        Tulsi (Ocimum sanctum), commonly known as Holy Basil is a revered herb with a rich history in traditional medicine systems, particularly in Southeast Asia. For its medicinal properties, Tulsi has been regarded as an “Elixir of Life” and has been used to treat various ailments. However, the comprehensive investigation of Tulsi extracts and their potential pharmacological benefits, specifically in relation to antioxidant activity remains limited. Hence, the objective of this study was to evaluate the antioxidant activity of Tulsi leaf and stem extract using various screening methods. We investigate the antioxidant activity exhibited by the extract using three different methods involved the utilization of the total polyphenol content assay, the ferric reducing power assay and 2, 2-diphenyl-1-14 picrylhydrazyl (DPPH) assays. The results revealed that the Tulsi leaf extract (TLE) exhibited significantly higher antioxidant activity when compared to the Tulsi stem extract (TSE) in all the performed assays. The higher content of phenolics in TLE may have contributed to its superior antioxidant activity. The HPLC (high performance liquid chromatography) analysis of TLE revealed the presence of eugenol, active compound for several therapeutic properties. These findings provide an understanding of the bioactive compounds present in Tulsi extracts and their potential antioxidant benefits.

        The leaves of Allium victorialis (AV) are known an edible perennial herb, which has been used in Korean traditional medicine. However, the beneficial pharmacological effects of AV extracts (AVE) on the antioxidant activity and atopic dermatitis (AD) have not been thoroughly examined. Therefore, the present study aims to investigate both antioxidant activity and anti-inflammatory effect of AVE on AD in vitro and in vivo. Antioxidant activity was evaluated by total polyphenol content and ferric reducing ability. AVE showed a level of polyphenol content and reducing power activity. The five-week-old BALB/c mice were used as an AD-like mouse model by treating them with 1-chloro-2, 4-dinitrobenzene (DNCB). Topical administration of AVE for 3 weeks to DNCB-treated mice significantly alleviated clinical skin lesion dermatitis severity and epidermal thickness. Histopathological analysis also demonstrated that AVE decreased eosinophil and mast cell infiltration into skin and ear tissue. These results suggest that topical application of AVE inhibits the development of AD-like skin lesion in mice by their antioxidant activity. Thus, AVE may be a potential therapeutic agent for AD.

        본 연구는 닭 진드기 구제를 목적으로 개발된 살비제인 와구잡이 II® (WGJB, 편백정유 : 계피정유 = 20 : 56)에 대하여 토끼와 기니픽을 이용하여 피부 자극성 및 감작성 평가를 각각 수행하였다. 일차피부자극시험에서 토끼의 피부에 WGJB를 24시간 동안 처리한 후 피부 자극성을 확인한 결과, WGJB는 비찰과 부위에서 홍반과 부종과 같은 어떠한 부작용도 일으키지 않았으나, 몇몇 토끼의 찰과 부위에서 매우 약한 홍반과 부종을 나타내어 WGJB의 1차 피부자극 지수는 0.625이었다. 따라서 WGJB는 약한 자극성이 있는 물질로 분류되었다. 피부감작성 시험에서 기니픽에 0.1 mL의 WGJB을 피내주사한 후 24시간 동안 감작시켰다. 감작 1주일 후 WGJB를 함유한 패취를 주사 부위에 부착하여 48시간 동안 처리한 다음, 2주 후에 WGJB 를 함유한 패취를 부착하여 감작을 야기시켰다. WGJB는 어떠한 알러지 반응도 나타내지 않았다. 따라서, 본 연구를 통하여 WGJB는 약한 피부 자극성을 가지며 감작성을 야기하지 않는 물질로 평가되었다.

        Jellyfish envenomation is a world-wide health problem, which often seriously affect the fishery and bathing activities. To date, few individual jellyfish venoms proteins have been thoroughly characterized yet. In this work, four species of scyphozoan jellyfish (Nemopilema nomurai, Cyanea nozakii, Aurellia aurita and Rhopilema esculentum) are compared according to their, cytotoxicity, hemolytic potency, brine shrimp toxicity and protein components. Jellyfish venoms showed higher cytotoxicity in H9C2 heart myoblast than in C2C12 skeletal myoblast, with the exception of C. nozakii venom. This result suggests that the selective cytotoxic effects may be possibly related to their in vivo effects of cardiac tissue dysfunction. On the other hand, hemolytic activity could be also observed from all tested jellyfish venoms. N. nomurai jellyfish venom displaying the greatest hemolytic activity. As an alternative method of evaluating the toxicities of jellyfish venoms, the toxicity on brine shrimp was examined with the four jellyfish venoms. From this, the venom of N. nomurai showed higher toxicity against brine shrimp than the other jellyfish venoms, which is consistent with the results of cytotoxicity assay as well as hemolysis assay of the present study. SDS-PAGE analysis of four jellyfish venoms showed the similar pattern with molecular weight of around 40 kDa, and appeared to be the major protein components. These results provided that N. nomurai jellyfish venom was potently toxic than other scyphozoan jellyfish venoms and may explain to some extent the deleterious effects associated with human envenoming.

        This study was to evaluate the skin sensitization of potassium hydroxide (KOH) by the guinea pig maximization test. In the preliminary test, the induction and challenge dose of KOH were determined as 1.0 and 0.2%, respectively. Guinea pigs were sensitized with intradermal injection of 0.1ml KOH for 24 h. After 1 week, 1.0% of KOH was treated on the site of injection and 0.2% of KOH was challenged 2 weeks later. The average skin reaction evaluated by erythema and edema on the challenge sites and sensitization rate in the KOH-treatment group at 24 h were 0.0 and 0%, respectively, which are substantially low compared with in the 2,4-dinitrochlorobenzene-treatment group (positive control group) (average skin reaction: 3.0, sensitization rate: 100%) and identical with in the distilled water-treatment group (negative control group), representing a weak sensitizing potential. The average skin reaction and sensitization rate observed at 48 h were 0.0 and 0% in the KOH-treatment group, respectively, and 2.0 and 100% in the positive control group, respectively. It was concluded that 1.0% of KOH classified to Grade I, induced no sensitization when tested in guinea pigs.

        Growth factors and cytokines play an important role in delaying skin aging. However, there has been no animal stability test reported for them yet. In the present study, we examined the plausibility of Growth factor and Cytokine Mixtures (GCM) as a cosmetic ingredient by assessing skin irritation and ocular irritation for hypersensitivity using New Zealand white rabbits. Skin safety study was performed to evaluate the potential toxicity of GCM based on the irritation test. For the irritation test, GCM was applied to the rabbit skin, and no adverse reaction, such as erythema and edema, had been observed from the exposed skin sites. In the ocular irritation test, the treatment of GCM did not cause any adverse reaction on rabbit eyes, including cornea, iris, and conjunctiva tissues. From the results, both tests proved that GCM makes no irritable reaction on skin and eye mucous membrane of rabbit. Hence, it may be suggested that GCM can be safely applied as a cosmeceutical ingredient without causing any significant hypersensitivity reaction.

        Despite the various effects on the health of Chamaecyparis obtusa essential oil (COEO), an adverse effect on the skin has recently been reported. Therefore, in this study, the skin sensitization test of COEO was conducted to confirm the skin safety of the essential oil by the guinea pig maximization test. Guinea pigs were sensitized with intradermal injection of 0.1ml COEO for 24 h. After 1 week, 5% of COEO was treated on the site of injection and 2% of the essential oil was challenged 2 weeks later. The average skin reaction evaluated by erythema and edema on the challenge sites and sensitization rate in the COEO-treatment group at 24 h were 0.0 and 0%, respectively, which are substantially low compared with in the 2,4-dinitrochlorobenzene-treatment group (positive control group) (average skin reaction: 3.0, sensitization rate: 100%) and identical with in the olive oil-treatment group (negative control group), representing a weak sensitizing potential. The average skin reaction and sensitization rate observed at 48 h were 0.0 and 0% in the COEO-treatment group, respectively, and 2.0 and 100% in the positive control group, respectively. It was concluded that 5% of COEO classified to Grade I, induced no sensitization when tested in guinea pigs.

        The use of bee venom (Apis mellifera L., BV) occasionally causes side effects such as inflammation and allergic reactions in the recipients. Several case reports also suggested the treatment of BV has some limitations in its clinical uses, due to the occurrence of dermal necrosis and anaphylatic reactions. It is generally understood that bee venom allergy is mainly the result of its allergic component, phospholipase A2 (PLA2). The present study was aimed to generate PLA2-free bee venom (PBV) and evaluate its efficacy as skin care and cosmetic preparation, comparing with original bee venom (BV). Our results showed that both BV and PBV exhibited significant protective effects in UVB-irradiated human keratinocyte (HaCaT) and human dermal fibroblast (HDF) cells and they also induced type I collagen synthesis in UVB-irradiated HDF cells except BV at 3 μg/ml. Furthermore, BV and PBV showed the inhibition of UVB-stimulated matrix metalloproteinase-1 (MMP-1), a major collagen degrading enzyme in skin. However, BV, unlike PBV, exhibited strong cytotoxicities in skin cells (both HaCaT and HDF) at its working concentrations of anti-wrinkle effect. The underlying cell signaling mechanisms of anti-wrinkle effects of BV and PBV were demonstrated by the activation of ERK1/2, and p38. Conclusively, PBV appears to be the bee venom of choice with less cytotoxicity and higher efficacy on UVB-irradiated skin cells in comparison with original bee venom (BV). Therefore, PBV can better be used as a cosmetic ingredient exhibiting excellent anti-wrinkle effect against photoaging than original BV.
