        To identify the key effects of human disturbance on terrestrial ecosystems is a major issue in the contemporary conservation. We compared the community structure of ground beetles among different forest patch sizes according to the different forest types. In addition, we examined how different functional groups and species responded to patch size, and habitat and geographical variables. We sampled ground beetles in 9 continuous forests and 18 patches including 6 deciduous, 6 Korean pine, and 6 Japanese red pine. Ground beetles were collected using 5 pitfall traps in each site, and replaced every month during May to October in 2013. Individual-based rarefaction curves indicated that higher species richness was found in continuous forests than forest patches irrespective of the forest types. Positive relationships were found between forest patch size and species richness of each functional group associated with forest habitat. When all patch size, geographical, and habitat variables were considered simultaneously for multiple regressions, patch size, longitude, latitude, elevation, organic matter, and litter depth were generally selected as significant predictor variables of the abundance and species richness of forest specialists, brachypterous, dimorphic, and large-bodied species, although longitude was only selected as a best predictor for 27 study sites in MRT. In conclusion, decreasing patch size is a major factor to the loss of biodiversity for ground beetles. To reduce biodiversity loss caused by habitat fragmentation, therefore, protecting as large as old-growth forests and improving habitat quality are critical for the biodiversity conservation and enhancement.

        This study was conducted to investigate the community structure and species distribution of ground beetle assemblages in western part of Yeongwol-gun, Korea. Ground beetles were collected by pitfall traps from 9 study sites during July to October in 2013. A total of 30 species were identified from 4,439 collected ground beetles. Two dominant species, Synuchus nitidus (1,916 individuals) and Synuchus cycloderus (1,900 individuals) were occupied 85.8% of total. Interestingly, Pterostichus orientalis orientalis and Synuchus melantho were only collected in relative high altitude above 350 m, such as Beopheung-ri, Unhak-ri, and Yongseok-ri. Large-sized beetles, such as Damaster, Leptocarabus, and Carabus species, were generally more collected from Suju-myeon (Beopheung-ri and Unhak-ri) and Jucheon-myeon (Docheon-ri and Yongseok-ri) as well-conserved area, but not occurred in Yeongwol-eup and Hanbando-myeon, which forests may have lower habitat quality for these species. These differences also lead to increasing of dissimilarity among study sites, which were diverged into 3 groups at 53.0% of similarity. In particular, species composition of Samok-ri was significantly different at 8.2% of similarity because of difference in habitat characteristics as river side. This study has an important value to provide the fundamental information of inventory and monitoring in Yeongwol-gun. For detail mapping of species distribution in Yeongwol-gun, however, further studies are necessary, especially eastern parts of Yeongwol-gun.

        Epigeic spiders (Araneae) were surveyed in apple orchards under different farming practices (organic and conventional) in Muju-gun, Jeollabuk-do, Korea. The study was conducted from April to October in 2013 with 2 weeks interval using pitfall traps. Collected spiders were identified as 20 species of 6 families from 819 individuals in organic orchardⅠ, 19 species of 10 families from 759 individuals in orchardⅡ and 16 species of 8 families from 209 individuals in conventional orchard for from 1,787 captured spiders. Biodiversity was analyzed with species richness, abundance and Shannon diversity index (H΄). Species richness was statistically higher in organic orchard than in conventional orchard. Organic orchardⅡ comprised the greatest abundance followed by organic orchard and conventional. But species diversity was the highest in organic orchardⅡ, intermediate in conventional orchard and the lowest in orchardⅠ. Lycosidae was the most dominant family in all orchards regardless farming practices. However, Linyphiidae dominated in organic orchards only. The most dominant species were Erigone koshiensis (Linyphiidae) in organicⅠ, Pardosa laura (Lycosidae) in organicⅡ and Arctosa pungcheunensis (Lycosidae) in conventional orchard.

        Urbanization is one of the leading causes of habitat loss, habitat degradation, and fragmentation. Urban development negatively affects biodiversity. This study aimed to clarify the change of butterfly communities on effect of urbanization in urban green areas. Butterfly survey was conducted using the line transect methods from April to October in 2012. A total of 59 species and 1,465 individuals of butterflies were observed in four urban green areas: Namsan Park (NS), Ewha Womans University (EW), Bukseoul Dream Forest (BD), and Hongneung Forest (HF), and natural forest: Gwangneung Forest (GF). The category of land use around study site was determined based on GIS data. Species richness and abundance of niche breadth and habitat type in urban green areas differed significantly from those in GF. Estimated species richness and species diversity (H’) in four urban green areas were significantly lower than those in GF. Species richness and abundance of forest interior species and specialist were positively correlated with paddy, field, and forest, whereas those of forest interior species and specialist were negatively correlated with urban area and road. Butterfly communities in four urban green area differed from that in GF. The result suggests that the decrease of paddy, field, and forest associated with increase of urban area and road negatively influences species composition and changes butterfly communities.

        The Diadegma fenestrale is known as parasitoid on potato tuber moth, Phthorimaea operculella and diamondback moth, Plutella xylostella. The Diadegma genus is reported to have symbiotic virus, ichnovirus, D. fenestrale Ichnovirus (DfIV) was identified from this species which is a first report. DfIV showed typical ichnovirus shape with two membranes surrounding the virus capsid. To identify DfIV genes, whole genome sequencing based on GS-FLX was conducted using purified total DfIV genomic DNA extracted from D. fenestrale calyx. About sixty ORFs were analyzed and several typical ichnovirus gene families were detected such as cys-motif, repeat element, vinnexin and vankyrin. Present study was focused on the gene expression patterns in two different lepidopteran hosts.

        The Diadegma fenestrale was known as parasitoid on potato tuber moth, Phthorimaea operculella and diamondback moth, Plutella xylostella. This species, genus Diadegma are first reported from Korea. DfIV showed typical ichnovirus shape which two membranes surround virus capsids. The genome contents of DfIV consist about sixteen double-stranded DNA segments ranging 2 to 6 kb. To identify DfIV genes, whole genome sequencing based on GS-FLX was conducted using purified total viral DNA extracted from D. fenestrale calyx. About sixty ORFs were analyzed and several typical polydna virus gene family detected such as cys-motif, rep, vinnexin and vankyrin. This is the first report of DfIV and these lepidopteran host immune suppression genes will be deeply identified.

        The purpose of this study is to identify which width of the base of support(BOS) is safer and more effective in lifting by comparing muscle activations and body sways when lifting objects under the width variation of the BOS. A total of fifteen healthy adults participated in this study. For the width variation of the BOS, the participants changed the width between their feet into three different types(10cm, 32cm, 45cm) and lifted a 10kg four times in each type after going up on a force plate. In order to measure body sways according to the width variation of the BOS, a motion analysis system was used. In addition, in order to measure the muscle activations of lower extremities, including the erector spinae, gluteus maximus, rectus femoris, and tibialis anterior, an electromyogram(EMG) analysis was employed. In addition, the Borg's scale was drawn by quantifying the subjective discomfort levels felt from each width of the BOS. In conclusion, no statistically significant differences according to the width variation of the BOS were observed(p=.295, .308)(p>.05). However, a statistically significant difference was exhibited between the Borg's scale, which indicates the discomfort levels from lifting performances, and the width variation of the BOS (p=.000*).

        에멀션 기반의 계면활성제를 이용한 주형합성법을 이용하여 산촉매로서 염산과 실리카의 전구체인 테 트라에톡시실란을 사용함으로써 메조다공성 실리카 마이크로스피어를 합성하였다. 테트라에톡시실란의 농도 증가에 의해 구형의 입자 형태가 파괴되었고, 기공구조도 크게 변하였다. 산촉매 농도 증가에 의한 구형의 입자형태 파괴 현상은 적었지만 상대적으로 작은 크기의 구형의 마이크로입자가 더 많이 생성되 었다. 하지만, 산성조건에서 입자들 간의 강한 응집현상이 나타남에 따라 낱개의 분리되어 있는 단일입자 를 얻기 위해서는 초음파 등의 후처리 과정이 필요하였다.

        Eleutherococcus senticosus (Siberian ginseng) is an important medicinal tree found in northeast Asia. In this study, we analyzed the genome-wide distribution of microsatellites in E. senticosus. By sequencing 711 clones from an SSR-enriched genomic DNA library, we obtained 12 polymorphic SSR markers, which also revealed successful amplicons in E. senticosus accessions. Using the developed SSR markers, we estimated genetic diversity and population structure among 131 E. senticosus accessions in Korea and China. The number of alleles ranged from 2 to 11, with an average of 7.4 alleles. The mean values of observed heterozygosity (HO) and expected heterozygosity (HE) were 0.59 and 0.56, respectively. The average polymorphism information content (PIC) was 0.51 in all 131 E. senticosus accessions. E. senticosus accessions in Korea and China showed a close genetic similarity. Significantly low pairwise genetic divergence was observed between the two regions, suggesting a relatively narrow level of genetic basis among E. senticosus accessions. Our results not only provide molecular tools for genetic studies in E. senticosus but are also helpful for conservation and E. senticosus breeding programs.

        The high quality of gene set is necessary to study the functional research of genes. Although perilla is cultivated as an oil crop and as a vegetable crop in Asian countries such as Korea, Japan, northeast China and Nepal, the reference genome is absent. To assembly perilla gene set, we sequenced the various tissues of perilla (Perilla citriodora) RNA-seq with Illumina HiSeq platform, generating 548,549,314 short reads. When de novo transcriptome assembly was performed with five samples, 86,396 and 38,413 transcripts were assembled as total and representative transcripts, respectively. Using 1,917,424 proteins at Phytozome ver. 9.1, we annotated the perilla assembled transcripts, and 66,139(76.55%) and 24,030(62.55%) transcripts showed the similarity with known plant proteins (E-value < 1e-10) as total and representative transcripts, respectively. Among the diverse molecular functions, we were interested in the regulatory components, such as transcription factor and transcription regulator. Using this data, we identified 499 transcripts annotated the putative transcription factor differentially expressed transcripts. 165 putative transcription factors were significantly expressed in perilla flower and 121 putative transcription factors in both leaf and flower. This study provides the perilla reference gene set and the understanding of the molecular regulation of transcription factor dependent on the tissue.

        Recent release of whole genome draft sequences in legume species have led comparative genome studies among legume plants including Glycine max, G. soja, Cajanus cajan and Medicago truncatula. The majority of comparative genomic researches have been conducted based on synteny of coding sequences and coding sequence variations may be one of major forces for speciation and evolution. However, non-coding sequences have been also reported to be important phenotypic regulators. Especially, since short sequence motifs in the promoter regions are highly conserved, they are suggested to be another resources of interests in comparative studies. In this study, we predicted the conserved short sequence motifs by BLASTN algorithm using dicot promoter database from Softberry (http://www.softberry.com). A total of 37,396 conserved short sequence motifs were identified onto 2 kb upstreams of 46,367 high confident gene model of G. max (cv. Williams 82). Meanwhile, whole genome of 7 soybean landraces (G. max) and 7 wild soybean genotypes (G. soja) were sequenced at low depth of less than ten using Illumina Hiseq 2000. Among these genotypes, nucleotide variations were identified in predicted conserved regulatory motifs by mapping of short reads to the reference genome sequence using the Samtools program (http://samtools.sourceforge.net/). Fifteen and two genes, which have SNPs in regulatory motifs and no SNP in coding sequence, were identified by comparisons of inter-species and intra-species, respectively. qRT-PCR experiments are in progress for investigating differences of these 17 genes expressions at transcriptional level.

        As soybean (Glycine max) is known for its high nutritional value of oil and protein, soybean has been domesticated and cultivated by one specific character trait based on human selection. Importantly, tracing back in time where G. max and G. soja, the undomesticated ancestor of G. max have diverged plays an important role in studying of genetic diversity and in investigating the common ancestor of soybean. In this study, we sequenced 6 G. max and 6 G. soja using Illumina’s Hiseq 2000 with a low coverage sequencing technology to estimate the divergence of times between genotypes and populations. A total of the 12 genotypes were sequenced at the average depth of 6.5 and resulted 892.5 Mb and 903.3 MB consensus sequences with the coverage of 91.54% and 92.65% for G. max and G. soja, respectively. The whole genome SNP analysis showed that G. max had lower frequency levels of polymorphism (~0.1%) than G. soja (~0.25%). And, a high number of SNPs located in introns were found among 6 G. soja genotypes as SNPs were approximately twice than those found in 6 G max. The number of SNPs in G. max intronic regions was 53,134, whereas a total of 133,329 SNPs were discovered in G. soja introns. Almost an equal number of SNPs were discovered in 5’ UTR and exon regions; however, different numbers of SNP in CDS and 3′ UTR were identified. By the rate of nonsynonymous change, divergence of time between G. soja and G. max would be investigated.
