        본 연구의 목적은 SNP 방법으로 벼 계통 및 품종의 전분특성을 이화학 특성 조사를 거치지 않고 빠르고 간편하게 판별하고자 프라이머를 제작하는 것이다. 실험을 위해 기존 육성된 100여 품종을 대상으로 아밀로스함량, 알칼리붕괴도, RVA 특성 및 단백질 함량을 조사하였고, SSⅡa 유전자와 SBE 유전자를 탐색하여 기존에 발표된 PCR Primer를 포함한 21개 SNP 마커 (SSⅡa 유전자 관련 18개, SBE 유전자 관련 3개)를 제작하였다.제작된 프라이머의 최적PCR 조건을 확립하였고, 아가로스젤상에서 품종별 전분특성과 제작된 마커의 관계를 분석한 결과 신규 제작한 프라이머 중 4330F-1, 4330F-2와 4198F-1, 4198F-2 프라이머의 통일벼 계통 품종 판별이 가능함을 확인할 수 있었다.

        Expressed sequence taqs (ESTs) have accepted to be a valuable tool for discovering single nucleotide polymorphism (SNP) in many species. For detection putative SNPs in soybean genome, approximately 90,000 EST sequences from genotype Williams 82 were downloaded from NCBI. The paralog sequences of these ESTs were distinguished by TGI clustering tools (TGICL) performing megablast and EST cluster analysis, and the EST clusters were used as reference sequences for detection putative SNPs by in silico. The EST clusters were aligned with EST sequence from other cultivars of soybean by Polybase (computer software). The results revealed that putative SNPs were distributed in 5,677 clusters with frequency of 1 SNP per 333 nucleotide sites. For SNP validation, 43 primer pairs were designed from EST clusters containing putative SNPs for sequencing genomic DNA of Williams 82, Harosoy, Peking, Pureunkong, Jinpumkong 2, Hwangkeumkong and IT182932. From results of sequencing PCR, we totally found 99 SNPs from 33 primer pairs. Twenty-three and 47 out of 99 SNPs showed polymorphisms between Pureunkong and Jinpumkong 2, and Hwangkeumkong and IT182932, respectively. The SNPs discovered from this study can be used for genetic mapping in the four genotypes of soybean.

        A reaction of 214 lines and varieties to BB korean race K1, resistance and moderately resistance was 33 varieties which were bred before in 1990, susceptible were 5 varieties (Milyang23, Dongjinbyeo, Sinseonchalbyeo, Changseongbyeo and Cheongmyeongbyeo). Among bred after in 1990 resistance appeared 58 varieties, 18 varieties were susceptible. Bred after in 2000 were 50 varieties appeared resistance and 5 varieties were susceptible (Manho, Goun, Jinbong, Josaengheugchal, and Manna). Accordingly most varieties bred in south korea seemed to possession resistance to BB korean race K1. As a result of whether or not having Xa1 gene of rice varieties by SNP marker appeared that 43 varieties and the other 10 lines and varieties have Xa1 gene. Varieties bred before in 1990 have 18 (62.1%) varieties out of 29 varieties possession Xa1 gene to showed resistance to BB korean race K1, 19 (47.5%) varieties bred in 1990~2000, 6 (12.8%) varieties (Boseogchal, Daepyong, Hanareumbyeo, Hopyeongbyeo, Jongnam, Kuman) bred after in 2000 seemed to possession Xa1 gene.
