        테트로도톡신(Tetrodotoxin)은 나트륨 통로를 차단하는 신경독소로 가장 오래되고 강력한 해양독소 중 하나이다. 전통적으로 아시아에서 복어 섭취로 인한 테트로도톡신 중독사고가 많이 발생했었다. 최근 아열대 복어 종과 그 잡종 섭취로 인한 테트로도톡신 중독사고가 아시아 외 지역에서도 산발적으로 발생하고 있다. 그러나 복어 종별 테 트로도톡신 및 유사체의 함량과 기관 내 분포에 대해서는 잘 알려져 있지 않다. 이 총설에서는 식품공전 복어독(테 트로도톡신) 분석법인 쥐를 이용한 생물학적 정량법, 효소 면역측정법, 고압 액체 크로마토그래피-형광검출기, 가스 크로마토그래피-질량분석기를 이용한 분석법의 장단점을 비교하였다. 또한 최근 주로 사용되는 액체 크로마토그래피-질량분석기를 이용한 테트로도톡신과 테트로도톡신 유 사체 분석법을 소개하고 이에 사용된 전처리법, 컬럼 및 이동상을 비교하였다. 또한 복어 종별 테트로도톡신과 그 유사체 함량과 다양한 기관에서의 분포를 정리하였다. 이 총설은 테트로도톡신 분석법과 다양한 복어 종의 테트로 도톡신과 유사체 함량 및 기관에서의 분포를 이해하는데 도움을 줄 수 있을 것이다.

        The mesophase pith was synthesized based on effects of a high-pressure thermal condensation and feed (PFO/FCC-DO) blending. The reaction conditions were designed by fluidity and reactivity of each feed during the thermal reaction. The high-pressure thermal condensation process disturbs distillation of the volatile compounds in pyrolysis fuel oil (PFO); the fluidity can be controlled. In the blending process, PFO, which has a high thermal condensation reactivity, and fluid catalytic cracking-decant oil (FCC-DO), which maintains fluidity, interacted with a synergistic effect. Thus, mesophase pitch with a large mesophase content was manufactured by the two above processes.

        알칼라인 수전해 공정에 사용되는 복합 분리막은 고분자와 나노 세라믹 입자로 구성되며 기계적 안정성과 높은 이온 전도성을 가지는 것이 필수적이다. 나노 세라믹 입자는 알칼라인 용액 내 수산화이온(OH-)의 전도성을 높인다고 보고되어 왔으나 세라믹 입자의 비율에 따른 OH-의 관계와 효과의 이해가 미흡한 실정이다. 본 연구에서는 여러 물성 측정(버블포인트, 이온 저항 등)을 통하여 분리막 성능에 입자의 비율이 미치는 영향과 원인을 연구하였다. 입자의 비율이 증가하면 이온 저항은 감소하며, 높은 버블포인트를 유지한다. 이는 분리막 내의 OH-증가로 이온 저항이 낮아지며, 입자의 밀집한 배열이 버블포인트를 유지하는 것으로 판단된다. 따라서 비율의 증가에 따라 OH-상호 작용으로 높은 전도성과 안정성을 가져온다.

        A membrane contactor technique is proposed to selectively remove the SO2 gas from the ship exhaust gas. Membrane contactor system operates with two independent streams of liquid and gaseous phase, with a large contact area between the streams, resulting in high mass transfer coefficient values. In this study, hydrophobic polypropylene substrate was used to greatly minimize the wetting of the membranes. The performance of three absorbents -distilled water, NaOH, and Na2SO3 aqueous solution- was investigated in terms of SO2 removal efficiency within a specific range of different parameters (gas flow rate, absorbent molarities, liquid flow rate, L/G ratio). Finally, optimum operating parameters for a high SO2 removal efficiency were suggested; and the overall mass transfer coefficient and HTU values were examined.

        To remove SO2 from flue gas, a thin film nanocomposite (TFN) hollow fiber membrane was decorated with Nafion/TiO2 nanoparticles. Morphological and structural analyses of the TFN membranes were performed using FTIR, SEM, EDX, TEM, and AFM. The gas permeation experiments were performed with pure gases and a mixed gas within a pressure range of 1-3 bar and feed gas flow rate of 0.03-0.15 L/min. The obtained experimental results suggest that the addition of Nf/TiO2 nanoparticles improved the membrane performance by introducing sulfonate and hydroxyl functional groups to the membrane, and thus increased SO2 permeability and selectivity. The SO2 permeability was found to be 411-1671 GPU, while the ideal selectivities achieved for SO2/N2 and SO2/CO2 were 2928 and 72, respectively. Overall, an SO2 removal efficiency of 93% was achieved by using the Nf/TiO2 incorporated TFN membrane.

        This research considers the effect of added mesophase pitch (MP) as an additive during the pitch synthesis reaction of pyrolyzed fuel oil (PFO). Two effects are generated by adding MP. One is an enhancement of thermal stability due to the high thermal property of the additive; the other is that the volatile compounds that were removed by vaporization of PFO during the thermal reaction can participate in the pitch synthesis reaction (PFO→pitch) more efficiently. The effect differs according to the amount of the additive. When the amount of the additive is less than 7 wt%, the first effect is dominant, whereas the second effect is dominant when the additive amount exceeds 10 wt%.

        A self-powered time-temperature indicator (TTI) was optimized to enhance the performance of the TTI by modifying a biofuel cell using different immobilization, redox mediators, and status of electrode. The performance of the TTI was measured by output voltage of the TTI. The enzymes and combinations of lacasse mediators (HBT (1-hydroxybenzotriazole), Rupy (Bis-(bipyridine)-(5-aminophenanthroline) ruthenium bis (hexafluorophosphate)), MB (Methylene blue), SDP (4,4- sulfonyldiphenol)) and glucose oxidase mediators (FA (Ferroceneboxaldehyde), DBQ(2,5-dihydroxybenzoquinone), HQS (8-hydroxyquinoline-5sulfonic acid hydrate), FHFP (Ferrocenium hexafluorophosphate)) were immobilized with stabilizers (pyrrole) on a glassy carbon electrode by electrodeposition by applying a square wave, cross-linking and physical immobilization. MWCNTs were used to modify glassy carbon electrodes. MWCNTs coated electrodes produced higher output voltage than uncoated electrodes. The optimum and stable performance of the self-powered TTI was that the output voltage of 64 mV and duration time was 3hr at 25°C, when the combination of Rupy, MB for laccase mediators and HQS, FHFP for glucose oxidase mediators were immobilized on MWCNTs coated electrodes by applying a square wave method. In the application, the concentrations of enzyme and glucose were adjusted to prolong the shelf-life of TTI at much lower output voltage.

        The pasteurization is employed for extending shelf-life and keeping the quality of products constant. However the pasteurization undesirably accelerates the oxidation of beer which renders volatile compounds concerning off-flavor. The pasteurization conditions was optimized to avoid off-flavor of beer during pasteurization. Under the isothermal condition, thermal destruction kinetics such as D and Z value of Lactobacillus brevis which is reported the most common beer spoilage, was determined. The binomial data (detected or non-detected of off-flavor) was treated with logistic regression to estimate off-flavor development (OFD) times. The temperature dependence of OFD times was established in terms of Arrhenius relationships. Optimized pasteurization temperature and time were found at which OFD times was not detected. The constraint for optimization was that the pasteurization degree should be larger than 5 decimal reduction time. The optimization was conducted through mathematical simulation using kinetics and temperature-dependent models for microbial death and OFD times. The optimized results were validated by the corresponding experimentations, which met the requirements that the concentration of Lactobacillus brevis was 5-Log reduction and the OFD times not detected.

        A novel nanocomposite LDPE film with UV protective properties was developed for active packaging applications. Initially, undoped and Mn-doped TiO2 nanoparticles (NPs) were synthesized by the sol-gel method and the resulting particles were characterized. Scanning electron microscopy (SEM) and transmission electron microscopy (TEM) images revealed an agglomerated nature and spherical morphology. X-ray diffraction (XRD) studies indicated that all products were crystalline and in the form of rutile. The reflectance spectrum of undoped TiO2 NPs demonstrated a characteristic sharp edge at 410 nm. Subsequently, nanocomposite (NC) LDPE samples were prepared with the NPs by solvent precipitation followed by film casting. The optical and thermal properties of the NC samples were investigated. Incremental increases in Mn concentration from 0.25 mol % to 1.00 mol % were associated with progressive decreases in light transmission in the UV region. The melting and maximum decomposition temperatures of all NCs were 107 and 442-449 °C, respectively. The UV protective LDPE-based NC films exhibited superior photostability. Absorption in the FTIR spectra at 1716 and 1734 cm-1 changed after 4-wk exposure to UV for all film samples as a consequence of photodegradation. Finally, the photooxidation of perilla oil was assessed as an example of a UV protective packaging application. After 12 days, protection with 1.00 mol% Mn-doped TiO2-LDPE was associated with a gradual increase in PV, while protection with TiO2-LDPE was associated with a significant increase and protection with the control treatment was associated with a dramatic increase in PV. Hence, a 1.00 mol% Mn-doped TiO2-LDPE NC showed promise for UV shielding packaging applications.

        Makgeolli is rich in nutrients such as beneficial microbial species, essential amino acids, oligosaccharides, and organic acids. The lactic acid bacteria contained in Makgeolli provides a healthy function as probiotics for customers. The purpose of this study was to enhance the nutritional properties, lactic acid bacteria propagation and alcohol contents of makgeolli by controlling redox potential balance. An automatic control system was created using ORP sensor, Labview control kit, and air supplier, in which air concentration was controlled by on & off mode. Makgeolli was fermented at three different redox potentials -50 mV, -100 mV, -150 mV. Regulating of aeration according to the redox potentials could give prominence to nutritional benefits of Makgeolli. The profiles of redox potential appeared bathtub curves, and related to lactic acid bacteria growth curve and byproducts. We could find the optimum redox potential balance that affects the factors such as LAB’s, alcohol contents and byproducts. In conclusion it was essential to control redox potential balance in order to produce nutrients rich Makgeolli.

        Time-temperature indicators or integrators (TTIs) indicate food quality changes based on time-temperature history. Whilst many types of TTIs have been developed and commercialized, educated consumers often refuse to purchase food products with attached TTI labels showing even a slight color change. In this study, a novel on-off diffusion-based TTI coupled with polydiacetylene/silica nanocomposites has been proposed. The prototype TTI tag has a multilayer structure comprised of a self-adhesive base layer, a middle microporous sheet, and an upper opaque white layer coupled with a square reservoir of Tween 20 attached to an activation stripe. At the end of the diffusion path, polydiacetylene/silica nanocomposites were injected into a loading site as a fine blue stripe. After activation, Tween 20 diffused and reached the loading site, where it rapidly changed from blue-to-red via solvatochromism. This alternative and innovative TTI continuously showed a blue color until reaching the end point, at which stage a red color rapidly appeared, indicating product rejection. Thus, this novel TTI it is of great benefit to the brand owner. The developed prototype was characterized and evaluated for its ability to monitor microbial quality based on published, isothermal, microbial growth data of modified-atmosphere packaged minced beef, Mediterranean fish, and ground pork. The diffusion of Tween 20 in the TTI system was measured under various isothermal conditions and a kinetic model, based on the association between diffusion and time-temperature, was investigated. The Gaussian-estimated activation energy value was 51.082kJ mol-1. Tween 20 diffusion of 6.10, 5.15 and 6.15mm along the TTI systems were considered to be end points and the 95% confidence interval between the times taken for TTI to display OFF and for the foods to reach their deterioration thresholds were 23.30-23.70, 23.00-23.50 and 23.44-24.05h for total aerobic bacteria, Shewanella putrefaciens, and Pseudomonas spp. respectively. The TTI performance test for reproducibility and accuracy revealed a normal frequency distribution with 35004.90, 1200.254.82 and 549.811.09min at 0, 11 and 25C, respectively in accordance with the investigation of diffusion in the TTI.

        We carried out DNA barcoding of five Korean Lymantria species to establish identification references library for quarantine inspection. Total of 118 samples including 34 samples obtained through quarantine inspection, two from USDA, and one collected from Philiphine were used for this study. And 30 sequences of 10 species from GenBank of NCBI were used as reference sequences. In a result of DNA barcoding of the Korean Lymantria species, sequence divergence of 148 DNA barcodes ranged from null to 17.0%, intraspecific divergence from null to 1.0%, and interspecific divergence from 5.1 to 17.0%. In NJ tree, L. dispar contained three clusters, which were identified as L. dispar asiatica, L. albescens, and L. xylina, respectively. L. xylina was collected through quarantine inspection on a foreign merchant ship in Yeosu port, and L. albescens was obtained by pheromone trap on L. dispar installed in Busan port. And L. monacha known as single species in Korea was revealed as species complex with three species, L. monacha, L. minomonis, and L. sugii. In subspecies level, L. dispar dispar (EGM) built single cluster, but L. d. asiatica (AGM) and L. d. japonica showed as multiple cluster. Therefore, DNA barcoding lead to rapid and accurate identification in species level, but in subspecies level, only a taxon showing geographically far distance was discriminated from the others. And the results could provide a taxonomic outline of the Korean Lymantria fauna and might be used as identification reference for Lymantria species in quarantine inspection.

        A taxonomic review on the Korean Lymantria Hübner, 1819 was conducted. In a result, a total of nine species under four subgenera including two new recorded species were detected as followings: L. dispar asiatica Vnukovskij, 1926, L. xylina Swinhoe, 1903, L. monacha (Linnaeus, 1758), L. minomonis Matsumura, 1933, L. sugii Kishida, 1986, L. lucescens (Butler, 1881), L. mathura Moore, 1865, L. fumida Butler, 1877, and L. bantaizana Matsumura, 1933. Of the two unrecorded species, L. minomonis was found only in Is. Bogildo of Jeollanam-do, the southern part of Korea, and the other one, L. sugii was collected in the middle part of Korea. On the two species, L. xylina and L. fumida, the Korean specimens could not be examined through this study. Therefore, we considered that the two species might be excluded from the Korean Lymantria fauna. Each species was identified on the basis of wing pattern and genitalia of male/female adult. We provided diagnosis, male/female adult habitus photos, male genitalia photos, and female ovipositor photos.

        This study aimed at investigating whether a porcine follicular fluid (pFF) supplementation positively affects the characteristics of donor cells and the developmental competence of porcine cloned embryos. Ear fibroblast cells (donor cell) from an Massachusetts General Hospital miniature pig were cultured in different culture methods: (1) Dulbecco's modified Eagle's medium (DMEM)+10% FBS (Control); (2) DMEM+0.5% FBS (SS); and (3) DMEM+10% FBS+10% pFF (pFF) for 72 h. In each conditioned medium, the concentrations of 4 amino acids (Thr, Glu, Pro, and Val) in the pFF group were significantly different from those in the control group (p<0.05 or p<0.01). The proliferation of the cells cultured in the SS group was significantly lower than that of the other treatment groups (p<0.01). The population of apoptotic and necrotic cells in the SS group was significantly higher than that of either the control or the pFF group (p<0.01). The number of embryos that cleaved (p<0.05) and developed into blastocysts (p<0.01) in the SS group was significantly lower than that of either the control or the pFF group. Compared to other groups, the blastocysts produced from the donor cells in the pFF group had higher total cells and lower apoptotic cells (p<0.05). It can be concluded that pFF supplementation in the donor cell culture medium positively affects cell death, cell cycle and quality of the cloned blastocyst.

        The shortage of human organs for transplantation has induced the research on the possibility of using animal as porcine. However, pig to human transplantation as known as xeno-transplantation has major problem as immunorejection. Recently, the solutions of pig to human xenotransplantation are commonly mentioned as having a genetically modification which include alpha 1, 3 galatosyl transferase knockout (GTKO) and immune-suppressing gene transgenic model. Unfortunately, the expression level of transgenic gene is very low activity. Therefore, development of gene overexpression system is the most urgent issue. Also, the tissue specific overexpression system is very important. Because most blood vessels are endothelial cells, establishment of the endothelial-specific promoter is attractive candidates for the introduction of suppressing immunorejection. In this study, we focus the ICAM2 promoter which has endothelial-specific regulatory region. To detect the regulatory region of ICAM2 promoter, we cloned 3.7 kb size mini-pig ICAM2 promoter. We conduct serial deletion of 5' flanking region of mini-pig ICAM2 promoter then selected promoter size as 1 kb, 1.5 kb, 2 kb, 2.5 kb, and 3 kb. To analyze promoter activity, luciferase assay system was conducted among these vectors and compare endothelial activity with epithelial cells. The reporter gene assay revealed that ICAM2 promoter has critical activity in endothelial cells (CPAE) and 1 kb size of ICAM2 promoter activity was significantly increased. Taken together, our studies suggest that mini-pig ICMA2 promoter is endothelial cell specific overexpression promoter and among above all size of promoters, 1 kb size promoter is optimal candidate to overcome the vascular immunorejection in pig to human xenotransplantation.

        Both iron-deficient and zinc-sufficient diets have been known to be associated with a decreased risk of colon cancer. We investigated that effect of dietary zinc on the formation of colonic aberrant crypt foci (ACF) induced by azoxymethane (AOM) followed by dextran sodium sulfate in iron-deficient mice. Five-week old ICR mice were acclimated for 1 week and fed on iron-deficient diet (4.50 ppm iron) with three different zinc levels (0.01, 0.1, and 1.0 ppm) for 12 weeks. The total number of aberrant crypt (AC) and ACF was measured in the colonic mucosa after methylene blue staining. The total ACF numbers of low Zn (LZn), medium Zn (MZn) and high Zn (HZn) diet groups were 10.00 ± 2.67, 8.78 ± 3.12, and 7.96 ± 2.44, respectively and there were no significant differences among the groups. However, the total AC numbers of HZn (27.07 ± 3.88) and MZn (26.39 ± 5.59) diet groups were significantly low compared to LZn (22.57 ± 5.09) diet group (p<0.01). Cytosolic SOD activity was the highest in LZn diet group. But thiobarbituric acid-reactive substances level in liver was also the highest in LZn diet group compared to other groups. There is no difference in cell proliferation in mucous membrane among the groups, while apoptotic positive cells were increased in the HZn diet group. The high zinc diet exhibited decreased β-catenin-stained areas on the mucous membrane of colon compared to the LZn or MZn diet group. These findings indicate that dietary zinc might exert a modulating effect on development of ACF/AC in the mice with iron-deficient status.
