        국내에서 유망 소과류 및 베리류로 농가에 보급되고 있는 과종 및 품종들을 대상으로 각 과실들 간의 안토시아닌, 폴리페놀 함량, 각 구성성분 및 항산화활성을 분석하였다. 총 안토시아닌 및 폴리페놀 함량을 분석한 결과, 같은 과종에서도 품종에 따라 안토시아닌과 폴리페놀 함량의 차이가 나타났으며, 총 안토시 아닌은 블랙 쵸크베리가 가장 많았으며(927.9~1,149.1 ㎎・100 g-1, F.W.), 총 폴리페놀 함량도 블랙 쵸크베리(502.2~530.5 ㎎・100 g-1, F.W.)에서 가장 많았다. DPPH 항산화활성에서도 블랙 쵸크베리가 가장 높은 것으로 나타나 안토시아닌, 페놀 함 량과 항산화활성과 고도의 상관관계를 나타냈다. 주요 폴리페놀 구성 성분을 조사한 결과 모든 과종에서 quercetin, catechin, epicatechin 함량이 높게 나타났다. 블루베리와 블랙 커런트의 주요 안토시아닌은 delphinidin-3-glucoside로 사스카툰 베리와 블랙 쵸크베리의 주요 안토시아닌은 cyanidin-3- galactoside였다.

        과피와 과육의 다양한 색은 복숭아 분류에 가장 널리 사용되는 상업적 기준 중 하나이다. 새로운 적색 과육 품종을 육성하기 위해서는 많은 교배 조합과 세대가 진전되어야 한다. 따라서 육종 효율을 높이기 위해서는 목적 형질을 가진 개체에 적용할 조기 선발 분자표지를 개발할 필요가 있다. 과육색이 다르게 발현되는 복숭아 품종의 유전자 발현을 비교하기 위해 2개의 cDNA library를 제작하였다. 적색 과육 품종인 ‘조생혈도’와 백색 과육 품종인 ‘미백도’의 유전자 발현 차이를 보기 위해 차세대 염기서열 분석(NGS) 기술을 사용하였고, 두 품종으로부터 얻은 EST의 염기서열을 결정하고 기존에 보고된 유전자와의 상동성을 분석하였다. ‘조생혈도’와 ‘미백도’의 EST database로부터 72쌍의 SNP 분자표지를 선발하였고, 적색 과육 품종 8개와 백색 과육 품종 24개를 구분할 수 있는 SNP 분자표지를 HRM 방법으로 분석하였다. 본 연구에서는 복숭아 EST database를 기반으로 HRM 분석 방법을 이용하여 복숭아 품종의 적육계와 백육계를 구분할 수 있는 효율적인 SNP 분자표지를 개발하였다. 이러한 SNP 분자표지는 복숭아 육종에 유용하게 사용할 수 있으며, 복숭아 품종의 다양한 색 변화에 관한 분자 기작 연구에 좋은 참고자료가 될 수 있을 것이다.

        Background : Solanum nigrum Lin. is an annual herbaceous plant which belongs to the family Solanaceae as known the important medicinal herb and health-benefiting plant. Phytochemical investigation of whole plant reported that contain total alkaloids, steroid alkaloids, steroidal saponins, flavonoids, tannins, glycosides, proteins, carbohydrates, coumarins and phytosterols, and glycoproteins, exhibiting antitumor activity. Methods and Results : The accumulation of anthocyanin known as synthesis by flavonoid biosynthesis pathway is increased by AtPAP1 transcriptional factor. To identify the effect of AtPAP1 in S. nigrum, we generate AtPAP1-overexpressed S. nigrum and investigate total anthocyanin in these transgenic lines. The transgenic lines resulted in increasing the total anthocyanin amount compared with control line. Thus, the transgenic lines appeared purple colored leave as well as light purple in flower petal and dark purple in flower anther. To identify that AtPAP1 affects the genes profiling of anthocyanin biosynthetic pathway in S. nigrum, we harvested the AtPAP1-overexpressed S. nigrum transgenic lines and proceeded to qRT-PCR analysis. The result indicated that AtPAP1 gene highly induces in transgenic lines and the most upstream pathway genes of flavonoid such as SnPAL, SnC4H, Sn4CL, SnCHI and SnHCT are induced by AtPAP1 overexpression, excepting UGT75C1. According to these results, those functional genes may be regulated by AtPAP1 transcription factor. Thus, the purple color appearing in AtPAP1-overexpressed S. nigrum plants may be regulated by those genes. Conclusion : When AtPAP1 is overexpressing, the most genes in flavonoid pathway upregulate and activate resulting in accumulation of anthocyanin compound in leave and flower organs in S. nigrum. Ultimately, our data will be helpful for further research in S. nigrum molecular and physiology and better strategies for exploiting its phytochemical properties.

        The study was performed to investigate basic tree growth and fruit characteristics of ‘Nero’ black chokeberry (Aronia melanocarpa) depending on the different cultivation regions and altitudes in 2014, 2015. Tree growth and change of developmental stages of ‘Nero’ were similar regardless of meteorological environment of the four cultivation regions (Danyang, Okcheon, Yeongdong, and Geumsan) in 2014. Fruit characteristics of ‘Nero’ were significant differences in berry weight and anthocyanin content among the four orchards due to difference in cultivation techniques and relatively wide daily temperature range just before harvest, respectively. Tree growth and change of developmental stages of ‘Nero’ grown at the two orchards with different altitudes appeared to be similar during the successive years 2014 and 2015. Soluble solids content of the berries cultivated at low altitude (117 m) was higher than at high altitude (342 m). Acidity showed an inverse pattern with soluble solids content. Anthocyanin content increased progressively until at the middle of August, 2014-15 and then it decreased. Our results showed that black chokeberry is a species adaptive to the domestic environment as there were no differences in tree growth and change of developmental stages of ‘Nero’. Considering fruit quality of black chokeberry such as soluble solids content, acidity, and anthocyanin content, our results suggest that optimal harvest period of black chokeberry ‘Nero’ is August 8 to 19.

        We report the development of new anthocyanin-rich grain corn hybrid variety ‘Saekso 2’. The seed parent, HA3 and the pollen parent, HA4 was developed from breeding materials collected from China in 2008 and from Canada in 2000, respectively. The hybrid was made in the winter of 2010/2011 and evaluated in Hongcheon for 3 years. After evaluation, the selected variety was named ‘Saekso 2’ and was approved for a variety registration in 2017. Anthocyanin content of Saekso 2 in grain was 685 mg/100g and was much higher than that of Saeko 1(0mg/100g). Since regional tests was conducted in only Gangwon province, it is recommended only in that region for commercial cultivation. For the highest content of anthocyanin, it is best to harvest grains at around 45 days after silking. Saekso 2 is the first hybrid bred specifically for processing for high anthocyanin production.

        Background: Purple potato contain sufficient phenolic compound and flavonoid which has high antioxidant capacity. Due to poor water solubility of phenolic compounds and quick oxidation of anthocyanin, we could not get maximum health benefits from purple potato. Therefore, we developed surfactant based aqueous nano suspension to enhance the solubility of phenolic compounds and protect the oxidation of anthocyanin from purple potato. Methods and Results: Two types of surfactant were used in this experiment based on hydrophilic-lycophilic balance viz. Brij and Span. In our study, lycophilic surfactant showed highest efficiency in TP extraction compared to water and lipophilic surfactant below 10 mM concentration. On the other hand, lipophilic surfactant showed highest efficiency in extracting flavonoid content. Conclusion: It is concluded that hydrophilic surfactant was significantly increased phenolic compounds five times, and lipophilic surfactant increased flavonoid two times, and anthocyanin three times compared to control. Therefore, total antioxidant capacity was increases two times compared to control.

        The purpose of this study was to obtain basic data for using Aronia as a functional food material. The composition of anthocyanin was characterized and quantitated by LC-MS/MS, HPLC, and UV-VIS spectrophotometer techniques, respectively. The anthocyanin content was analyzed by temperature, time, pH, and the addition of citric acid. The UV-VIS spectrophotometer used for analysis of anthocyanin is less accurate than the LC-MS/MS method used in recent years. In the past, cyanidin-3-Glucoside was reported to be a major anthocyanin that contains Aronia. However, LC-MS/MS analysis in this study confirmed cyanidin-3-galactoside to be the major compound. The anthocyanin content of the Aronia powder began to decrease sharply at a temperature of 65 ℃ or higher when heated for 24 hours. In an aqueous solution of Aronia, the anthocyanin content was reduced by 50% at 65 ℃ for 10 hours and decreased by 85% at 85 ℃ within 10 hours. Above pH 8, the anthocyanin content was reduced by more than 50%. The results of this study will provide useful information to maintain anthocyanin content in the manufacturing process of Aronia. It could also be used to ensure the stability of anthocyanins in similar species of berries.

        This study was carried out to create new waxy corn variety using anthocyanins germplasm including 13 ornamental maize stocks, purple corn and two parental lines of Mibaek2. First, we made anthocyanin husk population for sustainable breeding from 13 ornamental stocks for 6 years from 2001 and then crossed purple corn with a dark purple kernel in 2007. We crossed this hybrid with two parental lines of Mibaek2, a good edible quality waxy corn, in 2007/2008 dry season nursery for additional breeding in Thailand. The anthocyanin-rich waxy lines derived from these hybrids were self-fertilized seven times and made single cross hybrids until 2011. These hybrids were evaluated on 2 or 3 places in Gangwon-do for next two years. One selected elite hybrid was named Cheongchunchal and applied for registration of new waxy corn variety in March 2014. Total anthocyanins content is zero in Mibaek2, 737 in Miheugchal, 18,809 mg/kg in Cheongchunchal. This variety is meaningful as health food and high-value industrial material.

        Background : Mulberry (Morus alba L.), renowned for their medicine benefits and the leave as the sole food for silkworm (Bombyx mori). To understanding the molecular mechanism of color formation and nutritive value in different mulberry fruit varieties, we use high-throughput transcriptome sequencing technique to investigated the anthocyanin and betulinic biosynthesis pathway related functional genes. In addition, the total antosyanin and betuinic acid contend were also measured. Methods and Results : The resulting cDNA library was then sequenced using an Illumina HiSeq™ 2000 system. The clean reads were assembled using Trinity software, Then perform gene family clustering to get final unigenes. The pH differential method was used to determine the total anthocyanin content (TAC) of methanol extract from the red and white mulberry, and High-performance liquid chromatography (HPLC) analysis was used to quantify the triterpenes content. In this study, total 50,149 unigenes with an average length of 1,125 nt and N50 equaling 1,861 nt were generated. Using these transcriptome sequecing, cDNAs encoding anthocyanin biosynthetic genes and triterpene biosynthetic genes were isolated. In addition, total anthocyanins and betulinic acid content were analyzed. A great amount of total anthocyanins (59.16 mg/g) were found in fully ripe fruit of Cheongil. Accumulation of betulin and betulinic acid were also detected in all stages of Cheongil and Turkey fruits with small amount. Conclusion : The results of transcriptome sequencing provide useful information at molecular lever in mulberry research, such as interesting gene discovering, marker assisted molecular breeding, and interesting metabolic pathway investigate. The gene expression results could help us understanding of the molecular mechanisms of different fruit color determining factor.

        Background : The objective of this study was to investigate antioxidant activities, inhibitory activities against heme induced colonic epithelial cell proliferations, anti-inflammatory activities and anthocyanin profiles in the anthocyanin rich fraction (ARFAM) from fruits of Aronia melanocarpa, where these are considered functional substances and available food coloring agents in Korea. Methods and Results : Anthocyanins were identified by reversed-phase C18 column chromatography and HPLC-DAD-ESI/MS analysis. To compare the antioxidative and anti-inflammatory capacity of Aronia melanocarpa berries, recognized for their high content of anthocyanins, isolation method was developed to obtain high-purity anthocyanins in the extract. Anthocyanin-rich fractions (ARFAM) enriched in anthocyanins were found to be potent strong inhibitory activity towards heme induced colonic epithelial cell proliferations are associated with an increased risk of colon cancer than acidic ethanol extract (AME). The immunomodulation properties were assessed in growth of both human B and T cells, its cytokines secretion such as IL-6 (interleukin-6) and TNF-α (tumor necrosis factor-alpha). AME enhanced interleukin-6 and reduced tumor necrosis factor-a production, whereas ARFAM only had a effect in increasing of IL-6 expression. Conclusion : These results demonstrated that there was no major relationship between the antioxidative and immunomodulation capacities of AME and ARFAM.

        The purpose of this study was to determine the content of phenolics and various anthocyanin compounds, and antioxidant and anticancer activities of the extracts from pomergranates, which were produced in two different regions including Goheung, Korea and California, USA. These pomergrantes were divided into juice, pericarp, and seed parts and each part was extracted with 95% methanol. Content of total phenolics [mg chlorogenic acid equivalents/kg DW] was highest in pericarp, followed by juice and seeds from pomergrantes in both regions. The anthocyanins identified in pomegranate fruit were cyanidin-3,5-diglucoside chloride (Cy3,5G), cyanidin-3-O-glucoside chloride (Cy3G), delphini din-3,5-di-O-glucoside chloride (Dp3,5G), delphinidin-3-O-glucoside chloride (Dp3G), pelargonidin- 3,5-di-glucoside chloride (Pg3,5G), and pelargonidin-3-glucoside chloride (Pg3G). Among these, cyanidin-3, 5-diglucoside chloride (Cy3,5G) was the major anthocyanin in California pomegranate fruit juice and Goheung pomegranate fruit pericarp. DPPH (1,1-diphenyl-2-picryl hydrazyl radical) free radical scavenging activity was dose-dependently increased, and was higher in pericarp part than juice or seed parts from pomergrantes in both regions. By MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay, pericarp extracts from pomergrantes in both regions showed the highest anticancer activity, which was higher on Calu-6 for human pulmonary carcinoma than SNU-601 for human gastric carcinoma. Correlation between polyphenols and anticancer activity on Calu-6 was determined to be in the range of r2=0.8904 to 0.9706.

        There has been known C3G (Cyanidin-3- Glucoside), D3G (Delphinidin-3-Glucoside), and Pt3G (Petunidin- 3-Glucoside) were main anthocyanin pigments in black-seeded soybean. Anthocyanin contents of total 1,032 black-seeded soybean germplasms were analyzed by HPLC. Average of total anthocyanin content was 11.67 mg/g on the all materials ranged from 0.54 to 23.45 mg/g. Mean value of C3G, D3G, and Pt3G contents in all black-seeded soybeans were 8.81, 1.78 and 0.79 mg/g, respectively. Environmental conditions influenced anthocyanin contents during seed development. Delayed flowering, especially of later maturing germplasms, has been showed to result in increased anthocyanin content. So, prolonged maturation period germplasm is generally higher than that of shorten genotypes. It may be concluded that the higher levels of anthocyanin content was associated with the late dates of harvest maturity. Also larger seeds showed high anthocyanin contents than smaller. That inclination is similar in C3G’s occasion because C3G content contribute highly to total anthocyanin content than other pigments.

        Seed color is an important factor affecting physiological and developmental process in wheat. One of the plant pigments, anthocyanins are a group of flavonoid compounds well known as pigments responsible for blue, purple, red, or yellow coloration of plant tissues. In this study, we investigated the pigmentation of purple and yellow color seed according to wheat grain developmental stages. The contents of anthocyanin and chlorophyll in the purple and yellow seeds were measured. Chlorophyll contents were changed similarly in both purple and yellow color seed, and no significant difference was observed between them. In purple color seed, the content of anthocyanin was significantly induced compared with yellow color seed. The individual anthocyanin components were investigated by ultra performance liquid chromatography (UPLC). Cyanidine-3-glucoside (C3G) and peonidine-3-glucoside (P3G) were detected as predominant anthocyanin in purple color wheat. To investigate whether structural genes in anthocyanin biosynthesis were involved in the trait differences between purple and yellow color seed, we examined the expression of anthocyanin biosynthesis-related genes (CHS, CHI, F3H, DFR, ANS, UFGT) and MYB transcription factor in developing wheat grains by using qRT-PCR. This study indicates that the expression of anthocyanin biosynthesis-related genes and MYB transcription factors correlate with anthocyanin levels of grain.

        Anthocyanins, providing the bright red-orange to blue-violet colors, flavonoid-derived pigments with strong antioxidant activity that have benefits for human health. We isolated RsMYB1, which encodes an R2R3 MYB transcription factor (TF), from red radish plants (Raphanus sativus L.) that accumulate high levels of anthocyanins. RsMYB1 shows higher expression in red radish than in common white radish, in both leaves and roots, at different growth stages. regulatory genes. Transient expression of RsMYB1 in tobacco showed that RsMYB1 is a positive regulator of anthocyanin production. Also, the synergistic effect of RsMYB1 with B-Peru was larger than the effect of Arabidopsis plants stably expressing RsMYB1 produced red pigmentation throughout the plant, accompanied by up-regulation of the six structural and two regulatory genes for anthocyanin production. This broad transcriptional activation of anthocyanin biosynthetic machinery in Arabidopsis included up-regulation of TRANSPARENT TESTA 8, which encodes a bHLH-type TF. These results suggest that overexpression of RsMYB1 promotes anthocyanin production by triggering the expression of endogenous bHLH genes as potential binding partners for RsMYB1. In addition, RsMYB1-overexpressing Arabidopsis plants had a higher antioxidant capacity than did non-transgenic control plants. Taken together, RsMYB1 is an actively positive regulator for anthocyanins biosynthesis in radish plants and it might be one of the best targets for anthocyanin production by single gene manipulation being applicable in diverse plant species.

        Anthocyanin is known for positive health beneficial effects that including reduces age related oxidative stress and inflammatory responses. It was produced by vegetable crops and a lettuce is one of the crops. The general pathway of anthocyanin expression is well defined but it is not clear how environments effects on anthocyanin accumulation in a lettuce. Therefore we initiated to study interaction between anthocyanin expression and environment factors. Frist, we applied RGB leaf images in a lettuce to calculate anthocyanin areas in a leaflet with two different cultivars, different developmental stages, and different environments. Later, we attempted to capture RNA expression level with next generation sequence (NGS) RNA sequencing method called RNA-seq. As a result, combined two technologies showed that quantitate phenotypic data help to understand the gene expression of anthocyanin in lettuce cultivars.

        Anthocyanins are responsible for vivid colors of flowers, fruits and vegetative tissues and biosynthesis of it is primarily controlled by several structural and regulatory genes. The regulatory mechanism of this pathway is still unknown. This study identified 19 transcription factors of Brassica rapa and investigated their regulatory function in anthocyanin biosynthesis pathway genes and cold and/or freezing tolerance in B. rapa. Expression analysis of these genes in the pigmented and non-pigmented portion of leaves of different lines of B. rapa revealed that BrMYB2-2 and BrTT8 showed responses contrasting with anthocyanin accumulation and cold stress. Sequences of these genes were analyzed and compared with similar gene sequences from other species and a high degree of homology with their respective functions was found. Co-regulated cis -elements were found in promoters of BrPAL1, BrCHS, BrF3H1, BrF3’H1, BrFLS, BrBAN, BrDFR8, BrANS1, and BrMYB2-2 and BrTT8 had binding sites of the promoters of those structural genes. Thus, the above results suggest the association of BrMYB2-2 and BrTT8 with regulation of anthocyanin biosynthesis pathway genes and cold and freezing stress tolerance and might be useful resources for development of cold resistant Brassica crops with desirable colors as well.

        In sustainable viticulture, green manure represents a safe and non-polluting way to bring large quantities of organic matter into the soil. The cultivation of green manure crops plays an important role in soil quality and sustainability of agricultural system. This study was conducted to evaluate the effects of green manure fertilization on yield and quality of the Campbell Early grape variety in the organic vineyard. Hairy vetch was the more productive green manure crop, yielding higher dry materials. Average yields of grape were significantly greater in hairy vetch + rye (13.02 ton ha-1) than nature weed (11.65 ton ha-1), respectively. The concentrations of total as well as individual anthocyanins were consistently higher with hairy vetch treatment compared with rye and nature weed, thus making the green manure cultivation is an environmentally friendly cultivation to increase the yield and anthocyanin contents in organic grape.

        This study was carried out to determine environmental factors affecting the anthocyanin content of colorfleshed potato (Solanum tuberosum L.) tubers. After planting of two color-fleshed potato cultivars of ‘Hongyoung’ and ‘Jayoung’ in different 14 locations, their soil chemical properties and meteorological data were evaluated, and anthocyanin contents of tubers were analyzed after harvest, additionally their relationship among them was analyzed through correlation analysis. In comparison with two cultivars, purple-fleshed ‘Jayoung’ potatoes showed higher anthocyanin content than red-fleshed ‘Hongyoung’ in almost locations. When locations were divided to three categories (highland, sub-highland and lowland) according to altitude, in general, highland-grown tubers had the higher content of anthocyanin compared to those grown in lowland. An analysis of the results of chemical components of soil showed that anthocyanin content of color-fleshed potato tubers was negatively correlated with the pH of soil. In addition, mean temperature and minimum temperature from 80 to 100 days after planting most significantly affected on the accumulation of anthocyanin in color-fleshed potato tubers, that is, higher content of anthocyanin was observed in tubers produced in locations with lower mean temperature and minimum temperature from 80 to 100 days after planting. This information can be useful to producers and industries in selection of proper fields for the production of color-fleshed potato tubers having high quality in Korea.
