There are a number of methods to evaluate the quality of squid. However, when purchasing the fish, consumers and retails rely only on the sensory test and flavor in the field. Therefore, this study was aimed to prove relationship between scientific indicator and sensory test. Total viable cell count (TVC), viable cell count of Pseudomonas spp., pH and volatile basic nitrogen (VBN) were selected as scientific indicators and mesured during the storage of squid at different temperature. The squid was storaged at 3 different temperature (5oC, 15oC, 20oC). Off flavor determination time was measured by R-index, and kinetic modeling was conducted. Activation energies of offflavor determination time, TVC, Pseudomonas spp, VBN, and pH were 51.210 kJ/mol, 42.88 kJ/mol, 50.283 kJ/mol, 72.594 kJ/mol and 41.99 kJ/mol respectively. Activation energy of off-flavor determination time was approximated to viable cell count of Pseudomonas spp., TVC, pH and VBN as an order. Especially, viable cell count of Pseudomonas spp. had best match of the activation energy. Therefore, it was judged that indicator of off-flavor determine time was viable cell count of Pseudomonas spp..

This study on the intensive landscape management zone of Muju county in Korea was conducted for the purpose of first, developing landscape guidelines that take the current conditions of the landscape in the area and tourists’ requirements into account and second, exploring ways to implement them. The current conditions of the landscape resources in the area were analyzed by conducting a theoretical examination; a survey of the current status of the landscape; and a questionnaire survey of the local people, tourists, and public officials. Based on the result of those surveys, the guidelines for the landscape design of the intensive landscape management zone were set, and measures to implement them were explored. The results of this study are as follows: 1. For the building landscape of the Meeting plaza at Muju IC, a variable structure made of natural materials is suggested for the extended space in front of the mall, while an integrated design of street furniture and installation of a landmark and its design are suggested for the streetscape. 2. For the area of Bandi Land and Taekwondowon, improvement of the street structure for the main street and of the height of the buildings, taking the view of mountains into account, is suggested, and the design is also proposed. 3. For the Muju Resort and entrance to the Gucheondong valley of Muju, building planning that takes fine natural views into account, as well as reorganization of public and commercial signboards, are suggested, and the design is also proposed. 4. For the implementation of these plans, focus is placed on the current status of land use by zone. First, implementation of landscape projects and switching to a roundabout at the entrance are proposed for the Muju IC Meeting Plaza. Second, a district-unit plan is suggested for Bandi Land and Taekwondowon areas because new development projects are planned for them. Third, a landscape agreement is suggested for the Muju Resort and the area of entrance to the Gucheondong valley of Muju because those are already-developed areas where active commercial activities are performed.

This plan aims to seek ways of regeneration in active utilization of local assets’ potentials such as historic and cultural resources and human resources as well as natural landscape resources of Samnye-eup, which was prosperous as the largest station town during the Joseon dynasty period and as an agricultural center around Mangyeong plain during and after the Japanese colonial era. The plan consists of the theoretical investigation, analysis of current conditions, basic concept, and business plan in the order. As for the theoretical investigation, domestic researches on the use of local assets in the context of urban regeneration and existing plans for regeneration projects were examined to come up with ways of utilizing local assets for the Samnye-eup regeneration plan. In the analysis of current conditions, specific information on Samnye-eup was collected to verify the need for the general regeneration project that embodies original projects for local asset utilization in each district. As for the basic conception, this regeneration plan envisions the reutilization of three types of storage facilities, utilizing closed grain warehouses scattered about in that region. The focus of filling ‘Farming Storages’ is to strengthen the residents’ capability through creative education customized for each generation and class based on the characteristics of the education city and in association with the local university. Second, filling of ‘Sensible Storages’ has the objective of filling the largest posthouse town in Honam region, the historic place of food plundering during the Japanese colonial era, with sophisticated cultural arts. Third, filling of ‘Food Storages’ is to establish a cooperative economic community in harmony with the local food, natural landscape, and cultural environments to overcome the general agricultural decline. The business plan includes collecting opinions from and discussing with local residents and related divisions in addition to expert forums. The filling of three types of storages are classified to the physical regeneration project, social regeneration project, and economic regeneration project, each of which forms a cycle in mutual complementation. In addition, this plan seeks synergy effects by complementing existing projects conducted by the central division and Wanju-gun.

Proline (Pro) accumulation is a common physiological reaction in response to abiotic stresses in many plants. Accumulation of Pro is believed to play the important role in protecting cellular components from dehydrating effects due to such stresses. The study was performed to investigate the relationship between cold hardiness and Pro content or expression of related genes in peach cultivars during a constant experimental deacclimation. Changes in cold hardiness were determined using electrolyte leakage method in the shoots of 10 peach cultivars (Prunus persica ‘Aikawanakajima’, ‘Chiyomaru’, ‘Daewol’, ‘Janghowon Hwangdo’, ‘Kiraranokiwami’, ‘Mihong’, ‘Misshong’, ‘Soomee’, ‘Suhong’, and ‘Sun Gold’). Pro content was analyzed using the ninhydrin method and related gene expressions were examined using quantitative real-time RT-PCR. While cold hardiness of 10 peach cultivars decreased, Pro contents of those increased during the deacclimation. Notably, at the same time, expression of P5CS (Δ1-pyrroline-5-carboxylatesynthase) decreased in 10 peach cultivars, whereas expressions of P5CR (Δ1-pyrroline-5-carboxylatereductase) and OAT (ornithine-δ-aminotransferase) increased. Our results demonstrate that Pro responds positively to higher temperature in the shoots of 10 peach cultivars and expression of both P5CS and P5CR genes could show contrasting patterns during the deacclimation. Furthermore, our results suggest that ornithine pathway, which has been suggested to be important during seedling development, could serve as an alternative pathway in Pro synthesis process during the deacclimation in peach.

Changes in cold hardiness were confirmed with relative electrolyte leakage (REL) method in the shoots of two peach cultivars (Prunus persica Janghowon Hwangdo and Odoroki) during cold acclimation and deacclimation. Changes in proline (Pro) content and related gene expressions were also analyzed. Particularly, transcript accumulations of P5CS and P5CR were examined using quantitative real-time RT-PCR. REL in the shoots of two peach cultivars was significantly different during the entire experimental period. Cold hardiness of two cultivars increased gradually to December 2012, and then decreased to April 2013, whereas Pro contents of ‘Janghowon Hwangdo’ and ‘Odoroki’ were reduced from the beginning of the experiment to February 2012 and then increased in the spring. Interestingly, P5CS gene encoding an enzyme, which catalyzes conversion from glutamic acid (Glu) into glutamic-γ-semialdehyde (GSA) in the first step of Pro pathway, showed the contrasting patterns with Pro contents of two cultivars. On the other hand, P5CR gene encoding an enzyme, which catalyzes conversion from Δ1-pyrroline-5-carboxylate(P5C)intoPro in the final step of Pro pathway, showed the similar patterns to Pro contents in two cultivars. Our results demonstrate that Pro responds negatively to low temperatures in the shoot of different peach cultivars, including the supplemental 10 peach cultivars, and expression of both P5CS and P5CR genes could show contrasting patterns from each other. Our results suggest that identification of both P5CS and P5CR genes are required necessarily for accurate analysis of Pro biosynthesis because Pro accumulation is affected more by expression of P5CR gene.
