        2012~2017년까지 강원도 3개 시 ‧ 군(춘천시, 강릉시, 횡성군)에서 채집된 모기는 총 6속 13종, 654,362마리가 채집되었다. 채집된 모기는 분류하여 얼룩날개모기속을 제외하고 종별, 채집일, 채집 장소에 따라 최대 50마리를 1개 실험군으로 reverse transcription-polymerase chain reaction, 염기서열 분석방법으로 플라비바이러스 감염여부를 조사하였다. 채집모기 276,224마리에 대해 7,721개 실험군을 검사한 결과 68개 실험군(0.9%)에서 플라비바이러스 유전자가 검출되었다. 검출된 플라비바이러스의 염기서열 분석결과 4개 실험군은 일본뇌염바이러스 (Japanese encephalitis virus), 64개 실험군은 차오양바이러스(Chaoyang virus)로 확인되었다. 일본뇌염바이러스는 2012년 춘천시에서 채집 된 2,728마리의 빨간집모기 중 1개 실험군, 2013년 횡성군에서 채집된 19마리의 동양집모기 중 1개 실험군, 2017년 춘천시에서 채집된 1,111마리의 빨간집모기 중 1개 실험군, 강릉시에서 채집된 빨간집모기 724마리 중 1개 실험군에서 검출되었다. 검출된 일본뇌염바이러스의 유전형은 모두 Ⅴ형 바이러스였다. 차오양바이러스는 강원도 내에서 6년간 채집된 금빛숲모기 232,871마리, 5,055개 실험군을 대상으로 검사한 결과 63 개 실험군에서 검출되었으며, 춘천지역에서 채집된 빨간집모기 585개 실험군 중 1개 실험군에서 검출되었다. 채집지역별 금빛숲모기의 차오양 바이러스 감염률이 가장 높은 지역은 MIR (최소감염률) 0.32, MLE (최대우도법) 0.33 (CI 0.23~0.46) 감염률을 보인 춘천시였다. 그 뒤로 횡성군 MIR 0.30, MLE 0.30 (CI 0.16~0.52)과 강릉시 MIR 0.21, MLE 0.21 (CI 0.13~0.31)순이었다. 월별 금빛숲모기의 차오양바이러스 감염률은 10월에 MIR 0.38, MLE 0.38 (CI 0.07~1.25)로 가장 높은 감염률을 나타내었다.

        초음파 모기 기피 장치 ‘모스 윈드솔루션(Mos windsolution)’의 성능을 실내에서 평가하였다. 해당 제품은 120Hz~1.2MHz 사이의 주파수를 가진 초음파를 발생시켜 모기가 기피하도록 유도하는 기능을 갖고 있다. 기기의 성능 평가를 위하여 선택적 실험과 비선택적 실험을 실시하였다. 선택적 실험은 2m의 PVC 파이프 양 끝에 15cm cube 형태의 실험 케이지를 연결하고, 그 옆면에 쥐를 한 마리씩 구속하였다. 처리구 쪽에 모스 윈드솔루션을 가동시켜 초음파가 발생하도록 놓은 후 파이프 중앙에 흰줄숲모기(Aedes albopictus)를 방사시켰고 1시간 뒤 양 케이지를 밀봉, 냉동시켜 유입된 흰줄숲모기의 성별과 수를 파악했다. 비선택적 실험은 150x200x150cm의 모기장 안에 상부만 열린 아크릴 케이지를 넣고, 아크릴 케이지 내부에 철망 뚜껑이 있는 용기에 담긴 쥐를 넣어두었다. 처리구 쪽에는 모스 윈드솔루션을 가동시켜 놓았다. 모기장에 흰줄숲모기를 방사시키고, 시간당 아크릴 케이지에 유입된 모기의 수를 파악하였다. 그 결과 두 실험 모두에서 모스 윈드솔루션이 대조구 대비 최대 80%의 모기 기피 효과를 보였음을 확인할 수 있었다.

        Surveillance of the vectors were conducted at a total of 7 locations from March to November, 2018 in Gyeongnam province. In the surveillance, a total of 13 species belonging to seven genera were collected in a total of 15,935 females and 126.5/trap/night (TI; Trap Index) in 2018. Among the collection sites, a bird refuge in Busan, Eulsukdo recorded the highest TI 230.4 (Species Ratio; SR 60.1%) for Cx. tritaeniorhynchus females in 2018. A Cx. tritaeniorhynchus female collected firstly for 2018 in the nation at the pigsty of Busan at 1st April. In the results of isolation of viral RNA from 12 mosquito species including Cx. tritaeniorhynchus, Cx. pipiens, Ae. vexans, Ae. albopictus, and Oc. togoi. Chaoyang virus was positive at a pool of Ar. subalbatus collected at a bird refuge but negative in the other 543 pools (14,295 individuals) in 2018.

        Mosquitoes are primary medical insect pests due to their diseases transmission as vectors. In Korea, the insecticide-resistant populations of disease vector mosquito species, such as Anopheles sinensis, Culex pipiens and Culex tritaeniorhynchus, have constantly increased. Thus, management of insecticide resistance to major insecticides including pyrethroids and organophosphates is required for more efficient control of resistant populations. In this study, the quantitative sequencing (QS) protocols were established to detect the frequencies of three mutations (the L1014F on voltage sensitive sodium channel and the G119S and F331W on acetylcholinesterase 1) that are associated with either pyrethroids or organophosphates. Based on the QS protocol using newly designed non-polymorphic primers, resistance allele frequencies (RAFs) were estimated in field populations of An. sinensis, Cx. pipiens and Cx tritaeniorhynchus collected from an identical site in Korea. The dynamics of each resistance allele frequency over time in the same populations were also evaluated.

        Aerosol-type insecticides are one of the most widely used household insecticides to control mosquitoes. Not only the structural difference such as the size and shape of orifice, but also the difference in formulation including solvent system and proportion of propellent can affect the efficacy of those aerosol products. Orifice structure can be represented by nozzle diameter while solvent systems are divided into water-based and oil-based types in wide view. In the present study using Asian Tiger Mosquito, Aedes albopictus, we found that the orifice nozzle size made a significant difference in shoot-out range, followed by heterogeneous mortality by distance. Additionally, oil-based spray affected farther space than water-based one. Oil-based solvent system was also responsible for lowering the LT50 and KT50(knock-down time 50) values. Therefore, they are one of the definitive parts in insecticides formulation that determine the way of insecticide delivery. In terms of insecticide resistance management, we define efficiency of insecticide as the proportion of insecticide treated to environment that is solely acted as toxin to target pest. The higher efficiency of insecticide, the less overused chemicals we face, which may imply lower risk of resistance development. Thus, we argue that formulations, the specific route of insecticide discharge to environment, should be deliberately considered not only for consumer satisfaction, but also for insecticide resistance management.

        It is known that the growth and development of the mosquito are greatly affected by the change of the meteorological factors. In particular, temperature and precipitation are closely related to the life cycle of the mosquito, and their effects have different characteristics for each species of mosquito. Therefore, to develop a mosquito activity index based on mosquito density, it is essential to develop a prediction model based on weather data. In this study, we developed a functional formula that can estimate the change of mosquito density according to the change of meteorological factors using the mosquito classification data of Incheon region collected from 2011 to 2017. Also, using the data of the digital mosquito monitoring system (DMS) from April to October 2018, mosquito activity index according to characteristics of space in city was developed. In order to reflect the temporal characteristics of the mosquito life-cycle, we used data of temperature and precipitation prior to 1-2 weeks, and used land cover data to reflect the spatial characteristics of mosquito density. Density of Culex pipiens collected in the Incheon area were gradually increased when the average temperature increased two weeks ago after adjusting the precipitation. However, when the average temperature reached 22°C, the density of Culex pipiens marked a peak, and above the 22°C, the density was decreased. The predicted mosquito activity index calculated by applying the machine learning method to the DMS data of the Incheon area is designed to calculate from 1 to 10 grades. The accuracy of the mosquito activity index was 87% when the one grade error was allowed.

        One of major mechanisms of insecticide resistance is the reduced susceptibility caused by mutations on the main target sites, such as sodium channel and acetylcholinesterase. Bioassay is a useful method to diagnose insecticide resistance of mosquitoes; however, it is hard to establish regional/annual resistance database based on bioassay. Recently, various markers of mutation and copy number variation have been identified through insecticide mechanism studies. Thus, molecular detection of resistance based on the resistance marker is now feasible, which can be readily implemented as a novel resistance monitoring tool to complement or replace the conventional bioassay method. In Korea, the density of vector mosquitoes native to the subtropical areas has increased due to climate change. Therefore, it is required to establish an efficient resistance monitoring system based on molecular markers to facilitate the construction of a nation-wide resistance map for more effective control of mosquitos. In addition, alternative insecticides should be introduced to the areas where mosquitoes develop high levels of resistance to maximize control efficacy against resistant populations

        3년간(2016-2018년) 청주지역에서 채집된 모기의 발생분포를 채집지 3곳(주거지와 철새도래지, 축사)에서 2종의 트랩(BL트랩과 BG트 랩)을 이용하여 비교 조사하였다. 주거지에서 가장 많이 채집된 모기 종은 빨간집모기였으며, 철새도래지와 축사에서는 금빛숲모기가 가장 많이 채집되었다. 철새도래지와 주거지에서는 BG트랩이 BL트랩보다 모기가 더 많이 채집되었으나 축사에서는 BL트랩에서 더 많은 모기가 채집되 었다. 빨간집모기와 흰줄숲모기, 큰검정들모기는 BG트랩에서 많이 채집되었고 금빛숲모기, 얼룩날개모기류, 동양집모기는 비교적 BL트랩을 선호하는 것으로 조사되었다. 채집된 모기를 분류 ․ 동정한 결과 2016년에는 10종 22,679개체가 채집되었고, 2017년에는 8종 6,502개체가 채집 되어 2016년보다 3.49배 감소하였다. 2018년에는 6,803개체가 채집되어 2016년보다 3.33배 감소하였다. 이러한 결과가 강수량과 관련이 있을 것으로 보이며 기상조건과 여러 변인들(채집지역과 모기종류, 트랩종류)에 의한 모기밀도와의 관련성을 비교한 결과 특정 관련성을 확인할 수는 없었다. 채집된 모기의 병원체 감염을 확인한 결과, 2016년도 축사에서 채집한 금빛숲모기 2개 pool, 2018년에 축사에서 채집한 빨간집모기 1개 의 pool에서 차오양바이러스(chaoyang virus)가 검출되어 각각 0.088와 0.147의 최소감염율(MIR)을 보였다. 2017년에는 바이러스가 검출되 지 않았다.

        The eggs of Asian tiger mosquito, Aedes albopictus, possess high desiccation resistance, which contribute the rapid spread of this mosquito across the world. Melanization of eggshell appear to play a role in the resistance to desiccation. Dopachrome-conversion enzyme (DCE, Yellow) significantly accelerates the melanization of the eggshell. In this study, we demonstrated functional importance of two yellow genes, AalY-g and AalY-g2, in the chorion formation. Both genes were highly induced in the ovary at 48 h after blood meal. Injection of dsRNA for AalY-g or AalY-g2 into adult females had no effect on fecundity. However, the outermost colorless exochorion of the eggs obtained from both dsRNA-treated females was fragile and peeled off in places, and melanization of the endochorion was obviously delayed by several hours. In addition, unlike eggs from control females which acquired high desiccation resistance between 18 and 24 h after oviposition (HAO), 60-70% 24 HAO eggs from either AalY-g- or AalY-g2-deficient females were collapsed when they were moved to an air-dry condition, and the desiccation resistance was not increased in later stages of embryonic development analyzed. TEM analysis revealed that abnormal morphology and ultrastructure of the endochorion, particularly outer-endochorion, in the 24 HAO and older eggs from either AalY-g-and AalY-g2-deficient females. These results indicate that AalY-g and AalY-g2 are required for morphology and formation of the endochorion (outer-endochorion), a structure that appears to be critical for desiccation resistance of the Ae. albopictus eggs.         Autophagy is an important self-eating process to eliminate damaged or unused organelles. We identified nine autophagy-related genes (Atg) including AaAtg-1, -3, -4b, -4d, -5, -6, -8, -12 and -13 from the Asian tiger mosquito, Aedes albopictus. Developmental expression patterns indicate that mRNA levels of AaAtg-1, -3, -4b, -4d, -5, -6, -12 and -13 were highly expressed in egg, whereas expression of AaAtg8 was high in 1stand3rdinstarlarvalstages. TissuespecificexpressionofthesegenesindicatesthatAaAtg1 was highly expressed in thorax and midgut in blood-fed adult female mosquitoes (BF), and head and thorax in sugar fed adult female mosquitoes (SF). Transcript level of AaAtg3 was high in thorax in BF, but head, thorax and Malpighian tubules in SF. AaAtg4b, -4d mRNA levels were significantly high in Malpighian tubules in BF, and head in SF, respectively. AaAtg-5 and -6 transcripts were highly expressed in head in BF, and expression of AaAtg-8 was high in Malpighian tubules in BF. Levels of AaAtg-12 and -13 mRNAs were significantly high in head and midgut in BF. Induction patterns of AaAtg genes against pathogens showed that AaAtg-1, -3, -4b, -8, -12 and -13 were strongly induced at 6 h-post injection of S. aureus, and mRNA levels of AaAtg-1, -3 and -13 were significantly induced by E. coli challenge after 3 h-post injection in SF abdominal carcass. In SF midgut, AaAtg-1, -3, -4b, -4d, -5, -6, -12 and -13 transcripts were drastically induced at 9 h-injection of E. coli and S. aureus, while expression of AaAtg-8 was highly induced by S. aureus and C. albicans at 9 h-post injection. Each AaAtg gene was slightly induced by E. coli, S. aureus or C. albicans at different time points in abdominal carcass in BF. Interestingly, AaAtg-8 was not induced by microbial challenge. While eight other Atg genes except AaAtg-8 were highly influenced by S. aureus at 6 and 9 h-post injection, E. coli at 3 h-post-treatment, and 3, 6, and 9 h-post inoculation. In the future, we will characterize the functional roles of autophagy during mosquito-microbes interaction.

        This study investigated number of eggs, hatch rate, toxicities and LT (lethal time) values of Culex pipiens and Aedes albopictus using Aquatain™(Aqua mosquito film). Aquatain™ is a covering agent applied on the surface of water for the control of larva and pupa of mosquito. It forms molecule layer on the surface of the water to lower the surface tension of the water, to prevent flooding of the larva of the mosquito larvae, or to suffocate by obstructing breathing. Cx. pipiens and Ae. albopictus larvae observed 99.9% and 1% larvicidal effect after 24h, respectively. However, both mosquito pupae were showed 100% mortality after 24 h. The AMF™ was treated with three doses (0.5, 1, 2 folds) to observe the LT value in larvae and pupae stages of Cx. pipiens and Ae. albopictus. The LT values of Ae. albopictus pupa was similar regardless of dose, but the LT value of Cx. pipiens pupa decreased as the dose increased. However, Ae. albopictus larvae showed 18.6 folds longer LT90 time than Cx. pipiens larvae. Both species mosquito showed statistically significant differences in the number of eggs laid and hatch rate at recommended dose. Therefore, Aquatain AMF™ may be used to control mosquitoes, although the activity varies depending on the species of mosquito.

        기후온난화 현상이 지속되고 있는 제주지역에서 환경적으로 다른 지역 모기의 계절적 발생밀도를 조사하기 위해 제주시의 국제공항, 항만 구역과 축사 그리고 서귀포 도심지의 11지점을 선정하여 3월부터 11월까지 매달 2회씩 Black light trap과 BG sentinel trap을 이용하여 모기를 채집하였다. 채집된 모기는 5속 7종. 6,042마리였으며, 이 중 빨간집모기(Culex pipiens)가 4,159마리(68.8%)로 우점종이었으며 흰줄숲모기 (Aedes albopictus)는 1,348마리(24.4%)였다. Black light trap를 이용한 채집에서 중앙동주민센터는 트랩당 72.8마리를 채집하여 모기 밀도가 가장 높게 나타났으며 제주국제공항은 트랩당 1.4마리로 가장 낮게 나타났다. BG sentinel trap을 이용한 채집에서는 항만에서 트랩당 71.7마리로 가장 많았고 도심지의 걸매생태공원에서 28.3마리로 가장 낮았다. 시기별로 모기의 밀도는 5월부터 증가하기 시작하여 8월에 1,156마리 (19.1%)로 가장 높은 밀도를 나타내었다. 채집된 암컷모기를 종별, 시기별, 지점별로 나누어 pool당 50마리 이하로 설정하여 총 364 pools에서 flavivirus 존재여부를 real time RT-PCR로 검사하였으나, 검출되지 않았다.

        Insect growth regulators (IGRs) are insecticides that disrupt the normal development of target insects. Among the IGR insecticides, juvenile hormone (JH)-related IGRs are of particular interest because they stimulate or interfere with the formation of JH receptor complex. In the precious studies, novel JH-related IGRs with JH agonist (JHA) and antagonist (JHAN) activity were identified by using yeast two-hybrid system transformed with the Aedes aegypti JH receptor complex. In this study, the transcriptomic responses of Aedes albopictus were investigated upon JHA and JHAN, respectively. These results will provide important information about understanding of impact of JH-related IGRs in transcription level.
