        본 연구에서는 TCM-199, mSOF, NCSU-23 세 종류의 배지의 자연 단위 발생 유도로 형성된 돼지 배아의 초기 발육에 대한 효과를 조사하였다. 실험 1에서는 세 종류의 배지에서 체외 성숙된 도축장 유래 돼지 난자의 성숙율을 성숙 48 시간째 핵상 관찰로 조사하였다. 각 배지에서의 핵 성숙율은 mSOF군에서 83.12%, NCSU-23 군에서 78.03%, TCM-199군에서 83.52%로 세 배지의 난자 성숙율에 대한 유의적 차이는 없었다(P

        Recent observations have shown that coronal magnetic fields in the northern (southern) hemisphere tend to have negative (positive) magnetic helicity. There has been controversy as to whether this hemispheric pattern is of surface or sub-surface origin. A number of studies have focused on clarifying the effect of the surface differential rotation on the change of magnetic helicity in the corona. Meanwhile, recent observational studies reported the existence of transient shear flows in active regions that can feed magnetic helicity to the corona at a much higher rate than the differential rotation does. Here we propose that such transient shear flows may be driven by the torque produced by either the axial or radial expansion of the coronal segment of a twisted flux tube that is rooted deeply below the surface. We have derived a simple relation between the coronal expansion parameter and the amount of helicity transferred via shear flows. To demonstrate our proposition, we have inspected Yohkoh soft X-ray images of NOAA 8668 in which strong shear flows were observed. As a result, we found that the expansion of magnetic fields really took place in the corona while transient shear flows were observed in the photosphere, and the amount of magnetic helicity change due to the transient shear flows is quantitatively consistent with the observed expansion of coronal magnetic fields. The transient shear flows hence may be understood as an observable manifestation of the pumping of magnetic helicity out of the interior portions of the field lines driven by the expansion of coronal parts as was originally proposed by Parker (1974).

        We examined the role of SR 144528 (N-[-(1S-endo-1,3,,3-trimethyl-bicycle[2, 2, 1 ] heptan-2-y1]-5-(-4-chloro-3-mothyl-phenyl)-(4-methylbenzyl)-pyrazole-3- carboxamide) in the modulation of certain AC isoforms in transiently transfected COS-7 cells. We found that CB2 in COS cells has a constitutive activity, and thus leading to inhibition of AC-V activity even in the absence of agonist. In addition, this constitutive modulation of AC is reversed by SR144528. It is now well established that several G protein-coupled receptors can signal without agonist stimulation(constitutive receptors). Inverse agonists have been shown to inhibit the activity of such constitutive G protein-coupled receptor signaling. Agonist activation of the Gi/o/-coupled peripheral cannabinoid receptor CB2 normally inhibits adenylyl cyclase type V and stimulates adenylyl cyclase type II. Using transfected COS cells, we show here that application of SR144528, an inverse agonist of CB2, leads to a reverse action (stimulation of adenylyl cyclase V and inhibition of adenylyl cyclase II). This inverse agonism of SR144528 is dependent on the temperature, as well as on the concentration of the cDNA of CB2 transfected. Pertussis toxin blocked the regulation of adenylyl cyclase activity by SR 144528.

        Pregnancy block by male pheromones in mice differs in incidence depending on the combination of strains. Female mice of BALB/cA strain mated with BALB/cA males show a 100% pregnancy block when exposed to males of inbred strain DDK shortly after copulation (Chung et al., Biol Reprod 1997). In the present study, BALB/cA females mated with the males of other strains (CBA/J, C3H/HeN, C57BL/6Cr, and IXBL) showed higher pregnancy rates (66.6-87.5%) even when they were exposed to DDK males. In the pharmacological induction of pregnancy block with dopamine agonist (Bromocriptine, 4mg/kg BW), BALB/cA females mated with BALB/cA males showed a 100% pregnancy block. In contrast, BALB/cA females mated with CBA/J, C3H/HeN, and C57BL/6Cr males showed higher pregnancy rates (40-70%). These results suggest that the better pregnancy rate of BALB/cA females mated with alien males may be due to the stronger viability of F 1 embryos. This interpretation was confirmed by an embryo transfer experiment in which a higher implantation rate was observed when BALB/cA embryos grown in BALB/cA females exposed to BALB/cA males were transferred into recipient BALB/cA females exposed to DDK males. These results suggest that the embryonic genotype or viability of the embryo is one factor contributing to the occurrence of pregnancy block by male pheromones in mice.

        The present study was conducted for the production of transgenic cloned cows by somatic cell nuclear transfer (SCNT) that secrete human prourokinase into milk. To establish an efficient production system for bovine transgenic SCNT embryos, the offset was examined of various conditions of donor cells including cell type, size, and passage number on the developmental competence of transgenic SCNT embryos. An expression plasmid far human prourokinase (pbeta-ProU) was constructed by inserting a bovine beta-casein promoter, a green fluorescent protein (GFP) marker gene, and a human prourokinase target gene into a pcDNA3 plasmid. Three types of bovine somatic cells including two adult cells (cumulus cells and ear fibroblasts) and fetal fibroblasts were prepared and transfected using a lipid-meidated method. In Experiment 1, developmental competence and rates of GFP expression in bovine transgenic SCNT embryos reconstructed with cumulus cells were significantly higher than those from fetal and ear fibroblasts. In Experiment 2, the effect of cellular senescence in early (2 to 4) and late (8 to 12) passages was investigated. No significant differences in the development of transgenic SCNT embryos were observed. In Experient 3, different sizes of GFP-expressing transfected cumulus cells [large (>30 ) or small cell (<30 )] were used for SCNT. A significant improvement in embryo development and GFP expression was observed when small cumulus cells were used for SCNT. Taken together, these results demonstrate that (1) adult somatic cells could serve as donor cells in transgenic SCNT embryo production and cumulus cells with small size at early passage were the optimal cell type, and (2) transgenic SCNT embryos derived from adult somatic cells have embryonic development potential.
