        The purpose of this study was to investigate the antioxidant activity and β-glucan content of extracts extracted by varying the temperature at 30, 55 and 80℃ after hot air drying or freeze drying of Pleurotus ostreatus and Pleurotus eryngii. For the analysis antioxidant activity of each mushroom, β-glucan, total phenol, flavonoid contents, and DPPH․ABTS+․Nitrite assay were measured. Also, the β-glucan content, total flavonoid content and ABTS+ scavenging activity increased with freeze drying rather than hot air drying, and increased with increasing extraction temperature in both mushrooms. However, the total phenol and nitrite scavenging activity increased with hot air drying rather than freeze drying, and decreased with increasing temperature in both mushrooms. DPPH scavenging activity was not significant in both mushrooms, but decreased with increasing extraction temperature. Pearson’s correlations between total flavonoid content and antioxidant activities were r=0.719~0.753 (p<0.01). As a result, the β-glucan content, total flavonoid content, and ABTS radical cation scavenging activity were highest during freeze drying and extraction at 80℃. And the highest total phenol content, DPPH radical scavenging activity and nitrite scavenging activity were obtained during hot air drying and extraction at 30℃.

        This research investigated the quality properties of yogurts added to barley (1, 2, and 3%) and tagatose or sucrose. After inoculation of S. thermophilus, pH, titratable acidity (TA), viscosity, and viable cell count were measured at 4 h intervals for 16 h and color value, antioxidant activity, and sensory evaluation were determined after fermentation in triplicate. In the case of yogurt containing only tagatose or sucrose, the pH was hardly decreased, but when 2-3% barley was added, the pH reached below 4.5 before 16 h. In addition, TA and viscosity tended to increase as barely was added. Viable cell count increased slowly in yogurt with tagatose, but rapidly increased when barley was added. As barley content increased, yellowness, redness, and antioxidant activity tended to increase, and significantly higher antioxidant activity in barley tagatose yogurt. Sour strength increased and significant difference in the sweetness was not observed when barley amount increased. Although there was no significant difference in overall acceptability, tagatose yogurt containing 3% barley scored the highest in preference. In conclusion, the fermentation rate of yogurt inhibited by the addition of tagatose is improved by the addition of barley, and the growth rate of lactic acid bacteria is considered to play a role.

        In this study, sulgidduk was prepared by addition of varying amount of black doraji extract and the physicochemical quality characteristics and antioxidant activities were investigated in order to explore the commercialization potential of sulgidduk supplemented with black doraji extract. The moisture content of sulgidduck supplemented with black doraji extract exhibited a significant decrease compared to the control. The pH of sulgidduck tended to decrease with increasing concentration of black doraji extract, and the reducing sugar content increased significantly from 0.33% to 3.51%. With an increase in the content of black doraji extract, a decrease in lightness (L*) and increase in redness (a*) was observed. A significant increase in the total polyphenol content and antioxidant activity (DPPH and ABTS radical scavenging activity) was observed with the addition of the black doraji extract. Consequently, it is proposed that black doraji extract can be used for recipe development and commercialization of various rice cakes.

        The purpose of this study was to determine the content of polyphenols and flavonoids, vitamin C, and antioxidant activity for the extract from the Deodeok sprout. To accomplish this, the Deodeok sprout whole (CLS-W), above ground part (leaf, stem, CLS-L), and root (CLS-R) were individually extracted using 70% ethanol. The highest levels of total polyphenols and total flavonoids were observed in the Deodeok sprout extract CLS-L2. Similarly, antioxidant activities resulting in radical scavenging activities increased significantly in the extract of CLS-L2. In conclusion, these results indicate that Deodeok sprouts can be used as a viable, new natural antioxidant source.

        The purpose of this study was to investigate the antioxidant activity and tyrosinase inhibitory activity of Codonopsis lanceolata 50% ethanol extract, and its solvent fractions (n-hexane, ethyl acetate (EA), n-butanol, water). The main components of the EA fraction were qualitatively analyzed using UPLC Q-ToF/MS. Additionally, a quantitative analysis was performed using UPLC. As a result, the total polyphenol content was 113.36 mg gallic acid/g in the EA fraction, which contained the largest amount of the C. lancolata solvent fractions. Also EA showed the highest antioxidant activity than other fractions. The IC50 of DPPH(2,2-diphenyl-1- picrylhydrazyl) radical scavenging activity was 0.03 mg/mL and the IC50 of ABTS [2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonate)] radical scavenging activity was 0.049 mg/mL. The EA fraction showed tyrosinase inhibitory activity than other fractions and especially inhibited monophenolase oxidase reaction higher than diphenolase oxidase reaction. The monophenolase oxidase inhibited 55% when the concentration of the EA fraction was 0.25 mg/mL. As a result of Q-ToF/MS analysis, it was confirmed that tangshenoside I and lobetyolin were the main components of EA fraction. Thus, these results suggest that C. lanceolata may be used as a potent source of cosmetic agents.

        The purpose of this study was to investigate the quality characteristics and antioxidant activity by collecting four times on six samples of Jeonbuk regional soybean pastes (Doenjang) traditionally prepared for this study. The water content of samples decreased according to progress of the aging period and the water content of the fourth sample collected was 42.40~59.64% (p<0.05). The salinity of the fourth Doenjang samples was 11.80~18.60%. The amino-type nitrogen content was 122.67~540.33 mg% immediately after the preparation of Doenjang in the Jeonbuk region and the content of the fourth collection samples increased from 251.49 to 982.36 mg% (p<0.05). The isoflavone glycosides decreased but daidzein, genistein and glycitein, which are aglycones, increased during aging periods. The total polyphenol content of the fourth collected samples was 11.99~19.27 mg GAE/g (p<0.05). The DPPH radical scavenging activity, ABTS radical scavenging activity and FARP of the fourth Doenjang samples were 51.88~81.21%, 84.14~90.32%, and 1.08~3.11 mg Trolox/g, respectively. As a result of quality analysis on Doenjang, the superiority of traditional Doenjang has been proven by the increase of amino nitrogen content and antioxidant activity according to the aging period. However, factor analysis on quality differences of regional Doenjang should be conducted to ensure standardization and quality improvement.

        본 논문에서는 꽃송이버섯을 4종의 유산균(Lactobacillus plantarum subsp. plantarum, L. acidophilus, L. helveticus, L. delbrueckii subsp. bulgaricus)을 사용해 발효를 하였다. 발효된 꽃송이버섯 발효물을 항산화 활성(DPPH free radical scavenging activity, ABTS radical scavenging activity, Hydroxy radical scavenging activity), SOD activity, 총 폴리페놀 함량(TPC), HPLC 분석, protease 활성을 동해 발효 전 후 생리활성 변화를 확인하였다. 꽃송이버섯 발효물의 항산화 활성은 IC50 28.09-64.46 μg/ mL, SOD 활성은 IC50 15.75-39.42 μg/mL, TPC 는 57.66-100.43 GAE μg/mL 와 85.25-110.68 μg/mL 로 확인되었다. 꽃송이버섯 추출물의 항산화 활성보다 약 8배 이상 증가된 결과로 확인되었다. 또한 꽃송이버섯 발효물의 HPLC 분석 결과는 ascorbic acid, gallic acid, chlorogenic acid의 함량이 높게 확인되었으며, protease 활성 또한 발효물의 활성이 높은 것으로 확인되었다. Lactobacillus 균주들의 작용에 의하여 꽃송이버섯이 발효 되면서 폴리페놀류가 증가하여 생리활성이 꽃송이버섯 추출물에 비해 증가한 것으로 추측된다. Lactobacillus 균 중에서 L. helveticus 균주로 발효된 꽃송이버섯 발효물은 가장 높은 활성을 나타냈으며, 꽃송이버섯을 발효하는데 있어 L. helveticus 균주가 가장 적합한 균주로 확인되었다.

        본 연구에서는 기능성 식품원료로써 흰점박이꽃무지 유충의 이용 가능성과 흰점박이꽃무지 유충 사료자원으로써 큰느타리버섯수확후배지의 이용 가능성을 조사하기 위해서 참나무톱밥과 큰느타리수확후배지를 각각 식이한 흰점박이 꽃무지 유충 추출물의 항산화 활성을 비교하였다. 참나무톱밥과 큰느타리수확후배지를 각각 식이한 흰점박이꽃무지 유충 추출물의 총 폴리페놀 함량은 각각 57.02±1.73 mg GAE/g과 75.33±0.43 mg GAE/g이었고 플라보노이드 함량은 각각 24.6±0.28 mg/g과 25.4±0.75 mg/g이었다. DPPH에 의한 라디칼소거 활성은 추출물의 농도가 증가할수록 증가하는 경향을 나타내었으나 참나무톱밥을 식이한 흰점박이꽃무지 유충에 비해 큰느타리버섯수확후배지를 식이한 흰점박이꽃무지 유충 추출물의 DPPH 라디칼 소거능이 우수하였으며 32 mg/ml 이상의 농도에서는 두 시료 간의 유의적 차이가 나타나지 않았다. 참나무톱밥과 큰느타리버섯수확후배지를 각각 식이한 흰점박이꽃무지 유충 추출물의 농도별 환원력도 추출물의 농도가 높을수록 환원력은 증가하였으나 시료 간의 유의적인 차이는 보이지 않았다. 참나무톱밥과 큰느타리버섯수확후배지를 식이한 흰점박이꽃무지 유충 추출물의 ORAC 지수는 각각 66.55±0.99 uM TE/g과 76.32±0.48 uM TE/g로 참나무톱밥을 식이한 흰점박이꽃무지 유충 추출물에 비해 큰느타 리버섯수확후배지를 식이한 흰점박이꽃무지 유충 추출물의 ORAC 지수가 높게 나타났다. 참나무톱밥과 큰느타리 버섯수확후배지를 각각 식이한 흰점박이꽃무지 유충 추출물을 각각 0, 2, 4, 8, 16, 32, 64 mg/ml의 농도로 처리하 였을 때 RAW 264.7 세포는 90% 이상의 생존율을 나타 내었으므로 참나무톱밥과 큰느타리버섯수확후배지를 각각 식이한 흰점박이꽃무지 유충 추출물은 RAW 264.7 세포에 독성을 나타내지 않는 것으로 판단된다.

        This study assessed the quality characteristics of cookies made with different amounts (0, 5, 10, 15%) of black carrot powder. The density and pH value of cookie dough were constant for all groups. Compared to the control group, the moisture content was higher in groups containing the black carrot powder. Increasing the amount of black carrot powder resulted in decreased L-value but increased a-value. Furthermore, the hardness of cookies increased with increasing amounts of black carrot powder. The consumer acceptance test revealed that up to 15% black carrot powder addition was desirable. Total phenol content of the control group was 24.0 mg GAE/100 g, while black carrot powder groups ranged from 45.5 to 103.5 mg GAE/100 g. The DPPH radical scavenging activity was also augmented with increasing volumes of black carrot powder. Taken together, we recommend that cookies with 15% black carrot powder are desirable in terms of overall acceptability and antioxidant activity.

        The purpose of this study was to demonstrate the quality characteristics of Brassica juncea cultivated in Jeongseon (BJJ), South Korea. We analyzed the nutritional components and antioxidant activity of BJJ. As a result of the free sugar analysis, the contents of glucose and fructose in BJJ were 0.29±0.02 g/100 g and 0.10±0.00 g/100 g, respectively. The major fatty acids were palmitic acid, octadecenoic acid and stearic acid. The palmitic acid was the highest at 31.22% of all fatty acids. The major minerals were identified as Ca, P, K, Mg and Na. The contents of vitamin B1, vitamin B2, vitamin B6, vitamin C and vitamin E in BJJ were 0.02±0.00 mg/100 g, 0.087±0.01 mg/100 g, 0.02±0.00 mg/100 g, 0.56±0.06 mg/100 g and 0.20±0.03 mg α-TE/100 g, respectively. As a result of the free amino acid analysis, total amino acid contents in BJJ were 2,801.21±115.38 mg/100 g. L-proline content was the highest (744.30±119.06 mg/100 g) in BJJ. BJJ extract inhibits reactive oxygen species production in 3T3-L1 adipocytes. Also, BJJ extract exhibits a protective effect on oxidative stress in H2O2-induced human dermal fibroblast. These results indicate that BJJ comprises various valuable nutrients which can be used as functional food ingredients.

        오늘날 양돈의 사양이 증가 함에 따라 부존사료 자원으로서 활용가치가 높은 물질에 항생제를 첨가 하지 않고 친환경적으로 제조된 가축용 사료를 개발하여, 기능성 친환경 돈육 및 가공품을 생산하는 것이 중요하다. 이에 따라 본 연구에서는 항암, 항염, 항산화 등 다양한 생리활성 물질이 다량 함유되어 있는 복분자(Rubus occidentalis, RO) 부산물을 양돈사료의 원료로 확립하고자 버크셔 돼지를 대상으로 복분자 발효사료(Rubus occidentalis fermented fodder, ROFF)의 혈액 내 영양 운반인자와 항산화 효능을 연구하기 위해 수행하였다. 시험구는 일반사료(대조구)에 ROFF를 0.3% 첨가하였고, 발효사료의 효능을 확인 하기 위해 각 체중에 따른 버크셔 거세돈 및 암퇘지에게 43~73일간 급여하여 효능을 확인 하였다. ROFF의 복합적인 효능은 혈액 생화학, 철분 및 항산화 분석을 통해 확인하였다. 그 결과 ROFF의 섭취로 인해 거세 자돈 및 육성돈, 암컷 자돈, 육성돈, 및 110-150 kg 암컷 비육돈 에서 total cholesterol (TC), LDL-cholesterol (LDL-C), HDL-cholesterol (HDL-C) 모두 감소하거나 HDL-C이 증가하는 경향으로 보아 영양생리학적으로 개선되는 경향을 확인하였다. 암컷 비육돈에서는 생화학적 수치가 모두 증가하는 경향으로 보아 임신 가능성이 있는 비육기의 경우 이러한 수치 향상으로 자돈의 생산을 위한 원활한 영양 공급이 예상되며, 건강한 자돈의 생산에 도움이 될 것으로 판단된다. Transferrin (TFE) 함량은 거세돈에서는 변화가 크지 않았으며, 암컷 육성돈 및 110-150 kg 암컷 비육돈에서 ROFF 섭취로 인해 증가하는 경향이 나타남에 따라 ROFF가 철분 결핍으로 인한 부정적인 영향들을 최소화 할 수 있을 것이다. Glutathione peroxidase1 (GPx1)분석 결과 거세 비육 돈 및 110-150 kg 암컷 비육돈에서 ROFF의 섭취로 인해 GPx1 활성이 유의적이지는 않지만 크게 증 가한 것으로 보아 ROFF가 항산화능을 향상시키는 것으로 판단된다. 본 연구의 결과들을 종합할 때, ROFF의 급여가 전체적으로 시험 개체의 개선 효능에는 영향이 크지 않으나 RO의 함량을 고려 할 때 매우 긍정적인 원료사료 중 하나라 할 수 있으며, 특히 ROFF가 거세돈 보다는 암컷 버크셔의 영양 운 반 및 철분 함량 등에 긍정적인 영향을 줄 수 있을 것으로 판단된다.

        본 연구는 사람의 다양한 세포주를 이용하여 활성산소종(과산화수소수)이 세포의 노화에 미치는 영향을 비교 조사하였다. 여러 농도의 과산화수소수에 세포주를 일주일 동안 배양하여 MTT 방법으로 과산화수소수에 대한 세포 성장의 반억제농도를 구하였다. 그 결과, 50대에서 유래하는 피부 섬유아세포와 10대의 노화 유도 피부 섬유아세포와 비교하여 10대에서 유래하는 피부 섬유아세포에서 과산화수소수에 대한 반억제농도의 값이 유의적으로 더 높았고, 10대의 피부 섬유아세포보다는 10대의 여러 조직 기원하는 성체줄기세포에서 반억제농도의 값이 유의적으로 더 높게 관찰되었다. 또한, 50 ppm 과산화수소수를 1주일 동안 처리한 후, 50대의 피부 섬유아세포에서 다른 세포주에 비해 세포 성장이 현저히 억제되었고, 노화 관련 베타-갈락토시다아제의 활성이 증가되는 것을 관찰하였다. 또한, 활성산소의 세포 독성을 중화시키는 두 유전자, 글루타티온 과산화효소(GPX)와 카탈라아제(CAT)의 발현을 각 세포주에서 조사하였을 때, CAT의 발현은 모든 세포주에서 대체로 낮았지만, GPX 유전자의 발현이 50 대의 피부 섬유아세포보다 10대의 피부 섬유아세포와 성체줄기세포에서 현저히 높게 발현되는 것을 관찰하였다. 이상의 결과에서 활성산소는 세포 노화를 유도하고, GPX의 발현이 높은 10대의 피부 섬유아세포와 줄기세포보다는 50대의 피부 섬유아세포와 노화된 피부 섬유아세포에서 활성산소종에 대해 더 큰 민감성을 가지고 있는 것을 알 수 있었다.

        The antioxidant activity, total phenol content, and inhibitory effect of xanthine oxidase (XOase) for six different medicinal plant complexes such as Achyranthes japonica, Acanthopanax sessiliflorus, Carthamus tinctorius, Eucommia ulmides, Viscum album, and Caragana koreana were investigated in this study. The free radical scavenging activity values using DPPH(2,2'-diphenyl-1-picrylhydrazyl) and ABTS[2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radical cation] analyses were found to be 532.03±86.60 μg/mL and 1376.50±35.01 μg/mL, respectively, in 95% of ethanol extracts among six medicinal plant mixed compositions (6MPMC). The total phenolic content in 70% ethanol extract of 6MPMC was 125.19±1.34 mg GAE/g. The inhibitory activity against XOase was highest at 33.20±0.17% in the 70% ethanol extract of 6MPMC. In the antimicrobial activity test of 6MPMC, the minimum inhibitory concentration (MIC) value showed activities against V. litoralis and E. coli in 70% and 95% ethanol extracts, and these two microorganisms were created in clear zones by method of paper disc diffusion. These results suggest that the 6MPMC, composed of Achyranthes japonica, Acanthopanax sessiliflorus, Carthamus tinctorius, Eucommia ulmides, Viscum album, and Caragana koreana, have both antioxidant and antimicrobial activities and, therefore, may serve as a functional health food product.
