Enhanced UVB Protection and Skin Regeneration by PEG-L-PDRN via A2A Receptor-Mediated Signaling KCI 등재

Polydeoxyribonucleotide(PDRN)는 A2A 아데노신 수용체 신호를 통해 피부 재생을 촉진하지 만, 연어 유래 PDRN(S-PDRN), 식물 유래 미생물 PDRN(L-PDRN, Lactiplantibacillus plantarum에서 유래), 그리고 PEG-변형 PDRN 사이의 기능적 차이는 명확하지 않다. 본 연구에서는 UVB가 가해진 인 간 각질세포와 진피섬유아세포에서 이 세 가지 제형을 비교하였다. 모든 PDRN 제형은 세포 생존율을 향 상시키고, 염증성 사이토카인을 감소시키며, 활성산소를 저하시켰고, 콜라겐 합성을 회복시키며, 상처 치유 를 촉진하였다. PEGylation은 L-PDRN의 분자 안정성을 증가시키고, 세포 내 체류 시간을 연장하며, A2A 수용체 매개 생존 경로와의 결합을 강화하기 위해 적용되었다. 그 결과, PEG-L-PDRN은 모든 제형 중 가 장 강력한 세포 보호 및 재생 효과를 지속적으로 나타냈다. A2A 수용체 길항제인 DMPX는 이러한 효과를 유의하게 감소시켰으며, 이는 관찰된 작용이 A2A 수용체 의존적 신호에 의해 매개됨을 확인시켜준다. 이 러한 결과는, 향상된 안정성과 연장된 수용체 결합에 의해 뒷받침되는 PEG-L-PDRN이 UVB-유도 피부 손상을 보호하기 위한 차세대 PDRN으로서 더 우수한 후보가 될 수 있음을 시사한다.

Polydeoxyribonucleotide (PDRN) promotes skin regeneration through A2A adenosine receptor signaling, yet the functional differences among salmon-derived PDRN (S-PDRN), plant-origin microbial PDRN (L-PDRN, derived from Lactiplantibacillus plantarum), and polyethylene glycol (PEG)-modified PDRN remain unclear. Here, we compared these three formulations in human keratinocytes and dermal fibroblasts exposed to ultraviolet-B (UVB). All PDRN types improved cell viability, reduced inflammatory cytokines, lowered reactive oxygen species (ROS), restored collagen synthesis, and enhanced wound closure. PEGylation was applied to increase the molecular stability of L-PDRN, prolong its cellular retention, and enhance engagement with A2A receptor-mediated survival pathways. Consequently, PEG-L-PDRN consistently demonstrated the strongest cytoprotective and regenerative effects among all formulations. The A2A receptor antagonist DMPX significantly reduced these benefits, confirming that the observed actions were mediated through A2A receptor-dependent signaling. These findings suggest that PEG-L-PDRN, supported by enhanced stability and prolonged receptor engagement, may serve as an improved next-generation PDRN for protecting skin from UVB-induced damage.
