This study compared the antioxidant and anti-inflammatory activities of spinach (Spinacia oleracea L.) leaf extract (SOL) and stem extract (SOS) prepared using 70% ethanol and evaluated their potential as cosmetic ingredients. Antioxidant activity was assessed using ABTS⁺ and DPPH radical scavenging assays, together with measurements of total polyphenol and flavonoid contents. SOL exhibited stronger radical scavenging activity and higher polyphenol and flavonoid contents than SOS. Antibacterial activities against Pseudomonas aeruginosa (P. aeruginosa), Staphylococcus aureus (S. aureus), Escherichia coli (E. coli), and Cutibacterium acnes (C. acnes) were evaluated; however, neither SOL nor SOS showed significant antibacterial effects against the tested bacterial strains. The anti-inflammatory activities of SOL and SOS were investigated in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. Both extracts dose-dependently suppressed nitric oxide (NO) production and reduced the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), with SOL showing more pronounced inhibitory effects than SOS. In addition, microscopic observations revealed that SOL alleviated inflammation-induced morphological changes in RAW 264.7 cells. To evaluate cosmetic applicability, an emulsion containing SOL was formulated and subjected to stability testing. The SOL-containing formulation maintained stable pH and viscosity under room temperature and elevated temperature storage conditions without noticeable phase separation or precipitation. Taken together, these findings suggest that SOL possesses potent antioxidant and anti-inflammatory activities and may serve as a promising functional cosmetic ingredient.
