        The objective of this research was to evaluate the ability of water and ethanol extracts from mulberry fruit (Morus alba L.) to influence the inhibitory activity of angiotensin converting enzyme (ACE) and xanthine oxidase(XOase). The total phenol contents and sixteen phenolic compounds were investigated in water and ethanol extracts. In order to understand the factors responsible for the potent antioxidant and antihypertensive ability of mulberry, it has been evaluated for anti-oxidative activity using Fenton's reagent/ethyl linoleate system and for free radical scavenging activity using the 1,1-diphenyl-2-picryl hydrazyl free radical generating system. The total phenol contents and total of phenolic compounds in ethanol extract showed higher levels than water extract in mulberry fruit six phenolic compounds (chlorogenic acid, narigin, syringic acid, quercetin, naringenin, kampferol) has a higher individual phenolic compound content in the 60% ethanol extraction than 80% ethanol extract. The inhibitory activity on angiotensin converting enzyme (ACE) were highest in 80% ethanol extract (9.0%). Also, activity of xanthine oxidase(XOase) inhibition appeared highest in 80% ethanol extracts and correlated well with the total phenolic content, which was modulated by the concentration of individual phenolic compounds. This result revealed, that strong biological activity was caused by specific phenol compound contents. Utilization of water and ethanol extracts from mulberry fruit are expected to be good candidate for development into source of free radical scavengers and anti-hypertentive activity

        작약의 지상부인 잎과 줄기 부위가 함유하고 있는 주요성분인 paeoniflorin, methyl gallate, astragalin, kaempferol 등 4종의 화합물의 이용성을 높이기 위해 건조방법 및 건조 온도별 성분의 함량 변화를 검토한 결과를 요약하면 다음과 같다. 1. 건조방법별 성분 변화는 kaempferol, methyl gallate, astragalin, paeoniflorin 등 모든 성분은 열풍건조나 dry oven 보다도 원적외선 건조 또는 상온 음건에서 더 높았다. 특히 methyl gallate는 상온 음건에서 0.52%로서 가장 높았다. 2. 건조온도별 성분변화는 kaempferol, methyl gallate, astragalin, paeoniflorin 등은 40℃ 건조에서는 성분의 함량이 모두 높게 나타났으나 건조온도가 높아질수록 성분은 감소하는 경향이었다.

        A series of methanol extracts from leaf and root parts in spring- and winter-barley plants were assayed to determine their allelopathy and antioxidant activity. The methanol extracts applied on filter paper in a Petri-dish significantly inhibited root growth of Chinese milkvetch (Astragalus sinicus L.) seedlings. Leaf extracts at 25 and 50 g L-1 inhibited root growth of Chinese milkvetch seedlings more than root extracts. No difference in phytotoxic effects of spring- and winter-barley seedlings extracts on root growth of Chinese milkvetch was observed. Methanol extracts dose-dependently increased DPPH free radical scavenging activity in vitro. DPPH free radical scavenging activity was higher in the methanol extracts from winter-barley seedlings than in those from spring-barley seedlings, and from leaf extracts than from root extracts. The antioxidant potential of the individual fraction from the methanol extracts of spring-barley seedlings was in order of n-butanol>ethyl acetate>water>chloroform>n-hexane fraction. By means of HPLC analysis, spring-barley (200.62 mg 100g-1 ) had more amount of total phenol acid than winter-barley (114.08 mg 100g-1 ). Especially, ferulic acid was detected in spring-barley extract (183.46 mg 100g-1 ) as the greatest amount. These results suggest that early seedlings of barley plants had potent allelopathy and antioxidant activity, and their activities were differently exhibited depending on plant parts and growing condition.

        국내산 인삼의 부위별 quercetin, p-coumaric acid, maltol, esculetin의 함량을 측정한 결과 잎에서는 quercetin과 esculetin이 주근에서는 p-coumaric acid가 다량 함유되어 있었다. 이중잎의 EA 분획물은 melan-a 세포주에서 뛰어난 멜라닌 생성억제 활성을 보였으며, 이의 주요성분인 quercetin과 esculetin이 tyrosinase 활성억제도, melan-a 세포주에서의 멜라닌 생성억제도, UV-흡수도 측정에서 모두 우수한 활성을 나타내는 결과로 미루어 인삼의 잎은 피부미백을 목적으로 하는 기능성 소재로 이용될 수 있는 가능성을 가지는 것으로 사료된다.

        This objective of this study was to determined the activity of antioxidants of DPPH, SOD and total phenolic compounds in mushroom(P. eryngii spp.) extract. The antioxidant acitivity using DPPH showed that strain No. 2007(31.78~%) and strain No.2014(27.45~%) were the highest DPPH inhibition. Among the thirteen mushroom examined, strain No. 2014(58.30~%) and strain No. 2010(57.19~%) had the highest SOD activity. The highest total phenol content were in strain No.2013(1465.03~mug/g) and strain No.2005(1401.09~mug/g) . Our study suggest that it may be possible to improve mushroom with high natural antioxidant.

        본 연구는 단메밀과 타타리메밀의 페놀성화합물의 함량을 측정하고 분석, 평가한으로써 생리활성 연구의 기초 자료와 건강기능성 식품소재 개발에도 이용될 수 있는 유용한 기초 자료를 얻고자 수행하였다. 1. 총 페놀의 함량은 타타리메밀 종자가 단메밀 종자보다 약 2배정도 높게 나타났다. 총 페놀 중의 플라보노이드 함량은 단메밀이 약 50~% , 타타리메밀이 약 95~% 를 차지하고 있다. 단메밀과 타타리메밀의 총 페놀 및 플라보노이드 함량은 메밀쌀이 메밀껍질보다 높았으나, 단메밀의 플라보노이드 함량은 메밀껍질 더 높았다. 3. 단메밀의 rutin함량은 메밀껍질(25.2 mg/100g)>껍질을 벗기지 않은 종자(19.8 mg/100 g)>메밀쌀(12.8 mg/100 g)의 순으로 높았으나, 타타리메밀은 메밀쌀(2042.1mg/100g)>껍질을 벗기지 않은 종자(1375.8 mg/100g)>메밀껍질(138.7 mg/100 g)의 순으로 높게 나타났다. 식물체 부위별 rutin의 함량은 단메밀이 메밀싹>잎>줄기>종자의 순이고, 타타리메밀은 잎enli메밀싹>종자>줄기의 순으로 나타났다. 모든 부위에서 타타리메밀이 단메밀보다 높은 rutin함량을 나타냈다. 4. 단메밀과 타타리메밀 종자의 flavanols함량은 큰 차이를 보이지 않았으나, 잎, 줄기, 싹나물부위의 함량은 타타리메밀이 높은 경향을 보였고, 두 종의 flavonols의 함량도 단메밀보다 타타리메밀에서 월등히 높은 경향을 보였다. 5. 식물체 부위별 flavanols의 함량은 단메밀과 타타리메밀의 메밀싹>잎>줄기>종자의 순으로 높게 나타났다. 모든 부위에서 catechin> Epicatechin> Epicatechingallate의 순으로 높게 나타났으나, 메밀싹은 두 종 모두 Epicatechingallate의 함량이 종자의 약 30~~40 배, 잎, 줄기보다 약 15~~20 배가량 높았다.

        Common thistle contains water-soluble substances that are antioxidative to foods. Antioxidant activities measured by DPPH method for the ground samples were the greatest in leaves, although was less than that of commonly used antioxidants, BHT and ascorbic acid. Methanol extracts and fractions from Xanthium occidentale plants dose-dependently increased DPPH free radical scavenging activity, in vitro test. The extracts from leaves showed the strongest antioxidant activity. DPPH scavenging activity of the individual fraction was in order of n-butanol>water>ethyl acetate>n-hexane fraction. By means of HPLC analysis, leaf samples of Xanthium occidentale had the highest amount of phenolic compounds, related with antioxidant activity, and followed by stems and roots. Total content of these antioxidant phenolic com­pounds for leaves extracts were detected in water fraction (36.7 mg 100 g-1 ) as the greatest amount, especially chlorogenic acid (39.4 mg 100 g-1 ) was the greatest component. These results suggest that Xanthium occidentale plants had potent antioxidant activity, and their activities were differently exhibited depending on plant part and fraction.

        한국에 분포하고 있는 82종의 약용식물에 대하여 환경내성 및 기능성 소재의 개발을 위한 탐색을 위하여 항산화효소와 관련된 첫 번째 방어기작인 SOD 효소활성과 Phenol 화합물의 함량정도를 검정하여 본 결과 SOD 효소활성은 원지 (P. tenuifolia Willd.), 백편두 (D. lablab L.), 백개자 (S. alba L.), 대황 (R. palmatum L.), 감초 (G. uralensis Fisch), 연자육 (N. nucifera Gaertn), 지황 [R. glutinosa (Gaertner) Liboshitz], 현삼 (S. buergeriana Miq.), 인진 (A. capillaris Thb.)등에서 다른 약용작물보다 비교적 높은 SOD 효소 활성 능력을 나타냈다. Total phenol함량은 (3~249.731 μg/g)의 범위를 보였으며 지유(S. officinalis L.), 대황 (R. palmatum L.), 복분자 (R. chingii Hu)에서 각각 249.731 μg/g, 209.546 μg/g, 170.333 μg/g으로 가장 높은 함량을 나타냈다. 16종의 phenol 화합물의 경우에서는 82종의 약용식물이 각기 매우 상이한 농도의 분포를 나타내었다. 앞으로의 연구에서는, 항산화 효소 및 phenol 화합물이 많이 함유하여 우수한 기능성을 가지고 있는 것을 확인된 약용식물에 대해 그들의 이용성을 증대시킬 수 있는 구체적인 연구가 수행되어져야 할 것으로 사료된다. 또한 phenol성 물질을 많이 함유하는 약용식물 추출물을 이용한 기능성 식품, 화장품, 천연 보존재 및 의약품 등으로 개발 (Cook & Samman, 1996)할 수 있을 것으로 판단되어진다.

        The objective of this study was to determine the role of storing years with the variation of total phenol and individual phenolic compounds in soybean (Glycine max L.) seeds. The total phenol content varied from 0.36 to 0.42% over four years, with the highest value (0.42%) found at storage for two and three years. Among the nine soybean varieties examined, Daweonkong had the highest total average phenol content (0.58%). The total content of 11 phenolics varied from 730.0 to 1812.8 ~mu~textrmg~;~textrmg-1 over storage for four years, and the highest concentration (1812.8 ~mu~textrmg~;~textrmg-1 ) was found at storage for two years. Myeongjunamulkong (1465.4 ~mu~textrmg~;~textrmg-1 ) had the highest mean content among the nine soybean varieties. The total content of 11 phenolic compounds measured in this study occupied from 20.96 to 47.73% of the total phenol contents. The highest total phenol contents were in seeds with black coats (5279.4 ~mu~textrmg~;~textrmg-1 ), while the highest concentration of individual phenolic compounds were in seeds with green coats (1419.5 ~mu~textrmg~;~textrmg-1 ). Our study suggests that it may be feasible to improve soybean varieties with high functional substances such as phenolic compounds.

        Cereal grains are rich in phenolic compounds that give beneficial effect in human health. Although several research works have been reported on the effects of phytochemicals of plant origin, such as fruits, vegetables, few studies have examined the antioxidative effects of whole cereal grains. The objective of this study was to determine total antioxidant capacity of 80% ethanolic extracts of cereal grains by testing the ability of the extracts to inhibit UV -induced lipid peroxidation in vitro using linoleic acid in comparison to well-known antioxidant such as ascorbic acid and tannic acid. The total phenolic content of the cereal grain (80% ethanolic extracts) investigated in this study varied from 2.1 mg/g (wheat cv. Olgeurumil) to 10.4 mg/g (barely cv. Seodunchalbori). Highly positive relationship between total phenol compounds and antioxidant activity was found. When the antioxidant activities of all investigated extracts were measured with application of same quantity of phenol compounds, oat grain extracts showed similar antioxidant activity of barely cultivars. However, barely extract appeared as the most potent antioxidant activity of inhibition of UV -induced lipid peroxidation. This indicated that factors such as phenolic compound composition and their individual antioxidant activity could playa crucial role in the total antioxidant activity of cereal grains.

        The five phenolic compounds of peony root were isolated by Sephadex LH-20 column chromatography. Their chemical structures were identified by spectroscopic methods (UV, FT-IR, FAB-MS and 1H .&13C -NMR). The complete structures of these compounds were elucidated to be (+)-taxifolin-3-0-β -D-glucopyranoside, benzoic acid, gallic acid, (-)-epicatechin and (+)-catechin. The concentrations of five phenolic compounds in the peony root of three Korean cultivars (Youngchonjakyak, Euisungjakyak and Jomjakyak) were determined by reverse-phase HPLC. The constituents concentration in Youngchonjakyak were generally higher than in Euisungjakyak and Jomjakyak. The concentrations of (+)-taxifolin-3-O-β -D-glucopyranoside, benzoic acid, gallic acid, (-)-epicatechin and (+)-catechin in three different cultivars were ranged 0.23-0.52%, 0.20-0.30%, 0.26-0.28%, 0.09-0.12% and 0.34-0.63%, respectively.

        A supercritical fluid extraction was performed for the extraction of phenolics from grape seeds which up to now have been discarded. The optimum condition for extraction process was predicted through response surface methodology using central composit experimental design. The extraction amount of grape seed phenolics was increased by increasing extraction temperature, pressure, and concentration of co-solvent (ethanol). The optimum extraction conditions were 84.83C, 51.50MPa and 1.27% ethanol. The yield of phenolics using SFE was higher with 3 folds than ethanol and 4 folds than hexane but less than 80% methanol. In the respects of food poisoning, the approved solvents were restricted to ethanol and hexane. So, SFE for extraction of phenolics could be powerful alternative method for solvent extraction.

        Currently in Korea, standard operating procedure for the analyses of phenolic compounds in water is the spectrometric comparison of colors developed by 4-amino antipyrin with phenolic compounds. It is however that this method cannot identify individual compound and that some phenolic compounds do not react with 4-amino antipyrin. Spectrometric determinations of phenolic compounds were compared with chromatographic analyses of gas chromatography (GC) and high pressure liquid chromatography (HPLC) of various phenolic compounds. Individual phenolic compounds could be determined by both chromatographic methods but HPLC methods were more precise with lower detection levels in general.
