        This study was designed to develop and to qualify a coffee alternative beverage using a mixture of coffee beans and roasted black beans (Rhynchosia nulubilis). Therefore, the total isoflavone content (TIC), total phenol content (TPC), antioxidant activity, anti-inflammatory activity, NFATc1 (Nuclear factor of activated T-cells c1) expression in RANKL (receptor activator of nuclear factor kappa-B ligand)-stimulated RAW264.7 cells and sensory evaluation were measured for 5 different Cb (coffee bean)-RoS (roasted seomoktae) mixture extracts (Cb100RoS0, Cb75RoS25, Cb50RoS50, Cb25RoS75, and Cb0RoS100). Cb0RoS100 had the highest TIC (516.83±36.61 mg/100 g) and TPC (18.11±1.77 mg TAE/100 g) along with the highest antioxidant activity as measured by DPPH radical scavenging activity (73.55±8.11%) and ABTS radical scavenging activity (63.27±7.27%). Also, Cb0RoS100 showed the highest anti-inflammatory activity as measured by NO production (13.57±2.21 μM) and PGE2 production (3.25±0.21 ng/mL). The more the RoS ratio was increased in the mixtures of Cb-RoS, the more the NFATc1 protein expression was decreased in RANKL-stimulated RAW264.7 cells. In case of sensory evaluation, Cb50RoS50 had the highest scores for flavor, delicate flavor and overall quality, which were similar to those in Cb alone (Cb100RoS0). We suggest that the use of RoS replacement instead of Cb in/as a coffee alternative beverage may help to reduce the risk of caffeine-related bone loss and/or bone disease by effectively blocking NFATc1 expression in RANKLstimulated RAW264.7 cells compared with Cb alone.

        This study aimed to analyze the effect of demographic characteristics of consumers and quality recognition factors on purchase intention of organic wine, and to provide preliminary data necessary for the development of wine industry. By factor analysis of collected surveys, quality recognition factors of both the experienced and non-experienced group of organic wine environment/health, varieties of grapes, price, and four brand elements. First, among demographic characteristics, only income level hae a significant influence on the changes in quality recognition causes. Each of quality recognition causes of organic wine had a significant relation with purchase intent. The result of our analysis suggested that ‘environment/health’ was most influential for the experienced group, and ‘health’ was most influential for the non-experienced group. Therefore, this hypothesis is supported.

        In order to perform the protein analysis using the paraffin sections previous fixed with formalin, we applied the ImmunoMemBlot (IMB) method1) to detect the epitopes of target proteins with specific antibodies. In this study the protein extracts were obtained from the paraffin sections of each representative case of ameloblastoma, adenomatoid odontogenic tumor (AOT), and normal gingiva, and more a protein extract from fresh tissue of ameloblastoma was also compared to evaluate the IMB results used with 24 different antibodies. First of all, in the comparison between the paraffin section extract and fresh tissue extract of ameloblastoma, the latter consistently showed more positive IMB reaction than the former. Meanwhile, the paraffin section extract of ameloblastoma was more comparable with that of normal gingival, disclosing that most of proliferating genes, oncogenes, and apoptosis related genes, i.e., PCNA, CDK4, c-erbB2, CEA, p53, Bax, Bad, FLIP, FAS, Bcl-2, p21, N-ras, MMP-2, MMP-9, caspase-3, -8, -9, were highly expressed in ameloblastoma, but EGFR, HGF, and VEGF were similarly expressed both in the ameloblastoma and in normal human gingiva. On the other hand, the comparison between ameloblastoma and AOT both in the immunohistochemistry and IMB using their paraffin section extracts clearly demonstrated that the ameloblastoma showed more expression of proliferating genes and oncogenes while the AOT showed more expression of apoptosis related genes, i.e., Bax, Bad, FLIP, and caspase-9. Taken together, these data suggest that the IMB can be used for the primary screening of quantitative protein analysis using the paraffin section extract, and that the IMB results could be evaluated in conjunction with the immunohistochemical observation.

        Immuno-MemBlot is a technique for detecting, analyzing, and identifying proteins, similar to the Western blot technique but differing in that protein samples are not separated electrophoretically but are spotted through circular or slot templates directly onto the membrane. Recently we developed a new Immuno-MemBlot (IMB) method applying immunoreactions and coloring procedures directly in the wells of MemBlot apparatus, which were connected by canals to perform drainage for reagent application and buffer irrigation. This IMB method was designed to get theimmunoblot results more rapidly and clearly than the previous immunoblot ones. This study is aimed to evaluate the analytical accuracy of IMB using different biological assay. In the sensitivity test of IMB the monoclonal antibody can clearly detect the 30 ng (about 12 pM) of Mucocidin peptide (35 mer), and is also available to detect at least 10 ng (about 4 pM) of Mucocidin peptide (35 mer). The IMB was effective in the quantitative analysis of methothrexate (MTX) assay for cellular apoptosis. And more, this IMB is useful to screen large number of specific samples with ease and accuracy in a short time. In the screenings for the presence of Mucocidin in saliva the quantitative comparison is conspicuous among 48 persons depend on the different conditions ofgender, drinking and smoking habits, and oral diseases. Therefore, it is presumed that, even though the target proteins were partly degraded, a specific epitope can be detected if a monoclonal antibody was still reactive. Conclusively, these data suggest that the IMB can be useful in the primary qualitativeand quantitative analysis of proteins in various fluids, i.e., blood, saliva, tear, urine, etc.

        Background : It is well known that Alzheimer`s disease (AD) is associated with neuronal loss and accumulation of extracellular senile plaque, whose major constituent is β-amyloid peptide (Aβ). In cell cultures, Aβ can directly stimulate neuronal cell death and make neurons susceptible to excitotoxicity which may include glutamate release and N-methyl-D-aspartate (NMDA) receptor activation. There are numerous reports in the literature of Cedrela sinensis (CS) for pro-apoptotic effects. It was hypothesized that CS might protect neurons against neurodegeneration in AD due to its pro-apoptotic effects. The current study aimed to determine the protective effect of ethanol extract from the leave of CS on Aβ (25-35)-induced neuronal cell death in primary cultured rat cortical neurons. Methods and Results : Cerebral neurons were collected from embryonic day 15 SD rat fetuses and were cultured on DMEM with serum. Neurotoxicity experiments were proceeded on cultured neurons after 4-5 days in vitro. Cultured neurons were treated with 10 μM Aβ (25-35) for 24 h or 1 mM NMDA for 20 h to induce neuronal death. CS was applied 20 min before the treatment with Aβ (25-35) or NMDA and also present in the medium during the incubations. Colorimetric MTT assay and Hoechst 33342 staining were used to estimate viability of neurons. Western blot analysis was carried out to examine the expression levels of anti-apoptotic and pro-apoptotic proteins. CS (5 and 10 ㎍/㎖) significantly inhibited Aβ (25-35)-induced apoptotic neuronal cell death in cultured cortical neurons. CS also inhibited Aβ (25-35)-induced change of apoptosis-related protein expression in western blot analysis. Furthermore CS (5 and 10 ㎍/㎖) reuduced NMDA-induced neuronal cell death. This study demonstrated that NMDA glutamate receptor activation is related with Aβ (25-35)-induced neuronal apoptotic death. Conclusion : CS protected culterd neurons against Aβ (25-35)-induced neurotoxicity probably via inhibition of NMDA receptor activation. These results suggest that CS can prevent the progression of neurodegenerative disease such as Alzheimer's disease.

        Background : This study was carried out to survey the effect of IBA (indole-3-butric acid) concentration on root development characteristics in Viburnum dilatatum cuttings. Methods and Results : The cuttings were collected on July 28, 2016 from plants growing in Korea National Arboretum. Cuttings were treated with different IBA concentrations [0 (control), 10, 100, 1000, 2000 ㎎⋅L-¹] for 10 seconds. Rooting parameters (root number, the longest root length, root fresh weight, root dry weight, rooting percentage, and survival rate) were observed at 8 weeks after cuttings. Rooting percentage was 43, 33, 17, 47, and 40 % at 0, 10, 100, 1000, and 2000 ㎎⋅L-¹ of IBA, respectively. Root number (13.4) was highest under 1000 ㎎⋅L-¹ IBA. Root length (9.4 ㎝) and Root weight (fresh and dry) were highest under 1000 ㎎⋅L-¹ IBA. Conclusion : According to the results, 1000 ㎎⋅L-¹ IBA was the best treatment for increasing percentage rooting percentage and other root development characteristics of V. dilatatum cuttings.

        Hopyeongbyeo is an anther culture derived Japonica rice (Oriza sativa L.) cultivar from the cross between Hitomebore and Hwajinbyeo by the rice breeding team of Honam Agricultural Research Institute (HARI), NICS, RDA, in 2003. This cultivar has a about 126 days growth duration from transplanting to harvesting. The short and round grain shape of Hopyeongbyeo is translucent with non-glutinous endosperm. Amylose content of rice grain is about 17.8% and high palatability of cooked rice compared with Nampyeongbyeo. This cultivar shows resistant to bacterial blight pathogenes of K1. The milled rice yield of Hopyeongbyeo is about 5.13MT/ha at ordinary transplanting culture in the local adaptability test. Hopyeongbyeo would be adaptable for the southern plain area of Korea.
