Cotesia plutellae, an endoparasitoid wasp, parasitizes larvae of Plutella xylostella, and disrupt immune response of the host through parasitic factors. These immune disruption factors are maternal (venom proteins, polydnavirus, and ovary proteins) and embryonic (teratocytes) factors. In this study, we performed transcriptome analysis of venom glands of C. plutellae and identified neprilysin-1 (NEP1) known to be potential immunosuppression gene. Cp-NEP1 encoded 451 amino acid and belongs to hymeopteran NEP1 via phylogenetic analysis. Based on the structural comparison Cp-NEP1 lacks in conserved motifs such as substrate binding (NAYY/F), zinc-binding site (HExxH), zinc-binding site, protein folding and maturation (CxxW). To investigate function of Cp-NEP1, we constructed a recombinant Cp-NEP1 harboring N-terminally fused 6X His tag. Peptide sequencing revealed successful expression of the recombinant Cp-NEP1 in Escherichia coli. Pre-heated E. coli as antigen induced spike of nodule formation whereas co-injection of the recombinant Cp-NEP1 and pre-heated E. coli exhibited suppression of nodule formation in the host. Quantitative real-time PCR revealed that expression of phenoloxidase related to nodule formation was suppressed under co-injection of the recombinant Cp-NEP1 and E. coli. These results suggest that Cp-NEP1 contributes to immunosuppression of P. xylostella via phenoloxidase suppression and conserved motifs of neprilysin family are not required for host immune suppression.

Recently, many habitat corridors have been constructed in Korea, but the effect of corridor types, i.e. overpasses and underpasses, on arthropods was poorly examined. Therefore, we compared the effect of habitat corridor types in terms of abundance, species richness, and composition of carabid beetles. As a result, 3,737 ground beetles belonging to 60 species were collected by pitfall trapping across the northwestern forest–habitat corridor–southeastern forest transects in 2015. Abundance and species richness of total carabid beetles in underpasses were significantly lower than overpasses. And species composition of underpasses corridors was significantly different compared to other habitats, such as overpasses and forests. Although more samples are needed to understand the effect of corridor types, the current underpasses may be unfriendly structures to movement of ground dwelling arthropods as well as carabid beetles.

국화재배시 발생하는 꽃노랑총채벌레와 대만총채벌레는 꽃을 선호하는 특성에 의해 국화의 개화시기에 꽃을 가해함으로 써 경제적으로 가장 큰 피해를 야기하는 해충이다. 시설국화는 5월 초순경 심겨지시 시작하면서부터 재배농가들은 10일 또는 1주일 간격으로 화학약제를 살포하고 있고 이로 인하여 건강악화로 인한 고통을 호소하고 있다. 따라서, 본 연구에서는 국화재배시 발생하는 총채벌레를 유인하여 방제함으로써 방제효율을 극대화하고 화학약제의 사용을 최소화 하는데 목적을 두었다. 개화한 황색의 포트국화에 미끌애꽃노린재가 산란한 채송화 줄기를 식재하여 재배국화 정식 10일 후 20m 간격으로 투입한 결과 총채벌레의 밀도는 신초당 3마리 이하의 밀도를 유지할 수 있었고 투입하지 않은 무처리구에서는 신초당 10마리 이상의 밀도를 확인할 수 있었다. 꽃봉우리 형성 시기에 1회의 총채벌레 방제용 화학약제를 살포한 결과 수확시기 개화한 국화의 꽃당 총채벌레 밀도는 0.3마리 이하의 수준으로 총채벌레를 방제할 수 있었다. 따라서, 시설국화재배시 발생하는 총채벌레 방제를 위하여 황색개화국화를 총채벌레의 트랩식물로써 투입하고 동시에 천적인 미끌애꽃노린재를 투입하여 수확전까지 총채벌레의 밀도를 관리한 후 수확시기에 화학약제를 사용함으로써 총채벌레에 의한 경제적 손실을 줄일 수 있을 것으로 판단된다.

수출용 파프리카 재배지에서 발견되는 곤충 중 파리목 해충이 비교적 종수가 많으며, 위험평가가 제대로 이루어지지 않아 수출 검사 시 수출 상대국에서 검출되기도 하였다. 기존의 파프리카 재배지에서 해충 조사를 실시한 논문에서는 파리목에 속하는 해충은 아메리카잎굴파리, 흑다리잎굴파리, 작은뿌리파리 3종이었었다(류 등, 2011, 김 등, 2013). 전국의 10개시·군의 파프리카 수출 재배지에서 유인등 트랩과 끈끈이트랩을 이용한 조사 결과 6개과 11종을 확인하였으며, 물가파리 과의 Scatella sp. 와 검정날개버섯파리과의 작은뿌리파리인 Bradysia impaiense, 초파리과의 노랑초파리는 대부분의 파프리카 온실에서 공통적으로 발견되었다. 실제 파프리카 작물에 피해를 주는 해충은 굴파리과의 Liriomyza huidobrensis 흑다리잎굴파 리가 있었으며, 포장의 수분과 밀접한 관계가 있는 종으로 위생해충에 속하는 물가파리과 일종, 작은뿌리파리, 나방파리과 일종의 종들이 포함되었다. 또한 작은 곤충을 사냥하는 집파리과의 일종인 Coenosia spp. 가 천적으로서 온실에 정착하여 서식하는 것이 밝혀졌다.

Entomopathogenic fungi are facultative microorganisms, dwelling in soil or infecting host insects, and some of the genera have been used as biological control agents worldwide. Collection of fungal isolates should be a platform for the development of highly effective resources, thus in this work we constructed a fungal library using a mealworm (Tenebrio molitor) pathogenicity-based fungal collection method and further characterized some isolates with high virulence. A phylogenetic three was generated, and of the isolates 17 isolates’ biological features were characterized, such as morphology, spectrum of virulence, cultural characteristics, thermo-stability of fungi, production of biologically active materials, such as enzymes. This work reports an attractive entomopathogenic fungal library including the information of effective isolates in pest management.

Oriental fruit moth Grapholita molesta (Busck) (Lepidoptera: Tortricidae) is an important pest for stone fruits like apple, peach, pear etc. Growers usually depend on chemical insecticides to control this pest, but it causes problems like resistance, pest resurgence, and health hazard. Although various plant extracts have been known to be potential against other lepidopterans, their effectiveness against oriental fruit moth has rarely been studied. So, we evaluated toxicity of 28 methanolic plant extracts known from previous literatures in the laboratory condition. Residual toxicity against 1st instar larvae (<5 h old) were examined using glass scintillation vial method at the concentration of 2 mg/ml. Among the tested botanicals Nicotiana tabacum L. was most effective and showed 92% mortality within 20 h and LT50 value was 12.9 h followed by Allium sativum L., Zanthoxylum piperitum (L.), and Sapindus mukorossi Gaerten. which showed 88, 70, and 65% mortality within 20 h, respectively. Rhododendron micranthum Turcz., Humulus japonicus Seibold & Zucc., Nerium indicum Mill., and other extracts showed least effectiveness on 1st instar larvae of oriental fruit moth. Hence, we suggest N. tabacum, A. sativum, Z. piperitum, and S, mukorossi as botanicals to be used against oriental fruit moth.

Pear psyllids are of major economic concern in Korea. In the literature five nominal species, including two West Palaearctic taxa, are reported from the Peninsula. The Korean pear psyllids, member of the large genus Cacopsylla, are reviewed here to comprise four species. Previous Korean records of C. pyrisuga (Foerster) are misidentification of C. burckhardti Luo et al. whereas those of C. pyricola (Foerster) concern C. maculatili Li and probably C. jukyungi (Kwon) stat. rev., comb. nov. (from Psylla). The latter, that is commonly found in pear orchards, occurs also in Japan where it was misidentified as C. chinensis. The differences between C. jukyungi and C. cinereosignata syn. nov. reflect seasonal dimorphism. For this reason, we synonymise the two. C. maculatili and C. qiuzili Li are shown to represent the winter and the summer form of the same species and are, therefore, synonymised as C. maculatili = C. qiuzili syn. nov. C. jukyungi and C. sandolbaea (Park & Lee) are redescribed, a key to the adult of the four species and short biological notes are provided.

Psyllids have shifted lately into general awareness as vectors of pathogens causing serious plant diseases, as pests in agriculture and forestry or as potential control agents of alien invasive weeds. These small insects are plant sap feeders which are generally very host specific. In addition, related psyllid species tend to develop on related plant species. This makes them an ideal group for studies on insect–plant cospeciation. A sound taxonomic and phylogenetic base is a prerequisite for successful pest control and meaningful research on insect–plant interactions. Currently almost 4000 named species of Psylloidea exist worldwide of which half was described in the last three decades. Despite this tremendous progress there are at least another 4000 species which remain undescribed particularly in Africa, South America and tropical Asia. Since White and Hodkinson’s seminal paper in 1985 a series of studies tested their classification with additional taxa using morphological and molecular techniques. In 2012 Burckhardt and Ouvrard proposed a new classification. Five of their families (Calophyidae, Carsidaridae, Homotomidae, Phacopteronidae and Triozidae) are identical with or similar to those of White and Hodkinson but three differ fundamentally from previous classifications (Aphalaridae, Liviidae and Psyllidae). Many of the recognised families and subfamilies are restricted more or less exclusively to a single plant taxon, e.g. Calophyidae, Phacopteronidae and Rhinocolinae to Sapindales, Carsidaridae to Malvaceae, Homotomidae to Moraceae, Spondyliaspidinae to Myrtaceae, and many Psyllidae to Fabaceae. The 1000 described species of Triozidae, in contrast, have colonised many families of dicots and, a few, even monocots and conifers. Several analyses suggest that cospeciation may be less important than geographical vicariance to explain the observed species richness in psyllids, and that shifts to new host taxa are frequent.

Various techniques have been developed to monitor insect behavior in the field. Harmonic radar tagging is a promising method of tracking target insects because it can allow efficient detection of the insects with negligible effects on their behavior or physiology. However, availability of the light limits the effectiveness of the method as it is almost impossible to spot an insect in the dark. On the other hand, fluorescent-marking, when combined with strong light source, makes the organisms distinct at night in a non-destructive manner. Therefore, we conducted a field experiment to determine the effectiveness of the combination of the two methods. Riptortus pedestris (Fabricius) adults were marked with both harmonic radars and fluorescent paint. The marked insects were haphazardly arranged and pinned onto trees or the ground in the forest at night. Then, a group of paired researchers, one equipped with a harmonic radar and the other with a hand-held laser, attempted to find R. pedestris within 20 minutes. The detection rate showed a high variance ranging from 40% to 100%. Thus, with a proper training, harmonic radar system combined with fluorescent-marking can be a powerful technique to detect insects in the field. With these equipments, tracking insect behavior in the field will be more efficient in the dark.

Expression of hairpin RNA corresponding to the part of COPA transcript was done by agroinfiltration in soybean plants and was confirmed by qRT-PCR. In a pot experiment, T. urticae was infested on agroinfiltrated soybean plants and T. urticae mortality was observed and compared with control plants overtime. Significantly higher mortalities of T. urticae were observed in the COPA-agroinfiltrated soybean plants from post-infestation day 2 (15 ±5%), day 4 (50 ±10 %). At post-infestation day 6, mortality reached to (70 ± 15%). To validate the observed COPA silencing effect in T. urticae fed on the agroinfiltrated soybean plant expressing COPA hairpin RNAs, qRT-PCR analysis was performed. The transcript level of COPA gene was decreased in T. urticae fed on agroinfiltrated soybean plants expressing COPA hairpin RNA from post-infestation day 2. At post-infestation day 2, 4 and 6, COPA transcript levels were reduced by 23.8, 20.7 and 18.8 fold, respectively compared to post-infestation day 1 (control). The results obtained in this study also ruled that the plant mediated production and uptake of silencing (dsRNAs/siRNAs) is an effective way to trigger RNAi in the T. urticae.

Myeloid differentiation factor 88 (MyD88) is an intracellular adaptor protein involved in Toll signaling pathway. In this study, we monitored the response of 4 key genes of the insect immune system against Beauveria bassiana JEF-007 in Tenebrio molitor using RT-PCR. TmGPR, antimicrobial peptide Tenecin 1 and Tenecin 2 were up-regulated after fungal infection. To better understand the roles of Toll signaling pathway in mealworm immune system, TmGRP and TmMyD88 was knocked down by RNAi silencing. Target gene expressions were decreased at 2 days post-dsRNA injection, and dramatically reduced at 6 days post-dsRNA injection. Therefore, mealworms were compromised by B. bassiana JEF-007 at 6 days post-dsRNA injection. Silencing of the TmMyD88 and TmGRP resulted in reducing the resistance of the host to fungal infection. However, only dsTmMyD88 showed significant difference with dsEGFP by statistical analysis, which may be due to partial gene knock down of dsGRP. These results indicate that TmMyD88 is required in mealworms for survival against B. bassiana JEF-007.

본 실험은 유기농 신선초(Angelica utilis)포장에서 점박이응애(Tetranychus urticae)의 발생지점 및 확산 양상을 확인하고자 수행하였다. 실험은 17주 동안 강원도 원주시 신림면 용암리에 위치한 유기작목반 비닐온실(625㎡/동)에서 진행되었다. 온실 입구부터 5m 간격으로 확대경을 이용해 해충 밀도를 조사한 결과 A농가에서는 1주차에 입구에서 95m 지점의 해충밀도가 가장 높았으며, 시간이 경과함에 따라 앞쪽으로 확산되었다. 이는 A농가 뒤쪽 우거진 수풀에서 점박이응애가 유입되어 도로변이 있는 앞쪽으로 확산된 것으로 보인다. B농가의 경우 1주차에 입구에서 50m 지점의 해충밀도가 가장 높았으며 시간이 경과함에 따라 뒤쪽으로 확산되었다. 이는 B농가 앞쪽에 있는 신선초온실에서 해충이 유입되어 확산된 것으로 보인다. 또한 점박이응애의 천적을 1주일간격으로 3회 방사한 결과 A농가는 방사 6주차에, B농가는 방사 3주차에 100% 해충방제효과를 보였다. 신선초는 1년 상시 재배되는 작형인 만큼 계절별, 지리적 특성에 따라 해충 양상이 달라질 수 있으므로 추후 체계적인 모니터링을 통해 효과적인 천적방사시기 및 방사지점을 확인한다면 해충발생초기에 소량의 천적투 입으로 방제효과 극대화, 노동력과 방제비용의 절감을 기대할 수 있을 것으로 기대된다.

담배거세미나방(Spodoptera litura) 유충의 섭식 행동을 분석하기 위해 부화 1일 후부터 고치 형성 때까지 1분 간격으로 피망 잎 섭식흔을 측정하였다. 잎 면적 측정은 컴퓨터와 카메라를 이용하여 동영상을 촬영하고 영상 처리(image processing) 기법을 이용하여 분석하였다. 관찰 결과, 담배거세미나방 유충의 섭식 행동은 탈피 직후부터 즉시 섭식을 시작하였다. 섭식 행동은 섭식기(약 1800분)와 비섭식기(약 760분)가 뚜렷하게 구분되었으며 섭식기가 끝나면 비섭식기를 거쳐 탈피를 하고 다시 섭식기에 들어서는 것을 확인할 수 있었다. 1회 섭식 행동은 ‘bout’로 구분되며, 1회 평균 섭식 지속 시간은 약 15분으로 영기에 따라 차이가 나지 않았다. 영기 내 평균 섭식 회수(no. of bout)는 약 27.5회로 섭식 지속 시간과 마찬가지로 영기에 따라 차이가 나지 않았다. ‘Bout’ 간의 간격은 ‘interbout interval’로 구분되며 평균 54분마다 섭식을 하였고 영기에 따른 차이는 관찰되지 않았다. 하지만 영기에 따른 섭식량은 지수적으로 증가하였다. 일주기(circadian rhythm) 행동은 관찰되지 않았다.

Integrin is a heterodimer protein that locates on cell membrane to interact with neighboring cells or extracellular matrix. A transcriptome analysis of the brassica leaf beetle, Phaedon brassicae, midgut identified both α and β subunits of integrin. RNA interference of β subunit genes significantly impaired survival of both larvae and adults of P. brassicae. A recombinant bacteria expressing double-stranded RNA specific to β integrin of P. brassicae were constructed and showed significant oral toxicities.

Bean bug, Riptortus pedestris (Hemiptera: Alydidae) is an agriculturally serious pest in East Asian countries. Chemical pesticides have been contributed to the management of the pest, but nowadays insect resistance limits the use of chemical pesticides, thus alternatively new pesticides with different mode of actions such as entomopathogenic fungi are considered. Herein entomopathogenic Beauveria bassiana JEF isolates were collected, identified and assayed against bean bugs in laboratory conditions. Some isolates showed >80% virulence by contact-exposure and spray methods. The Agrobacterium tumefaciens-mediated transformation of B. bassiana JEF-007 generated random transformants and some mutants showed reduced virulence against Tenebrio molitor (Coleoptera: Tenebrionidae) larvae and R. pedestris nymph. Compared to the wild-type, the two transformants showed remarkably different morphology, conidial production, and thermotolerance. To figure out pathogenicity-related genes, thermal asymmetric interlaced (TAIL) PCR of the random transformants was performed and possibly some virulence-related genes were predicted. This work can be a strong platform for the functional genetics of bean bug-pathogenic B. bassiana.

곤충 발육기간분포모형은 어느 특정발육단계가 완료되는 시기의 변이를 나타내는 것으로 시간에 따라 발육이 완료되는 개체들의 비율로 나타낸다. 개체군 모형 작성시 발육단계전이모형의 기본 구성요소로 사용되며, 발육기간 누적빈도분포 자료를 Weibull 함수에 적용시켜 사용하고 있다. 곤충 발육기간분포모형을 추정하려면 발육실험의 원자료가 필요하기 때문에 원자료가 없는 경우 대체할 수 있는 방법의 개발이 필요하다. 따라서 본 연구에서는 곤충의 발육기간 빈도분포가 정규분포를 따른다는 가정 하에 해당 곤충의 발육기간 평균 및 표준편차를 이용하여 가상의 발육기간 빈도분포를 조성하고 매개변수를 추정하는 방법을 제시하였다. 정규분포에 준한 발육기간 누적빈도분포로 Weibull 함수의 모형 매개변수를 추정하고, 이것을 실제 발육기간 빈도분포를 이용하여 추정한 모형 매개변수와 비교하였다. 총 8종 (18개 발육단계) 곤충을 대상으로 분석한 결과 곤충 발육기간 빈도분포가 정규분포를 따르지 않음에 따라 원자료 값과 편차가 발생하였다. 일부 매개변수를 보정함으로써 원자료 값에 가깝도록 조절 할 수 있었으며, 이 방법을 통해 어떤 곤충의 발육기간 빈도분포자료 없이도 발육기간 평균과 표준편차만을 이용하여 발육기간분포모형을 추정할 수 있을 것으로 기대되었다.

The spined soldier bug, Podisus maculiventris (Hemiptera: Pentatomidae), is a generalist predator commonly found in North America. The spined soldier bug can feed on more than 70 species of insects including immature stages of Lepidoptera and Coleoptera. The spined soldier bug can inhabit various agricultural ecosystems (i.e. crop fields, forests, and grasslands) and plays a key role in the control of major insect pests. The spined solider bug has high the potential for augmentative biological control because it has a high reproduction rate. In 2016, 500 eggs of the spined soldier bug were introduced to South Korea from U.S.A. under the cooperative research project between West Virginia University and the National Institute of Agricultural Science (NIAS), Rural Development Administration. Currently the spined soldier bug is being reared in a quarantined facility at NIAS. In this presentation, we will describe the natural history of the spined soldier bug in North America including its feeding ability as a generalist predator, its potential for biological control of various agricultural and forest pests, and its response to aggregation pheromone. In addition, current research at West Virginia University including mass rearing and aerial release using unmanned aerial vehicles (a.k.a. drones) of the spined soldier bug will be presented.

Beauveria bassiana (Bb) is an entomopathogenic fungus with a wide host range, and is commonly used as an environment-friendly biopesticide. However, the molecular mechanisms of Bb-host interactions are not well understood. Here, RNA isolated from a highly virulent strain of B. bassiana (Bb JEF-007) and Riptortus pedestris (Hemiptera: Alydidae) (bean bug) infected with this strain were subjected to high throughput next generation sequencing (NGS) to analyze and compare transcriptomes. Differentially expressed gene (DEG) analysis showed that 2,381 genes were up-regulated and 2,303 genes were down-regulated upon infection. Most DEGs were classified into the categories of single-organism, cellular and metabolism processes by gene ontology (GO) analysis. Carbon metabolism-related enzymes in the glyoxylate cycle were significantly up-regulated, suggesting a possible role for them in Bb growth in the host. This work provides insight into how entomopathogenic B. bassiana occupies agriculturally harmful bean bug at the late stage, which might be essential during fungal infection.

Cotesia plutellae bracovirus (CpBV) is a bracoviral polydnavirus and a parasitic factor of an endoparasitioid wasp, C. plutellae, which parasitizes the diamondback moth (DBM) larvae. CpBV genome was reported to consist of 35 circular segments and encode 157 predicted open reading frames (ORFs). This study revisited and re-annotated the CpBV genome. Total ORFs consist of 15 groups of known gene or gene families as well as hypothetical (HP) group. All 74 HP genes are matched to other PDV genes. RNASeq analysis indicates that 147 genes out of 157 predicted ORFs are expressed in the parasitized DBM at the first day or 7 days after parasitization. At the early parasitic stage, CpBV genes annotated as ELP, BEN, E94K, DUFB, PTP, and HP are highly expressed compared to late parasitic stage. Interestingly, highly expressed genes at the late parasitic stages are HPs.

The brown marmorated stink bug (BMSB), Halyomorpha halys (Hemiptera: Pentatomidae), is an invasive insect pest from Eastern Asia. Since its invasion into North America during the late 1990s, frequent outbreaks of BMSB have caused considerable economic damage to orchards, field crops, and vegetables. Currently, management practices in the United States rely on insecticides to control BMSB. Therefore, identification of potential natural enemies of BMSB is crucial to justify integrated pest management (IPM). We investigated the viability of using the spined soldier bug, Podisus maculiventris (Hemiptera: Pentatomidae), as a potential biological control agent for BMSB. In this study, we used choice and non-choice tests to determine which life stages of the spined soldier bug can effectively kill specific stages of BMSB using videography. Prey searching and feeding behavior of the spined soldier bug and defensive behavior of BMSB were analyzed. Our study showed that BMSB eggs and nymphs attacked by the spined soldier bug had a mortality rate of 100%. We also observed that all first instar BMSB remained on the egg cluster allowing the spined soldier bug to attack and kill many BMSB during one feeding session. The results indicate that the spined soldier bug has the potential for killing BMSB
