We conducted long-term field monitoring of hard ticks collected in Andong, South Korea, from 2019 to 2022, in four environments (grassland, mountain trail, graveyard, and copse) with CO2 -baited trap. The collected ticks were identified morphologically and molecularly, and pooled RNA samples were tested for severe fever with thrombocytopenia syndrome virus (SFTSV) (Dabie bandavirus, Phenuiviridae, Bunyavirales). A total of 2,491 ticks were collected, with Haemaphysalis longicornis comprising 88.4% of specimens, followed by H. flava (10.4%), Ixodes nipponensis (0.6%), Haemaphysalis larvae (0.5%), and Ixodes spp. (0.1%). Population peaks of nymphs, adults, and larvae were observed in April, June, and August, respectively, indicating univoltine life cycle of H. longicornis. Among the 287 RNA pools tested, 18 were positive for SFTSV, yielding a minimum infection rate (MIR) of 1.43%. The MIR was highest in October (16.67%) and highest by species in H. flava (10.0%). All viral sequences belonged to genotype B-2. The integration of quantitative species identification with pathogen detection enhances tick surveillance strategies and facilitates early identification of emerging tick-borne diseases in endemic regions.
