Monochamus saltuarius Gebler are the insect vector of the pine wood nematode (PWN), Bursaphelenchus xylophilus (Steiner & Buhrer) mostly in the mid-northern areas of South Korea. In 2017, we installed sentinel logs to lure efficient parasitic wasps of the vector and to evaluate ecological characteristics of parasitoids in prior to the development of biological control strategy for the PWN transmitting insect vector. As a result of the survey, we found Spathius verustus Chao (Hymenoptera: Braconidae) as a predominant parasitoid with the highest parasitism rate among six species of larval parasitoids. The parasitism rate was higher along the edge (0m of the forest depth) of the survey sites than the inner sides (20m and 40m of the forest depth). Also, the parasitoid showed highest parasitism rate in late-May at one of Gapyeong_sites, where was supposed to be the oldest infected areas. Lastly, the parasitism rate along the trap heights was higher at height of 0m than the 1.8m above the ground.

In order to identify the specific antigens for pine wood nematode (PWN), we confirmed that one of the genes commonly found in the transcriptome, proteome and secretory proteins of PWN belonged to the Aldose Reductase (AR) family protein. 36.5 kDa PWN-AR1 was expressed and purified using Baculovirus Expression System. Total 1,546 hybridoma fusion library was generated and screened for specificity to PWN-AR1 by Enzyme Linked Immunosorbent assay (ELISA). Nine clones showed strong immunoreactivity to PWN-AR1 were limited-diluted. Total 864 limited-diluted clones were further screened using PWN-AR1 by ELISA and 34 monoclonal antibody (Mab) clones were selected. 34 Mab clones were further screened using PWN extracts and a standard PWN-infected pine tree extract by ELISA. Finally nine clones were selected and their immunoreactivities to 4 different nematodes were examined by ELISA. Seven clones pecifically recognized PWN while two clones recognized 4 nematodes. Our data suggested that PWN-AR1 is a PWN secretory enzyme while PWN is invading pine trees, Thus, PWN-AR1-Mabs could be used to develop diagnosis tools for PWN and its infected pine trees. Two species in Monochamus (Coleoptera: Cerambycidae) are wellknown vector species of the pine wood nematode (PWN) in South Korea. In 2018, the surveys using sentinel logs with two Monochamus species larvae were made from early June to last September in Pinus densiflora forests of Pohang city, South Korea. The experiments were repeated 10 times in order to investigate parasitism patterns of the larval parasitoids of M. saltuarius and M. alternatus. Three plots were chose at interval distance of 20 m (0 m, 20 m, 40 m) from the edge to the center of the forests (i.e. forest depths) in one damage area of the pine wilt disease for each of three different locations. For each time of the exposure ten sentinel logs were hung along the tree heights (i.e. sentinel logs positions) (height: 0 m, 1.8 m, 3.6 m, 5.4 m, 7.2 m). As the results of the surveys six larval parasitoids in total were identified, and most abundant parasitoids were Spathius verustus and Cynopterus flavator. Analyses showed that the parasitism rate on larvae of two Monochamus species was not affected according to forest depth, but the parasitism rate was the highest in the third plot. Also the parasitism rate was the highest at 5.4 m height of a tree. Lastly, parasitisms rate on M. alternatus was higher than that of M. saltuarius.
