        Recently, consumer's demand for natural preservatives is increasing because of residual toxicity, mutagenicity and etc. of synthetic preservatives and the study of natural preservatives is being done. In this study, antimicrobial activity of spices was investigated Fungi occurred in bread and a noodle without mustard after 3days and 4days, respectively. However, they didn't occur in bread and noodle with mustard ball. Temperature of the water used in mixing-up mustard powder didn't affect antimicrobial activity of mustard. Fungi occurred in bread with cinnamon, Japanese pepper or horseradish after 4days, 4days or 6days, respectively. However, there wasn't the occurrence of fungi in bread with mustard after 8days, yet. Mustard and horseradish of extracts by water of spices had a strong antimicrobial activity. But the extracts by ethanol had low antimicrobial activity.

        V. parahaemolyticus possessed an extracellular alkaline protease activity during the stationary growth phase. Various factors such as initial pH of medium, incubation temperature and shaking rate were investigated for optimizing the production of allcaline protease from V. parahaemolyticus ATCC 17802. Maximal activity of the protease was obtained when the bacteria were grown in 2% skim milk medium in 0.1 M tris/HCl buffer (pH 7.6). Maximal activity of the protease was obtained when the bacteria were grown at initial pH of 7.6, incubation temperature of 37℃ and shaking rate of 250 rpm.

        Nine strains of pathogenic Vibrio sp. of clinical and environmental origin were examined for the degradation of gelatin, casein and hemolysin which is important to the virulence of this bacterium. Culture filtrates of all nine strains of Vibrio exhibited proteolytic activities. Especially, four strains of V. parahaemolyticus and one V. alginolyticus showed strong gelatin-degrading activity. In terms of hemolytic activity, three V. parahaemolyticus and V. mimicus showed strong β-hemolysis whereas those of strains of V. alginolyticus, V. fluuialis, V. uulnificus examined lacked this activity.

        The number of viable microorganism in drinking water was monitored according to storage temperature and storage period. The number of general bacteria in underground water was 10²-10^5CFU/ml and that of coliform group was decreased after 7days. The number of general bacteria in bottle water was increased until 7days but decreased after that day. Coliform group in bottle water were detected only 1 sample. The number of general bacteria in purified water was 10³-10⁴CFU/ml and 10²-10⁴CFU/ml at 8 and 25, respectively. Coliform group were not detected in purified water.

        Klebsiella sp. L-10 의 N-methyl-N`-nitro-N``-nitrosoguanidine 변이주인 Klebsiella sp. L-10 NTG-50으로부터 생산된 고점성의 히알우론산 복합체를 이상계 추출법으로 정제하여 물리화학적 성질을 조사하였다. 정제된 히알우론산 복합체의 점도는 온도와 염농도가 높아짐에 따라 낮아졌고 pH 5.0 이하와 11.0 이상에도 매우 낮았다. 정제된 히알우론산 복합체의 굳기(hardness)는 1.20kg, 응집성(cohesiveness)은 1.92, 점착성(adhesiveness)은 0.62 이었다. 또한 보수력은 g당 6.9ml이었다.

        명태단백질의 이용성을 향상시키기 위한 방법으로 pronase를 이용한 가수분해를 제조하였다. 명태로 부터 고기풀을 제조하여 pronase를 기질 g당 1, 000 units에 해당하는 양을 가하여 pH 7.0, 40℃에서 4시간 가수분해하여 89%의 가수분해도를 보이는 명태단백질의 가수분해물을 제조하였으며 총질소량과 아미노태 질소량은 가수분해가 진행될수록 증가하는 경향을 보였다. 또한 고미와 소수성도 역시 반응초 3.3와 299.3에서 4시간 가수분해시 1.8과 229로 감소하였다. 고미와 밀접한 관계를 가지는 소수성 아미노산(Tyr, Met, Ala, Ilu, Leu, Phe)는 1, 2시간 가수분해시 42.6%와 42.9%로 증가하다가 가수분해가 진행될수록 감소하여 4시간 가수분해시 36.7%로 감소하였다. 정미성 아미노산(Asp, Glu, Pro, Ser, Thr, Gly)은 가수분해가 진행될수록 증가하는 경향을 보여, 4시간 가수 분해시 46.4%의 함량을 보였다.

        The optimum conditions and mechanisms for the plasmid-mediated genetic transformation of intact cells of Bacillus brevis P176-2, an extracellular protein producing bacterium, by electroporation were investigated. It was found that pUB110 plasmid DNA can be introduced into intact bacterial cells by electroporation. The frequency of transformation by this electroporation system depended upon the initial electric field strength, the capacity of the electric discharge capacitor, growth stage, number of successive pulses and composition of electroporation buffer. It was effective for transformation that cells were harvested, washed and resuspended with HSM [7mM HEPES(pH 7.4), 272mM sucrose, 1mM MgCl_2] electroporation buffer when cell growth was attained to 1.2 at OD_660. A maximum frequency of transformation of 2.40×10 exp (4) transformants per ㎍ plasmid DNA was obtained by two succesive pulses with an initial electric field strength of 12.5㎸/㎝ and with a capacitance of 7.3㎌.

        In order to investigate the extraction mechanism of barley tea in cylindrical packed column, pilot plant scale packed column was designed and constructed. And extraction conditions for steady flow in the packed column were searched. The main results of this study are as follows : 1. Circulation of the stock barley tea before the extraction running was indispensable in consideration of inequality, gas, particles existed in packed bed. Solid-liquid equilibrium was initially maintained after the circulation. 2. Flow direction of solvent must be up-flow for maintaining the constant bed height and flow rate during the extraction. Paessure drop in pucked bed was similar to decrease ratio of extract concentration. 3. The porosity of packed bed was in the range of 0.24∼0.36 according to the particle size. And it was decreased as the particle size became smaller.

        A computer program for automatic correlation of marine magnetic profiles is developed and tested. The algorithm involves an iterative stretching the short series selected from the reference profile and cross-correlating with the long series chosen from the other profile. This method gives superior results for correlating near-by profiles and relatively reliable results even for very distant profiles located in different oceans. To increase the effectiveness of the method, it is essential to choose proper window size for the long series and an assumed range of stretching between profiles.

        해상운송 서비스의 특수한 환경적 특성으로 인해 해운기업은 불확실한 경영환경에 노출되기 쉬우며, 환경 불확실성에 대응할 수 있는 적절한 경쟁전략을 활용하여 기업의 경영성과를 달성하는 것은 중요하다. 따라서 본 연구는 해운기업에서 높은 경영성과 달성을 위해 환경 불확실성에 따라 어떠한 전략을 취할 것인지를 분석하는데 목적이 있다. Porter의 본원적 경쟁전략인 원가우위전략과 차별화전략을 바 탕으로 하여 우리나라 외항해운기업을 대상으로 환경 불확실성과 기업의 추구하는 경쟁전략의 관련성을 분석하고, 경쟁전략의 활용정도가 경 영성과에 미치는 영향력에 대해 검증하였다. 연구목적 달성을 위해 선행연구와 전문가 개별 인터뷰를 통하여 측정항목을 설정하였고, 설문을 통하여 실증분석을 수행하였으며, 가설 검증에는 OLS회귀분석과 Tobit모형분석을 이용하였다. 본 연구 결과 첫째, 환경 불확실성의 요인 중 변동성은 원가우위전략과 차별화전략의 활용에 영향을 미치고, 다양성은 차별화전략에 유의한 영향력이 있음을 도출하였다. 둘째, 기업의 경 영성과는 원가우위전략의 활용을 통해서 달성할 수 있으므로, 해운 서비스를 제공하는데 비용절감의 노력이 요구된다.
