        깔따구 (Diptera: Chironomidae)는 저서성 대형무척추동 물로 환경 및 수질 변화에 민감한 영향을 받는 중요한 환경 지표생물이다. 이러한 깔따구 유충이 정수장에서 본 연구에서는 제주 정수처리장에서 발견된 깔따구 유충의 종을 분류하기 위해 형태 사진 및 COI (cytochrome c oxidase subunit I) Primer로 증폭시킨 DNA의 염기서열을 계통수 분석을 통해 분석을 실시하였다. 정수장 내 수도꼭지와 소화관 등에서 채집된 17개체는 둥근깃깔따구 (O. tamarutilus) 14개체, 타마긴털깔따구 (P. tammaater) 3개체 총 2종으로 확인되었다. 각 깔따구 종의 형태적 특징은 두부, 하순기절, 대악, 안테나, 발톱의 형태적 특징의 분류기로 종 동정하였다. NCBI Genbank에 등록된 깔따구 19종 COI 염기서열을 기반으로 본 연구에서 조사된 17개체의 계통진화적 분석 결과 채집된 깔따구 COI 염기서열이 둥근깃깔따구 (O. tamarutilus)와 타 마긴털깔따구 (P. tammaater) 2종으로 각각 계통군 (clade)를 이루는 것이 확인되었다. 이러한 결과는 정수장 환경별 발견 되는 깔따구 유충의 다양성의 확인과 형태적-유전적 종 동정 분류정보를 바탕으로 수환경 관리 및 평가를 위한 기반 정보로 활용될 것이다.

        A single postlarva (9.72 mm in standard length) specimen of Crossorhombus azureus (Alcock, 1889) belonging to the family Bothidae, was collected using a bongo net from the southern sea of Korea on December 10, 2019. This species is characterized by having spines on post basipterygial processes. It has two eyes located at the opposite side in head, which accordingly belongs to pre-metamorphosis stage. Melanophores are distributed on the dorsal and anal fin base on the right side (blind side), which is regarded as a useful identification key distinguishing C. azureus from congeneric species in their postlarval stage. A molecular analysis based on mitochondrial DNA COI sequences showed that our specimen was closely matched to adult C. azureus (K2P distance = 0.017). As there is no Korean name for the genus Crossorhombus in spite of presence of Crossorhombus kobensis and its Korean name “Go-be-dung-geul-neob-chi” in Korea, we proposed a new Korean name “Dung-geul-neob-chi-sog” for the genus Crossorhombus and “Pa-lang-dung-geul-neob-chi” for the species C. azureus.

        During the ichthyoplankton survey around the Chilsan Island (previously known as the biggest spawning ground of the Sciaenidae) in the southwestern sea of Korea from April to June in 2019 using ring nets, we collected a total of 12 individuals belonging to the family Sciaenidae. Using molecular methods, we identified five sciaenid species (Collichthys lucidus, Collichthys niveatus, Johnius grypotus, Nibea albiflora, and Pennahia argentata), and described and compared them on the basis of the preflexion larval stage. C. lucidus was well distinguished by the presence of occipital crests at preflexion stage. Although there were no occipital crests, preflexion larva of C. niveatus was distinguished by the absence of melanophore except for the upper part of the abdominal cavity. J. grypotus and N. albiflora were very similar morphologically, but were distinguished by myomere height (15.22-15.53% in J. grypotus vs. 11.66-12.78% in N. albiflora) in the percentage of notochord length, and eye diameter (32.58-33.37% in J. grypotus vs. 40.32-42.53% in N. albiflora) in the percentage of head length between specimens of similar size (J. grypotus: 3.22-3.23 mm, N. albiflora: 3.04-3.13 mm). P. argentata were distinguished by distribution of ventral caudal melanophore (one row of small spot in P. argentata vs. irregular patches on the central part of caudal in J. grypotus and N. albiflora). Comparative morphological studies using more diverse species must be conducted for more comprehensive understanding of the morphogenesis of Sciaenidae.

        A single juvenile malacanthid specimen (Hoplolatilus chlupatyi) was collected from the South Sea of Korea and identified by DNA barcoding. This species is readily distinguished from other malacanthid species in having well developed rostral spine and elongated spines in the posttemporal and preopercle. A molecular analysis based on mitochondrial DNA COI sequences showed that this species is matched to adult H. chlupatyi (genetic distance = 0.005). Therefore, this is the first record of H. chlupatyi in Korea. We propose new Korean names, “Mu-ji-gae-og-dom-sog” for the genus Hoplolatilus and “Mu-ji-gae-og-dom” for the species H. chlupatyi.

        깔따구 (Diptera: Chironomidae)는 저서성 대형무척추동 물로 환경오염 및 수질 모니터링에 이용되는 중요한 지표생물이다. 본 연구에서는 인천 수돗물 정수장에서 발견된 깔따구류의 정밀한 종 동정을 위해 형태적 분류와 미토콘드리아 DNA에서 cytochrome c oxidase subunit I (COI) 유전자의 염기서열을 이용하여 분석하였다. 정수장 6곳의 20 개체는 안개무늬날개깔따구 (Chironomus kiiensis) 12개체, 노랑털깔따구 (Chironomus flaviplumus) 6개체, 등깔따구 (Chironomus dorsalis) 1개체, 용산무늬깔따구 (Polypedilum yongsanensis) 1개체 등 4종으로 확인되었다. 각 깔따구 종의 형태적 특징은 두부, 하순기절, 대악, 안테나, 발톱의 형 태적 특징을 살펴보았다. NCBI Genbank에 등록된 깔따구 17종 21개체의 COI 염기서열을 바탕으로 본 연구에서 조사된 20개체의 계통진화적 분석한 결과 각 4종의 깔따구 COI 염기서열은 등록된 동인 종과 높은 상동성을 보이며 (99~100%) 같은 계통군 (clade)으로 나타났다. 이러한 결과는 국내 깔따구의 종 동정을 위한 형태적 - 유전적 정보를 통합적으로 제공함으로 담수생태계의 모니터링을 위한 주요한 정보로 활용될 것이다.

        We found ten species of larvae belonging to the family Pleuronectidae as a result of analysis on ichthyoplankton collected monthly from the East Sea, Yellow Sea, Korea Strait and East China Sea between February 2016 and May 2018 using bongo net. The ten species of pleuronectid larvae were divided into three groups in morphology. Group A had three or four bars on the lateral side of the tail: Glyptocephalus stelleri, G. kitaharae, Pseudopleuronectes yokohamae, and Hippoglossoides dubius. Group B had a row of melanophores along the dorsal and ventral side of tail: Cleisthenes pinetorum, Eopsetta grigorjewi, Dexistes rikuzenius, and Platichthys bicoloratus. Group C had dense melanophores which are covering trunk and tail entirely except caudal peduncle: Platichthys cornutus and Platichthys japonicus. The three groups did not associate with previous molecular phylogenetic studies except for the G. stelleri and G. kitaharae of the group A.

        From August to October 2013, a powdery mildew was found on Japanese artichoke(Stachys sieboldii) in Jinju, Korea. White colonies were present on leaves and petioles. Severely infected lesions were discolored, being brown. In the present study, the morphological features of anamorphic and teleomorphic Korean specimens were studied. To complete the identification, the sequence of the internal transcribed spacer region of the ribosomal RNA gene(ITS rRNA) was analyzed. Based on morphological characteristics, including anamorphic and teleomorphic features, as well as analysis of the ITS rRNA gene region, the fungus associated with the symptoms was identified as Neoerysiphe galeopsidis. Although the host ranges of powdery mildew caused by N. galeopsidis has been previously recorded, no full description or illustration of fungal symptoms and signs on Japanese artichoke has yet appeared. To the best of our knowledge, this is the first full description of powdery mildew disease on Japanese artichoke featuring molecular identification, morphological features, symptoms, and signs in Korea.

        Root-lesion nematodes, Pratylenchus sp., are recognized as one of the most important plant parasitic nematodes. They infest various crop plants and cause severe root damages which lead to the yield loss of agricultural crops. There are 72 species recorded in the world, but 14 species in Korea. Soil samples were collected from chrysanthemum cultivars in various regions. Morphological and molecular diagnosis firstly recognized two species as Pratylenchus kumamotoensis and P. pseudocoffeae in Korea. Morphologically, P. kumamotoensis characterized by pharyngeal gland lobes overlapping with intestine dorsally. P. pseudocoffeae, which is similar with P. coffeae, but pharyngeal gland lobes are longer than P. coffeae. In molecular diagnosis, nucleotide sequences of D2-D3 region (700-800 bp) within 28S rDNA and ITS region including ITS1, 5.8S and ITS2 (700-1,000 bp) between 18S and 28S rDNA were determined from collected samples. NCBI database comparison of D2-D3 of P. kumamotoensis and P. pseudocoffeae were 99% similar with those of corresponding species, respectively. ITS of P. pseudocoffeae was 99% similar within species. ITS sequence of P. kumamotoensis was firstly determined in this study. Distribution of P. kumamotoensis was only reported in Japan but P. pseudocoffeae is present in Japan and USA. Here we firstly report the presence of these two species in Korea.

        Root knot nematodes were extracted from five different plants (indian spinach, papaya, sweet gourd, okra and jute) collected from kurigram, gazipur, jessore and patuakhali districts of Bangladesh. Firstly, Meloidogyne spp. were identified morphologically mainly based on perineal pattern of adult female. Secondly, female were collected from plant roots and used directly in PCR for amplification of mitochondrial DNA (mtDNA) region between COII and lrRNA genes followed by hinfI digestion and sequencing. The root knot nematodes in indian spinach, jute and sweet gourd were identified as M. incognita whereas in okra and papaya were M. javanica.

        Tetranychid mites are one of the most diverse group including at least 1,200 species in the world. Species identification is difficult due to the small size and similar morphology within the group. We collected 17 species of spider mites from various host plants in different regions of Korea and determined species identity by the comparison of morphological characters and nucleotide sequences of internal transcribed spacer 2 (ITS2) and the cytochrome oxidase subunit I (COI). In addition, we report three new species that were firstly identified in Korea. Amphitetranychus quercivorus (Ehara and Gotoh) was collected from Mongolian oak plant in Daegu, Schizotetranychus miscanthi Saito was from the common reed plant in Ulleungdo, and S. cercidiphylli Ehara was from Bamboo plant in Jeju. Morphological identification of three species were similar with those of Japanese samples, but the ITS2 and COI sequences of A. quercivorus and the COI sequence of S. miscanthi were different with Japanese species at the rates of 1/419, 2/331 and 3/332 nucleotides, repsectively. S. cercidiphylli can be identified by the aedeagus shape of males but we firstly sequenced ITS2 and COI of this species. Our results can be used for the identification of spider mites which are important in plant quarantine.

        Acquisition of a reference Tetranychusstrains that are completely susceptible to acaricides and retain identical genetic backgrounds to acaricide-resistant strains is an essential step in elucidating mechanisms of resistance. To establish both completely susceptible and acaricide-resistant strains for this purpose, I collected Tetranychus mite populations from various regions in South Korea including both heavily cultivated and remote regions. Suitability as a susceptible or resistant reference strain was tested by determining species identity as Tetranychus urticae along with baseline susceptibility to common acaricides. The majority of mite populations collected from cultivated areas belonged to a monophyletic group of the previously reported green-type T. urticae as determined by mtCOI- and ITS2-based phylogenetic analysis. These strains showed relatively reduced levels of susceptibility to the acaricides tested, suggestive of the development of resistance. Among them, the AbaR strain was classified as a minor group in the T. urticae complex. The UD strain, originally collected from a remote island region, was found to be susceptible to the acaricides tested and generated an independent phylogenetic branch. The UD strain was also considered a minor group in the T. urticae complex. Phenotypic analysis based on morphological characters confirmed that both the AbaR and UD strains were statistically undistinguishable from the major green-type T. urticae. Taken together, I propose that the UD strain be used as a susceptible reference strain as it provides baseline susceptibility to acaricides and a wild-type genetic background for the resistance studies.

        본 연구는 유기농업에서 생물자원으로서 담수 클로렐라의 활용 가능성을 연구하고자 유기농 생태계로부터 담수 녹조류를 분리, 동정하고, 생물학적인 특성을 조사하였다. 조사 지역의 수온은 12.4~28.2℃, pH는 6.1~8.5이었다. 담수 녹조류를 분리할 때 고체배양법이 액체배양법보다 오염도가 낮고 분리 빈도가 높았다. 전국 9개 지역, 6개 담수 녹조류 서식처로부터 총 115개의 균주를 분리하였다. 담수 녹조류의 분리 및 배양을 위해 질소원으로는 NaNO3과 KNO3, 탄소원으로 Na2CO3를 사용하였고, macro media의 구성 성분 중 MgSO4 · 7H2O과 CaCl2 · 2H2O를 분리하여 제조한 BGMM(BG11 Modified Medium)배지를 고안하였다. 담수녹조류는 배양 후 4일째부터 급격히 흡광도가 증가하였고 8일째부터 흡광도가 감소하였다. 공시한 7개의 균주 중 CHK008 균주가 배양 7일째에 가장 높은 흡광도를 보였다. 담수 녹조류 배양에 적합한 BGMM 배지의 pH는 6~7이었고 조사되는 빛이 강할수록 생육이 증가하였으며 5종류 당류 중에서 Glucose와 Galactose를 첨가하였을 때 클로랄라의 생육이 좋았다. 순수 분리한 16개 녹조류 균주의 균총색은 녹색, 진녹색, 연녹색을 나타내었고, 11개의 균주가 형광현미경하에서 강한 형광 빛을 나타내었다. 녹조류 16개 균주의 형태적인 특징을 조사한 결과 C. vulgalis, C. sorokiniana, C. pyrenoidosa, C. kessleri, C. emersonii, and Chlamydomonas sp.의 2개 속 6개종으로 동정되었다. 담수 녹조류의 세포 크기는 종마다 다양한 변이를 보였다. 대부분의 담수 녹조류의 세포형태는 구형이었다. Chlamydomonas sp.는 타원형이었고 Chlorella sorokiniana는 구형과 타원형이 섞여 있었다. 6개 녹조류 종류 중 Chlamydomonas sp.를 제외한 모든 균주는 편모가 없었다. Chlamydomonas sp. 1개 균주와 C. sorokiniana 5개 균주는 세포에서 점질물을 분비하였다.
