        목적 : 본 연구는 보호자의 인터뷰 없이 환자의 평가를 통해 다양한 인지영역을 빠르게 평가할 수 있고, 경도 인지장애의 치료적 중재 제공을 결정하는 노인 경도 인지장애 선별 검사 항목 개발하고자 한다. 연구방법 : 2014년 10월 23일부터 2014년 12월 16일까지 총 3회에 걸쳐 전문가 패널로 선정된 30명의 참여자에게 전자우편을 통해 델파이 조사를 실시하였다. 이후 경도 인지장애 환자 28명을 포함하여 총 81명의 환자에게 한국판 간이 정신상태 검사(Korean version of Mini-Mnetal State Examination; MMSE-K)와 본 평가도구를 평가하여 타당도를 검증하였다. 결과 : 최종 델파이 조사결과 평균 적합도 4.24와 중요도 4.30으로 분석되어 경도인지장애 선별평가 항목 으로 적절한 것으로 분석되었다. 적합도의 내용타당도 비율 0.84, 중요도의 내용타당도 비율 0.93으로, 적합도와 중요도 모두 내용타당도 비율이 높게 나타났다. 최종적으로 델파이 조사를 통해 노인 경도 인 지장애 선별검사 항목 개발에 관한 24개의 항목이 최종적으로 도출 되었다. MMSE-K와 높은 상관성을 보이고, Cronbach’s α는 .80으로 높은 타당도를 보였다. 결론 : 전문가의 의견을 수렴한 델파이방법을 통해 만들어진 본 평가도구는 기존의 평가도구와 높은 상관 성을 보이고 타당도가 높은 것으로 나타났다. 이에 본 도구는 경도 인지장애를 평가함에 있어 효과적이 라고 사료된다. 주제어 : 경도 인지장애, 노인, 델파이 조사, 선별평가, 항목개발

        A positional scanning synthetic peptide combinatorial library (PS-SCL) was screened in order to identify antimicrobial peptides against the cariogenic oral bacteria, Streptococcus mutans. Activity against Streptococcus gordonii and Aggregatibacter actinomycetemcomitans was also examined. The library was comprised of six sub-libraries with the format O(1-6)XXXXX-NH2, where O represents one of 19 amino acids (excluding cysteine) and X represents equimolar mixture of these. Each sub-library was tested for antimicrobial activity against S. mutans and evaluated for antimicrobial activity against S. gordonii and A. actinomycetemcomitans. The effect of peptides was observed using transmission electron microscopy (TEM). Two semi-mixture peptides, RXXXXN-NH2 (pep-1) and WXXXXN-NH2 (pep-2), and one positioned peptide, RRRWRN-NH2 (pep-3), were identified. Pep-1 and pep-2 showed significant antimicrobial activity against Gram positive bacteria (S. mutans and S. gordonii), but not against Gram negative bacteria (A. actinomycetemcomitans). However, pep-3 showed very low antimicrobial activity against all three bacteria. Pep-3 did not form an amphiphilic α-helix, which is a required structure for most antimicrobial peptides. Pep-1 and pep-2 were able to disrupt the membrane of S. mutans. Small libraries of biochemically-constrained peptides can be used to generate antimicrobial peptides against S. mutans and other oral microbes. Peptides derived from such libraries may be candidate antimicrobial agents for the treatment of oral microorganisms.

        목 적: 20대 대학생을 대상으로 McMonnies 및 Ocular Surface Disease Index(ODSI) 건성안 설문지를 이용한 건성안 유병률을 비교하고 그 차이를 확인하고자 하였다. 방 법: 전국 10개 대학교 안경광학과에 재학 중인 대학생 1,157명을 대상으로 McMonnies 및 OSDI(Ocular Surface Diseease Index) 설문지를 이용하여 건성안 정도를 조사하였다. 증상의 정도를 정량화하여 두 설문지의 유병률을 비교한 후 카파 계수(kappa coefficient)를 활용하여 유병률 일치도가 가장 높은 분류기준점을 찾고자 하였다. 결 과: McMonnies설문은 기준점이 14.5점 기준일 때 유병률이 7.1%였고 OSDI설문은 23.5점 기준일 때 41.6%였으며 일치도(kappa coefficient)는 매우 낮았다(k=0.292, p

        This study was carried out to analyze the differences in p-hydroxylbenzyl alcohol (HBA) content, antitumor and anti-obesity activities and tyrosinase inhibitory activity between non-fermented G. elata (NFGP) and fermented G. elata powder. The HBA content, which is an index-component of G. elata decreased from 1.58 mg/g before fermentation to 1.07, 0.32, and 0.13 mg/g after the 1st fermentation (1st FGP), 2nd fermentation (2nd FGP) and 3rd fermentation (3rd FGP), respectively. The anti-proliferation effects on the cell lines HT29 and AGS were significantly higher for the fermented G. elata than the NFGP. The antitumor activity was also increased in a fermentation number-dependent manner. During adipocyte differentiation, the ethanol extract of the 3rd FGP inhibited lipid accumulation in 3T3-L1 cells significantly better than NFGP and the 1st FGP, treated at the concentration of 10μg/mL. The tyrosinase inhibitory activity of the 2nd FGP at 600μg/mL over was higher than that of kojic acid. At the concentration of 1,000μg/mL, the tyrosinase inhibitory activity was increased in a fermentation number-dependent manner. From these results, the fermented G. elata, especially the 3rd FGP, is expected to be good candidate for the development of functional food and agents with antitumor, anti-obesity, and tyrosinase inhibitory potential.

        The purpose of this report is to discuss the obtained findings gathered from ultrasound screenings of the liver. After running diagnostic tests health, screens were then conducted to analyze and compare the gained results. This data was then charted and used to strengthen our theorized hypothesis. From January 2013 to June 2013 a recorded 2906 people over the age of 20 visited Health Promotion Centers in various areas throughout Daejeon. Of those 2906 participants 1789 underwent screenings for abdominal ultrasonography; these participants as well as their ultrasound results were used as the bases of our study. For the establishment of our baseline and the comparison of our results, we gathered case-controlled studies from various reputable sources. Both the control and the experimental groups were tested to measure the following liver enzymes (AST, ALT, r-GTP, ALP, and etc.). Kidney functions were measured as well enzymes such as BUN, Creatinine, and Uric Acids levels were analyzed and recorded to see if any relationships existed between the levels documented in the liver and those in the kidneys. It was shown that the two primary causes of fat build up within the liver were significantly connected to obesity BMI(OR=4.14) and waist circumference(OR=3.88).

        A number of screening tools have been developed to evaluate the human balance capability. Many of them were designed to identify the elderly with falling risk. Three different screening tools, which have been well used many clinical fields, were used in this study. The purpose of this study was to evaluate balance capability in facilities maintenance workers in korea. There were no statistical significance between male and female when evaluated with three different screening tools. However, significant differences were found among the age groups irrespective of gender when evaluated with three different screening tools. The results of three different screening tools in korea showed poor values compared with previous results. These results revealed that facilities maintenance workers faced more critical falling risk in korea.

        Silkworm transgenesis is now a routine method leading to a satisfactory yield of transformed animals and the reliable expression of transgenes during multiple successive generations. However, the screening of G1 transgenic individuals from numerous progeny has proved to be difficult and time-consuming work. Previously, we characterized the promoter of heat shock protein 70 from Bombyx mori (bHsp70), which is ubiquitously expressed in all tissues and developmental stages. To investigate the utilization of the bHsp70 promoter to screen transgenic individuals, the EGFP marker gene was inserted into the piggyBac vector under the control of the bHsp70 promoter. Mixtures of the donor and helper vectors were micro-injected into 3,060 eggs of bivoltine silkworms (Keomokjam). EGFP fluorescence was observed in 17 broods of transgenic silkworms under a florescence stereomicroscope. Interestingly, this fluorescent marker protein was detected not only in parts of the embryo segments on the seventh day of the G1 embryonic developmental stage but it was also detected in a part of the body of G1 hatched larvae, in the middle silk gland of G1 fifth instar larvae, and in the wings of seven-day-old G1 pupae and G1 moths. Therefore, we suggest that the bHsp70 promoter can be used for the rapid and simple screening of transgenic silkworms.

        Bacterial soft rot is one of the major disease of Zantedeschia species (Calla lily) caused by Erwinia carotovora. The objectives of this study were i) to screen the most efficient method to determine resistance level against E. carotovora and ii) to evaluate the genetic variability in Zantedeschia genotypes by inoculation of Ecc NHRI-3 and PD 1784 isolates. Four screening tests i.e. leaf disk, petiole, tuber and tuber slice tests were used to determine the resistance level in calla lily. Eleven genotypes from section Zantedeschia were used for variation studies against E. carotovora by using the leaf disk test. It was observed that all genotypes showed variation in resistance level and could be categorized in 3 groups on the basis of their resistance level. Four of the genotypes were resistant against this pathogen whereas, 6 genotypes were moderately resistant and only one genotype was found susceptible. Within section Aestivae, ‘Galaxy’, ‘Florex Gold’, ‘Treasure’ and ‘Mango’ were found very susceptible cultivars, whereas ‘Coral Sunset’, ‘Hazel Marie’ and ‘Neroli’ were less susceptible genotypes. Most of the cultivars from section Aestivae were susceptible to bacterial soft rot and the cultivar ‘Florex Gold’ was identified as susceptible control. It is suggested to perform pre-screening through leaf disk method which proved to a non-destructive test. The pre-screening evaluation can discriminate susceptible cultivar and resistant cultivar. Tuber slice test is more useful in screening subsequent genotypes at a later growth stage in any breeding program.

        The aim of the current study was to analyze the active ingredients and to screen the pharmacological properties of freshwater laver, Prasiola japonica, the only species grown in Korea. According to results of gas chromatography- mass spectrometry assay, components from P. japonica were more diverse than those from sea laver. Of particular interest, our results indicated that ethanol extract of P. japonica (PJE) contained loliolide, sorbitol, mannitol, and alverine, which were known to have an anti-oxidant, anti-oral microbial, osmotic diuresis, and smooth muscle relaxant, respectively. In addition, five solvent fractions of PJE (water, butanol, chloroform, ethyl acetate, and hexane) significantly inhibited the production of lipopolysaccharide-induced nitric oxide and a higher amount (>100 μg/mL) of chloroform, ethyl acetate, and hexane fraction were considered to play a specific role in cancer cell death. PJE and its solvent fractions found to be effective scavengers of free radicals, particularly, hydroxyl radicals. Glucose uptake in L6 myoblast cell line that stably expresses the glucose transporter type 4 (GLUT4) proteins was also remarkably enhanced upon treatment with solvent fractions, remarkably chloroform fraction. Taken together, we concluded that P. japonica may have potent pharmacological properties and thus contribute to development of novel natural candidates for various disease targets.

        Beet armyworm, Spodoptera exigua are difficult to control using chemical insecticides because of the development of insecticide resistance. For eco-friendly beet armyworm managements, various control agents are required. Entomopathogenic fungus is one of promise control agents as an alternative to chemcal control agent. We conducted bioassays with entomopathogenic fungi to select high virulence isolate to larva of beet armyworm. The bioassay was used 150 entomopathogenic fungal isolates which were isolated from soil samples of nine provinces by insect-bait method using Galleria mellonella and Tenebrio molitor. . Three isolates, Metarhizium spp. FT83, FT89 and FT90 had high virulence as 84.6%, 100% and 100%, respectively, against S. exigua. The medial lethal time(LT50) of conidia of three isolates was 5.01, 2.99 and 2.92days respectively.

        Rapid identification of pest species found under quarantine is an important factor in preventing an economic loss of agricultural commodities. In this study, we analyzed RNA-Seq of the larvae of C. sasakii, G. molesta and G. dimorpha, which are serious pests in several fruits in Korea and are difficult to discriminate by species in their larval stage because of lack of a morphological character. To select immunological diagnostic markers, discriminating the larvae of C. sasakii from the G. molesta and G. dimorpha, RNA-Seq was performed for the larvae of the three insects. The 454 pyrosequencing generated 3,058-4,686 contigs for each three pest species, which assembled into 2,584-3,970 isotigs with average lengths of 829-1,244 bp. Functional annotation of the sequencing results generated 774 orthologs for the three pest species, and 12 isotigs were finally registered as candidate markers for species discrimination through bioinformatical screening, literature search, and gene expression study. The selected candidates include serine proteases, serpins, 27 kDa glycoprotein and storage protein with a constitutive gene expression in their larvae, pupae and adult stage.

        Entomopathogenic fungi are natural pathogens of insects and contribute to the regulation of host insect populations in the environment. Several these fungi produce a wide range of secreted enzymes, secreted protein toxins and secondary metabolites to overcome host defenses and ultimately kill the host, and to defend host resources against competing pathogens and saprophytes. This study was performed to evaluate the antimicrobial activity of 207 entomopathogenic fungi form Korea soils against plant pathogenic bacteria Ralstonia solanacearum and plant pathogenic fungi Botrytis cinerea using dual culture technique on SDYA. As results, twelve isolates (5.7%) and six isolates (2.8%) showing the greatest inhibition against R. solanacearum and B. cinerea, respectively. The culture supernatant of these selected isolates completely suppressed the growth of the pathogen, indicating that suppression was due to the presence of antimicrobial compound in the culture filtrate. The stability test of the culture filtrate showed that the antimicrobial component was heat stable and not protein. These entomopathogenic fungal metabolites may be a good feature to be used in the development of a new biocontrol method of R. solanacearum and B. cinerea.

        The exposure of food to ionizing radiation has been recognized as a safe and effective mode of food preservation in more than 55 countries. The benefits include eradication of insect pests, inactivation of food pathogens, extension of shelf-life, and improvement in food hygiene. Regulatory authorities around the world have emphasized the implementation of various national and international regulations to facilitate trade and development of consumers’confidence in purchasing irradiated foods. Therefore, the need for reliable irradiation detection methods has increased to enforce these regulations. At present, a number of promising analytical approaches have been developed and evaluated. Moreover, about 10 European Standards have been adopted as General CODEX Alimentarius methods for the detection of irradiated foodstuffs. However, most of these methods demand relatively expensive equipment and prolonged sample preparation. Therefore, simple and cost-effective approaches would be advantageous for rapid screening of foodstuffs. The suspected samples need to be analyzed further with more validated techniques to confirm the screening results. In this review, existing screening methods (i.e. physical, chemical, and biological) for the identification of irradiated foods have been outlined along with their principles, scopes and limitations.

        This study investigated the genes involved in the dif- ferentiation of odontoblasts derived from human dental pulp stem cells (hDPSCs). hDPSCs isolated from human tooth pulp were validated by fluorescence activated cell sor- ting (FACS). After odontogenic induction, hDPSCs were analyzed investigated by Alizaline red-S staining, ALP assay, ALP staining and RT-PCR. Differential display-poly- me rase chain reac tion (DD-PCR) was pe rformed to s c re en differentially expressed genes involved in the differentia- tion of hDPSCs. By FACS analysis, the stem cell markers CD24 and CD44 were found to be highly expressed in hDPSCs. When hDPSCs were treated with agents such as β- glycerophosphate (β-GP) and ascorbic acid (AA), nodule formation was exhibited within six weeks. The ALP activity of hDPSCs was found to elevate over time, with a detectable up-regulation at 14 days after odontogenic induc- tion. RT-PCR analysis revealed that dentin sialophospho- protein (DSPP) and osteocalcin (OC) expression had inc- reased in a time-dependent manner in the induction culture. Through the use of DD-PCR, several genes were diffe- rentially detected following the odontogenic induction. These results suggest that these genes may possibly be linked to a variety of cellular process during odontogenesis. Further-more, the characterization of these regulated genes during odontogenic induction will likely provide valuable new in- sights into the functions of odontoblasts.

        국내야생종인 새머루로부터 내병성 포도품종 육성에 필요한 육종소재와 유용정보를 획득하고자, 포도 새눈무늬병균과 줄기혹병균에 감염된 새머루에서 식물 방어관련유전자의 발현양상을 분석하였다. Thaumatin (TLP), glutathione peroxidase (GPX), glutathione-S-transferase (GST), chalcone synthesis (CHS), protein kinase regulator (14-3-3) 및 β-glucanase (Glu) 등의 방어관련유전자, leucine reach-repeat (LRR)와 lipoxygenase (LOX) 등의 신호전달관련유전자, polygalacturonase-inhibiting protein (PGIP)과 같은 세포벽관련 유전자의 발현이 포도 새눈무늬병균 접종에 의해 유도되었다. 포도새눈무늬병균의 포자접종 및 배양여액처리에 의해 WRKYtranscription factor 10 (WRKY), fatty acid elongase (FAE)의 발현이 유도되었으며 proline-rich protein (PRP) 유전자의발현은 억제되었다. 발현이 증가한 15개 유전자 중에서 LRR, LOX, TLP, GST 등은 포도새눈무늬병균 포자현탁액 접종에 의해 발현이 크게 증가하였으나, CHS와 tonoplast intrinsicprotein (TIP)는 배양여액처리에 의해 증가하였다. 포도 줄기혹병균의 접종에 의해 LRR, CLP, LOX, TLP, GPX, 14-3-3,GST, PGIP, FAE, TIP, Glu, WRKY 등은 발현이 증가하였고, CHS와 PRPs는 발현이 억제되었다. 포도 줄기혹병균 접종에 의해 활성산소의 축적 및 분해와 관련된 GPX와 GST는 발현이 크게 증가하였다.

        The purpose of this study is to determine the possibility of using Agarum cribrosum as a natural health food source. To accomplish this purpose, the contents of the general and antioxidant activities of Agarum cribrosum were measured. The contents of carbohydrate, crude protein, crude lipid and ash were 45.4%, 15.0%, 2.3% and 33.1%. The calories of Agarum cribrosum were 262.3 kcal and total dietary fiber of Agarum cribrosum was 34.0%. The protein contained a total of 18 different kinds of amino acids. The content of amino acids was 12, 402.42 ㎎/100 g. The K was the largest mineral followed by Ca, Na and Mg, implying that Agarum cribrosum is an alkali material. The antioxidant activity of Agarum cribrosum was assessed by various radical scavenging assays using DPPH (2, 2-diphenyl-1 picrylhydrazyl), FRAP (ferric ion reducing antioxidant power), reducing power, and ABTS (2, 2'-azino-bis(3 ethylbenzothiazoline-6-sulphonic acid)). All antioxidant activity of Agarum cribrosum extract increased the concentration of the dependents. Total phenolic contents of Agarum cribrosum extract were 34.1±2.56 ㎎/g, and total flavonoids contents were estimated at 4.9±0.26 ㎎/g. General nutrients and other antioxidant bioactive materials in Agarum cribrosum were also potential materials for good health food. It is expected that a follow-up study of Agarum cribrosum through developing processed food and evaluation of their functional properties would provide useful information or sources of functional foods.

        The purpose of this study is to determine the possibility of using cultured wild ginseng roots as a natural health food source. To accomplish this purpose, the contents of general and antioxidative nutrients of cultured wild ginseng roots were measured. The contents of carbohydrate, crude protein, crude lipid and ash are 61.72%, 17.36%, 0.23% and 10.90%, respectively. Further, the calories of cultured wild ginseng roots were 323.97 kcal. Total dietary fiber was 82.13%. The protein contained a total of 18 different kinds of amino acids. The contents of amino acids were 16.15 g. The K was the largest mineral followed by P, Ca, and Mg, which means cultured wild ginseng roots is alkali material. The contents of saturated fatty acids and unsaturated fatty acids were 0.23 g, and 0.62 g, respectively. Crude saponine content was 25.87 ㎎/g. Total phenolic contents of cultured wild ginseng roots were 11.2 ㎎/g, and total flavonoids contents were estimated as 4.2 ㎎/g. The electron donating ability of cultured wild ginseng roots were 24.7~31.6%. The nitrite scavenging activity was pH dependent, and was highest at pH 1.2 and lowest at pH 6.0. The cultured wild ginseng roots extract showed the highest reducing power (0.06) at the concentration of 1, 000 ㎍/㎖. Based on the above results, we deemed that the cultured wild ginseng roots might have potential antioxdant activities.

        Decisions on reliability screening rules and burn-in policies are determined based on the estimated reliability. The variability in a semiconductor manufacturing process does not only causes quality problems but it also makes reliability estimation more complicated. This study investigates the nonuniformity characteristics of integrated circuit reliability according to defect density distribution within a wafer and between wafers then develops optimal burn-in policy based on the estimated reliability. New reliability estimation model based on yield information is developed using a spatial stochastic process. Spatial defect density variation is reflected in the reliability estimation, and the defect densities of each die location are considered as input variables of the burn-in optimization. Reliability screening and optimal burn-in policy subject to the burn-in cost minimization is examined, and numerical experiments are conducted.
