In Korea, most temporary storage facilities for spent nuclear fuel are nearing saturation. As an alternative to this, the 2nd basic plan for high-level radioactive waste management specified the operation plan of dry interim storage facility. Meanwhile, the NSSC No. 2021-19 stipulates that it is necessary to evaluate the possibility and potential effect of accident before operating interim storage facility. Therefore, this study analyzed the categories of accident scenarios that may occur in dry storage facility as part of prior research on this. We investigated the case of categorization of dry storage facility accident scenarios of IAEA, NRC, KAREI, and KINS. The IAEA presented accident scenarios that could occur in on-site dry storage facility operated with silo and cask method. NRC has classified accident scenarios in dry storage facility and estimated the probability of accidents for each. KAERI and KINS selected major accident scenarios and analyzed the processes for each, in preparation for the introduction of dry storage facility in Korea in the future. Overall, a total of 10 accident scenarios were considered, and the scenarios considered by each institution were different. Among 10 scenarios, cask drop and aircraft collision were included in the categorization of most institutions. The results of this study can be used as basic data for cataloging accidents subject to safety evaluation when introducing dry interim storage facility in Korea in the future.

We investigated the pharmacokinetics of ferulic acid, a potential antioxidant agent, after intravenous (i.v.) bolus administration in rats. To analyze ferulic acid levels in the plasma, bile, urine and tissue samples, we developed an HPLC-based method which was validated for a pharmacokinetic study by suitable criteria. After i.v. bolus administration of ferulic acid, it rapidly disappeared from blood circulation within 15 min. The mean plasma half-lives at α phase (t1/2α) when administered at doses of 2 and 10 mg/ kg were 1.10 and 1.39 min, respectively. The values of t1/2β at the corresponding doses increased 40% (from 5.02 to 7.01 min) with increasing doses. The total body clearance (CLt) values significantly decreased as the ferulic acid dose increased. On the other hand, steady-state volume of distribution (Vdss) values did not show the significant difference with the increase in dose. Of the various tissues, ferulic acid mainly distributed to the liver and kidney after i.v. bolus administration. The ferulic acid concentrations in various tissues at 2 hr after i.v. bolus administration were below 1.0 μg/g tissue. Ferulic acid was excreted in the bile and urine after i.v. bolus administration at the dose of 10 mg/kg. The cumulative amount of ferulic acid in the bile 2 hr after dosage was comparable with the amount excreted in the urine after 72 hr, indicating that i.v. administered ferulic acid was mainly excreted in the both bile and urine. In conclusion, ferulic acid was rapidly cleared from the circulating blood and transferred to tissues such as the liver and kidney after i.v. bolus administration. Moreover, the majority of ferulic acid appears to be excreted in the bile and urine after i.v. bolus administration.

Chronological aging and photoaging affect appearance, causing wrinkles, pigmentation, texture changes, and loss of elasticity in the skin. Phragmites communis is a tall perennial herb used for its high nutritional value and for medicinal purposes, such as relief from fever and vomiting and facilitation of diuresis. In this study, we investigated the effects of ethanol extract of P. communis rhizome (PCE) on skin aging. The total flavonoid and total phenolic content in PCE were 2.92 ± 0.007 ㎍ of quercetin equivalents (QE) and 231.8 ± 0.001 ㎍ of gallic acid equivalents (GAE) per 100 ㎎ of dried extract (n = 3). The half-maximal inhibitory concentration (IC50) values of PCE for 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) and hydrogen peroxide scavenging activities were 0.96 and 0.97 ㎎/mL, respectively. PCE showed inhibitory effects on tyrosinase when L-tyrosine (IC50 = 1.25 ㎎/mL) and L-3,4-dihydroxyphenylalanine (IC50 = 0.92 ㎎/mL) were used as substrates. PCE treatment up to 200 ㎍/mL for 24 h did not cause any significant cytotoxicity in B16F10 melanocytes, human dermal fibroblasts (HDFs), and HaCaT keratinocytes. In B16F10 melanocytes, PCE (25 and 50 ㎍ /mL) inhibited melanin production and cellular tyrosinase activity after challenge with α-melanocyte-stimulating hormone (α-MSH; p < 0.05). In HDFs, PCE suppressed the mRNA expression of matrix metalloproteinase-1 (MMP-1) and reduced the activity of elastase (p < 0.05). In addition, ultraviolet B (UVB)-mediated downregulation of hyaluronic acid synthase-2 gene expression in HaCaT keratinocytes was also effectively suppressed by PCE treatment. Overall, our results showed that PCE has potential anti-skin aging activity associated with the suppression of hyperpigmentation, wrinkle formation, and reduction in dryness. PCE is a promising candidate for the development of an anti-skin aging cosmetic ingredient.

Stevia rebaudiana (Asteraceae), a perennial plant, has been used as a low-calorie sweetener and is being developed as a therapeutic agent for diabetes, hypertension, myocardial diseases, and microbial infections. Despite the common use of its leaves and stem, the bioavailability of the components present in S. rebaudiana flowers, when used as ingredients of cosmetics, has not been well investigated. Herein, we investigated the antioxidative and antimelanogenic effects of an aqueous extract of S. rebaudiana flowers (Stevia-F). Total flavonoid and phenolic content in Stevia-F were determined to be 8.64 ± 0.23 ㎎ of quercetin equivalents/100 g and 631.5 ± 2.01 ㎎ of gallic acid equivalents/100 g, respectively. The IC50 values of Stevia-F for reducing power, and 2,2-diphenyl-1-picryl-hydrazyl-hydrate radical, hydrogen peroxide, and nitric oxide scavenging activities were 5541.96, 131.39, 466.34, and 10.44 ㎍/mL, respectively. Stevia-F showed inhibitory effects on the tyrosinase (IC50 = 134.74 ㎍/mL) and α-glucosidase (IC50 = 114.81 ㎍/mL) activities. No significant cytotoxicity of Stevia-F was observed in B16F10 cells, treated with up to 100 ㎍/mL of the extract for 24 and 48 h (p > 0.05). Stevia-F (1–100 ㎍/mL) suppressed α-melanocyte stimulating hormone-induced melanin production in B16F10 cells (p < 0.05) and also inhibited the cellular tyrosinase activity (p < 0.05). Overall, our results show that Stevia-F possesses potential for inhibiting tyrosinase and α-glucosidase activities and has significant antioxidant capacity. The antimelanogenic potential of Stevia-F should extend the usage of S. rebaudiana flowers in the development of skinwhitening products.

Angelica tenuissima, also known as Ligusticum tenuissimum, is classified as a food-related plant and has been used as traditional medicines treating headache and anemia in Asia. However, its anti-melanogenic effect has not been reported in detail. When the extract of Angelica tenuissima (ATE) was prepared by the extraction with 70% EtOH at 80°C (final yield = 22%), the contents of decursin and Z-ligustilide in ATE were determined 0.06% and 8.43%, respectively. Total flavonoid and phenolic content in mg ATE were 5.52±0.07 ㎍ quercetin equivalents and 237.27±13.24 ㎍ gallic acid equivalents, respectively. Antioxidant capacity of ATE determined by DPPH and ABTS assay was increased with a dose dependent manner up to 1000 ㎍/㎖. The amount of melanin synthesis followed by α-melanocyte stimulating hormone on B16F10 cells were significantly reduced in the presence of ATE (250 to 1000 ㎍/㎖, p<0.05). ATE (125 to 1000 ㎍/㎖, p<0.05) suppressed the tyrosinase activity but did not show any significant effect on α-glucosidase activity at the same condition. Taken together, ATE possesses tyrosinase inhibitory potential with significant antioxidant capacities. These effects of ATE might be involved in suppression of melanin synthesis, at least, in B16F10 cells. The anti-melanogenic potential of ATE will provide an insight into developing a new skin whitening product.

Numerical Weather Prediction (NWP) models such as the Weather Research and Forecasting (WRF) model are essential for forecasting one-day-ahead solar irradiance. In order to evaluate the performance of the WRF in forecasting solar irradiance over the Korean Peninsula, we compared WRF prediction data from 2008 to 2010 corresponding to weather observation data (OBS) from the Korean Meteorological Administration (KMA). The WRF model showed poor performance at polluted regions such as Seoul and Suwon where the relative Root Mean Square Error (rRMSE) is over 30%. Predictions by the WRF model alone had a large amount of potential error because of the lack of actual aerosol radiative feedbacks. For the purpose of reducing this error induced by atmospheric particles, i.e., aerosols, the WRF model was coupled with the Community Multiscale Air Quality (CMAQ) model. The coupled system makes it possible to estimate the radiative feedbacks of aerosols on the solar irradiance. As a result, the solar irradiance estimated by the coupled system showed a strong dependence on both the aerosol spatial distributions and the associated optical properties. In the NF (No Feedback) case, which refers to the WRF-only stimulated system without aerosol feedbacks, the GHI was overestimated by 50–200 W m-2 compared with OBS derived values at each site. In the YF (Yes Feedback) case, in contrast, which refers to the WRF–CMAQ two-way coupled system, the rRMSE was significantly improved by 3.1–3.7% at Suwon and Seoul where the Particulate Matter (PM) concentrations, specifically, those related to the PM10 size fraction, were over 100 g m-3. Thus, the coupled system showed promise for acquiring more accurate solar irradiance forecasts.
