        건조방법을 달리한 초석잠(Stachys sieboldii Miq.)을 열수 및 70% 에탄올 용매로 추출하여 제조한 후 추출물 농도에 따른 총 flavonoid 및 총 polyphenol 함량, DPPH radical 소거활성, ABTS radical 소거활성, 산화적 스트레스로부터의 신경세포 보호효과에 미치는 효과를 조사하기 위하여 실시 하였다. 초석잠 추출물들의 총 flavonoid와 총 polyphenol 함량은 동결건조 열수 추출물에서 각각 가장 높았다(p<0.05). DPPH 및 ABTS radical 소거활성은 농도의존적으로 증가하였으며, 1,000 ug/mL 농도에서는 동결건조 열수추출물에서 유의적으로 가장 높게 나타났으나, 대조군인 비타민 C에 비해 유의적으로 낮았다(p<0.05). 신경세포 보호효과는 열품 및 동결건조 열수추출물에서 농도의존적으로 높게 나타났으며, 대조군인 비타민 C에 비해 낮았다(p<0.05). 본 연구를 통하여 열풍건조에 비해 동결건조 초석잠의 열수추출물이 총 flavonoid와 총 polyphenol 함량이 더 높게 함유되어 있어 항산화 활성 및 산화적 스트레스로부터의 신경세포 보호효과가 우수하게 나타내는 것이 관찰되었다.

        From August to October 2013, a powdery mildew was found on Japanese artichoke(Stachys sieboldii) in Jinju, Korea. White colonies were present on leaves and petioles. Severely infected lesions were discolored, being brown. In the present study, the morphological features of anamorphic and teleomorphic Korean specimens were studied. To complete the identification, the sequence of the internal transcribed spacer region of the ribosomal RNA gene(ITS rRNA) was analyzed. Based on morphological characteristics, including anamorphic and teleomorphic features, as well as analysis of the ITS rRNA gene region, the fungus associated with the symptoms was identified as Neoerysiphe galeopsidis. Although the host ranges of powdery mildew caused by N. galeopsidis has been previously recorded, no full description or illustration of fungal symptoms and signs on Japanese artichoke has yet appeared. To the best of our knowledge, this is the first full description of powdery mildew disease on Japanese artichoke featuring molecular identification, morphological features, symptoms, and signs in Korea.

        Background : Domestic use of cosmetics has largely been dependent on imports for the opening of cosmetics raw materials, but research has been progressing actively on natural ingredients to develop new materials. Methods and Results : The extracts of hot–water and ethanol extracts from Stachys sieboldii were tested cytotoxicity, inhibition of melanin biosynthesis and NO production activity. The cytotoxicity of the hot water extract of S. sieboldii were 98.4, 95.0, 93.9 and 90.6%, respectively, when treated with 10, 50, 100, 500 ㎍/㎖. Cytotoxicity of S. sieboldii ethanol extract was 97.1, 95.3, 94.6 and 94.4%, respectively, at the concentrations of 10, 50, 100 and 500 ㎍/㎖. Melanin biosynthesis inhibitory effect of α-MSH at 100 μM increased to 118.1% in melanin, but decreased to 92.3% in S. sieboldii ethanol extract. NO production inhibition increased to 109.2% when treated with LPS and was 103.3% at 500 ㎍/㎖ of hot water extract and 105.9% at 500 ㎍/㎖ of ethanol extract of S. sieboldii. Conclusion : The ethanol extract of S. sieboldii did not showed the cytotoxicity, and reduce NO production. Therefore, ethanol extract of S. sieboldii had an potentials for developing whitening cosmetics.

        This study investigated the antioxidant and antidiabetic activities of Stachys sieboldii Miq. extracts by solvents (water, ethanol, butanol, chloroform, and hexane). The contents of total polyphenols (7.18-37.25 mg/g) and flavonoids (0.21-5.21 mg/g) in extracts from Stachys sieboldii Miq. showed a significant difference dependent on the extraction solvents, butanol > ethanol > water > chloroform > hexane. Antioxidant activities by DPPH and ABTS radical scavenging were increased in a dose-dependent manner. These activity trends associated with the extraction solvent were different at each concentration, but resembled phenolic compound contents trend, generally. FRAP value increased in a dose-dependent manner, but there was a difference in radical scavenging activities when comparing between extraction solvents by butanol > ethanol > hexane > chloroform > water on all concentrations. The trend of α-amylase inhibition of extracts from 1,000 μg/mL to 2,000 μg/mL was not affected as enzyme activity is promoted and not inhibited. The inhibition of α-glucosidase was increased in a dose-dependent manner without water extracts, the activity on hexane extracts was higher than others per the extraction solvent. α-Glucosidase inhibition of hexane extracts showed 57.76% at 250 μg/mL, which is 2.8 times higher than the second highest chloroform extract (20.65%). From these results, we presume that the active ingredients of Stachys sieboldii Miq. is different according to the extraction solvent and also the activity is different by these major functional groups.
