To understand antioxidant enzyme response of two contrasting Arundinella hirta ecotypes to drought stress, drought-tolerant Youngduk and drought-sensitive Jinju-1, were comparatively analyzed changes in the enzymatic activities of peroxidase (POD), ascorbate peroxidase (APX), catalase (CAT), superoxide dismutase (SOD), and glutathione reductase (GR). Two ecotypes, drought-tolerant Youngduk and drought-sensitive Jinju-1 were subjected to drought stress by withholding water for 12 days. ROS accumulation level and electrolytic leakage were significantly increased in both A. hirta ecotypes by drought stress treatment but less in Youngduk than Jinju-1. The RWC significantly decreased in both the drought stress-treated ecotypes as compared to control, but less in Youngduk than Jinju-1. Soluble sugar and protein content were increased more in drought stress-treated Youngduk as compared to Jinju-1. The activities of antioxidant enzymes such as SOD, CAT, POD, APX, and GR increased significantly in both the drought stress-treated ecotypes Youngduk and Jinju-1 as compared to control. The increase in antioxidant enzyme activity level was more prominent in drought stress-treated Youngduk as compared to Jinju-1. Taken together, these results suggest that Youngduk was more tolerant to drought stress than Jinju-1, and seem to indicate that tolerance of A. hirta to drought stress is associated with increased activity of antioxidant enzymes.

Marine macroalgae are important in coastal ecosystems and interact with marine microorganisms. In this study, we isolated fungi from seven types of marine macroalgae including Cladophora sp., Gloiopeltis furcate, Gracilariopsis chorda, Hydroclathrus clathratus, Prionitis crispata, Sargassum micracanthum, and Ulva lactuca collected in Korea. Morphological and phylogenetic analyses identified the isolates as four Aspergillus spp. (A. fumigatus, A. sydowii, A. tamarii, and A. terreus), three Penicillium spp. (P. crustosum, P. jejuense, and P. rubens), and Cladosporium tenuissimum. Among them, A. fumigatus TOPU2, A. tamarii SH-Sw5, and A. terreus GJ-Gf2 strains showed the activities of all enzymes examined (amylase, chitinase, lipase, and protease). Based on the enzymatic index (EI) values in solid media, A. terreus GJ-Gf2 and C. tenuissimum UL-Pr1 exhibited the highest amylase and lipase activities, respectively. Chitinolytic activity was only observed in A. terreus GJ-Gf2, A. tamarii SH-Sw5, and A. fumigatus TOP-U2. Penicillium crustosum ULCl2 and C. tenuissimum UL-Pr1 showed the highest protease activities. To the best of our knowledge, this is the first report of lipolytic and proteolytic activities in a marine-derived C. tenuissimum strain. Overall, the fungal strains isolated from the marine macroalgae in this study actively produced industrially important enzymes.

Macrophages secrete various cytokines and inflammatory mediators, resulting in playing critical roles in inflammation and immunity. In this study, we investigated anti-inflammatory and immune enhancing properties of PB203, which is a water-soluble extract powder from the fruit of Actinidia polygama, in macrophages. A. polygama is a medicinal plant traditionally known to treat abdominal pain, stroke and rheumatoid arthritis. However, the molecular mechanism for the immune modulation of PB203 is still unclear. Therefore, we assessed the effects of PB203 on the lipopolysaccharide (LPS)-induced inflammation and immune activation, and elucidated its action mechanism in mouse macrophage, RAW264.7 cells. PB203 significantly suppressed not only the levels of nitric oxide (NO), prostaglandin E2, tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β), but also the mRNA expression of inducible NO synthase, cyclooxygenase-2, TNF-α and IL-1β in LPS-stimulated RAW264.7 cells. We also found that these anti-inflammatory activities of PB203 were mediated through the inhibition of toll-like receptor 4 and nuclear factor kappa B (NF-κB) induced by LPS. On the other hand, in normal macrophages, PB203 dose-dependently elevated the gene expression of immunomodulators including granulocyte-macrophage colony-stimulating factor, granulocyte colony-stimulating factor, monocyte chemoattractant protein-1 and TNF-α in a statistically significant manner. The expression of IL-10, IL-1β, IL-6, and interferon-γ were also remarkably upregulated by the treatment of 500 μg/mL PB203. In addition, PB203-mediated production of NO and TNF-α was attenuated by NF-κB inhibition in RAW264.7 cells. Interestingly, PB203 promoted the production of nuclear factor erythroid-2-related factor 2, resulting in the increased level of heme oxygenase-1, which is a representative antioxidant enzyme, in both LPS-stimulated and normal RAW264.7 cells. Taken all together, these results suggest that PB203 may have great potential as the candidate of anti-inflammatory agent for improving inflammatory diseases or immune enhancing agent for preventing infectious diseases. Keywords: Actinidia polygama extract (PB203); macrophages; immunomodulator; nuclear factor kappa B (NF-κB); heme oxygenase-1 (HO-1)

Safety-related items in the decommissioning Nuclear Power Plants (NPPs) can largely consider safety for workers and residents. At this time, the effects of radioactive contamination on the Systems, Structures, and Components (SSCs) are caused by the performance of work related to Decontamination and Dismantlement (D&D) activities. Classification according to dismantling activities will be important, and the decay factor of radionuclides and the impact of contaminations due to plant characteristic (thermal and electrical capacity) in estimation of exposure dose from such activities will be considered compared to other overseas NPPs. Therefore, this study will consider some factors to consider for comparison with overseas cases in estimating worker exposure dose. To assess worker exposure doses, the classification of decommissioning activities must first be made. It should be classified including large components that can be generally considered, and the contents should be similar to compare with overseas cases. In case of decommissioned NPPs with prior experience, it is possible to predict worker’s exposure with respect to plant capacity, but this does not seem to have a specific correlation when reviewing the related data. Depending on the plant capacity, the occurrence of contamination of radioactive materials may have some correlation, but it cannot be determined that it has causality with the worker’s dose when dismantling. In addition, it is expected that the effects of workers’ exposure doses will vary depending on when the highly contaminated SSCs will be dismantled from permanent shut down. Therefore, the decay correlation coefficient for this high radiation dose works should be considered. If the high radiation dose work is performed before the base year, a correlation coefficient larger than 1 value will be applied, and in the opposite case, a value less than 1 will be applied. Whether or not to perform Full System Decontamination (FSD) is also an important consideration that affects worker dose, and correlation factors should be applied. In this study, the matters to be considered when estimating worker dose for dismantling NPPs were reviewed. This suggests factors to be reflected in the work classification and dose results for comparison with overseas NPP experiences. Therefore, when doing the workers’ dose estimation, it is necessary to derive a normalized doses considering each correlation factor when comparing with overseas cases along with dose estimation for the dismantling activities.

For licensees who face the decommissioning project for the first time, even if they can utilize their experience in operation, they should be well prepared and assessed for the risks of dismantling activities reflecting the characteristics of decommissioning. This can be included in the risk management of the decommissioning project, but what we want to discuss in this study is the evaluation of the industrial risk of the actual work before the dismantling work is carried out. We would like to focus more on the review of dismantling activities subject to industrial risk assessment and a series of processes for risk assessment. The dismantling work plan will need to obtain approval from the supervisory department before work on the Systems, Structures, and Components (SSCs) can be carried out. At this time, risk assessment may be included among many safety-related required documents, which are divided into radiological and non-radiological risks. The target activities at Level 1 level can include preparation for dismantling and maintenance of facilities, dismantling big components, removing the contamination of concrete structures, managing radioactive waste, etc. In addition, it can be composed of preparation work, removal of connections, lifting/installation, cutting, radiation/radioactivity measurement, and withdrawal as detailed work stages of each item’s activities. For domestic nuclear decommissioning projects, two major performance organizations, licensees and contractors, must be considered. Regarding risk assessment, the licensee will have a supervisory department controlling decommissioning activities and an HSE department at the site, and a process will need to be established in consideration of the contractor’s work organization. Therefore, activities in the risk assessment process may be established. In this study, risk assessment was reviewed as safety-related matters to be considered when carrying out the dismantling work. Safety-related risk assessment is a necessary procedure for performing practical dismantling activities, and this should be considered well in advance. Therefore, work activities and criteria were established for risk assessment, and the performance process was assumed to apply them. In terms of the performance organization and the responsibilities and roles of the processes to be performed by each organization were constructed, and this can be referred to in the process of preparing for the decommissioning project.

Since the National R&D Innovation Act was enacted in 2022, it became a crucial issue how to qualify or improve R&D activities and disseminate their outcomes. Many organizations have referred to various quality management standards such as the American National Standards Institute/American Society for Quality (ANSI/ASQ) Z1.13, International Organization for Standardization (ISO) 9001, and the American Society of Mechanical Engineers Nuclear Quality Assurance-1 (ASME NQA-1), as a means to set up their own quality system. ISO is the international standard for implementing a quality management system (QMS), which provides a framework and principles for managing an organization’s QMS, with the aim of ensuring that the organization consistently provide products or services that meet regulatory requirements. ISO 9001 can cover all aspects of an organization’s operations, and it can also be expanded to include R&D areas. The introduction of ISO 9001 to R&D aims to improve R&D practices and establish a standardized process framework for conducting R&D. ANSI/ASQ Z1.13 provides quality guidelines for research and consists of 10 sections covering various aspects of research quality, emphasizing ethical conduct, clear objectives, reliable data collection, and analysis. ASME NQA-1 is one of quality assurance standards for nuclear facility applications, but it has been extended and applied to R&D activities in the nuclear fields. It just focuses on planning, procedures, documentation, competence, equipment, and material control. KINAC has conducted extensive research on verifying and regulating nuclear activities while providing support for national nonproliferation technologies and policies. In addition to the quantitative growth achieved so far, efforts are being made to establish a qualitative and integrated management system. As a first step to achieve this goal, this study reviewed international standards and methodologies for research quality and derived the key components for R&D quality management. Moreover, the appropriate outline of quality management system framework was proposed for R&D as a regulatory support process, based on the ISO 9001. The implementation of quality management standards and procedures for R&D in KINAC, which could lead to improved research practices, more reliable data collection and analysis and increased efficiency in conducting R&D activities.

Satellite imagery is an effective supplementary material for detecting and verifying nuclear activities and is helpful in areas where access and information are limited, such as nuclear facilities. This study aims to build training data using high-resolution KOMPSAT-3/3A satellite images to detect and identify key objects related to nuclear activities and facilities using a semantic segmentation algorithm. First, objects of interest, such as buildings, roads, and small objects, were selected, and the primary dataset was built by extracting them from the AI dataset provided by AIHub. In addition, to reflect the features of the area of interest (e.g., Yongbyon, Pyongsan), satellite images of the area were acquired, augmented, and annotated to construct an additional dataset (approximately 150,000). Finally, we conducted three stages of quality inspection to improve the accuracy of the training data. The training dataset of this study can be applied to semantic segmentation algorithms (e.g., U-Net) to detect objects of interest related to nuclear activities and facilities. Furthermore, it can be used for pixelbased object-of-interest change detection based on semantic segmentation results for multi-temporal images.

Background: The gluteus maximus (GM) muscle comprise the lumbo-pelvic complex and is an important stabilizing muscle during leg extension. In patients with low back pain (LBP) with weakness of the GM, spine leads to compensatory muscle activities such as instantaneous increase of the erector spinae (ES) muscle activity. Four-point kneeling arm and leg lift (FKALL) is most common types of lumbopelvic and GM muscles strengthening exercise. We assumed that altered hip position during FKALL may increase thoraco-lumbar stabilizer like GM activity more effectively method. Objects: The purpose of this study was investigated that effects of the three exercise postures on the right-sided GM, internal oblique (IO), external oblique (EO), and multifidus (MF) muscle activities and pelvic kinematic during FKALL. Methods: Twenty eight healthy individuals participated in this study. The exercises were performed three conditions of FKALL (pure FKALL, FKALL with 120° hip flexion of the supporting leg, FKALL with 30° hip abduction of the lifted leg). Participants performed FKALL exercises three times each condition, and motion sensor used to measure pelvic tilt and rotation angle. Results: This study demonstrated that no significant change in pelvic angle during hip movement in the FKALL (p > 0.05). However, the MF and GM muscle activities in FKALL with hip flexion and hip abduction is greater than pure FKALL position (p < 0.001). Conclusion: Our finding suggests that change the posture of the hip joint to facilitate GM muscle activation during trunk stabilization exercises such as the FKALL.

This study aimed to identify and characterize lactic acid bacteria strains with antimicrobial activity against Proteus mirabilis, a gram-negative bacterium associated with Parkinson’s disease, Crohn’s disease, and nosocomial infections. Among the 987 lactic acid bacteria strains isolated from various sources, strain no. CHK903 showed high antimicrobial activity against P. mirabilis. Phylogenetic tree analysis based on the 16S rRNA gene and scanning electron microscope analysis identified the selected strain as the rod-shaped Weissella cibaria. The culture supernatants of W. cibaria CHK903 showed antimicrobial activity against some pathogens. Two antimicrobial compounds with molecular weights 189 and 365 Da were partially purified from the culture supernatants of W. cibaria CHK903 using Biogel P2 gel permeation column chromatography. The culture supernatants of W. cibaria CHK903 also showed significant antibiofilm properties, inhibiting biofilm formation by 90% and removing pre-formed biofilms by 60%. These findings suggest the potential therapeutic use of W. cibaria CHK903 as a natural antimicrobial against P. mirabilis-related infections.

The objective of this study is to evaluate the antioxidant components, elastase inhibition activities, and melanin synthesis rates of Korean traditional Actinidia (Actinidia arguta) fruits and leaves depending on the ethanol extraction concentrations. The total polyphenol content was the highest in the 50% ethanol extract of both fruits and leaves, with values of 634.1 mg GAE/100 g and 3,985.2 mg GAE/100 g, respectively. The total flavonoid content was the highest in the fruit 90% extract and leaf 50% extract at 191.9 mg/100 g and 2655.6 mg/100 g, respectively. The vitamin C content was the highest in the 50% extract of leaves at 2990.3 mg/100 g. Elastase inhibition was the highest at 56.9% in the leaf 50% extract at a concentration of 1,000 μg/mL. Melanin synthesis inhibition showed the highest melanin synthesis inhibitory effect among the extracts, as the leaf 50% extract showed an inhibitory rate of 65% or more. Therefore, the antioxidant components, elastase inhibition activities, and melanin synthesis inhibitory rate were better in leaves than in fruits. The leaf 50% extract was particularly the best among the extracts. Korean traditional Actinidia leaves can be considered as potential sources for new functional materials.
