Antioxidant and Anti-inflammatory Activities and Cosmeceutical Potential of a Multi-origin Complex Extract of Helianthus tuberosus, Cistanche deserticola, and Lespedeza cuneata
본 연구는 예루살렘아티초크(Helianthus tuberosus L.), 육종용(Cistanche deserticola), 야관문 (Lespedeza cuneata)으로 구성된 복합추출물의 항산화 및 항염 활성을 평가하고, 이를 함유한 크림 제형의 안정성을 검토함으로써 코스메슈티컬 소재로서의 활용 가능성을 확인하고자 수행되었다. 항산화 활성 평가 결과, ABTS 라디칼 소거능은 모든 처리 농도에서 97% 이상의 높은 활성을 나타냈으며, DPPH 라디칼 소 거능과 SOD 유사 활성은 농도 의존적으로 증가하였다. 또한 elastase 및 collagenase 저해 활성을 통해 피 부 관련 효소 반응에 대한 조절 가능성을 확인하였다. 항염 활성을 평가하기 위하여 lipopolysaccharide (LPS)로 자극한 RAW 264.7 세포에 복합추출물을 처리한 결과, nitric oxide (NO) 생성량과 inducible nitric oxide synthase (iNOS) 및 cyclooxygenase-2 (COX-2)의 발현이 농도 의존적으로 감소하였다. 반 면 세포 생존율은 유의하게 유지되어, 관찰된 항염 효과가 비특이적 세포독성에 기인하지 않음을 시사하였 다. 한편 복합추출물을 함유한 크림 제형은 5, 25 및 45℃에서 28일간 저장한 조건과 0↔45℃ 열순환 조건 에서 pH, 점도 및 외관 변화가 크지 않아 양호한 제형 안정성을 나타냈다. 이상의 결과를 종합할 때, 본 복합추출물은 산화스트레스 및 염증 관련 경로, 특히 NO 생성 및 iNOS, COX-2 발현 조절과 연관된 항 산화 및 항염 활성을 나타냈으며, 다산지 유래 식물자원의 복합화라는 원료 차별성과 함께 제형 안정성까 지 확보되어 항산화·항염 기반 코스메슈티컬 원료로서의 활용 가능성을 제시한다.
This study was conducted to evaluate the antioxidant and anti-inflammatory activities of a complex extract composed of Helianthus tuberosus L., Cistanche deserticola, and Lespedeza cuneata, and to examine the stability of a cream formulation containing the extract for its potential application as a cosmeceutical ingredient. In the antioxidant assays, ABTS radical scavenging activity remained above 97% at all tested concentrations, while DPPH radical scavenging activity and superoxide dismutase (SOD)-like activity increased in a concentration-dependent manner. The extract also showed inhibitory activities against elastase and collagenase, suggesting its regulatory potential toward skin-related enzymatic responses. For the evaluation of anti-inflammatory activity, RAW 264.7 macrophages stimulated with lipopolysaccharide (LPS) were treated with the complex extract. As a result, nitric oxide (NO) production and the expression levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) decreased in a concentration-dependent manner. In contrast, cell viability was maintained, indicating that the observed anti-inflammatory effects were not attributable to nonspecific cytotoxicity. In addition, a cream formulation containing the complex extract showed minimal changes in pH, viscosity, and appearance after storage at 5, 25, and 45℃ for 28 days and under thermal cycling conditions between 0 and 45℃. Taken together, these findings suggest that the complex extract exerts antioxidant and anti-inflammatory effects associated with modulation of oxidative stress- and inflammation-related pathways, particularly the regulation of NO production, iNOS expression, and COX-2 expression. In combination with the differentiated concept of using plant resources from multiple geographical origins and the confirmed formulation stability, the present results support the potential applicability of the extract as a cosmeceutical ingredient with antioxidant and anti-inflammatory properties.
