        A new barley(Hordeum vulgare L.) cultivar ‘Dachung’ having high forage yielding and good silage quality was developed at National Institute of Crop Science, RDA in 2015. This cultivar was derived from a cross of the ‘Sunwoo’ and ‘Keunalbori1ho’ in 2002. And it’s promising line showed both high yield and lodging resistance through the preliminary and advanced yield trials(PYT, AYT) at Iksan from 2010 to 2012. It was designated as the ‘Iksan479’. ‘Iksan479’ was conducted to regional yield trials(RYT) in six locations around Korea for three years from 2013 to 2015. And it was released as the name of ‘Dachung’. It has erect plant type, growth habit of IV and green leaf. In the paddy field its heading date was April 24 and maturing date was May 26, same day with ‘Youngyang’. Plant height of ‘Dachung’ was 99cm. Dachung’s spikes per m2 was 625. It has high rate of leaf blades, resistance to BaYMV(Barley Yellow Mosiac Virus) and better winter hardiness than that of ‘Youngyang’. The average dry matter of ‘Dachung’ was about 11.9 ton ha-1 in paddy field. And average feed quality of ‘Dachung’ was 9.0% of crude protein content, 31.3% of ADF (Acid Detergent Fiber), 54.4% of NDF (Neutral Detergent Fiber), 64.0% of TDN (Total Digestible Nutrients). ‘Dachung’ had grade I of silage quality. This cultivar would be suitable for the area above the daily minimum temperature of -8℃ in January in Korean peninsula.

        This study was carried out to investigate the optimal harvesting time, feed value and fermentation quality of barley and wheat for the making of chopped whole crop silage substitute for formula feed. As a result, the moisture content of barley and wheat decreased with a late harvest, and barley progressed faster than wheat. The plant height was similar with harvesting time, and the number of spikes decreased with prolonged period after heading. The dry matter yield and TDN yield of barley harvested at 35 and 40 days after heading were significantly higher than those at 30 days after heading and wheat was significantly higher at 40 and 45 days than at 35 day after heading(P<0.05). Crude protein content of barley and wheat were increased with later harvesting time, and crude fiber, crude fat and crude ash were slightly decreased, but not statistically significant. NDF and ADF content of barley decreased with later harvesting time, and those showed similar level in wheat. TDN content of barely was slightly increased but there was no difference in wheat. Comparing the effects of fermentation on feed value of chopped whole crop silage, the approximate compositions were slightly increased after fermentation, but the difference was not significant. Fermentations resulted in increasing the pH value of barley silage with late harvesting time, but decreasing the lactic acid content(p<0.05). A pH value of wheat silage showed similar level in different harvest time, and lactic acid content was decreased. Considering the quantity and quality of fermentation, barley and wheat can be used for making chopped silage of whole crop silage when they were harvested at 35 days and 40~45 days after heading, respectively.

        This study was conducted to examine the effect of corn (Zea mays L.) - soybean (Glycine max L.) silage prepared by intercropping method on the nutritive value of the silage, in vitro rumen fermentation characteristics, dry matter degradability, as well as milk yield and milk composition of dairy cows. In a couple of experiments intercropped corn-soybean silage (CSBS) was compared with corn silage (CS) and/or Italian ryegrass hay (IRG). Numerically, CSBS had higher crude protein, ether extract, and lactic acid contents compared to CS. In vitro rumen fermentation analysis demonstrated that up to a 24-h incubation period, both CS and CSBS showed higher total gas production, ammonia N concentration, and dry matter degradability compared to IRG (p<0.05). The investigation on animals was conducted in a commercial dairy farm located in Gyeongju, South Korea, employing 42 Holstein cows that were divided into 2 group treatments: CS and CSBS in a completely randomized design. Although no significant difference was observed in milk yield, animals fed on CSBS showed significantly higher milk protein (p<0.05) and milk fat content (p<0.01), compared to animals fed on CS. Taken together, our findings indicate that corn-soybean silage that is cultivated, harvested, and prepared through intercropping can improve the protein content of the silage, and can also enhance in vitro rumen fermentation, dry matter degradability, and performance of dairy cattle.

        The present study analyzes the role of Lactic Acid Bacteria Mixture (LBM) on improving rye silage quality. Rye of four different stages (Booting, Heading, Flowering, and Late flowering) was collected and silage was prepared. The nutrient profile analysis of experimental silage groups showed no significant changes between control and LBM inoculation. Interestingly, the pH of rye silage in LBM treatments showed significant reduction than control (p<0.05) in all stages of rye silage. However, lowest pH (3.69) resulted on booting stage among other stages of rye. Subsequently significant lactic acid production was noted in all stages of LBM inoculation than control. Conversely maximum lactic acid production of (5.33%DM) was noted at booting stage followed by (4.86%DM) in heading stage. Further the lactic acid bacterial (LAB) count in LBM inoculated group showed significant increase than control. Similarly, the silage of booting stage group registered maximum LAB population (63.7 ×106CFU/g) after that heading stage (32.3 ×106CFU/g). Further significant reduction in yeast growth and no fungal growth was noted in all LPM treatment groups. Hence, LBM inoculants could be a better additive for improving rye silage quality.

        This study was conducted to estimate the effect of home or hetero fermentative lactic acid bacteria(LAB) on chemical composition, fermentation quality, and aerobic stability of rye silage. Rye forage was harvested at dough stage(28.9% of dry matter), chopped to 3-5 cm length, and divided into 4 piles for different inoculations as treatment, following 1) No additives(CON); 2) Lactobacillus plantarum at rate of 1.5 x 105 cfu/g of fresh forage(LP); 3) L. buchneri at rate of 1.2 x 105 cfu/g of fresh forage(LB); and 4) Mixture of LP and LB at 1:1 ratio(MIX). Rye silage was ensiled into 20 L bucket silo in quadruplicate for 0, 1, 4, 7, and 100 day periods. After 100 days of ensiling, the silage treated with LB had lower acid detergent fiber content(p<0.05), but higher in vitro dry matter digestibility(p<0.05). The LB and MIX reduced (p<0.05) pH more rapidly than CON and LP across the ensiling days, but had no difference on 100 days. Silage treated LP had lowest(p<0.05) acetic acid, but highest(p<0.05) propionic acid. In contrast, LB treated silage had highest(p<0.05) acetic acid, but lowest(p<0.05) propionic acid with the absence of butyric acid. On microbial count, LP treated silage had lowest(p<0.05) LAB, yeast, and aerobic stability, whereas LB and MIX treated silages had highest(p<0.05). Mold was not detected across all silages. Therefore, it could be concluded that heterofermentative LAB solely or combo with homofermentative LAB might improve in vitro dry matter digestibility, fermentation characteristics, and aerobic stability of rye silage harvested at dough stage.

        본 연구는 팽이버섯 수확 후 배지를 첨가하여 호밀 사일리지 제조 시 팽이버섯 수확 후 배지를 에너 지원으로서 사용하기 위하여 in vitro 반추위 발효실험이 수행되었다. 공시 사일리지는 출수기의 호밀 에 팽이버섯 수확 후 배지 첨가비율(0%, 20%, 40%, 60%)에 따라 제조하여 6주일간 발효시켰다. In vitro 배양액 제조를 위한 반추위액은 농후사료와 볏짚을 40:60의 비율로 급여한 반추위 cannula가 시 술된 Holstein 수소 2두로 부터 채취하였다. In vitro 실험은 발효시간대를 3, 6, 9, 12, 24 및 48시간 으로 설정하고, 각 처리구별로 3반복으로 발효특성과 건물소화율을 측정하였다. In vitro 배양액의 pH 는 발효시간이 길어짐에 따라 낮아지는 경향이었으며, 48시간 경과 시에는 버섯수확 후 배지 60% 첨가 구가 타 처리구에 비해 유의적(p<0.05)으로 낮았다. 미생물 성장율은 배양시간이 경과함에 따라 증가 하는 경향이었으며, 발효 48시간 경과 시에는 버섯수확 후 배지 20% 첨가구가 타 처리구에 비해 유의 적으로(p<0.05) 높았다. Gas발생량은 48시간 발효 시에 대조구가 타 처리구에 비해 유의적(p<0.05)으 로 높았다. 건물소화율은 버섯수확 후 배지의 첨가비율이 높을수록 높았는데, 발효 24시간 및 48시간에 는 R-60구가 처리구 중에서 가장 높았으며(p<0.05), 대조구에서는 전 발효기간 동안 건물소화율이 현 저히 낮은(p<0.05) 상태에 있었다. In vitro 반추위내 발효실험의 결과와 버섯수확 후 배지의 활용성을 고려할 때, 호밀사일리지 제조 시 팽이버섯 수확 후 배지 첨가비율을 원물기준으로 60%수준이 가장 긍 정적인 것으로 나타났다. 향후 대사시험이나 사양시험을 통하여 가축사료로써의 최적 대체 비율을 규명 해야 할 것으로 사료되는 바다.

        Single nucleotide polymorphisms (SNP) markers allow rapid screening of crop varieties in early growth stages. We developed a modified SNP PCR procedure for assaying SNPs in maize. For SNP marker development, we chosen 200 SNP sites from MaizeGDB database, and designed two base pair mismatch primers based on putative SNP site of B73 genome sequence. PCR products size was from 200 to 500 bp or was not shown in the case of SNP site existing in Korean silage corns. Using previously discovered 16 primer sets, we investigated distinctness of 50 silage F1 hybrid corns including 10 Korean silage corns developed by RDA such as Gangdaok, Kwangpyeongok, Dapyeongok, Andaok, Yanganok, Singwangok, Jangdaok, Cheongdaok, Pyeonggangok, and Pyeonganok as well as 40 foreign commercial silage corns. From cluster analysis, we confirmed that 10 Korean silage F1 hybrid corns were clearly distinguished except for Singwangok, P1395, and several foreign commercial corns, and selected minimum SNP primer combination for Gangdaok, Jangdaok, Pyeonggangok, and Pyeonganok. Therefore, development of SNP marker sets might be faster, cheaper, and feasible breed discrimination method through simple PCR and agarose gel electrophoresis.

        This experiment was conducted to comparison study on the productivity for certified varieties of import adaptability of silage corn in Pyeongchang area. Total eight varieties (Gangpyeongok, 31N27, 32P75, 32W86, P3156, P3394, DK 689 및 DK 729) were evaluated. The experimental design was 8 treatment of randomized block with three replications. Corn varieties were cultivated in experimental field of Pyeongchang campus, SNU from 1 May to 2 September, 2015 and plot size was 15㎡. Plant height of 32W86 and ear height of Gwanpyeongok was the highest (p<0.05). Tasseling and silking date was 27 July-3 August and silking occurred after 1-3 day of tasseling. Average day to silking was 92 days and that of 31N27 variety was short (p<0.05). The varieties of DeKalbo Company (DK 689 and DK 729) required more times to silking. Average GDD (growth degree day) of eight varieties was 1,023℃ and P3352 was the lowest GDD. In the trials of resistance evaluation, P3394 was strong in disease, P3156 was the highest in insect. All varieties did not show the lodging and variety of DK 729 showed the highest stay green score (p<0.05). Average dry matter content was 30.77%, it showed higher trend in DM. 32W86 was the highest DM content among the varieties, but there was no significant difference among varieties (p>0.05). The weight per ear was the highest in 32W86 and the lowest in Gwanpyeongok. The ration of ear to whole plant was higher in 32W86 and P3394, but it was not found the significant difference (p>0.05). Average yield of fresh and DM was 59,017 and 13,476 kg/ha, respectively. DK 689 showed higher DM and TDN yield than others, but there was not significant difference (p>0.05). According to results, the difference of productivity was not found among certified variety of import adaptability of silage corn. The varieties Gwanpyeongok, 32W86 and 32P75 would be recommendable in Pyeongchang area for stable cultivation.

        This study was conducted to evaluate the possibility of expanding the usage of whole crop silage from beef cattle and dairy cow to hogs and chickens. For this purpose, a crushing device was developed to crush whole crop silage. The crushed silage was sealed, and analyzed for its feed value. The silage varieties used for the experiment included Saessal barley and Geumgang wheat. Whole crop barley and wheat were crushed in the crushing system as a whole without separating stems, leaves, grains, etc.. When the crushed whole crop silages (CWCS) were analyzed, full grain, grains above 10 mm in size, grains 5~10 mm in size, and grains below 5 mm in size accounted for, 20%, 4%, 27%, and 49 %, respectively. In order to facilitate the fermentation of CWCS, inoculated some fermenter into each CWCS sample (barley or wheat). As control, another set of sample was not inoculated. Crude protein (CP), ether extract (EE), crude fiber (CF), neutral detergent fiber (NDF), acid detergent fiber (ADF), lignin, cellulose content, total digestible nutrient (TDN), and relative feed value (RFV) of fermenter-inoculated Saessal barley were 2.45 %, 1.61%, 8.95%, 16.94%, 9.52%, 1.01%, 8.51%, 81.38%, and 447.5%, respectively. The CP, EE, CF, NDF, ADF, lignin, cellulose content, TDN, and RFV in the other sample of Saessal barley without inoculation of fermenter were 2.57%, 1.62%, 9.61%, 18.25%, 10.13%, 1.10%, 9.04%, 80.90%, and 412.9%, respectively. The CP, EE, CF, NDF, ADF, lignin, cellulose content, TDN, and RFV of fermenter-inoculated Geumgang wheat sample were 2.43%, 1.27%, 10.99%, 19.49%, 11.23%, 1.46%, 9.77%, 80.03%, and 382.6%, respectively. The CP, EE, CF, NDF, ADF, lignin, cellulose content, TDN, RFV of the other set sample of Geumgang wheat sample without the inoculation of fermenter were 2.28%, 1.44%, 10.08%, 18.02%, 10.44%, 1.26%, 9.18%, 80.65%, and 416.9%, respectively. The TDN and RFV content in the fermenter-inoculated Saessal barley were 81.38% and 447.5%, respectively, while the one in the fermenter-inoculated Geumgang wheat were 80.03% and 382.6% respectively. When the feed value of whole crop barley and wheat silage without crushing process was compared to the feed value of whole crop barley and wheat silage made from crushing system, the latter appeared to be higher than the former. This could be due to the process of sealing the crushed silage which might have minimized air content between samples and shortened the golden period of fermentation. In conclusion, these results indicate that a crushing process might be needed to facilitate fermentation and improve the quality of silage when making whole crop silage.

        The objective of this study was to optimize analytical methods for ochratoxin A (OTA) and zearalenone (ZEA) in rice straw silage and winter forage crops using ultra-high performance liquid chromatography (UHPLC). Samples free of mycotoxins were spiked with 50 μg/kg, 250 μg/kg, or 500 μg/kg of OTA and 300 μg/kg, 1500 μg/kg, or 3000 μg/kg of ZEA. OTA and ZEA were extracted by acetonitrile and cleaned-up using an immunoaffinity column. They were then subjected to analysis with UHPLC equipped with a fluorescence detector. The correlation coefficients of calibration curves showed high linearity (R2 ≧ 0.9999 for OTA and R2≧0.9995 for ZEA). The limit of detection and quantification were 0.1 μg/kg and 0.3 μg/kg, respectively, for OTA and 5 μg/kg and 16.7 μg/kg, respectively, for ZEA. The recovery and relative standard deviation (RSD) of OTA were as follows: rice straw = 84.23~95.33%, 2.59~4.77%; Italian ryegrass = 79.02~95%, 0.86~5.83%; barley = 74.93~97%, 0.85~9.19%; rye = 77.99~ 96.67%, 0.33~6.26%. The recovery and RSD of ZEA were: rice straw = 109.6~114.22%, 0.67~7.15%; Italian ryegrass = 98.01~109.44%, 1.65~4.81%; barley = 98~113.53%, 0.25~5.85%; rye = 90.44~108.56%, 2.5~4.66%. They both satisfied the standards of European Commission criteria (EC 401-2006) for quantitative analysis. These results showed that the optimized methods could be used for mycotoxin analysis of forages.

        The study was conducted to evaluate the effects of microbial culture supplements on ruminal fermentation and fermentative quality of Italian ryegrass silage (IRGS) both in vitro and in situ. Three species of microbes (Lactobacillus casei (LC), Bacillus subtilis (BS), and Saccharomyces cerevisiae (SC)) were used in this study. They were applied to IRGS at 30 days after silage manufacture. Various items were measured using in vitro and in situ incubation technique after each microbial supplement was inoculated into IRGS at 0.5×104 CFU/g. In the first experiment, in vitro ruminal fermentation characteristics of IRGS were evaluated at 0, 12, 24, 48, and 72 hours after microbes were inoculated into IRGS. In the second experiment, in situ fermentation characteristics were investigated at 0, 1, 3, and 5 days after the inoculation of each microbial supplement. In vitro ruminal NH3-N content was significantly (p<0.05) increased in LC-, BS-, and SC-IRGS at 12 hrs post incubation compared to that in control IRGS. In vitro ruminal total VFA concentration and dry matter digestibility (DMD) of IRGS were not significantly difference among LC-, BS-, and SC-IRGS, although they were numerically increased in LC-IRGS than those of the other IRGS. In addition, this study evaluated the fermentation characteristics and in situ DMD of IRGS with the lapse of incubation time up to 5 days. Throughout the incubation times from 1 day to 5 days, the pH value was significantly (p<0.05) lower in BS-, LC-, and SC-IRGS than that in control IRGS. Lactate was significantly (p<0.05) higher, and significantly (p<0.05) butyrate was lower in LC-IRGS than that in other treatments at 0 day. It was higher (p<0.05) in control IRGS than that of BS-, LC-, and SC-IRGS at 1-5 days. In situ DMD tended to increase in BS-, LC-, and SC-IRGS compared to that in control IRGS. Especially, DMD was higher in SC-IRGS than that in other treatments at 0 day. It tended to be higher in LC-IRGS at all incubation time. Taken together, these results suggest that it might be useful to select a microorganism by considering the feeding time of IRGS to ruminants because organic acids and DMD of IRGS were affected by the incubation time of each microorganism with IRG silage, especially for L. casei decreased the content of acetate and butyrate in IRGS.

        The present study investigated the effect of enzyme inclusion on silage quality using meta-analysis tool. A total of 16 research papers reporting the effect of enzyme application on silage quality were employed in the meta-analysis of this study. Mixed model for integrating quantitative results from multiple studies was used first to calculate the predicted error of each study. Individual error from the estimated model was the applied into standard deviation of each study to calculate the mean difference. Finally, summary effect was determined using standard mean difference (SMD) and inversed variance weighting. Mixed model analysis and SMD analysis showed the same effect patterns in all analysis items. Enzyme inclusion in silage significantly (p < 0.05) altered all silage quality characteristics investigated compared to control when enzyme was not included. Our results showed that enzyme treatment increased dry matter content, preserved crude protein effectively, and elevated water soluble carbohydrate content. However, the pH value, acetic acid, propionic acid, neutral detergent fiber, and acid detergent fiber contents in silage with enzyme inclusion were lower than those of the control.
