This work was conducted to identify sweet potato [(Ipomoea batatas (L.) Lam.)] cultivars showing high yield and processing suitability in the Jeonbuk region of Korea. Shoot-slips (30 cm in length) of 14 cultivars were planted with a planting density of 75×25 cm on May 30, and cultivated with black film mulching. Vine elongation at 30 days after planting was significantly increased (> 90 cm) in ‘Dahomi’ and ‘Sinhwangmi’, and was excellent (> 250 cm) 120 days after planting in ‘Sinhwangmi’, ‘Shinyulmi’, ‘Daeyumi’, ‘Jinhongmi’, and ‘Yeonhwangmi’. The branch number per plant showed a dramatic increase in ‘Shinzami’ 30 and 120 days after planting. The leaf number per plant showed the highest increase in ‘Shinzami’ 30 days and in ‘Sinhwangmi’ 120 days after planting. Total weight, mean weight, and number of marketable tubers per plant were significantly higher 150 days after planting than that 120 days after planting. Marketable tuber yield at 120 days was high, over 2.5 ton/10a in ‘Daeyumi’, ‘Dahomi’, and ‘Jeonmi’, and was considerably increased, from 3.2 ton/10a to 3.5 ton/10a, 150 days after planting. Total weight of marketable tubers per plant and marketable tuber yield 120 and 150 days after planting had significant positive correlations with vine length and number of nodes 30 days after planting. Starch value was significantly higher 150 days after planting than that 120 days after planting. ‘Daeyumi’ and ‘Jeonmi’ had good growth vigor, high yield, and high starch content; ‘Dahomi’ also exhibited beneficial traits such as good growth vigor, high yield, and bright orange-colored flesh.
Small-sized tuber of sweet potato (Ipomoea batatas Lam.) is more preferred by Korean consumers, because it is convenient to eat with hands, and can easily be steamed or roasted in small pan as a healthy snack for a small family. The present study was aimed to investigate the effect of highdensity plantings and harvesting times on production of smallsized tuber (50~200 g). Four varieties (‘Annobeny’, ‘Daeyumi’, ‘Shinzami’, and ‘Yeonhwangmi’) were planted in 2-row zigzag high-density planting (75×30 cm, A; 75×25 cm, B) and 1-row planting (75×25 cm, control) covered with black vinyl film on May 23th, 2014. Marketable yields and small-sized tuber yields were compared between 120- and 150-day harvesting. Vine length and fresh weight per plant were significantly decreased in 2-row planting A and B plots compared to the control at 120 days after planting. Weight of tuber per plant, mean weight of tuber and number of tuber per plant were significantly decreased in 120-day harvesting of A and B plots than in the control. Marketable yields per 10a were increased by 17% (2.4 ton‧10a -1 ) and 8% (2.6 ton‧10a -1 ) in 120-day harvesting of A and B plots compared to the control (2.4 ton‧10a -1 ), and those were largely increased by 29% (4.0 ton‧10a -1 ) and 26% (3.9 ton‧10a -1 ) in 150-day harvesting than in the control (3.1 ton‧10a -1 ). Proportions of small size tuber (50~200 g) were increased by about 65% in 150-day harvesting of A and B plots compared to the control (55.3%). Small-size tuber yield and marketable yield were highly increased in 150-day harvesting of 2-row zigzag high-density planting A (75×30 cm).
바이러스 무병묘의 만기재배를 통한 우량 씨고구마의 안정적 생산과 적응품종 선발을 위하여, 9 품종의 바이러스 무병묘와 농가묘를 7월 10일 75 × 25 cm로 정식하여 흑색비닐로 멀칭재배를 한 다음, 생육특성 및 수량 등을 조사하였다.1. 정식 30일째의 초기생육은 농가묘보다 무병묘에서 줄기 길이, 줄기 직경, 마디수, 곁가지수 등이 유의하게증가하였고, 품종 간에도 유의한 차이를 보였으며, ‘고건미’, ‘신천미’, ‘신황미’, ‘신율미’, ‘연황미’ 등에서 초기생육이 양호하였다. 2. 정식 110일째의 지상부 생육에서 줄기 길이, 줄기 직경, 마디수, 분지수 및 생체중 등은 품종간에 유의한차이를 보였으나, 무병묘와 농가묘간에는 마디수 외에다른 형질들은 유의한 차이를 보이지 않았다.3. 만기재배에 따른 무병묘 수량은 품종에 따라 농가묘보다 12-49% 증수되었으며, 무병묘와 농가묘간 평균수량은각각 1,625 kg/10a과 1,230 kg/10a 으로 무병묘에서 유의하게 증가하였다.4. 씨고구마로 이용할 수 있는 40 g 이상의 상저비율은 무병묘 65.6%, 농가묘 57.8%로 무병묘에서 유의하게 증가하였으며, 무병묘와 농가묘간 상저수량은 각각 1,067 kg/10a과710 kg/10a 이었다.5. 공시된 9품종 중에서 1,300 kg/10a 이상의 상저수량을보인 ‘신자미’, ‘신천미’, ‘신율미’ 등 3품종의 만기재배적응성이 높았다.
Haploid system by anther culture allows the development of homozygous lines when doubled. The response of anther culture to Basta (glufosinate) resistance was investigated on transgenic plants (cv. Anjungbyeo) in order to identify inheritance of bar gene associated with Basta. Most of the regenerated transgenic plants were sterile, and only a few plants produced viable seeds (A1) in the greenhouse. The bar gene was analysis by PCR in basta resistant transgenic plant (TA0). The transgenic seeds (A1) were significantly germinated in Basta solution compared with non-transformed seeds. As a result of anther culture, in regenerated haploid plants, segregation ratio was 1:1 in five of eight cross combinations. In diploid plants, segregation ratio was 1:1 in seven of eight cross combinations. Although there was some differences in the cross combinations, most of the combinations had 1:1 segregation ratio which supports the theory. The difference may be a result of the small sample size or the difference of anther culture response caused by genotypic difference. Hence, when many cross combinations were anther-cultured the results would support the theory.
This study was conducted to investigate the crossability, seed dormancy and overwintering ability of rice plant in GM (glufosinate ammonium-resistant lines. Iksan 483 and Milyang 204) and non-GM (their parents) or red rice (Andongaengmi). Seed-setting rate was not significantly different between GM and non-GM rice varieties. Iksan 483 and Milyang 204 showed the similar level of seed germination rate from 30 to 50 days after heading as compared to non-GM rice varieties. After overwintering in paddy field, seed germination rate of GM and non-GM rice varieties ranged from 14.3 % to 57.6 % in dry soil condition, but there was no germination in wet-soil except red rice. The result in wet-soil condition may help to set up a strategy for reducing the risk of gene flow of transgene via dispersal of seeds of GM plants. The crossability, seed dormancy and seed overwintering of Iksan 483 and Milyang 204, herbicide resistant GM rice varieties, were not significantly different compared to non-GM rice varieties. The results might be helpful to reduce the risk of transgene dispersal from GM crop via seeds and pollens.
Aloe in vitro culture was attempted to induce callus and regeneration ability from different explant sources onto MS medium with 0.5mg/l NAA plus 1.0mg/l BA. Anthers that no developed any callus and plant regeneration, while only four out of 274 filament explants induced calli at cut edge without regenerated plants. Twenty ovary explants regenerated four direct plantlets without via callus from the base of epidermal tissues. Regenerated plants on the root tip gave 2n=14 of chromosome numbers.
