        Chrysanthemum boreal, C. indicum, and C. indicum var. albescens are well-known wild Chrysanthemum species used for traditional medicine in Korea. In this study, volatile compounds from three wild Chrysanthemums were identified according to four different flowering stages and analyzed using HS-SPME-GC-MS to determine the temporal variation of the volatiles. As a result, 132, 151, and 142 peaks were identified from C. boreale, C. indicum, and C. indicum var. albescens, respectively. Furthermore, 70 out of 132 peaks were identified in C. boreale with a matching ratio of >90% from library search. In addition, 85/151 and 76/142 peaks were identified from C. indicum and C. indicum var. albescens. Forty-nine volatile compounds were found commonly in all three wild Chrysanthemums through all four different flowering stages. However, six, seven, and five unique compounds were detected only in C. boreale, C. indicum, and C. indicum var. albescens, respectively. One hundred volatile compounds were selected for multivariate analysis considering volatile compounds overlapped with each other. The one-way ANOVA (p < 0.05) detected significant differences from 77 out of 100 volatile compounds. In addition, PLS-DA showed the different profiles of volatile compounds according to four different flowering stages in each wild Chrysanthemum. PC1 of each Chrysanthemum accounted for 45.8 56.9, and 11.9% in C. boreale, C. indicum, and C. indicum var. albescens, respectively. PC1 of C. boreale and C. indicum clearly separated the BF stage and the other three stages. Conversely, PC1 of C . indicum var. albescens showed a difference in the composition of volatile compounds between the BF/BO and HO/FO stages. In addition, the different profiles of volatile compounds could be visualized using a heatmap from three wild Chrysanthemums according to four different flowering stages. This study will help improve particular volatile compounds in three wild Chrysanthemums both in quality and quantity.

        The change in volatile compound composition of three wild chrysanthemum species (Chrysanthemum boreale, C. indicum, and C. indicum var. albescens) was identified and analyzed according to four flowering stages using HS-SPME-GC-MS (headspace solid-phase microextraction coupled to gas chromatography–mass spectrometry). The top five compounds of each flowering stage were selected because those main compounds accounted for 43.25%, 44.14%, and 54.20% of the total relative content of volatile compounds from C. boreale, C. indicum, and C. indicum var. albescens, respectively. Nine compounds (1S-α-pinene, α-thujone, chrysanthenone, umbellulone, thymol, caryophyllene, germacrene D, α-zingiberene, and α-patchoulene) in C. boreale were ranked in the top five compounds through the whole flowering stages. In C. indicum, camphene, eucalyptol, camphor, umbellulone, bornyl acetate, caryophyllene, β-farnesene, germacrene D, and α-zingiberene were ranked in the top five compounds. However, only five compounds (camphor, bornyl acetate, β-farnesene, germacrene D, and α-zingiberene) were ranked in C. indicum var. albescens showing a more stable composition rather than C. boreale and C. indicum. Flowerheads of three wild Chrysanthemums showed a different profile of volatile compounds according to different flowering stages, varying compositions, and relative content in the top five volatile compounds. This study illustrates how main volatile compounds in wild chrysanthemums change dynamically during the flowering regarding compositions and their relative contents, suggesting that it should provide a useful index for harvesting or blending certain target compounds from wild chrysanthemums.

        Ornamental chrysanthemum (Dendranthema × grandiflorum Ramat.) belongs to genus Chrysanthemum in the Asteraceae (Compositae) family and is a top leading ornamental plant worldwide. During the last two decades, advances in biotechnology (genetic engineering) and nucleotide sequencing techniques have enabled a deep understanding of biological processes of many ornamental plants at a molecular level. The blue-colored chrysanthemum has been generated and commercialized, and various DNA markers were developed using advanced techniques. Ornamental chrysanthemums are hexaploidy (2n = 2x = 54) and self-incompatible, making it challenging to generate a pure line as breeding resources and a model platform for chrysanthemum research. However, recently, self-compatible pure lines of chrysanthemum (C. seticuspe) were developed, and whole-genome sequencing datasets have been open to the public. Other datasets of whole-genome sequences and transcriptome have been elucidated from wild chrysanthemums, C. nankingense and C. morifolium. In this review, recent progress in the study of ornamental chrysanthemums and the potentiality and importance of wild chrysanthemums as bioresources for ornamental chrysanthemum studies and breeding resources is discussed briefly. Many important resources for studying chrysanthemums are now available and easily accessible. Comprehensive studies with important resources will shed light on chrysanthemum research and breeding of ornamental chrysanthemums at a molecular level. Additionally, this review provides comprehensive information and should pave the way for successful research on ornamental chrysanthemums.

        Dental caries is the most common chronic disease in the dental field. Streptococcus mutans (S. mutans) is the most important bacteria in the formation of dental plaque and dental caries. In a previous study, we confirmed that the essential oil of Chrysanthemum boreale has antibacterial activity against S. mutans. Alpha-pinene is one of the major chemical components of Chrysanthemum boreale essential oil. In the present study, we investigated the inhibitory effects of ɑ-pinene on cariogenic properties such as growth, acid production, biofilm formation, and bactericidal activity on S. mutans. Alpha-pinene at a concentration range of 0.25-0.5 mg/mL significantly inhibited the growth of S. mutans and acid production of S. mutans. Biofilm formation was significantly inhibited at > 0.0625 mg/mL ɑ-pinene, similar to the data from scanning electronic microscopy. Under confocal laser scanning microscopy, the bacterial viability was decreased by ɑ-pinene in a dose-dependent manner. These results suggested that ɑ-pinene may be a useful agent for inhibiting the cariogenic properties of S. mutans.

        본 연구는 산국 생균제가 비육후기 거세한우의 성장율과 육질에 미치는 효과를 조사하기 실시하였다. 거세한우 15두(565±17kg)를 시험구별 개시체중이 유사하도록 산국 생균제 무첨가구(CON), 농후사료의 0.2% 첨가구(LCB) 및 0.4% 첨가구(HCB)에 각 5두씩을 배치하고 150일간 사양시험을 실시하였다. 농후사료와 조사료를 8:2의 비율로 설정하고 생체중의 약 2.5%를 오전(8시)과 오후(5시)로 나누어 급여하였다. 미네랄 블록과 음수는 자유로이 섭취할 수 있도록 하였다. 사양시험 종료 후에는 모든 공시동물은 도축하여 도체특성을 조사하고 등심육을 채취하여 육질분석에 이용하였다. 개시체중과 종료체중은 처리구간 차이가 없었으나, 일당증체량, 사료섭취량 및 사료효율은 산국 생균제를 급여한 모든 시험구에서 대조구에 비해 개선되었다(p<0.05). 분 중 황화수소와 암모니아 가스는 시험구간 차이가 없었다. 신선육 관능평가에서 등심육의 육색은 산국 생균제를 급여한 시험구가 개선되었으나(p<0.05), 등심육의 물리화학적 특성과 기타 신선육 관능평가는 시험구간 차이가 없었다. 등심육의 지방산 조성은 산국 생균제를 급여한 시험구에서 C14:0과 C18:0 함량이 증가하였으며(p<0.05), HCB 시험구에서 C18:2n-6와 polyunsaturated fatty acid 함량은 HCB 시험구에서 가장 높게 나타났다(p<0.05). 따라서 비육후기 거세한우에 산국 생균제를 0.4% 수준으로 급여하는 것은 사료효율 개선, 육색 및 등심육 지방산 품질개선에 유리할 것으로 사료된다.

        Chrysanthemum is one of the most popular and economically important ornamental plants due to its huge diversity in growing habits, wide range of colors, and different patterns of flower. In the present study, we conducted the karyotype analysis in four naturally occurring genotypes of Chrysanthemum boreale. Karyotype studies based on the fluorescence in situ hybridization (FISH) using 5S and 45S rDNAs. Four chrysanthemum genoteyps showed an aneuploid chromosome number of 2n=18+2 (111016 and 111021) or a diploid of 2n=18 (121001 and 121002). All the genoteyps had the same karyotype formula of 14 metacentrics and 4 submetacentrics. In 111016, the chromosome length during somatic metaphase ranged from 3.11 ± 0.26 μm (shortest) to 3.94 ± 0.20 μm (longest), with a total length of 32.94 μm. The chromosome length at the mitotic metaphase ranged from 3.11 to 6.46 μm, with a total length of 32.94 μm in 111016 and 51.05, 32.81, and 46.00 μm in 111021, 121001, and 121002, respectively. The 5S rDNA and 45S rDNA signals recorded different in all four wildly grown genotypes of C. boreale. This information can be useful in cultivar improvement, as well as in elucidation of the evolution of the chrysanthemum.

        이연구에서재식밀도, 적심, 그리고예취가생육과수 량에 영향을 미쳤다. 재식거리 90cm × 30cm에서 최고 의생장과 수량을얻었는데, 이재식거리에서는최고초 장이 140cm, 식물체당 1차, 2차 및 3차 분지수가 각각 32개, 164개및 367개로자랐다. 이처리에서건조한꽃 의무게는 10a당 98kg에달해재식거리 120cm × 30cm 에비해 40% 증수되었다. 경경, 지상부 건물중및건조 한 꽃 무게는 적심 하였을 때 무적심에 비해 증가되었 다. 7월 10일 적심시 건조한꽃 무게가 10a당 102kg으 로무적심에 비해 57% 증가되었다. 생육과 수량특성은 무예취와 6월 10일예취처리간에비슷하였지만 7월 10 일예취에서는초장, 경경및분지수가무처리에비해유 의하게감소되었다. 누적 지상부 건물중은모든처리에 서차이가없었지만 6월 10일예취시건조한꽃무게와 꽃수가 10a당각각 123kg과 2,592개로가장많았다.

        Chrysanthemum boreale Makino (C. boreale) is widely distributed in Asian countries, and has traditionally been used to treat various inflammatory diseases including bronchitis. In this study, we aimed to isolate biologically active compounds from leaves and stems of C. boreale. Chemical components were purified by column chromatograpy and recyclic HPLC, and characterized from their spectral data (IR, MS, NMR). Biological activity experiments were conducted for Farnesyl-protein transferase (FPTase) activity, apoptosis and nitirc oxide (NO) release. As a results, three sesquiterpene lactones were isolated. Compound 1 (4-methoxy-8-O-acetyl-10-hydroxy-2,11(13)-guaiadiene-12,6-olide) showed strong cytotoxic activities having an average growth inhibition of 50% (GI50) value of 1.89 ㎍/㎖against human colon adenocarcinoma cells. Compound 1 also showed a low half maximal inhibitory concentration (IC50) value of 10 ㎍/㎖ for NO release. In the caspase 3 activity, compound 1 and compound 2 (8-O-(2-carbonyl-2-butyl)-3,10-dihydroxy-4,11(13) -guaiadiene-12,6-olide) exhibited 94% and 90% apoptosis inhibition activity, respectively. Compound 3 (4,8-O-diacetyl -10-hydroxy-2(3),11(13)-guaiadiene-12,6-olide) showed a strong inhibitory effect on FPTase activity with 90% inhibitory activity at a concentration of 100 ㎍/㎖. These results clearly show the presence of lactone compounds in the leaves and stems, which may partially contribute to the pharmacological activity of C. boreale.

        Chrysanthemum boreale Makino is widely distributed in Korea, China, Japan and Southeast Asian countries. C. boreale is one of the herbs used for treating various inflammatory diseases in oriental medicine. The present study was conducted to identify biologically active compounds from the leaves and stems of C. boreale. We isolated two sesquiterpene sactones from the leaves and stems of C. boreale using silica gel column chromatography and recyclic high perfomance liquid chromatography. The lactones were characterized by their spectroscopic data (NMR, IR, MASS). These compounds were subjected to Farnesyl Protein Transferase (FPTase) inhibition, Nitric Oxide (NO) release inhibition and apoptosis inhibition. The structur of the following isolated compound were elucidated 8,10-o-Acetyl-2-methoxy-10-hydroxy-3,11(13)-guaiadiene-12,6-olide and 4,10-dihydroxy -8-o-Acetyl-2,11(13)-guaiadiene-12,6-olide. In the NO release inhibition assay, compound 2 showed strong activities, with an IC50 value of 7 μg/mL, whereas compound 1 did not exhibit significant activity with an IC50 value of over 14 μg/mL against murine macrophage.

        Acyltransferase (AT) catalyzes the transfer of an acyl moiety from acyl-coenzyme A (acyl-CoA) to an acceptor. ATs play important roles in the maintenance of homeostasis in the human body and have been linked to various diseases; therefore, several ATs have been proposed as potential targets for the treatment or prevention of such diseases. The AT family includes acyl-CoA:cholesterol AT (ACAT), diacylglycerol AT, and monoacylglycerol AT for the metabolism of lipids. Furthermore, recent molecular biological studies revealed the existence of their isozymes with distinct functions in the body. ACAT plays a critical role in the formation of cholesteryl esters from cholesterol and fatty acids, and is a potential target for treating hypercholesterolemia. During an experiment designed to discover biologically active compounds from herbal medicines, we isolated two known guaianolide sesquiterpene lactones from Chrysanthemum boreale Makino (Compositae). The lactones were characterized from their spectroscopic data (NMR, IR, MASS). These compounds were subjected to ACAT inhibition assay. Here, we report the isolation and structural elucidation of the compounds 8-o-acetyl-2-methoxy-10-hydroxy-3,11(13)-guaiadiene-12,6-olide and 8-acetyl-3,10-hydroxy-4(15),11(13)-guaiadiene-12,6-olide. In the ACAT inhibition assay, compound 1 showed strong inhibitory activity, with an IC50 value 45 μg/mL, whereas compound 2 did not exhibit significant inhibitory activity with an over 100 μg/mL.

        본 연구는 수증기 증류법을 이용하여 추출한 산국화 유래 hydrosol의 피부각질형성세포(HaCaTs) 증식 및 이주 유도 활성 평가를 통해 피부재상피화 및 상처치유 활성을 확인하였다. 산국화 hydrosol은 HaCaTs의 증식과 이주를 농도 의존적으로 유도하였으며, 특히 1 μg/mL에서 음성 대조군(control)에 비해 143.71 ± 3.37%의 증식과, 139.98 ± 5.72%의 이주 유도 활성을 나타내었다. 또한, 산국화 hydrosol은 extracellular signal-regulated kinase 1/2 (Erk 1/2)와 serine/threonine-specific protein kinase (Akt)의 인산화를 유 의하게 증가시켰을 뿐만 아니라 collagen sprout out growth를 농도 의존적으로 유도하였다. 이러한 결과들을 통해서 산국화 hydrosol은 정상적인 재상피화 과정과 상처치유 등의 활성이 있음을 확인하였고, 향후 화장품 소재로서 응용가능성을 검증하였다.

        In the genus Chrysanthemum, repetitive DNA sequences, the dominant part of a genome, are still to be elucidated. To explore the matter, the present study applied fluorescent in situ hybridization (FISH) to the mitotic metaphase chromosome of Chrysanthemum boreale with C0t DNA as probes. Based on DNA re-assotiation kinetics, three kinds of C0t DNA exhibiting different degrees of repetitive nature were fractionated and used as FISH probes to map the repetitive sequences. Signals from all C0t DNAs were successfully observed but their coverage on the chromosomes was different among C0t-1, C0t-10, and C0t-100. C0t-1 FISH signals resulted to have its intensity on the telomeric region and were also dispersed on both chromosome arms except for some distal regions. In C0t-10, signals were observed in all parts of the chromosome with greater intensity around pericentromeric regions. FISH with C0t-100 DNA was observed in bright signals all over the chromosome. Signals of C0t FISH found in this study covered the regions where ribosomal DNAs and telomeric repeats of C. boreale have been distributed (previous report), thus signifying their repetitive attributes. The present results could enhance the efficiency of studying genomes, chromosomes and repetitive sequences of C. boreale and subsequently hasten the realization of the genetic scheme of Chrysanthemum.

        광원에 따른 식물의 생육은 새싹채소나 실내 작물재배에 여러 용도로 연구되어왔다. 최근 여러 분야에서 그 이용용도가 연구되고 있는 발광 다이오드 LED (Light Emitting Diode)를 광원으로 사용하여 생육반응을 알아보고자 용외국화와 산국을 이용하였다. 용뇌국화는 마키노국화, 뇌향국화라고도 하며, 산국과 비슷하지만 잎의 뒷면에 솜털이 빽빽이 나는 것이 다르다. 땅속줄기가 옆으로 벋어서 나무처럼 단단해지고 가지가 많이 갈라진다. 잎은 어긋나고 달걀 모양이며 3개로 얕게 또는 중간까지 갈라진다. 갈래조각은 다시 1∼2개로 갈라져서 국화잎 비슷하게 보인다. 꽃은 가을에 피며, 두상화(頭狀花)는 지름 3∼5cm이고 흰색이며, 관상화는 노란색이다. 열매는 수과로 젖으면 끈끈한 성질이 생겨 서로 붙는다. 산국은 개국화라고도 하며, 꽃은 진정·해독·소종 등의 효능이 있어 두통과 어지럼증에 사용한다. 관상용으로 식재하며 어린순은 나물로 먹는다. 이와 같은 기능성 산채류로 활용이 가능한 용뇌국화 및 산국을 공시재료로 LED 광질 종류에 따른 생육차이를 분석하였다. LED 광원으로 red와 blue 혼합구, red, blue, white 혼합구를 사용하였고, 광도를 각각 2처리구로 구분하였다. 형광등 처리구 및 대조구와 함께 조광 15시간, 암조건 9시간, 온도는 주간 25℃, 야간 18℃로 조절하여 60일간 생육시켜 그 차이를 알아보았다. 용뇌국화는 대조구에 비해 처리구에서 엽생장이 감소하였고, 광원별로는 red+blue 처리에 비해 red+blue+white 조합처리구의 생육이 더 좋았다. 산국은 고광도의 red+blue+white 처리구에서 생육이 양호한 것으로 나타나 생산증가 효과를 기대할 수 있었다.

        산국은 개국화라고도 하며, 산국 꽃은 진정·해독·소종 등의 효능이 있어 두통과 어지럼증에 사용한다. 관상용으로 심으며 어린순은 나물로 먹는다. 구절초는 구일초(九日草), 선모초(仙母草), 고봉(苦蓬)이라고도 불린다. 꽃이삭에 1.1%, 잎과 줄기에 0.01%의 정유가 있으며 전초는 알칼로이드 반응을 나타낸다. 항균성이 있고, 민간요법으로는 전초와 꽃 이삭을 해열, 폐렴, 기관지염, 기침, 감기, 인후염증, 방광질병, 무월경, 고혈압 치료 등에 사용했다. 본 연구는 산국과 포천구절초의 질소형태별 생육습성을 조사하여 자생식물의 활용도를 증진시키기 위하여 실시하였다. 배양액은 Hoagland 배양액을 변형 처리하였으며, 질소비료의 형태를 NO3-N, NH4-N, NO3-N/NH4-N 혼용처리구로 구분하였다. 비닐하우스에서 EBB 시스템을 이용하여 재배하였으며, 60일 후 초장, 엽수, 엽장, 엽폭을 측정하였고, 엽록소 함량을 조사하였다. 산국과 포천구절초 모두 초장, 엽장, 엽폭의 경우 NO3-N와 NH4-N 혼합처리구에서 가장 많았으며, NH4-N처리구의 엽수감소가 뚜렷하였다. 엽록소 함량은 산국은 처리 간 큰 차이가 없었으나 포천 구절초는 NO3-N, NH4-N, 혼합처리구 순으로 증가하였다.

        The flowers of Chrysanthemum boreale Makino were extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with n-hexane, EtOAc, n-BuOH and H2O. Two compounds from the n-hexane fraction and one glucoside from the n-BuOH fraction were isolated through the repeated silica gel and ODS column chromatographies. From the result of physico-chemical data including NMR, MS and IR, the chemical structures of the compounds were determined as β-sitosterol (1), phytol (2) and zingerone 4-O-β-D-glucopyranoside (3). Compounds 2 and 3 were isolated for the first time from this plant.

        우수한 자생식물자원인 산국의 수량과 유효성분을 높이기 위해 식물생장조절제 ABA와 SA를 처리한 결과를 요약하면 다음과 같다. 1. 꽃의 건물수량은 ABA, SA과 ABA+SA 처리구에서 무처리구보다 증가되었으며, ABA 처리구보다는 SA 처리구가 증수되었다. 2. 무기성분의 함량변화는 SA 처리구가 다른 처리구들에 비하여 질소, 인산, 마그네슘의 함량이 증가하였으나 칼륨의 흡수는 오히려 감소하였다. 3. 아미노산함량은 SA 처리구와 ABA+SA 처리구는 ABA 처리구보다 높았다. 4. 생장조절제처리는 정유함량을 증가시켰으며, 그 반응은 monoterpenoid계 화합물보다는 sesquiterpennoid계 화합물에서 더 민감하였다. 5. Cumambrin A의 함량은 ABA 처리구에서 증가하였고, SA 처리구에서는 차이가 없었다.

        돈분퇴비 시용시 산국 (Chrysanthemum boreale)의 수량과 유효성분들의 함량변화를 알아보고자 토양 16 kg이 충진된 와그너포트에서 돈분퇴비가 각각 0, 2000, 4000, 6000, 8000, 12000 kg 10a-1을 처리한 결과, 8000 kg 10a-1처리구에서 대조구보다 전체 수량과 꽃의 수량은 각각 5.9배 (195.0 g plant-1)와 24배 (21.7 g plant-1)가 증가한 것으로 조사되었다. 질소양분 이용율은 4000 kg 10a-1처리구가 66.6%로 가장 높았다. 꽃에서의 주요 아미노산은 proline (7.4~9.2 g kg-1)이였으며, 돈분퇴비의 시용량이 증가할수록 정의 상관관계를 보였다. Cumambrin A함량을 측정한 결과, 돈분퇴비 시용량에 의한 질소의 함량이 증가할수록 부의 상관관계가 나타났으나 (R2=-0.723, p〈0.01), 전체 수량면에서는 오히려 돈분퇴비의 시용량이 증가할수록 cumambrin A의 수량이 오히려 증가하였다. 이로서 돈분퇴비의 시용이 산국의 수량을 향상시킬 수 있을 것으로 기대되었다.

        천연물로부터 항산화 활성물질을 목적으로 DPPH 자유 라디칼 소거활성 검색법을 이용하여 검색을 실사하고, 유의한 활성을 나타낸 산국 메탄올 추출물로부터 2종의 화합물을 분리하고 기기분석을 이용하여 화합물 1과 2의 구조를 apigenin과 acacetin-7-O-rutinoside (Linarin)으로 동정하였다. 이들 화합물은 각각 13.3 및 42.1μg의 농도에서 DPPH 라디칼을 50% 소거하는 활성을 나타내었으며, 화합물 1은 양성 대조양물인 L-ascorbic acid (RC50 = 13.1μg)에 비하여 강한 활성을 나타내었다.

        The stability of carotenoid pigments extracted from Chrysanthemum petals against pH, sugar, and organic acid was investigated. The contents of total carotenoids in C. boreale and C. morifolium were 3.37mg% and 4.56mg% Per fresh weight, respectively. The elect of pH on the stability of extracted carotenoids showed that the periods reaching 50% of pigment retention were more than 5 to 6 days in pH 4, and the longer periods in pH 6 to 7 in both samples. The addition of sugars such as sucrose, sorbitol, glucose and fructose resulted in the reduction in pigment stability of C. boreale, but C. morifolium was contrast to C. boreale. However, citric acid and ascorbic acid were found to have a protective effect on both carotenoid samples extracted from C. boreale and C. morifolium.
