimicrobial activity was not found against five strains of yeasts (Saccharomyces cerevisiae, Saccharomyces pastorianus, Torulaspora delbrueckii, Zygosaccharomyces rouxii, and Zygosaccharomyces bailii) while the activity was confirmed against five strains of bacteria (Bacillus subtilis, Staphylococcus aureus, Bacillus licheniformis, Escherichia coli, and Pseudomonas aeruginosa) at the RVS extract concentration of 0.2-1.0% (w/v) by plate cultural method. In the investigation for 24 hours by turbidometry, the antimicrobial activity showed a relatively high growth inhibition of 79.7% against Bacillus subtilis. The highest growth inhibition of 82.6% was against Bacillus licheniformis at the RVS extract concentration of 2,000 μg/mL. The lowest growth inhibition was 25.7-44.1% against E. coli at the extract concentration of 250 μg/mL and 2,000 μg/mL. The total polyphenol and flavonoid contents of the RVS extract were 42.48 mg/g and 5.42 mg/g, respectively. The highest antioxidative activity at 79.15% was observed at the RVS extract concentration of 0.5 mg/mL by DPPH. Also the antioxidative activity by ABTS demonstrated the highest level at 81.56% at the same concentration. It can be gleaned from these results that the hot water extract of RVS has a great potential to be a natural antioxidant and antimicrobial material against gram positive bacteria(Bacillus subtilis and Bacillus licheniformis).

        The purpose of this study is to evaluate antimicrobial effects of organic acid and some natural occurring antimicrobials against Staphylococcus aureus isolated from various origins (vegetables, peanut, pea leaf, kim- bab, person, perilla leaf, and animal) and to calculate their MIC and MBC values. Five organic acids (acetic, lactic, citric, malic, and propionic acid), three essential oils (carvacrol, thymol, and eugenol), and two other natural antimicrobials (nisin and cinnamic acid) were evaluated for their antimicrobial effects against 113 strains of S. aureus using combination treatments. Propionic acid (7%), nisin (1%), thymol (1%), carvacrol (1%) showed antimicrobial activities against S. aureus strains in agar disc diffusion test. And, carvacrol, thymol, and nisin were found to be the most effective with the lowest MIC values of 0.0313%, 0.0625%, and 0.0625% against S.aureus, respectively. Propionic acid (0.2313%) and citric acid (0.6000%) were the most effective among organic acids tested. Therefore, these five antimicrobials were selected for next combination treatments. Combination of propionic acid and citric acid were showed the strongest inhibitory effectiveness against S. aureus among combination treatments. These results suggest that organic acid such as propionic and citric acid, and natural occurring antimicrobial such as nisin, carvacrol, and thymol might be possibly used as preservatives for inhibiting S. aureus in foods.

        목적: Polyhexamethylene biguanide (PHMB)와 Epigallocatechin-gallate (EGCG) 혼합물의 각막염 유발 균주에 대한 항균효과를 검사하고 콘택트렌즈 관리용액의 항균제로서의 가능성을 판단하고자 하였다. 방법: 국제표준화기구(International Organization for Standardization, ISO)가 규정한 황색포도상 구균(Staphylococcus aureus), 메티실린내성 황색포도상구균(Methicillin-resistant Staphylococcus aureus), 녹농균(Pseudomonas aeruginosa), 세라티아 마르세센스(Serratia marcescens), 칸디다 알비캔 스(Candida albicans), 푸사리움 솔라니(Fusarium solai)를 각각의 액체배지에 접종한 후 각 균주들의 적정 배양온도에서 24시간동안 활성화시켰다. 활성화된 균주는 96-well plate를 사용하여 PHMB와 EGCG를 각 각 0.0000125~0.0004%, 0.0025~0.3% 함유한 액체배지에서 배양하였고, PHMB/EGCG 혼합물은 PHMB 최소저해농도(MIC, 0.00005%)와 0.0125~0.1%의 EGCG가 포함되도록 하여 12시간 동안 배양하였다. 배양 후 항균력은 600nm에서 흡광도를 측정하여 확인하였다. 결과: ISO 규정 안질환 균주에 대한 EGCG 최소저해농도는 PHMB보다 높아(100~1,500배) 항균력은 낮은 것으로 평가되었다. PHMB/EGCG 혼합물의 ISO 규정 안질환 균주에 대한 최소저해농도는 0.00005%/ 0.05%로 PHMB 최소저해농도의 25% 수준에서도 충분한 항균효과를 나타내었다. 결론: PHMB와 EGCG 혼합물은 ISO 규정 안질환 균주에 대하여 항균 시너지 효과를 나타내어 다목적 용액에 사용하는 PHMB 농도를 낮출 수 있을 것으로 보이며 이 결과는 콘택트렌즈 관리용액 개발에 기초자료로 활용될 수 있을 것으로 사료된다.

        This research was carried out to find herbal preservatives for Makgeolli, as Makgeolli loses its commercial value due to overproduced acidic materials. When Makgeolli was kept at 25℃ to find the changes in acidity, total microbial cell number, yeast cell number, and bacterial species variety, a sudden increase of acidity as well as the disappearance of yeast cells occurred at day 6, and Makgeolli was changed to complete off-flavor. Acetobacter pasteurianus is the main acidifier in Makgeolli and shows a synergy effect in acid formation when cultured in combination with Lactobacillus casei. Among 12 herbs, the ethanol extract of Sutellaria baicalensis showed antimicrobial activity against A. pasteurianus, whereas the ethanol extract of Coptidis rhizoma showed antimicrobial activity against L. casei. Makgeolli added with Sutellaria baicalensis extracts demonstrated a lower acidity than that with Coptidis rhizoma extracts, which indicates that the inhibition of an acetic acid former is more important than that of a lactic acid former in Makgeolli preservation. Sutellaria baicalensis extracts prolonged the shelf life of Makgeolli by 1~2 weeks at a minimal inhibitory concentration (0.63 ㎎/㎖) during storage at 10℃.

        We sought to study the qualities and scientific benefits of Dongchimi, a traditional Korean food. We compared and analyzed ingredients used for the appearance and storability of dongchimi - honggot (Brassica juncea czerniak et coss), cockscomb (Celosia cristata L.), and beet Beta vulgaris L.). We specifically examined the antioxidative and antibacterial activity of pigments from extracts of these ingredients. Distilled water (H2O) and 1% citric acid were used to safely extract pigments. The antioxidative activity of the pigments was then measured for total phenolic compounds, SOD (Super Oxide Dismutase), and EDA (Electron Donation Ability) by DPPH. The antibacterial activity of was also assessed by a Paper disc solution. Our results show that the pigments had sufficient antioxidative activity and had antibacterial properties against Gram positive and negative bacteria. In particular, Cockscomb (used for enhancing color) contained the highest amount of polyphenol compounds and had the most efficient antioxidative activity.

        국내 마늘의 품종과 관련한 연구는 산지에 따른 비교로 한정되어 있어 마늘의 성분 및 특성에 대한 기 초자료를 확보하고자 품종별 마늘 시료 12종을 대상으로 이화학적 특성과 항균활성을 분석하였다. 품종별 마늘의 구폭은 캘리포니아 만생종(60.44mm, 53.73g)이 가장 크고 무거웠으며, 창녕종(44.04mm, 25.15g)이 가장 작고 가벼웠다. 품종별 마늘의 표면색 중 L값은 84.13∼90.56, a값은 –1.10∼0.77, b값은 18.24∼26.61의 범위였다. 전단가는 캘리포니아 조생종이 가장 낮았으며, 94-12-2종은 4211.35 cm/kg2 으로 타 시료에 비하여 전단가가 가장 높았다. 가용성 고형분 함량은 6.40∼11.33 %brix의 범위였는데, 창녕종에서 유의적으로 높았다. 품종별 마늘의 pH는 5.57∼6.53의 범위였다. 총 thiosulfinate 함량은 이 태리종이 93.23 mM/g로 가장 낮았고, 캘리포니아 만생종이 146.05 mM/g로 가장 높았다. 총 pyruvate 함량은 유고종(162.50μM/g)에서 가장 높았고, 캘리포니아 조생종(147.41μM/g)에서 가장 낮았다. 품종 별 마늘의 추출물로 항균활성을 측정한 결과, Streptococcus mutans균에서 활성이 가장 낮았으며, Escherichia coli균에 대한 항균 활성이 높았

        목적: 합성보존제 성분인Polyhexamethylene biguanide (PHMB)와 PHMB가 첨가된 다목적용액의 국제표준화기구(International Organization for Standardization, ISO)가 규정한 균주에 대한 항균력을 비교하고, 콘택트렌즈 관리용액에 적합한PHMB 농도를 확인하기 위해 본 연구를 수행하였다. 방법: 항균력 측정을 위하여 배양된 황색포도상구균(Staphylococcus aureus), 메치실린내성 황색포도상구균(Methicillin-resistant Staphylococcus aureus), 녹농균(Pseudomonas aeruginosa), 세라티아 마르세센스(Serratia marcescens), 칸디다 알비캔스(Candida albicans), 푸사리움 솔라니(Fusarium solai)를 각각의 액체배지에 접종한 후 각 균주들의 적정 배양온도에서 24시간동안 활성화시켰다. 활성화된 균주는 96-well plate를 사용하여 PHMB 용액과 PHMB를 첨가한 다목적용액의 PHMB 농도가 0.0000125%~0.0004%인 배양액에서 12시간 배양한 후 600nm에서 흡광도를 측정하여 항균력을 비교하였다. 결과: 생육억제를 나타내는 최소저해농도는 순수한 PHMB가 PHMB를 첨가한 다목적용액에 비하여 황색포도상구균, 메치실린내성 황색포도상구균, 녹농균, 세라티나 마르세센스, 칸디다 알비캔스에서 2배 높게 나타났다. 다목적용액에 포함된 0.0002% PHMB 농도는 국제표준화기구에서 규정한 균주에 대한 충분한 항균력을 나타내었다. 결론: 다목적용액에 포함된PHMB의 농도는 순수한 PHMB 최소저해농도보다 낮지만, 용액에 포함된 다른 첨가물에 의해 항균시너지효과를 나타냄을 확인하였다.

        물(water, TW), 에탄올(ethanol, TE) 및 핵산(n-hexane, TH)을 이용하여 탱자씨 추출물(trifoliate orange seed extracts, TSEs)을 각각 제조한 후 그람양성 식중독균 6종(Bacillus cereus KCCM 11341, Bacillus subtilis KCTC 1022, Listeria monocytogenes ATCC 12692, Staphylococcus aureus ATCC 19111, Streptococcus mutans KCTC 3065 및 Yersinia enterocolitica KCCM 41657)에 대한 항균활성과 Lactobacillus acidophilus IFO 3025에 대한 프레바이오틱 효과(prebiotic effect)를 측정하였다. 그 결과, 탱자씨 핵산추출물(TH)은 S. aureus ATCC 19111에 대해 배양시간이 증가함에 따라 대조군에 비해 강한 증식저해활성을 보였으며, 탱자씨 에탄올 추출물(TE)은 약간의 증식저해활성을 보였다. 특히, 배양 81시간 후 대조군은 90.31%의 증식활성을 보인 것에 반해 탱자씨 핵산추출물과 에탄올추출물은 64.95%와 75.50%의 증식활성을 각각 보였다. 탱자씨 추출물의 Lb. acidophilus IFO 3025에 대한 프레바이오틱 효과는 대조군에 비해 증식활성을 보이지는 않았으나, 적어도 생육저해효과를 보이지는 않았다. 따라서 탱자씨의 핵산 및 에탄올 추출물은 S. aureus ATCC 19111에 대한 항균활성물질로서의 가능성을 제시하였다

        The study indicated that antimicrobial activity about gram positive and gram negative bacteria of ginger-oleoresin(GO) extract with the condition of ethanol and supercritical fluid extractions. As the concentration of extraction increases, the clear zone of GO ethanol extract also increased dependently. This led the antimicrobial activity of gram positive bacteria to take bigger place than gram negative bacteria especially in Listeria monocytogenes. There was a high antimicrobial activity in E-III treatment where the ratio of the ginger powder extract to ethanol extraction was 1:6. It was quite effective to treat the antimicrobial activity of GO ethanol extract under 80oC and there was not big difference in the intervals which were the extraction time - 1 to 7 hours. The antimicrobial activity of supercritical fluid extract seemed to take the biggest place in Listeria monocytogenes. From the supercritical fluid extract, it was shown the strong ability of antimicrobial activity in the condition with 100 bar 35oC, 250 bar 35oC and 250 bar 65oC. Furthermore, according to the case of solvent extract, there was not any significant difference in the antimicrobial activity with condition of extraction. However, there was significant antimicrobial activity in E-III treatment of 100 bar and 500 bar of extraction pressure, and 35oC and 65oC of extraction temperature.

        미강으로부터 분리 정제된 피틴산을 식육에 적용하여 식육에서의 항균활성과 항산화능을 조사함으로써 식품보존제로서의 가능성을 확인하였다. 피틴산이 첨가된 Tryptic soy broth에 S. Typhimurium KCCM 11806을 배양하였을 경우 1%(w/v) 피틴산 첨가 시 S. Typhimurium의 생육을 완전히 저해시켰다. E. coli O157:H7에 대한 항균 효과는 1.0%(w/v) 피틴산 첨가 시 약 3.0 log 단위의 생육저해효과를 확인하였다. 신선한 닭고기, 돼지고기, 소고기에 S. Typhimurium을 접종하고 피틴산을 첨가하여 37oC에서 24시간 배양하여 피틴산의 항균효과를 조사한 결과 모든 식육에서 항균효과가 피틴산 농도에 의존적으로 나타났으며 1%(w/v) 피틴산 첨가 시 약 3.0 log 단위의 항균효과를 나타내었다. 생육인 닭고기, 돼지고기, 소고기에 피틴산을 여러 농도로 첨가한 결과 E. coli O157:H7에 대한 항균효과는 모든 생육에서 0.5% 피틴산 첨가 시 약 2.0 log 단위의 생육저해 효과를 보여 주었으며, 가열 처리된 식육에서도 첨가된 피틴산 농도에 의존적으로 유의성 있게 증식을 억제하였다. 생육에 피틴산을 첨가하여 4oC에서 3일간 저장할 경우 저장기간이 증가함에 따라 TBARS(thiobarbituric acid-reactive substances) 값이 증가하였으며 피틴산을 5 mM 첨가하였을 때 TBARS 값이 유의적으로 감소하여 식육에서의 항산화능을 확인하였다.

        본 연구는 probiotics 균주 선발을 위해 동치미로부터 식중독균에 대해 높은 항균활성을 보이는 유산균을 분리동정하여 배양조건을 확립하고, 위산, 담즙산, 췌장액에 대한 저항성 실험을 비교, 분석하여 probiotics 균주로 사용 가능성을 탐색하고자 하였다. 2% CaCO3를 첨가한 MRS agar를 이용하여 김치 무리로부터 식중독균의 저해능이 우수한 8 균주를 분리하였다. 식중독균에 대해 항균활성실험 결과 J4가 B. cereus는 19.85±0.53 mm, L. monocytogenes는 18.37±0.38 mm, S. aureus는 22.49±0.37 mm, E. coli는 17.90±0.64 mm, S. enterica는 16.28±0.53 mm, V. parahaemolyticus는 24.59±0.29 mm의 생육저해환을 형성하여 가장 높은 항균력을 나타내었다. J4 균주의 16S rDNA 염기서열을 분석한 결과 L. sakei와 99% 상동성을 나타내어 L. sakei J4라 명명하였다. L. sakei J4의 최적배양조건은 pH 8.5, 30, 정치배양시 가장 활발하게 생육이 진행됨을 확인하였다. L. sakei J4는 인공위액에서의 생존율 24.3±3.0%, 담즙산에서의 생존율 25.8±0.4%, 췌장액에서의 생존율 101.2±4.5%로 다른 균주보다 상대적으로 생존율이 높았으며, 향후 alginate를 이용한 캡슐화, chitosan을 이용한 재코팅 등의 연구방법을 정립하여 인공위액에 대한 저항성을 높인다면 프로바이오틱스 균주로서 조건을 충족시킬 수 있으리라 사료된다.

        The present study was evaluated the antibacterial effect of the combination of Coptidis rhizoma, Glycyrrhiza uralensis Fischet, Schizandra chinensis and Corni Fructus(1:1:1) extracts(CGSC10). Furthermore, the effectiveness of CGSC10, sodium chlorate, and the combination of CGSC10 and sodium chlorate(CGSCS10) against E. coli O157:H7 infection was studied using ICR female mice. During the incubation period, the dose of 5, 10, and 20% CGSC10 was inhibited the growth of E. coli O157:H7 by 34.7, 60.2, and 76.4%, respectively. For 7 days after single challenge with E. coli O157:H7, forty female ICR mice were divided into four experimental groups which were administered in drinking water with saline, 10% CGSC10, 15 mM sodium chlorate, and CGSCS10, respectively. On the 3rd day, the number of E. coli O157:H7 in mouse feces was significantly decreased by administration of CGSC10, 15 mM sodium chlorate, and CGSCS10 (p < 0.001). On the 7th day-after administration, CGSC10, sodium chlorate, and CGSCS10 were decreased the number of E. coli O157:H7 by 27.1, 67.7, and 83.3%, respectively. According to the results of the present study, administration of CGSCS10 to mice can reduce the severity of E. coli O157:H7 infection. In addition, it is suggested that CGSCS10 represents a good candidate for the treatment of enteric infections in domestic animals.

        In recent days, there is much interest in the biocidal activity of silver since silver is known to be safe and effective as disinfectant and biocidal material against bacteria and viruses. There is also increased interest in the nano-sized silver particles which can be used as effective bactericidal material. The purposes of this study were to investigate the effect of silverized elastomers on growth of oral pathogenic microorganisms, including Streptococcus mutans, Fusobacterium nucleatum, Porphyromonas gingivalis by comparing with conventional non-silverized elastomers. The antibacterial efficacy of silverized elastomer against oral pathogenic microbes was investigated by modified disk diffusion test and growth inhibition test. In modified disk diffusion test, silverized elastomer showed no antibacterial activity against S. mutans, F. nucleatum and P. gingivalis, In culture medium, the growth of S. mutans, F. nucleatum, P. gingivalis was not inhibited by silverized elastomer. Silverized elastomer exhibited no bacteriostatic and/or bacteriocidal effects against some experimental strain.nd clinical indications and diseases.

        This study was carried out to investigate efficacy of aerosol sanitizer with natural antimicrobial and organic acids against Escherichia coli O157:H7, Salmonella Typhimurium and Listeria monocytogenes. The artificially inoculated pathogens on stainless steel coupon were treated with grapefruit seed extract (GFE), acetic acid,citric acid and lactic acid in model cabinet for 5 min. The number of three foodborne pathogens with individual treatment was reduced by 0.34-3.77 log units, treatment with GEF + organic acid was reduced by 1.72-3.89 log units and treatment with GEF + organic acid + alcohol was reduced by 1.46-5.05 log units. By treatment with GEF + lactic acid + alcohol in scale-up model system for 10 min. Populations of E. coli O157:H7, S. Typhimurium and L. monocytogenes were reduced by 3.42, 2.72 and 2.30 log units from the untreated control respectively. From the above result,aerosol sanitizer with natural antimicrobial agents and organic acid can be used as an environmental sanitation method with satisfying the consumer demand on safe food.

        This study was focused on the determination of antibacterial activity of Scutellaria baicalensis extract against antibiotic resistant bacteria(Salmonella Enteritidis, Staphylococcus aureus and enteroaggregative E. coli). Extract of Scutellaria baicalensis were tested for antibacterial activity by paper disc methods. The Scutellaria baicalensis extract in 0.1 g/㎖ and 0.2 g/㎖ showed a significant antibacterial activity against antibiotic resistant bacteria. Minimum inhibitory concentration (MIC) of Scutellaria baicalensis extract were appeared to 2, 048 ㎍/㎖ at S. Enteritidis, S. aureus and enteroaggregative E. coli. Finally, the growth incubation curve was determined using Scutellaria baicalensis extract against S. Enteritidis, S. aureus and enteroaggregative E. coli. The growth of S. Enteritidis was significantly inhibited within 10 hours by the addition of at least 10, 000 ppm of Scutellaria baicalensis extract. The 10, 000 ppm of Scutellaria baicalensis extract retarded the growth of S. aureus and enteroaggregative E. coli more than 10 hours. In conclusion, Scutellaria baicalensis extract might be useful to control antibiotic resistant bacteria in vitro.

        This study was performed to investigate the effect of Portulaca oleracea extract on the antioxidant and antimicrobial activities against Helicobacter pylori. The each solvent extracts prepared from Portulaca oleracea were investigated by measuring total phenolic compounds, electron donating ability, superoxide dismutase-like ability and thiobarbituric acid reactive substances The herb extractor extract yielded the highest content of total phenolic compounds(72.2 ㎎%). The electron donating abilities(EDA) of ethyl acetate and methanol extracts showed high antioxidant activity. The superoxide dismutase (SOD)-like abilities of ethyl acetate and petroleum ether extracts also showed some activity. The antioxidant activity of thiobarbituric acid reactive substances was not significant. The petroleum ether extract of Portulaca oleracea showed the highest antimicrobial activity at 10, 000 ppm concentration.

        Salmonellosis is a major bacterial zoonosis that causes self-limited enteritis to fatal infection in animals and food-borne infection and typhoid fever in humans. Multidrug-resistant strains of Salmonella spp. has increased over the last several decades and recently causes more serious problems in public health. The present study was investigated bacteriocidal effects of sodium chlorate, sodium azide, sodium cyanide, and sodium salts mixture containing sodium chlorate, sodium azide, and sodium cyanide on infection with S. typhimurium in macrophage RAW 264.7 cells, and antibacterial effects of sodium salts mixture for murine salmonellosis. In infection assay of S. typhimurium in RAW 264.7 cells, bacterial survival rates within macrophage in all treated groups was significantly reduced comparing to that of the control group with the passage of incubation time. Administration of sodium salts mixture showed a therapeutic effect for S. typhimurium infected ICR mice. The mortality of mice treated with sodium salts mixture was 70% until 12 days, while that of control mice was 100% until 9 days after S. typhimurium infection. The results of this study strongly indicate that sodium salts mixture has a potency treatment for murine salmonellosis.

        The present study was investigated the antibacterial effect of several sodium salts and the sodium salt mixture composed with sodium chlorate, sodium azide and sodium cyanide on Salmonella gallinarum (S. gallinarum) infection in murine derived macrophage RAW 264.7 cells. In the infection assay of S. gallinarum in macrophage cells pretreated with 15 mM sodium chlorate, 0.3 mM sodium cyanide, 0.3 mM sodium azide and the sodium salt mixture (15 mM sodium chlorate, 0.3 mM sodium cyanide, 0.3 mM sodium azide), respectively, the numbers of S. gallinarum in all treated-groups tended to decrease in the process of time after treatment, but the group treated with sodium cyanide was no significant difference compared with control. After 24 hours of treatment, the number of S. gallinarum in sodium azide (p<0.05), sodium chlorate (p<0.001) and the sodium salt mixture (p<0.001) treated-group was significantly decreased compared with control, and that in the sodium salt mixture treated-group was decreased the higher than all groups. The results of this study demonstrated that the sodium salt mixture composed with sodium chlorate, sodium azide and sodium cyanide, has the antimicrobial activity for S. gallinarum and may be beneficial on the control of intracellular pathogens.
