        Preservation of sperm is essential for long-term storage of valuable animal genetic resources and for the conservation of threatened mammalian species undergoing progressive extinction. In this study, using pig as a model system, we evaluated the feasibility of germ-plasm preservation via sperm cell lyophilization. We show that, pig sperm can be successfully lyophilized and stored in a liquid nitrogen-free condition for at least 6 months. Intracytoplasmic injection of lyophilized sperm (ICSI), stored at 4℃ for four months, into in vitro matured pig oocytes could successfully develop up to blastocyst stage (13.0±3.0%). Lyophilized sperm could also be stored at room temperature for at least three weeks without further compromising their in vitro development up to the blastocyst stage (14.6±3.2 vs. 16.6±5.1%; p>0.05). Blastocysts produced from ICSI of lyophilized sperm stored at 4℃ or room temperature contained similar number of cells per blastocyst (44.9±3.2 vs. 44.0±4.3; p>0.05) but was significantly lower than those produced from non-lyophilized fresh sperm (52.1±5.8 p>0.05). Interestingly, use of a custom-designed HEPES-buffered, calcium-free, defined medium for the lyophilization resulted in normal post-ICSI embryonic development up to blastula stage (23.4±2.8 vs. 24.0±2.9%) and, the resultant blastocysts contained similar number of cells per blastocyst (47.9±4.3 vs. 50.6±7.0) compared to those generated from non-lyophilized fresh sperm (p>0.05). These lyophilized sperm could also be stored at room temperature for at least three weeks with slight reduction in post-ICSI embryonic development (19.6±1.4%). Therefore, these results suggest that, pig sperm could be successfully and efficiently lyophilized for their long-term storage at 4℃. Lyophilization of sperm could be a practical option for long-term storage of mammalian germ-plasm.

        진딧물 포식성 무당벌레의 성충을 대상으로 저온처리에 의한 생식적 휴면을 유도하였다. 성충의 인위적인 생식적 휴면의 유지는 4℃에서 5개월 정도까지 가능하며 먹이의 공급이 필요하지 않아 경제적인 무당벌레의 저장법이라 할 수 있다. 하절기를 포함한 계절에 구분 없이 무당벌레의 성충의 저온저장이 가능하여 필요시 어느 때라도 진딧물 발생지에 무당벌레를 공급할 수 있게 되었다. 휴면 후 25℃에서 수명과 산란력이 우수하며 생식호르몬인 juvenile hormone(JH)의 합성 유사체인 methoprene은 암컷의 초반기 생식력을 증진시키는 효과를 나타낸다.

        To develop a kimchi with high sensory quality and storage stability, the optimum pasteurization time and temperature were investigated. Sliced kimchi with different acidity (0.3, 0.5, and 0.8%) was prepared and pasteurized at 65 or 75oC, then the microbiological (number of total aerobic bacteria, lactic acid bacteria, and yeasts), physicochemical, and sensory changes were measured. The viable counts of total aerobes, lactic acid bacteria, and yeasts as well as the values of pH and acidity changed rapidly in control. However, the growth of microorganisms was controlled by pasteurization treatments, and the changes of pH and acidity in the treated kimchi were much smaller than those of control, although acidity of the samples was different at the initial state. The sensory results indicated that the overall quality of kimchi samples pasteurized at 0.3 or 0.8% acidity was relatively lower than that for the samples at 0.5% acidity. Particularly, kimchi pasteurized at 65oC were high scored in quality attributes compared to the other samples, with preserving a desirable flavor during whole storage period.

        Background : Aralia cordata and Polygonum multiflorum GAP cultivation requires a stable drying and storage settings after harvesting. therefore, this experiment was performed in order to effectively manage the physical, chemical and biological hazards. Methods and Results : Test materials were used biennial Aralia cordata, Polygonum multiflorum harvested from the medicinal testing ground. The drying temperatures were treated with 40, 50, 6 0℃ and natural drying. Storage containers were stored in plastic boxes, styrofoam boxes and kraft paper containers, examined the color and quality changes for eight months. Aralia cordata and Polygonum multiflorum drying temperature is dry it took natural drying 720 hours, 40℃ hot air drying 180 hours, 50℃ hot air drying 168 hours and 60℃ hot air drying 108 hours. However, the difference chromaticity of the Lab value corresponding to the temperature does not appear, it was good to dry in a short time at 60℃. Aralia cordata and Polygonum multiflorum stored in a styrofoam box storage method but can be stored at room temperature for up to four months, began to decay caused by moisture content it continues to increase. In plastic box in case of Aralia cordata and kraft vessel in case of Polygonum multiflorum can be stored for eight months in room temperature without decay. Styrofoam boxes stored at 5℃ cold storage were higher water absorption such as room temperature, but decay did not occur. Plastic box and styrofoam box were a tendency such as room temperature. Conclusion : Aralia cordata and Polygonum multiflorum are thought that the color change is not large depending on the drying temperature the lower the water content. Styrofoam storage box, the air permeability is higher than plastic boxes and containers Kraft vessel, decay occurs expected increase.

        Geneally, rice seeds regardless indica or japonica are showing low germination ratio or completely lost germination ability together with lost of good eating quality under high temperature and humidity conditions. Thus, this study was designed to evaluate a longevity for conservation of good eating quality during long term storage in rice. For the longevity evaluation, germination ability was studied after 5 days of high temperature and humidity stress (50℃/RH 95%). Dharial, originated from Bangladesh and showing weedy type with red pericarp, was selected as a good donor for longevity genes. A mutant was developed from Dharial through EMS mutagenesis and two populations of Dharial/4*Ilmibyeo and Dharial/4*Gopumbyeo were also developed for genetic study. In the 2-DE analysis followed by MALDI-TOF MS with wild and mutant lines, several candidate genes were identified. In the longevity test of two populations, a few lines showing good germination ability after high temperature and humidity stress were selected and subjected to confirm the relationships between longevity and conservation of good eating quality under long term storage.

        본 연구는 장기저장이 가능한 김치제조를 위해 사용된 20kGy의 선량으로 감마선 조사된 김치와 동결건조된 김치의 미생물학적,조직학적 및 관능적 품질특성을 비교하였다. 김치의 총균수는 감마선조사 후 검출한계 이하로 나타났으나 동결 건조된 김치는 비조사 대조구에 비해 1 log 수준의 미생물 감소를 나타내었다. 감마선조사 김치의 경도는 동결건조 후 재수화된 김치에 비해 유의적으로 높았다. 전자현미경을 통한 김치 조직 관찰 결과 동결건조 후 복원된 김

        체세포배발생을 이용한 가시오갈피 대량증식체계 확립을 위하여 저온처리에 의한 체세포배의 장기저장 실험을 수행하였다. 현탁배양 체세포배의 4℃에서의 저온저장에는 구형배 이전의 조기단계의 체세포배가 적합하였고 2 g/l 이하의 낮은 접종밀도에서는 36개월 이상 저장이 가능하였다. 장기간 저온 저장한 체세포배는 상온에 옮긴 후 2,4-D를 첨가한 배지에서만 성장을 회복하여 배발생캘러스를 형성하였으며 배발생캘러스의 증식속도는 배양온도를 32℃로 높였을 때에 가장 효율적이였다.

        Sweet com seeds deteriorate faster due to low starch content than field com seeds when stored for a long tenn. This study had been conducted to observe the seed deterioration of four different sweet corns in a long tenn storage conditions in room temperature. Four kinds of sweet com genes (sh2, bt, su, and se) were harvested from 15 days to 50 days after silking with 5-day intervals. These seeds were stored in the room temperature and tested for germination percentages from 3 months to 18 months period with 3-month interval. su seeds germinated better than other types of gene. Hybrid Mecca which is sh2 gene germinated better when stored for 3 months to 18 months. For all genes, mean regression equations in relation to storage periods showed linear responses. For regression equation, the slope of sh2 gene was lower than that of su gene. The highest slope value was observed in bt gene showing faster deterioration rate. The rate at which seed deteriorates seems to be affected by the date at which it was harvested. The seeds that were harvested at the optimum time deteriorated more slowly than those which were not

        In an attempt to test experimental condition of preparing grape powder, grapes having less commercial value was used and tried. With drying method, spray and freeze drying were satisfactory to produce power. Moisture content and odor retention were better by the latter method. Three grape strains stored for 40 days contained more odors than those stored for 5 days. Maltose 90% plus dextrin 10% was suitable for drying support. To increase odror sense, citric acid and vitamin C can be added up to 0.1 and 0.2%, respectively. Considering these conditions, grape complex powder prapared from grape powder 20% comprising drying support, glucose 79.7%, citric acid 0.1%, vitamin C 0.2% with freeze drying was the best by overall evaluation including sensory test. When campbell and neomuscut were mixed by 15:5 or 10:10, sensory evaluation was also ameliorated.

        Chemical, physical md microbial analysis of salted baechu(Chinese cabbage) and its exudate were conducted during storage in large plastic bags(LDPE, HDPE, PVC-box) at 0. Salinity was slightly lowered for 2 weeks storage and maintained that level thereafter in salted haechu, but in its exudate increased rapidly for 2 weeks storage and decreased thereafter. In all treatment pH decreased to 5.0 in salted baechu and to 4.5 in its exudate for 6 weeks storage. Reducing sugar content of salted baechu was 1.5∼2.0 fold higher than that of its exudate, and decresed slowly in both. Total viable cells increased rapidly for 2 weeks but maintained that level thereafter and lactic acid bacteria increased continuously until 6 weeks. Lightness of exudate decreased rapidly in PVC-box, but decreased slowly in LDPE. Compression force of salted baechu increased continuously untill 8 weeks except for PVC-box.

        양곡창고에 정조상태로 장기저장한 벼 종실의 이화학적특성 변화를 검토하기 위하여 1987년부터 1992년까지 해마다 생산되어 64개월부터 4개월까지 저장되어 있던 통일형품종 삼강벼를 시험재료로 하여 저장기간이 다른 쌀의 식미관련 이화학적 특성을 조사, 비교한 결과를 요약하면 다음과 같다. 1. 쌀의 지방산도는 저장기간이 길어질수록 증가하였는데 16개월 저장시까지 급격하게 증가하였고 그 이후는 완만한 증가추세를 보였다. 2. 백미의 아밀로스, 마그네슘 및 칼륨함량과 K/Mg 비율은 저장기간의 장단에 따른 변화가 없었고 단백질함량은 저장기간이 길어짐에 따라 감소하였다. 3. 상온 및 가열흡수율은 저장기간이 길어짐에 따라 증가하였는데 상온흡수율은 침지초기에 그리고 가열흡수율은 침지시간이 경과할수록 고미와 신미간 차이가 컸다. 4. 저장기간이 연장됨에 따라 쌀의 알칼리붕괴도는 높아졌으며 호응집성은 감소하는 경향이었다. 5. 쌀가루의 아밀로그램특성 중 최고점도, 최저점도, 냉각점도, breakdown 및 setback은 저장기간이 길어짐에 따라 증가하는 경향이었다. 6. 취반후 팽창용적은 저장기간이 긴 쌀이 짧은 쌀에 비하여 상대적으로 컸으며 용출고형물질과 요드정색도를 나타내는 흡광율은 저장기간이 길어질수록 감소하였다.
