        Myrosinases (thioglucosidases) catalyze the hydrolysis of a class of compounds called glucosinolates, of which the aglycones show various biological functions. It is often necessary to minimize the loss of myrosinase activity during thermal processing of cruciferous vegetables. Myrosinase was isolated from a popular spice, white mustard (Sinapis alba), and its thermal inactivation kinetics was investigated. The enzyme was extracted from white mustard seeds and purified by a sequential processes of ammonium sulfate fractionation, Concanavalin A-Sepharose column chromatography, and gel permeation chromatography. At least three isozymes were revealed by Concanavalin A-Sepharose column chromatography. The purity of the major myrosinase was examined by native polyacrylamide gel electrophoresis and on-gel activity staining with methyl red. The molecular weight of the major enzyme was estimated to be 171 kDa. When the consecutive step model was used for the thermal inactivation of the major myrosinase, its inactivation energy was 44.388 kJ/mol for the early stage of destruction and 32.019 kJ/mol for the late stage of destruction. When the distinct two enzymes model was used, the inactivation energy was 77.772 kJ/mol for the labile enzyme and 95.145 kJ/mol for the stable enzyme. The thermal inactivation energies lie within energy range causing nutrient destruction on heating.

        To identify and compare the venom components and expression patterns of some bees/wasps, venom gland-specific transcriptome analyses were conducted for 14 Aculeate bees/wasps. Most of the allergens and pain-producing factors showed extremely high expression levels in social wasps, implying that social wasps have evolved to use venom to defend the colony against intruders. Acid phosphatase and tachykinin, which are known as allergens and neurotoxic peptides, were found with high frequencies in the venom glands of solitary wasps. This suggests that solitary wasps might use their venom for catching and preserving prey. In the venom glands of bumblebees, little or no transcripts of major allergens or pain producing factors were identified, implying that bumblebees venoms are relatively less toxic than those of social or solitary wasps. Taken together, the differential expression patterns of venom genes in some Aculeate bees/wasps implies that bees/wasps have unique groups of highly expressed venom components, which appear to have evolved in response to both ecological and behavioral influences.

        To identify the venom components and their expression patterns of some Aculeata bees/wasps, venom gland-specific transcriptome analysis was conducted. FPKM values were normalized with the average of the transcription level of reference gene (a-tubulin). Common components in both solitary and social wasp venoms include hyaluronidase, phospholipase A2, metalloendopeptidase, etc. Although it has been expected that more diverse bioactive components with the functions of prey inactivation and physiology manipulation are present in solitary wasps, the information on venom compositions of solitary wasps obtained in this study was not sufficient to generalizae this notion. Nevertheless, some neurotoxic peptides (e.g., pompilidotoxin and dendrotoxin-like peptide) and proteins (e.g., insuline-like peptide binding protein) appear to be specific to solitary wasp venom. In contrast, several proteins, such as venom allergen 5 protein, venom acid phosphatase, and various phospholipases, appear to be relatively more abundant in social wasp venom. In the venom gland trancsriptome of bumblebees, major allergens or pain producing factors were barely identified, implying that bumblebee venoms are relatively less toxic than those of social or solitary wasps.

        Insect growth regulators (IGRs) are insecticides that disrupt the normal development of target insects. Among the IGR insecticides, juvenile hormone (JH)-related IGRs are of particular interest because they stimulate or interfere with the formation of JH receptor complex. In the precious studies, novel JH-related IGRs with JH agonist (JHA) and antagonist (JHAN) activity were identified by using yeast two-hybrid system transformed with the Aedes aegypti JH receptor complex. In this study, the transcriptomic responses of Aedes albopictus were investigated upon JHA and JHAN, respectively. These results will provide important information about understanding of impact of JH-related IGRs in transcription level.

        Insect growth regulators (IGRs) are insecticides that disrupt the normal development of target insects by inducing symptoms such as premature molting or supernumerary larval stages. Juvenile hormone (JH)-based IGRs are of particular interest because they simulate or interfere with the formation of juvenile hormone receptor complex. Insect larvae generally have been poorly susceptible to JH agonists (JHAs) because endogenous JH titer is already high in the larval stage. In contrast, JH antagonists (JHANs) might be more effective for control of target pests in larval stages through interference in the status quo action of JHs. In this study, we tested 2,354 chemical compounds using the yeast-two hybrid system to identify JHANs. Among them, K265674 sample was selected and its insecticidal activity was evaluated against Aedes albopictus larvae. These results may be useful for development of novel IGR insecticide.

        Bacillus thuringiensis (Bt) produces a variety of insecticidal crystal proteins and widely used as one of the most successful biological control agents. Recently, studies that introduce cry genes into crops to create pest resistance have made much progress, and the total area of land planted with Bt crops has increased substantially. In this study, pest resistance of 8 transgenic Bt rice events with a synthetic cry1Ac gene linked to rice rbcS-tp sequence were assessed under laboratory conditions. Bioassays were performed against Cnaphalocrocis medinalis, which is a significant pest of rice in Asia. C. medinalis larvae were shown to be susceptible to all eight events, even though there were differences between the causes of death. The results differed between developmental stages of the larvae, despite the fact that all 8 events led to high mortalities. These results may be a significant foundation for the evaluation of improved transgenic Bt rice.

IGRs are categorized into three types based on their mode of action. One of the them is classified as juvenile hormone agonists (JHAs). In recent studies, we identified juvenile hormone antagonists (JHANs) from plant extracts. In order to identify a novel juvenile hormone agonists and antagonists, we tested 2,354 chemical compounds using the yeast-two hybrid system. Among them, 2 JHAs and 4 JHANs were selected and their insecticidal activities were evaluated against Aedes albopictus larvae. These results may be useful for development of novel IGR insecticides.

        Bombolitin is a venom peptide originally isolated from bumblebees and possesses various biological activities, including hemolytic activity. Bombolitins exhibit amphipathic α-helical structure in lipid-membrane-mimicking environments. To investigate their pharmacological and toxicological properties, anti-tumor, anti-microbial and cytotoxic activities of bombolitins from Bombus ardens and Bombus ussurensis were evaluated. Bombolitins of the two species exhibited extremely high anti-tumor activity against ovarian tumor cells SK-OV-3 and NIH; OVCAR-3 at 25-50 μM, which is 2-fold more potent than other wasp venom peptides studied to date (Yoon et al., 2015; Yoon et al., 2016). The two bombolitins showed significantly high antimicrobial activity. However, bombolotin of B. ussurensis showed no antimicrobial activity against Escherichia coli. In addition to their high levels of anti-tumor activity, bombolitins showed considerable levels of hemolytic activity. Thus, to utilize bombolitins as a potential candidate for anti-tumor peptide drugs, further studies for reducing cytotoxic properties of bombolitns is essential.

        To obtain information on the sanitary of indoor environment in greenhouses used for shiitake cultivation, bacteria associated with larvae and imagoes of Sciaridae flies, pest to shiitake mushroom, were isolated and identified. A total of 1,048 bacterial colonies were isolated from the flies’ larvae and 984 bacterial colonies were isolated from the flies’ imagoes. Based on molecular analysis of the 16S rDNA sequence, Achromobacter xylosoxidans and Tetrathiobacter kashmirensis of β-proteobacteria, Enterobacter asburiae and Raoultella ornithinolytica of γ -proteobacteria, Curtobacterium sp. and Microbacterium thalassium of Actinobacteria, and Penibacillus taichungensis of Firmicutes were identified from the colonies of the flies’ larvae. While, Bacillus megaterium, B. thuringiensis, Lysinbacillus sphaericus and L. fusifomis of Firmicutes, Microbacterium thalassium and Citricoccus parietis of Actinobacteria, and Enterobacter asburiae of γ-proteobacteria were identified from the flies’ imagoes. Some of the isolated bacterial species were known be human pathogens. Overall, the results of this study suggested that mushroom fly carrying human pathogenic bacteria is one of sources impact on the sanitary of indoor environment of greenhouses used for shiitake cultivation.

        Pronuclear DNA microinjection has been the most universal method in transgenic animal production but its success rate of transgenesis in mammals are extremely low. To address this long-standing problem, we used retrovirus- and lentivirus-based vectors carrying the enhanced green fluorescent protein (EGFP) gene under the control of ubiquitously active cytomegalovirus (CMV) promoter to deliver transgenes to bovine embryos. The rate of transgenesis was evaluated by counting EGFP positive blastocysts after injection of concentrated virus stock into the perivitelline space of the bovine oocytes in metaphase II. Among two different types of lentivirus vectors derived from FIV (feline immunodeficiency virus) and HIV (human immunodeficiency virus), the former scored the higher gene transfer efficiency; almost 100% of the blastocysts developed from the oocytes infected with FIV-based vector were EGFP positive. As for the vectors derived Com HIV lentivirus, the transgenesis rate of the blastocysts was reduced to 39%.

        Mitis-salivarius sucrose bacitracin(MSB) medium is widely used in the selective isolation of mutans streptococci(MS), a designation for a group of oral cariogenic species. Recently, we have isolated three bacterial strains grown on MSB agar from human dental plaques. The three strains exhibited biochemical characteristics similar to those of the biotype IV of MS, with the exception that they manifested a positive reaction for arginine deaminase. The objective of this study was to identify and characterize these three clinical isolates. The bacteria were identified with biochemical tests as well as by 16S rDNA cloning and sequencing. In order to compare the antibiotics susceptibility of the clinical isolates with that of type strain, the minimum inhibitory concentrations of 9 antibiotics were determined using broth dilution assays. The results identified all of our three clinical isolates as Enterococcus faecalis. All E. faecalis strains were found to be susceptible to penicillin G, amoxicillin, augmentin, and vancomycin, but were resistant to ciprofloxacin, cefuroxim axetil, and clindamycin. Our findings indicate that E. faecalis is capable of growing on MSB agar, and suggest that the MSB medium be improved so that only MS should be recoverable on the medium, as originally devised for their selection.

        Background : Recently, there is a urgent demand for development of new varieties with enhanced resistance to various biotic/abiotic stresses. In order to develop ginseng varieties with such traits, systematic breeding program and comprehensive field studies are prerequisite. Methods and Results : ‘Cheonmyeong' seeds were collected in 2008 from the farmer field of Buyeo. Physiological investigation and propagation were conducted from 2009 to 2011. It was given the name 'Eumseong No. 8 through the observed yield trial from 2012 to 2013 and local adaptability was carried out from 2014 to 2015. All phenotypes including agronomic characteristics, seed yield, and physiological response to biotic/abiotic stresses were investigated according to the ginseng GAP and UPOV guidelines. Yellow-red leaf and pink berry at maturing stage were observed. The time of emergence, flowering and berry maturity of the ‘Cheonmyeong’ were faster than those of ‘Chunpoong’. Stem length of ‘Cheonmyeong’ was shorter than that of ‘Chunpoong’, whereas stem diameter was thicker than that of ‘Chunpoong’. Main root length was shorter but main root diameter is thicker than that of ‘Chunpoong’. Number of seeds of ‘Cheonmyeong’ was fewer than that of ‘Chunpoong’ but 1,000-seeds weight and stratification rate were higher than those of ‘Chunpoong’. The yield performance of this variety was 661 kg/10 a in local adaptability test for two years, which is 22% higher than that of ‘Chunpoong’. ‘Cheonmyeong’ showed strong resistance to phytophthora blight, mulberry mealybug and nematode and moderate resistance to alternaria blight. ‘Cheonmyeong’ did almost not occur yellow spot of aerial part and rusty skin of root, show moderate resistance at high temperatures. Conclusion : Our study demonstrated that ‘Cheonmyeong’ is an ideal variety with heavier root weight and enhanced stress resistance and contribute will enhance biotic/abiotic stress resistance and increase the farmers' income.
