        동소동속종인 팥나방(Matsumuraeses phaseoli)과 어리팥나방(M. falcana)(나비목: 잎말이나방과) 사이에 교잡 가능성을 알아보기 위해서 실험실에서 인위적으로 두 종을 교잡시켰다. 두 종의 암수를 교차하여 교미시켰을 때, F1 잡종세대가 발생하였다. 두 잡종세대 집단을 집단 내 및 집단 간 암수를 교차하여 교미시킨 경우들에서, 팥나방 암컷과 어리팥나방 수컷이 교잡되어 생성된 F1 잡종세대 집단(H집단)의 암컷이 같은 집단의 수컷 혹은 다른 집단의 수컷과 교미되었을 때, F2 후대를 거의 생성하지 못했다. 다른 집단으로 만들어진 F2 세대는 집단 내 교미에서 F3 세대를 생성시켰다. F1 잡종세대 집단 암수와 팥나방 혹은 어리팥나방 암수를 각각 교차하여 교미시킨 역교잡 8개 집단 중에서도 H집단의 암컷과 짝지어진 어미세대 수컷 집단 2개는 전혀 산란하지 못했다. 후대가 생성된 다른 6개 집단은 모두 집단 내 암수 교미에서 역교잡 F2 세대를 생성하였다. 이 결과는 팥나방 암컷과 어리팥나방 수컷이 교미하였을 경우 F1 후대잡종을 생성할 수 있으나, 이 F1 후대잡종 암컷은 불임이 되는 것을 나타냈다. 결과적으로 두 종 사이에 접합후 생식단계에서 부분적인 생식격리가 발생할 수 있는 것을 나타냈다.

        A karyotype analysis of five wild rose species (Rosa helenae, R. mulliganii, R. multiflora, R. rubus, and R. solieana) was performed using bicolor fluorescence in situ hybridization (FISH). All five species were found to be diploid (2n = 2x = 14). The chromosome length in the metaphase stage ranged from 1.29 to 2.05 μm in R. helenae, 3.32 to 6.82 μm in R. mulliganii, 1.58 to 2.24 μm in R. multiflora, 2.05 to 3.46 μm in R. rubus, and 1.62 to 2.46 μm in R. solieana. The chromosomes were either metacentric or submetacentric, with no subtelocentric or telocentric chromosomes being observed. Each p air of 5 S and 4 5S rDNA sites was detected at the proximal region of the long arm and the terminal region of the short arm of chromosome #7, respectively.

        Mammals have 3 pairs of major salivary glands i.e., the parotid, submandibular, and sublingual glands. Saliva secretion of these glands is modulated by taste perception. Salivary glands are composed mainly of acinar and ductal cells. Primary saliva is secreted by acinar cells and modified during ductal flow. Recently, of the murine 35 bitter taste receptors, Tas2r108 was expressed at highest levels in the submandibular gland by qPCR. Further, Tas2r108-transfected cells respond to a range of bitter compounds, such as denatonium, quinine, colchicine, diphenidol, caffeine and dapson. The objective of the present study was to characterize the expression of Tas2r108 mRNA in acinar and/or ductal cells of the submandibular gland using in situ hybridization (ISH). Male 42-60 days old DBA2 mice were used in the study. Messenger RNAs were extracted from the submandibular gland for generating digoxigenin (DIG) labeled-cRNA probes. These probes were transcribed in anti-sense and sense orientation using T7 RNA polymerase. Dot blot hybridization was performed using DIG labeled-cRNA probes, in order to estimate integrity and optimal diluting concentration of these probes. Subsequently, ISH was performed on murine submandibular gland to detect Tas2r108 mRNA. Dot blot hybridization data demonstrated that Tas2r108 DIG labeled-cRNA anti-sense probes specifically detected Tas2r108 cDNA. ISH results showed that the anti-sense probes labeled acinar and ductal cells in the submandibular gland, whereas no staining was visible in sense controls. Interestingly, the Tas2r108 expression levels were higher in acinar than ductal cells. These results suggested that Tas2r108 might be more associated with primary saliva secretion than with ductal modification of saliva composition.

        A new Hibiscus cultivar ‘Woolred’ having vigorous growth, uniform plant habit, upright, compact branches, and unique flower shape was developed through interspecific crosses between H. sinosyriacus ‘Seobong’ (♀) and H. syriacus ‘Samchully’ ( ♂ ). Hibiscus breeding program was initiated in 2002 and Hibiscus × ‘Woolred’ was preliminarily selected as ‘R-161’ in 2004 due to its vigorous growth and stable flower quality with rich pink flower. The selected line was further evaluated for different growth characteristics such as leaf shape, leaf size, flowering characteristics, and tested for distinctness, homogeneity, and stability during 2007-2009. The new cultivar ‘Woolred’ had violet pink color flower (RHS N74C) with red eye spot, medium in size having fan petals. The size of flower was 11.5 cm and size of red eye was 3.2 cm. Leaves were 7.7 cm long and 4.2 cm wide. After the plant characteristics evaluation for 3 years (2007 ~ 2009), it was registered as a cultivar ‘Woolred’ (3702, No. of plant variety protection rights) in 2011. This newly developed cultivar ‘Woolred’ having vigorous growth and unique flower shape can be used as specimen plant in landscape.

        This research was conducted in order to understand the hybridization between Quercus aliena Blume and Q. serrata Murray in Korea which show wide range of morphological variations within species and interspecific variations of diverse overlapping characteristics caused by hybridization. Morphological analysis (principal components analysis; PCA) of 116 individuals representing two species and their intermediates were performed. As a result, two species were clearly distinguished in terms of morphology, and intermediate morpho-types assumed to be hybrids between the two species were mostly located in the middle of each parent species in the plot of the principal components analysis. There was a clear distinction between two species in trichome distribution pattern which is an important diagnostic character in taxonomy of genus Quercus, whereas intermediate morpho-types showed intermediate state between two species’ trichome distributions. Forty-two individuals representing two species and their intermediates were examined for leaf flavonoid constituents. Twenty-three flavonoid compounds were isolated and identified: They were glycosylated derivatives of flavonols, kaempferol, quercetin, isorhamnetin and myricetin. The flavonoid constituents of Q. aliena were five glycosylated derivatives: kaempferol 3-O-galactoside, kaempferol 3-O-glucoside, quercetin 3-O-galactoside, quercetin 3-O-glucoside, and Isorhamnetin 3-O-glucoside. The flavonoid constituents of Q. serrata had 20 diverse flavonol compounds including five flavonoid compounds found in Q. aliena. It was found that there is a clear difference in flavonoid constituents of Q. aliena and Q. serrata. Flavonoid chemistry is very useful in recognizing each species and putative hybrids. The flavonoid constituents of intermediates were a mixture of the two species’ constituents and they generally showed similar characteristics to morpho-types. The hybrids between Q. aliena and Q. serrata showed morphologically and chemically diverse characteristics and it is assumed that there are frequent interspecific hybridization and introgression.

        A new Hibiscus cultivar ‘Tohagol Red’ with unique flower shape and color, uniform growth habit and hardiness was developed through interspecific hybridization between Hibiscus sinosyriacus and H. syriacus. A new cultivar 'Tohagol Red' (Hibiscus × ‘Tohagol Red’) developed after crossing between Hibiscus sinosyriacus ‘Seobong’ ( ♀) with pink flower and Hibiscus syriacus ‘Samchully’ with pink flower to improve flower quality and growth habit in 2002. Hibiscus ‘Tohagol Red’ was preliminarily selected as ‘R 202’ in 2004 for its stable flower quality with rich pink flower and ruffled petals. This cultivar had violet purple, ruffled flowers accented by an intense red eye. Large and showy, the overlapping flower petals formed a complete circle and then named ‘Tohagol Red’ in 2010. The tree showed medium growth habit with slightly upright branch. The selected line was further evaluated for different growth characteristics, leaf shape, leaf size, flowering characteristics, and tested for distinctness, homogeneity, and stability during 2004-2009. In The flowers are medium in size (11 ~ 13 cm) with fan (II-type) petal having width 9.2 cm. Leaves are 7.2 cm long and 5.4 cm wide. After the plant characteristics evaluation for 6 years (2004 ~ 2009), it was registered as a cultivar ‘Tohagol Red’ (3803, No. of plant variety protection rights) in 2011. ‘Tohagol Red’ having violet-purple flower and red eye with ruffled petals is an excellent garden plant.

        In this study, we report for the first time a hybridization between Caretta caretta and Chelonia mydas in South Korea. On 7th May 2012, a sea turtle (Jeju sea turtle 01, JST01) was found in the east coast of Jeju Island, South Korea. The morphological characteristics of JST01 were compared with those of C. caretta and C. mydas that were frequently observed in Korean Seas. Interestingly, JST01 showed similar morphological characters such as body color, appearance of carapace, shape of upper beak, serration on marginal scute, and two claws with those of C. caretta. On the other hand, JST01 showed also similar patterns in several characteristics including the numbers of marginal, inframarginal and costal scute, smooth carapace without keel, no contact between costal scute and nuchal scute with those of C. mydas. From these results, we suggest that JST01 is one of hybrid animals bred between C. caretta and C. mydas, because JST01 has speciesspecific characters from both species. We anticipate that our study would be useful and have significant impact onto study inter-specific hybridization in the groups of sea turtle.

        The objective of this study was to develop PCR primers that are specific for Streptococcus sanguinis, Streptococcus parasanguinis, and Streptococcus gordonii. We designed the S. sanguinis-, S. parasanguinis-, and S. gordonii- specific primers, Ssa21-F3/Ssa21-R2, Spa17-F/Spa17-R, and Sgo41-F1/Sgo41-R1 respectively, based on the nucleotide sequences of the Ssa21, Spa17, and Sgo41 DNA probes that were screened using inverted dot blot hybridization (IDBH). The species-specificity of these primers was as- sessed against 43 strains of mitis group streptococci, in- cluding clinical strains of S. sanguinis, S. parasanguinis, and S. gordonii. The resulting PCR data revealed that species-specific amplicons had been obtained from all strains of the target species tested, and that none of these amp- licons occurred in any other strains from other species. These results suggest that the Ssa21-F3/Ssa21-R2, Spa17- F/Spa17-R, and Sgo41-F1/Sgo41-R1 primers may be useful in detecting S. sanguinis, S. parasanguinis, and S. gordonii at the species level, respectively.

        본 연구에서 한국에 자생하는 12종의 민자주방망이버섯과 13종의 프랑스, 스위스, 포루투갈, 일본 등지에서 버섯을 수집하였다. 이들 한국, 외국 품종의 민자주방망이버섯은 단핵균사를 얻기 위해 이들 버섯으로부터 단포자를 얻어 YB배지에 배양하였다. 또한 민자주방망이버섯 육종을 위하여 자실체의 형태적 특성과 단핵균주의 특성 등을 고려하여 교배 단핵균주를 선별하였다. 그 결과 선별된 11개의 교배계통(K1xK2, K1xK3, K1xK4, K1xK6, K1xK5, K2xK4, K2xK3, K2xK6, K3xK4, K4xK5, K4xK6)에서 clamp connection을 관찰하였으며, 또한 RAPD 패턴 분석을 통해 교배 여부를 판단하였다. 교배된 2핵균주를 다시 PDA 배지에 배양하여 교배균주의 양상을 관찰한 결과 선별된 11개의 국내 야생 민자주방망이버섯으로 이루어진 교배계통은 야생품종 2핵 균사 또는 외국 민자주방망이버섯 품종 보다 성장속도가 빠른 것을 확인 하였다. 이는 앞으로 민자주방망이버섯의 한국 고유 품종개발 시 우수 교배형 선택에 있어, 기초자료로서 유용할 것으로 판단된다.

        Evolutionary processes such as recombination and natural selection usually develop over extended periods of time; however, they get accelerated in cases of hybridization. Interspecies hybrids exhibit novel genome which are exposed to natural selection, thus provides a key to unravel the ultimate causes of adaptation and speciation. On the other hand, with rapid upsurge in demand for stronger food security has been confronting us on daily basis. Lately, the scientists have started to gather are now attracted by specific extraordinary traits of different animals by way interspecies hybridization combining the superior genetic material from different animals. Besides, this technology could also explore new life saving vaccines and medicines and also would find beneficial in conserving wild heritage of endangered and critical species. Several researchers have also done commendable work on interspecies hybridization with some degree of success. Nevertheless, lots of obstacles and hurdles still pose for this phenomenal technology to reach to its final destination. This review will point out the hurdles on interspecies hybridization in reference to the natural mating and somatic cell nuclear transfer (SCNT).

        A new cultivar "KNR11-1" of Pleurotus eryngii was developed by the method of mono-mono crossing between monokaryotic strains derived from KNR2598 and KNR2610. The optimum temperature of mycelial growth was 25℃ and that of fruiting body development was 15 - 16℃. The period of harvesting including primordia formation of "KNR11-1" was one day faster than that of control strain KNR2312. The color of pileus and stipe surface was neutral-brown and pure white, respectively. The shape of pileus was dome and has the teeth of a comb. The yield was 90.4 g/850cc plastic bottle. The fruit body has a longer shelf life than control strain KNR2312 at 4℃. It is able to store up to 67.5 days in a cold room. Analysis of the genetic characteristics of the new commercial strain "KNR11-1" showed a different profile as that of the control strain KNR2312 when Random Amplified Polymorphic DNA primers were used. The new cultivar of Pleurotus eryngii is characterized by the improved storability after harvesting.
