        This study was conducted to examine the utilization potential of grape stems as nutritional supplements, and the physiological functionalities of 70% ethanol extracts from grape fruit stem (GFS) were investigated. Each experimental group was prepared with different methods and included GFSF (GFS prepared with freeze drying), GFSI (GFS prepared with infrared drying), GFSH (GFS prepared with heat air drying), and GFSS (GFS prepared with sun drying). The respective yields of freeze-dried powders for the GFSF, GFSI, GFSH, and GFSS were 59.27%, 57.13%, 58.57%, and 58.87%, respectively. Total polyphenol contents in the GFSF were significantly greater than those in the other extracts, whereas total flavonoid contents in the GFSI were higher than those in the other extracts. The contents of proanthocyanidin-related substances were ranked in the order of GFSF > GFSI > GFSH > GFSS. The thin layer chromatograph (TLC) analysis of catechin showed that the GFSF, GFSI, GFSH, and GFSS were detected in the same band. The electron donating ability with 500 μg/mL (w/v) solutions of GFSF, GFSI, GFSH, and GFSS amounted to 93.14%, 93.07%, 92.64%, and 86.95%, respectively, and the reducing powers (OD 700) were 1.933, 1.765, 1.455, and 1.200 absorbance units, respectively. Additionally, the ABTS radical scavenging ability showed the same tendency that was observed with the electron donating ability and reducing power. The angiotensin-converting enzyme (ACE) inhibitory activity and tyrosinase inhibitory activity with 500 μg/mL (w/v) solutions of GFSF and GFSI were higher than those of GFSH and GFSS. In conclusion, the infrared drying technique is the superior method for the enhancement of biological activity for by-product utilization.

        The physiological activities of 70% ethanol extracts of grape seed (GS) prepared by freeze-drying (GSFD), infrared drying (GSIR), hot-air drying (GSHD), or sun-drying (GSSD) were investigated. The moisture contents of GSFD, GSIR, GSHD and GSSD powders were 4.53, 6.71, 6.91 and 5.55% respectively. Hunter’s color value analysis revealed that the L* value of GSIR was lower, and the a* and b* values of GSIR were higher, than those of GSFD, GSHD, and GSSD. The total polyphenol and proanthocyanidin contents of GSFD were significantly higher than those of the other extracts. The flavonoid related substance contents were in the order of GSFD (7.68 g/100g) = GSSD (7.59 g/100g) = GSHD (7.33 g/100g) > GSIR (6.45 g/100g). The electron donating abilities of 500 μg/mL solutions of GSFD, GSIR, GSHD and GSSD were 88.71, 52.62, 65.20, and 65.22%, respectively, while their reducing powers (OD700) were 1.633, 1.097, 1.217 and 1.054 absorbance units, respectively. Additionally, the same trend was observed for the ABTS radical-scavenging abilities of the extracts as that observed for their electron-donating abilities and reducing powers. These results suggest that GSFD is the best method for preparing GS extracts with enhanced antioxidant activities, and that GS extracts may be used as a natural antioxidant material for use in health foods.

        본 논문은 미국 남부지역 특화작물인 머스커다인 포도의 부분별 추출물의 항산화능 및 항균활성을 구명하기 위한 목적으로, Higgins, Jumbo, Noble 3종의 머스커다인 품종을 대상으로 하여 포도의 과피/과육부분과 종자부분을 추출하 여 실험에 사용하였다. 각 추출물에 대하여, 항산화능은 총페놀함량 및 라디칼 소거능(Scavenging activity 및 EC50) 을, 항균활성은 E.coli K12에 대한 최소생육저해농도 (minimum inhibitory concentration, MIC)를 조사하였다. 종 자 추출물의 총페놀함량은 231.24~294.81 mg/mL GAE로 과피/과육 추출물에 비하여 높은 함량을 보였다. 또한 Higgins품종의 종자 추출물이 가장 우수한 라디칼소거능 (EC50=0.026 mg/mL)을 나타내, 종자추출물이 과피/과육추 출물보다 우수한 항산화능을 보유하였다. 반면, E.coli K12 에 대한 항균활성은 Higgins 품종의 과피/과육추출물이 40 mg/mL의 MIC를 보임으로써 가장 우수하였다. 따라서, 본 연구는 머스커다인 포도 추출물이 천연 유래의 항산화 및 항균 기능성을 보유한 소재로 활용될 수 있는 잠재적 가치 를 제시하였다.

        To investigate the anti-diabetic activity of ethanol and aqueous peel and seed extracts obtained from three different grape species (Cambell Early, MBA, and Kyoho), alpha-glucosidase inhibitory activity was examined. All extracts showed anti-diabetic activity, especially aqueous extract exhibited inhibitory effect above 70%. Thus, we used aqueous extract to check the potential hypoglycemic effects in a streptozotocin (STZ)-induced diabetic mice model. The results showed that the blood glucose level of STZ-induced diabetic mice decreased drastically after 3 hr when the aqueous extract of Cambell Early seed was treated to the mice model. The aqueous extract of Kyoho seed lessened blood glucose level by 60%. Together, these data indicate that extracts of grape peel and seed (aqueous or ethanol) may have potential in improving hypoglycemic effects in the diabetic symptoms, suggesting that further investigation on biomarker expressions should be rewarding.

        In order to compare what kinds of transcription factors are associated with the inhibition of preadipocyte cell proliferation, we prepared several grape extracts and tested the expression patterns by reverse transcription-polymerase chain reaction. As a result, 50% ethanol extract of Campbell early seed inhibited adipogenesis derived from the MDI solution. Extract of Campbell early seed was significantly inhibited lipid droplet formation and expression of molecular factors C/EBP-alpha and delta in 3T3-L1 cells. It is suggested that grape extracts of fractions would be a good candidate for the development of regional skin fat modulator.

        포도씨의 총 카테킨 함량과 추출물의 항산화성을 극대화하기 위한 마이크로웨이브 추출조건의 최적화를 시도하였다. 중심합성계획에 따라 추출조건(microwave power , 에탄올 농도 , 추출시간 )을 설계하고, 종속변수로서 추출물의 수율, 카테킨 함량 및 전자공여능을 회귀 분석함으로써 최적 추출조건을 예측하였다. 모든 회귀식의 는 0.9 이상으로 1% 수준에서 유의성이 인정되었다. 총 카테킨 함량의 최대 추출 값은 434.16 mg%로 예측되었으
