본 연구에서는 국산과 중국산 능이버섯과 송이버섯의 일반성분과 아미노산 조성을 분석하여 채취지역에 따른 능이버섯과 송이버섯의 식품학적 품질을 비교하고자 하였다. 송이버섯의 조회분 함량은 중국 티벳산이 5.52%, 운남 5.40%, 연길 6.95%, 한국 양양 6.40%로 시료 간에 차이를 나타내었다. 조지방 함량은 한국 양양산이 1.19%로 가장 낮았고, 조단백질 함량은 중국 티벳산이 16.83%로 가장 낮아 시료 간에 차이를 나타내었다. 송이버섯의 총아미노산 함량은 중국 연길 11,490.14±892.07 mg%, 중국 운남 8,000.03±2072.65 mg%, 티벳 6,815.48± 771.82 mg%, 한국 양양 6,074.74±814.86 mg% 순으로 나타났다. 능이버섯의 조회분, 조지방, 조단백질, 총아미노산 함량은 한국산과 중국산 시료 간에 차이를 나타내지 않았다. 본 실험결과 조회분 함량을 중국 남방지역(티벳, 운남) 송이버섯과 한국 양양 및 중국 북방(연길) 송이버섯의 구분지표로 활용할 수 있을 것으로 사료되며 무기질 성분에 대한 추가 실험을 통해 지표 무기질 도출이 필요한 것으로 판단된다.
본 연구에서는 이산화탄소 흡수/재생 공정에 효율적으로 적용할 수 있는 아미노산염 흡수제의 연속재생을 통해 재생효율을 확인하였다. 재생효율은 공정적용에 있어 경제성에 큰 영향을 끼치는 인자로, 보다 경제성 있는 이산화탄소 흡수/재생 공정 확립을 위해 연속재생 실험을 진행하였다. 실험에 사용한 아미노산염은 Potassium L-lysinate와 Potassium L-alaninate이며, 각 아미노산과 Potassium hydroxide(KOH)를 1:2 몰비로 혼합하여 사용하였다. 흡수제의 재생 효율을 확인하기 위해 두 물질에 이산화탄소를 충분히 흡수시킨 후 가열을 통해 이산화탄소 탈리실험을 진행하였다. 반응초기에는 L-alanine의 반응속도가 빠르게 이루어졌으나, 시간이 지남에 따라 흡수량이 보다 큰 L-lysine이 높은 농도의 이산화탄소를 배출하였다. 두 물질의 재생효율을 비교하였을 때, L-alanine은 47.26%, L-lysine 은 62.11%로 L-lysine이 더 높은 재생효율을 나타내었다. 흡수량 및 재생효율이 좋은 L-lysine을 이용한 연속재생 실험결과, 재생횟수가 증가함에 따라 재생효율이 감소하는 것을 확인할 수 있었다.
The objective of this study was to determine the changes in content of polyphenolics, flavonoids, amino acids, especially γ-aminobutyric acid (GABA), and the antioxidant activities in the methanol extracts of four different pigmented rice seeds (white, black, red, and green color) at different germination stages, dry seeds (DS), imbibed seeds (IS), and germinated seeds for 1 (GS1), 2 (GS2) and 3 days (GS3). Total phenolics content at 1000 mg/kg extract was highest in black rice, ranging from 208.3 to 257.3 mg/kg, and followed by red rice 144.7 to 185.7 mg/kg, brown rice 151.3 to 194.2 mg/kg and green rice 125.7 to 160.7 mg/kg (lowest). Total flavonoid level had same tendency to total phenolics among colored rices with significant differences, showing highest amounts (45.2 to 56.8 mg/kg) in black rice and lowest (18.8 to 23.0 mg/kg) in green rice. Total phenolics and total flavonoids levels in black and brown seeds were highest when soaked, and in red rice when nonsoaked, and in green rice when germinated for 1 day. These results showed that content of physiologically- active substances were differ depending on seed coat color, soaking or germinating degree. Content in total amino acids during germination was ranged from 136.5 to 232.0 mg/kg in green rice (highest), and 98.9 to 176.0 mg/kg in red rice (lowest). Total content of amino acids and GABA level in all seed colors were high in order of GS3 > GS1 > IS > DS. The antioxidant activity of the methanol extracts from all the seed colors concentration-dependently increased. DPPH free radical scavenging activity at 2,000 mg/kg was highest in black rice by 83.0 to 87.7%, and lowest in green rice by 44.3 to 65.7%.
Compound specific isotope analysis of amino acids (CSIA-AAs) is being highlighted as an alternative approach for overcoming some restrictions in application of stable isotope analysis of bulk tissue (SIA) for trophic position (TP) estimation. However, this approach has rarely been applied in Korea. The present study determines TP of two Polychaeta (Nephtyidae and Glyceridae) and two fish species (Platycephalus indicus and Lophius litulon) collected from the Geum River estuary using nitrogen isotope ratio of amino acid and compared with the TP values estimated by SIA. The Polychaeta species, sampled in two sites, showed similar TP between SIA (2.7 and 3.1) and CSIA-AAs (2.6 and 3.1). However, for both fish species, TP values displayed a large difference between SIA (3.1 and 2.3) and CSIA-AAs (3.8 and 3.7). In this study TP values estimated by CSIA-AAs showed more similar to the previously reported gut content analysis of both fishes compared with the results of SIA. Current study suggests the applicability of nitrogen isotope ratio of amino acid to understand coastal ecosystem structure and trophic ecology.
This study was conducted to investigate the proximate composition, free sugars, amino acids, and minerals in five Lentinula edodes cultivars collected in Korea. No significant differences in the content of crude fat and crude fiber were found between the samples; however, there was significant variation in the contents of nitrogen free extract, ash, and crude protein. Three kinds of free sugars (fucose, arabinose, and glucose) were identified by HPLC, with the glucose content ranging from 5.94% to 12.08%. Sixteen kinds of free amino acids were identified: the highest content of amino acids was found in ‘Sanlim5ho’ (13,768.33 mg%), while the highest content of free amino acids was found in ‘Sanlim4ho’ (6,790.57 mg%). The minerals detected in the L. edodes cultivars were potassium, calcium, magnesium, and sodium, the contents of which ranged from 567.16–2,356.09 mg%, 3.85–7.42 mg%, 9.79–20.88 mg%, and 19.60–22.62 mg%, respectively.
The objective of this experiment was to investigate amino acids and mineral compositions of the four major muscles (LD: longissimus dorsi, PM: psoas major, SM: semimembranosus, and GM: gluteus medius) from Korean native black goat (KNBG). Five uncastrated male KNBGs of 36 months of age were commercially slaughtered and the four muscles were sampled to determine concentrations of myoglobin, collagen, amino acids and minerals. There were significant differences (p<0.05) in myoglobin and collagen content among the four muscles. Myoglobin content of PM was significantly higher (p<0.05) than those of SM and GM. Collagen content of SM was significantly higher (p<0.05) than that of LD. There were significant differences (p<0.05) in cystine and leucine concentrations among the four muscles, and PM had the highest concentrations of the two amino acids compared to other muscles (p<0.05). Also, there were significant differences (p<0.05) in K, P, Mg and Fe contents among the four muscles from KNBG. PM had the highest mineral content while the lowest mineral content was observed in LD. In addition, Fe contents of PM and GM were higher than those of LD and SM (p<0.05). These results indicated that amino acids and mineral content vary considerably with the anatomical location of muscles of Korean native black goat.
The present study investigated the effects of dietary supplementation of purified amino acid (PAA) isolated and purified from animal slaughter house blood on growth performance and immune response of weanling piglets. A total of fifteen, 28 days old, weaned pigs were randomly distributed into three treatment groups, 5 pens per treatment (one head per pen). Each experimental diet was provided to five piglets per group (T0, 0% PAA; T1, 0.1% PAA; T2, 0.5% PAA). The feed consumption was calculated daily and body weight on a weekly basis. The blood samples were analyzed for plasma concentration of biochemical parameters and cytokines using ELISA Kit assay. Pigs fed with the 0.5% PAA have greater average daily weight gain (ADG) and feed efficiency (FE) as compared to those of T0 and T1. However, ADG and FE of T0 and T1 have no remarkable differences. The plasma cytokine levels were lower in T2 as compared to T0 and T1. The blood parameters like total bilirubin, blood urea nitrogen (BUN), glutamate oxalate transaminase (GOT) and glutamate pyruvate transaminase (GPT) were within the normal range in all the treatment groups. The study indicated that supplementation of 0.5% PAA have positive effect on the growth performance of the weanling pigs. Moreover, supplementation of the amino acid isolated and purified from animal slaughter house blood has no adverse effect on palatability of the feed and health of the animals.
감은 우리나라 주요 과실의 하나로 무기질과 비타민 A, B1, C가 풍부할 뿐 아니라 기능성 페놀화합물이 다량 함유되어 있다. 이러한 영양학적 가치가 높음에도 불구하고 주로 생과와 곶감으로만 소비되고 있는 형편이다. 최근에 감을 활용한 새로운 용도 창출을 위한 노력이 시도되고 있으나 여전히 미비한 실정이다. 따라서 본 연구는 감의 이용성 확대를 위한 기초자료로 감의 품종별(상주둥시, 상감둥시, 도근조생, 청도반시), 부위별(과피, 과육)에 따른 유리당, 유기산, 유리아미노산, 카로테노이드 함량을 분석하였다. 그 결과, 감의 주 유리당은 과당과 포도당이었으며 과당 보다는 포도당의 함량이 높은 편이었다. 감 품종별 유리당 총 함량은 과육과 과피 모두 청도반시가 가장 높은 반면에 상감둥시가 가장 낮은 함량을 보였다. 감의 유기산 함량은 사과산이 가장 높은 함량을 보였고 과피에서는 옥살산이 검출되지 않았다. 감 품종별 과육 및 과피의 유리아미노산은 시료간의 차이를 보였으나 일정한 경향을 보이지는 않았다. 감의 카로테노이드를 분석한 결과 caxpanthin, zneaxanthin, β-cryptoxanthin, β-carotene, lycopene이 검출되었고 카로테노이드 종류에 따라 감 품종별, 부위별로 그 함량의 차이를 보였다. 이와 같은 결과로 보아 감의 품종이나 부위에 따라 영양성분의 차이를 보이는 것을 확인하였고 향후 감의 식품소재화를 위한 자료로 활용될 것으로 생각된다.
고압균질화 기반 nanosuspension화은 난용 혹은 불용인 기능성 물질의 가용성을 높이는 기술로서 식품산업체로부터 관심을 받고 있다. 그러나 nanosuspension은 나노 크기로 분산된 난용성 입자들은 응집 또는 Ostwald ripening 현상에 쉽게 노출되기 때문에, 저장안정성이 낮다. 그러므로 이러한 nanosuspension에 유화제 및 고분자 물질 등의 분산안정제를 도입함으로써 위의 현상을 지연시키거나 방지하기 위해 반드시 필요하다. 이 연구는 분산안정제의 특성이 nanosuspension화된 분지쇄아미노산의 용해도 및 저장안정성에 미치는 영향을 평가했다. Tween 80, Span 80 또는 lysolecithin을 10 mM 인산 완충 용액(pH 7)에 녹이고, pH를 3 또는 6으로 보정하여 안정제 용액을 제조하였다. 분지쇄아미노산 혼합물(L-leucine:L-isoleucine:L-valine=2:1:1)을 최종 농도가 5%(w/v)가 되도록 안정제 용액에 첨가 하였다. 모든 분지쇄아미노산 현탁액을 25°C에서 2 시간 동안 교반하였다. 분지쇄아미노산를 함유한 나노서스펜션을 제조하기 위해서 70°C로 예열된 분지쇄아미노산 현탁액을 고압균질화기를 사용하여 100 MPa에서 5 회 균질화를 진행하였다. pH에 관계없이 고압균질화를 통해 분지쇄아미노산의 가용성을 증가되었지만, 가용성이 장기간 동안 유지 되지 않았다. 분산안정제에 의해 분지쇄아미노산의 포화 농도는 증가하였고, 분지쇄아미노산의 포화 농도 증가에 대한 안정제의 영향은 pH 6에서 더 확연했다. pH 6에서 Tween 80은 분지쇄아미노산와의 혼합 비율과는 무관하게 분지쇄아미노산 nanosuspension의 저장안정성을 크게 증가 시켰지만, pH 3에서는 그렇지 않았다. 그러나 lysolecithin과 Span 80을 적용한 분지쇄아미노산 nanosuspension의 저장안정성은 pH와 안정제의 혼합 비율에 따라 영향을 받았다.
To date, many researches have been done for maize QPM lines, there is little maize QPM varieties in Korea. In this reason, to develop maize QPM cultivars, investigation of the growth characteristics and amino acid composition among QPM inbred lines was carried out. For this study, QPM inbred lines were donated from CIMMYT. Analyses of growth characteristics changes for each line were investigated. The range of siliking days were ranged from 60 to 69 days. Silking days for 4 QPM lines such as CML140, CML141, CML152, and CML493 were 60 days among donated QMP lines, CML557 was 69 days. In plant height, CML492 was shortest(152 cm) while CML143 was the longest(295 cm). The ear height of CML492 was 66 cm and 175 cm for CML143. Protein content of each maize QPM grains was from 9.07% to 12.67%. CML556 have the highest proteins content, while CML493 was the lowest among maize QPM grains. Content of fat was from 3.38% to 5.25%, and CML141 was the lowest and CML147 was the highest. The content of ash ranged from 1.22% to 1.78%, CML555 was the least and CML142 was the highest. The content range of carbohydrate was from 71.7% to 75.5%, CML557 was the least and CML493 was the highest. The calories of each inbred line ranged from 346 to 365 Kcal, CML140 was the least and CML492 was the highest. Major amino acid composition in maize QPM grain is glutamic acid (16.34%), proline (9.80%). The mean of lysine composition was 3.64%, and CML140 showed the highest ratio of 4.34%.
This study investigated the crude protein and amino acid contents of local agricultural products widely and specifically grown in Korea, including 25 vegetables and 13 fruits. The crude protein content of vegetables and fruits ranged from 0.46 to 6.53% and 0.29 to 2.23%, respectively. Totally, 17 types of amino acids were found in most samples. The total amino acid content of vegetables and fruits ranged from 457.38 to 9,303.18 mg% and 368.82 to 3,118.75 mg%, respectively. The total amino acid contents of garlic and passion fruit was higher compared to other vagetables and fruits. The calibration curves of the standard components showed good linearity (r2>0.99), except Met (r2=0.989). The limits of LOD and LOQ were in the range 0.034 to 0.991 μg/mL and 0.009 to 0.474 μg/mL, respectively. The results of the study can serve as a fundamental source of information regarding crude protein and amino acids contents in food, for diet planning.
Sparassis latifolia (formerly S. crispa) is used in food and nutraceuticals or dietary supplements, as rich in flavor compounds and β-glucan. Some previous studies have reported the effects of mushroom on brain function, including its neuroprotective effect. Thus, for this mushroom to be used as an effective nutraceutical for brain function, it would be desirable for it to contain other compounds such as γ-aminobutyric acid (GABA) in addition to β-glucan. In this study, the enhancement of growth and GABA production in the mycelium of medicinal and edible mushroom S. latifolia was investigated. Amino acids were added externally as the main source of nutrition, and the effects of amino acids were investigated using liquid medium, specifically amino acid-free potato dextrose broth (PDB). The amino acids added were L-glutamic acid (named PDBG medium) and L-ornithine (named PDBO medium). The growth of mycelia was determined to be 0.9 ± 0.00 g/L, 2.2 ± 0.16 g/L, and 1.93 ± 0.34 g/L PDBG respectively. The GABA content was 21.3 ± 0.9 mg/100 g in PDB medium, and it in PDBG 1.4% medium, at 115.4 ± 30.2 mg/100 g. However, the PDBO medium was not effective in increasing the GABA content of mycelia. Amino acids had little effect on the β-glucan content of mycelia. The β-glucan content was 39.7 ± 1.4 mg/100 mg, 34.4 ± 0.2 mg/100 mg, and 35.2 ± 9.2 mg/100 mg in PDB, PDBG 1.8% and PDBO 1.4% media, respectively. Addition of glutamic acid and ornithine positively affected the growth of S. latifolia mycelia, and glutamic acid positively affected GABA production; no degradation of GABA was observed with addition of glutamic acid.
In this study, we investigated the amino acids content of shiitake mushrooms grown in eight different media and under three different temperature conditions. The sawdust media were supplemented with 4 kinds of nutrients - rice bran, barley, bean curd refuse, and red ginseng refuse in an 8:2 ratio with or without 0.5% calcium chloride, and the fruiting temperature was set to low (13–15oC), medium (18–20oC), or high temperatures (24–26oC). Seventeen amino acids were detected, and the content varied depending on the compositions of the media. The highest total amino acid content (209.7 mg/g) was measured in mushrooms from the media with red ginseng refuse, and the glutamic acid content was the highest (51.3 mg/g) in mushrooms from the media with barley. In terms of fruiting temperature, the amino acid content was the highest (38.2%) at medium temperature followed by the content at low (32.5%) and high temperature (29.3%). The amino acid composition of shiitake mushrooms was similar between the medium and low temperature conditions.
The objective of this study was to investigate the effect of amino acid and vitamin C supplementation on the quality attributes of top round (TR) muscle from Korean cattle (Hanwoo). Sixteen-month-old steers were divided into three different feeding groups and raised until they were 28 months old. A control group (9 heads) was fed a basal diet and rice straw, whereas two experimental groups (8 heads/group) were fed a basal diet and rice straw supplemented with rumen-protected amino acids (≥16.0% L-lysine monohydrochloride, ≥8.0% DL-methionine) and vitamin C at a dose of 100 g/head/day (AA+VC group) or only vitamin C at 100 g/head/day (VC group). After slaughter, TR muscle (M. semimembranosus) was collected from carcasses and used to measure the quality attributes. Crude fat content was significantly higher (p<0.05) in the AA+VC group than in the VC group. No significant difference was observed for water-holding capacity and shear force value between the control, AA+VC, and VC groups. Contents of the free amino acids (FAAs) glycine, aspartate, and glutamate were significantly higher (p<0.05) in the VC group than in the control group. Sensory evaluation of tenderness and flavor preference revealed significantly higher scores (p<0.05) in the AA+VC group than in the control group. These findings suggest that vitamin C supplementation increases the contents of some FAAs, and the combined supplementation of amino acids and vitamin C improves the marbling and sensory attributes of TR muscle in Hanwoo.
The purpose of this study was to investigate the chemical contents (sugar, nitrogen, and amino acids) and sensory characteristics of traditional soy sauce according to three different methods. The samples were taken from four different periods (0, 20, 40, and 60 days after fermentation). Total sugar contents increased in all groups according to ripening period, and Gyupjang (G) showed the highest sugar content among the groups. Total nitrogen and amino acid contents of all groups increased after 60 days of ripening, and Gyupjang (G) and Gyupjang mixed with salt water (SG) had higher total nitrogen content compared to Chungjang (S) at the same period. The results from the sensory evaluation show that preferable sensory characteristics, such as color preferences, sweetness, umami taste, and overall preference, were significantly higher in Gyupjang (G) and Gyupjang mixed with salt water (SG) than in Chungjang (S). Preferable sensory characteristics had significantly high positive correlation with most amino acids, total nitrogen, and sugar contents, except for glutamine. Based on the significant difference in preferable sensory characteristics between Gyupjang mixed with salt water (SG) and Chungjang (S), Gyupjang mixed with salt water (SG) can be used to improve sensory characteristics. This research implies that adding salt water during the manufacturing Gyupjang process is advisable to yield high quality soy sauce.
본 연구결과 glucose, NH3, AST, ALT는 4℃일 때 가장 높게 나타났으며, glucose와AST는 4℃일 때 다른 염분구보다 26 psu에서 유의하게 높았다. 참전복에는 총 18가지 FAA가 존재하였으며, taurine, arginine, glutamic acid, alanine, glycine, lysine이 전체의 90% 가량을 차지하는 우점 FAA 로 나타났다. Taurine은 30 psu 이상에서는 수온 간 유의한 차이가 없었으나, 26 psu에서는 6, 8℃일 때 유의하게 증가하였다. Glycine은 수온이 낮아질수록 유의하게 감소하여 6℃일 때 가 장 높게 나타났다. 따라서 참전복은 30~34 psu, 8~10℃에서의 조건으로 수송 시 스트레스를 최소화할 수 있을 것으로 여겨진다.
국내외에서 수집한 표고 품종을 대상으로 영양성분 및 β-glucan 함량을 분석한 결과 수집한 표고의 유리당 함량 을 분석한 결과 총 4종의 유리당이 검출되었으며, 그 중 trehalose 함량은 시료별로 0.83 ~9%까지 큰 차이를 보였 다. 구성아미노산 함량을 분석한 결과 총 아미노산함량은 중국 톱밥배지 표고 JMI10050에서 17,672 mg%로 가장 높게 나타났으며, 필수아미노산의 함량은 국내 원목재배 표고 JMI10059에서 6,211 mg% 가장 높게 나타났다. 유 리아미노산 함량을 분석한 결과, 총 16종의 아미노산이 검출되었으며, 주요 유리아미노산으로는 histidine, glutamic acid 및 arginine으로 나타났다. 필수 유리아미노산인 threonine, valine, methionine, isoleucine, leucine, phenylalanine, histidine 및 lysine의 함량은 중국 톱밥배 지 표고 JMI10052에서 521.06 mg%로 가장 높게 나타 났다.
중국에서 수집한 표고에 비하여 국내에서 재배된 표고 의 β-glucan이 약 10% 가량 높게 검출되었고, 그 중 국내 원목재배 표고 JMI10066이 31.74%로 가장 높은 함량을 나타냈다.
