        In this study, we investigated the morphological characteristics and antioxidant ability of mushroom cultivar Lentinula edodes ‘Nongjingo’ fruiting bodies after exposure to various light conditions. Color differences between mushrooms treated with mixtures of LED light revealed that mushrooms displayed lighter color shades when compared to the control group (fluorescent light treated mushrooms). Redness increased and yellowness decreased after exposure to all treatments other than the fluorescent control. Measurement of growth characteristics of ‘Nongjingo’ fruiting bodies showed increases after exposure to all mixed LED treatments. In addition, the uniformity of fruiting bodies was higher when using LED light compared to fluorescent light. The measurement of stem diameters did not show a significant difference between the treatments, however, diameters were slightly larger with exposure to white-green LED. Moreover, stem length was longer in the mixed LED treatments when compared to those exposed to fluorescent light. Examination of the ratio of stem diameter to stem length revealed that the diameter of the stem was greater than the length. The antioxidant activity of water extracts made from Nongjingo fruiting bodies grown under mixed LED conditions was compared to those from mushrooms grown under fluorescence light conditions. The highest antioxidant activity was observed from mushrooms treated with white LED; however, no significant difference was found between mushrooms exposed to white-green LED compared to white-blue LED. The treatment showed higher antioxidant ability than vitamin C. Our results confirm that treatment of white LED and white-blue LED affects the growth and antioxidant ability of Nongjingo mushroom fruiting bodies.

        This study was conducted to investigate the proximate composition, free sugars, amino acids, and minerals in five Lentinula edodes cultivars collected in Korea. No significant differences in the content of crude fat and crude fiber were found between the samples; however, there was significant variation in the contents of nitrogen free extract, ash, and crude protein. Three kinds of free sugars (fucose, arabinose, and glucose) were identified by HPLC, with the glucose content ranging from 5.94% to 12.08%. Sixteen kinds of free amino acids were identified: the highest content of amino acids was found in ‘Sanlim5ho’ (13,768.33 mg%), while the highest content of free amino acids was found in ‘Sanlim4ho’ (6,790.57 mg%). The minerals detected in the L. edodes cultivars were potassium, calcium, magnesium, and sodium, the contents of which ranged from 567.16–2,356.09 mg%, 3.85–7.42 mg%, 9.79–20.88 mg%, and 19.60–22.62 mg%, respectively.

        We measured the antioxidant activities of Oak mushroom (Lentinula edodes) in browned sawdust medium under conditions of varying LED sources and amounts of light. Consequently, exposure to 200Lx blue LED resulted in highly efficient browning; the most efficient browning was shown at 200Lx, regardless of the type of luminous source. We identified that quantities obtained with the blue luminous source increased compared to those in other treatment plots. The DPPH radical scavenging test conducted to examine antioxidant activity revealed that the red luminous source caused high radical scavenging compared to efficient browning. The fruiting body for Nongjin-go, as a treatment plot with the highest scavenging, under a 400Lx red luminous source was 34.3±1.80% and that for ‘Sanjo 701’ at a 300lx red luminous source was 32.99±1.58%. The polyphenol content, reported to be correlated with DPPH radial scavenging, showed no similar correlation in the ‘Nongjin-go’ variety. By contrast, ‘Sanjo 701’ showed a similar association.

        Lentinula edodes are mainly used for cultivation and food in Korea. Consumption is gradually increasing with the characteristic smell and taste. In addition, anti-cancer effect, hypertension, atherosclerosis and other effects have been proven to be a functional food. The cultivation method of highland rice changed from lumber to sawdust bag. Thus, a browned step is required so that the surface of the medium can act like a tree epidermis. The browned medium is not easily contaminated with other bacteria even when in contact with the outside air. It is also a necessary step because it suppresses evaporation of water in the medium and helps mushroom production. But it takes a very long time to browning. Therefore, it is necessary to study the browning of the medium during the cultivation of the altitude. Therefore, the color of the Lab color was measured using a colorimeter to determine the browning efficiency of the medium by LED light source type and intensity. The varieties were Nongjingo and Sanjo-701. The LED light sources were divided into upper, middle, and lower by setting the blue, green, white, and red colors at 100 to 500 ㏓. L values of light source showed the greatest decrease in blue color, followed by white, green and red. It was confirmed that the L value of Sanjo 701 was lower than that of Nongjingo. The decrease of L value by the type of light source showed the same tendency as Nongjingo, and blue was the most decreased, followed by white, green and red.

        This study was conducted to investigate the temperature and accumulated temperature of Lentinula edodes strains from primordial occurrence to fruit body maturity. The experimental strains were Sanjo 108 (high temperature type), Sanjo 302 (middle temperature type) and Sanjo 502 (low temperature type). When the Sanjo 108 and Sanjo 302 occurred, the daily mean temperature inside the L. edodes inoculated logs was 17.9 ~ 20.5 °C, which was lower than air temperature by 0.7 ~ 1.15 °C. When the Sanjo 502 occurred, the daily mean temperature inside the L. edodes inoculated logs was 3.6 ~ 16.6 °C, which was lower than air temperature by 0.1 ~ 0.7 °C. The difference between the maximum and the minimum temperature when the fruit body occurred was 4.4 °C inside the inoculated logs and 6.9 °C in the air temperature in Sanjo 108 and Sanjo 302, and in Sanjo 502 was 5.5 °C inside the inoculated logs and 9.1 °C in the air temperature. The number of days from primordial occurrence to fruit body maturity in Sanjo 108, Sanjo 302 and Sanjo 502 was 3.9 days, 5.1 days and 10.6 days, and accumulated temperatures of these strains were 104.9 °C, 129.2 °C and 141.4 °C, respectively. The number of days and accumulation temperatures were 518 days and 8,361 °C in Sanjo 108, 444 days and 6,475 °C in Sanjo 302, and 560 days 9,263 °C in Sanjo 502.

        At present, oak sawdust and rice bran are mainly used as the medium materials for bag cultivation of Lentinula edodes. This experiment was conducted to develop the optimum medium of ‘Nongjingo’, a new variety of Lentinula edodes. We investigated oak sawdust, oak fermented sawdust, poplar sawdust, and douglas fir sawdust as main materials, and rice bran and wheat bran as nutrient resources, respectively. The medium composition was consisted of main material 80%+nutrient 20% based on the volume ratio. Eight treatments of medium composition were tested. The period of mycelial incubation in ‘oak sawdust 40%+douglas fir sawdust 40%+rice bran 20% was 86 days (39 days in dark incubation and 47 days in browning) shortened up to 18 days compared to the control. The 1st yield of commercial fruiting bodies in this treatment was 48.1 g/bag which was 44.8% higher than that in control. We expect that the cultivation period of ‘Nongjingo’ will be shortened, and the yield increased in the medium replaced half of oak sawdust by douglas fir sawdust.

        The purpose of this study was to expand the utility of the Polygonum multiflorum. Also, we fermented P. multiflorum by mushroom mycelial, and analyzed for chemical compositions and biological active of fermented P. multiflorum root. The crude protein content did not show a significant difference between control and fermented P. multiflorum root, crude fat, ash and crude fiber content of fermented P. multiflorum root were lower than control. The content of soluble nitrogen free extract of P. multiflorum root was significantly higher than fermented P. multiflorum root. The major amino acids of P. multiflorum root were determined the arginine. The content of arginine and glutamic acid were 586.67 mg%, and 283.78 mg%, respectively. Sixteen kinds of amino acids were detected in fermented P. multiflorum root, and the major amino acids were determined the arginine and threonine. The total amino acid contents of control and fermented P. multiflorum root were 3,469.03 mg%, and 3,630 mg%, respectively. As the results of antioxidant activity test, the antioxidant capacity of fermented P. multiflorum had a higher than the control. As the mushroom fermentation progresses, it is confirmed that the amino acid content and antioxidant capacity were increased, and it is expected to develop the product using the fermented P. multiflorum using mushroom mycelials.

        Lentinula edodes, a member of the Pleurotaceae, is a blood pressure and cholesterol lowering effect. Recently, research the reduction of irritation and the enhancement of function have been cultivation of natural resources using mushroom mycelium. For the development of food fermented with L. edodes mycelium of natural medicinal herb remnants, determined to be 20%, and the product were used after lyophilization. In this study, we were analyzed the contents of β-glucan and amino acids of fermented herb remnants by L. edoeds mycelials. β -Glucan is a type of polysaccharide that has immune-enhancing and is known to inhibit cancer cells. The content of β-glucan and total amino acids of fermented herb remnants by L. edoeds mycelials were 35.13% and 8,697.36 mg%, respectively. The content of essential amino acid from fermented herb remnants by L. edoeds mycelials was 3,226.85 mg%. Collectively, our findings suggest that fermented herb remnants by L. edoeds mycelials extract could be used as an chemo-preventive material for consumers.

        β-glucan is a safe and highly potent biological response modifier that nutritionally activates the immune response through the Macrophage, Dendritic and additional immune cells to yield various therapeutic effects. Shiitake mushrooms (Lentinula edodes) contanining β-glucans may be beneficial for human health; they have been used in the treatment of cancer, hypertension, and high cholesterol levels. The effect of different substrates and various developmental stages (mycelium growth, primordium appearance, and fruiting-body formation) on β-glucan production in the edible mushroom L. edodes was studied. The cap of the mature mushroom showed the highest β-glucan activity, and β-glucan activity seemed to be influenced somewhat by some well-known inducers or sawdust. In this study, we utilized five strains (JMI 10022, 10036, 10077, 10079, 10080) of L. edodes regardless of origin and growth conditions. This experiment showed that the expression of β-glucan was induced by glucose bond, and increased with the growing of L. edodes. Quantitatively, reverse transcription PCR utilized pairs of primers specific for β-glucan gene expression shows that expressed genes were most commonly indentified during the process of fruiting-body formation. We suggest that the results will provide valuable information to assist L. edodes industry.

        For centuries, Lentinula edodes called “Pyogo” in Korea has been cultivated as a high-grade edible mushroom in Asia. This is known as medicinal mushroom for cold prevention in oriental medicine. In the world market, this mushroom of Lentinula edodes is introduced as representative edible and medicinal mushroom in Asia. As consumer interest increases, Asian Lentinula edodes market is also growing. There is a record that Korea has already been used as a gourmet food on the royal meal table for the king in the 15th century Joseon Dynasty. In Korea's famous oriental book, it is cooked with many traditional foods because of the pharmaceutical efficacy of this mushroom. For long time ago, difference in cultivation as well as in mushroom growth of Lentinula edodes has studied in Korea. Recent advances in cultivation of selected high quality mushroom studied from substrate optimization using log and sawdust of oak tree. Cultivation of Lentinula edodes strain research controlled the production of large quantity of various substrate conditions. This mushroom mycelia and fruit-body were cultivated in log of oak tree and sawdust substrate cultivation in types of bioreactors. According to compared cultivation method, it presented the optimistic condition for the amount production of various Lentinula edodes strain in Korea.

        This study was carried out to investigate the management characteristics and techniques by comparing and analyzing the process of mixing, sterilization, cooling, inoculation and incubation for bag cultivation of L. edodes. The medium was sterilized after mixing it for 30 minutes, 1 and 2 hours respectively. The general moisture content of the sawdust substrate has been adjusted to 65%, while the case of the L. edodes substrate was 55%. Only 5% of wood chips of 5-8 mm in particle size were mixed to secure the space of the sawdust particles. As a result, mycelial growth was 1.4cm faster and the density was better(+++) than control(++). Wood chips are soaked in advance for a week during winter and 4 days during summer. There is an average number of 1.6x108 (cfu) of microorganisms in the sawdust that has been piled for six months outdoor. In summer, it has to be used immediately after mixing sawdust. High-pressure sterilization should be performed to use as a mushroom spawn, and to improve physical properties, it was great to sterilize the medium at a normal-pressure. There are height and width type for bags to be consumed for bagging. When the height was reduced into 17cm and the width was increased into 13cm, the browning period was shortened by 30 days and the period of mycelial growth was shortened by 25 days. The sterilized medium had an average of 4x103(cfu) of microorganisms at 25°C and 1.3×102 (cfu) at 15°C. After 25 days from inoculation in vitro, the growth condition of sawdust was the best with 13.2 cm, followed by grain spawn and liquid respectively. When inoculated with liquid spawn, the moisture content of substrate should be adjusted to 55% to 50% in advance.

        Lentinula edodes shows atypical mating behaviors deviated from tetrapolar mating in outcrossing. To better understand the mating behavior, we investigated variations in the mating type genes, residing in A and B mating loci, of 129 dikaryotic strains from East Asia. Through sequence analysis of A locus, we found that hypervariable region spanning N-term of HD2-intergenic region-N-term of HD1 could represent A mating type of L. edodes. Mating and hypervariable region analyses revealed 50 unique A mating types: 27 from 98 cultivated strains, 33 from 31 wild strains, and 10 commonly found. It was also revealed that only a few A mating type alleles such as A1, A4, A5, and A7 were prevalent in cultivated strains. The A mating type in wild strains were highly diverse: 23 unique A alleles were discovered in small mountainous area in Korean peninsula. The B locus was assessed by allelic variations in pheromone (PHB) and pheromone receptor (RCB) pairs. Sequence analyses revealed 5 alleles of RCB1 with 9 associated PHBs in B sublocus and 3 alleles of RCB2 with 5 associated PHBs in B sublocus. Each RCB was primarily associated with two PHBs. This allowed us to propose 15 B mating types via combinations of five B and three B subloci. Thus, the total number of mating types in L. edodes became 750 through a combination of 50 A and 15 B. This number will further increase because of rapid diversification of A mating type and possible emergence of variants of RCBs and PHB-RCB combinations.

        The choice of strain, substrate, and environmental conditions are fundamental factors that determine the ability of a fungus to develop and produce mushrooms with high quality. The objective of this study was to compare the strains of Lentinula edodes and Ganoderma lucidum originated from South Korea and Brazil in relation to their vegetative development in different culture media, to obtain isolates with high quality. The strains of the two mushroom species from Brazil and South Korea grew slowly, possibly because of abiotic factors or storage technique used. However, it may be concluded that temperature, humidity, light, pH, and nutrients from the substrate, if not regulated appropriately may affect the vegetative species.

        표고버섯은 담자균강 주름버섯 느타리과 잣버섯속에 속하는 식용버섯으로 봄과 가을에 주로 참나무류 등 활엽수의 나무토막 등에서 자란다. 본 연구에서는 수종에 따른 표고버섯 자실체의 생리활성을 비교 분석코자 하였으며, 본 연구에서 사용된 수종은 상수리나무, 구실잣밤나무, 개서어나무, 메타세쿼이아, 두충나무 이다. 연구결과 대표적으로 사용되는 상수리나무 보다 구실잣밤나무에서 재배한 표고버섯이 페놀성화합물, DPPH 항산화활성, 주름억제 실험에서 약 2배정도 높은 효과를 보였다. 하지만 개서어나무에서 재배한 표고버섯은 효과가 떨어지는 경향을 보였다. 이러한 결과를 이용하여 향후 표고버섯 재배시 원목선택의 폭을 넓혀주고 기능성 버섯 개발을 통해 농가 소득에도 도움이 될 것으로 기대된다.

        버섯은 영양적인 면에서 뛰어난 기능성 식품이며 인체에 중요한 영양 성분을 함유하고 있어 오래전부터 식용은 물론 약용으로 널리 이용되어 왔다. 표고버섯은 항종양, 항당뇨, 혈압조절 등의 성인병 예방 효과가 있는 것으로 알려져 있고 현재 배지에 기능성 소재를 첨가하여 기능성이 증진된 버섯을 생산하려는 연구가 많이 시도되고 있다. 본 연구는 수종별로 상수리나무, 메타세쿼이아, 두충나무, 개서어나무, 구실잣밤나 무의 원목 재배에 의한 표고버섯의 기능성 성분 함량을 분석하였다. 총폴리페놀 함량 분석결과 보편적으로 쓰이는 상수리나무 5.9ug/g 보다 구실잣밤나무에서 9.6ug/g 약 1.6배 높았고 총플라보노이드 함량은 상수리 나무의 경우 69.3ug/g, 구실잣밤나무에서 83.9ug/g 약 1.2배 높았다. 구실잣밤나무의 경우 gentistic acid, chlorogenic acid, esculin 함량이 높게 나타났다. 이러한 결과를 통해 향후 기능성 버섯 등 목적에 맞는 맞춤 형 재배, 농가 등에 유용한 자료로 활용하고자 한다.

        In this study, we investigated the amino acids content of shiitake mushrooms grown in eight different media and under three different temperature conditions. The sawdust media were supplemented with 4 kinds of nutrients - rice bran, barley, bean curd refuse, and red ginseng refuse in an 8:2 ratio with or without 0.5% calcium chloride, and the fruiting temperature was set to low (13–15oC), medium (18–20oC), or high temperatures (24–26oC). Seventeen amino acids were detected, and the content varied depending on the compositions of the media. The highest total amino acid content (209.7 mg/g) was measured in mushrooms from the media with red ginseng refuse, and the glutamic acid content was the highest (51.3 mg/g) in mushrooms from the media with barley. In terms of fruiting temperature, the amino acid content was the highest (38.2%) at medium temperature followed by the content at low (32.5%) and high temperature (29.3%). The amino acid composition of shiitake mushrooms was similar between the medium and low temperature conditions.

        Antioxidant activities of 80% methanol, water, and 70% ethanol extracts of Lentinula edodes GNA01 were compared and estimated. The yield of Lentinula edodes GNA01 was identified to be in the following order: water>70% ethanol>80% methanol, but there was no significant difference between 80% methanol and 70% ethanol extracts. The highest total phenolic content (TPC) and total flavonoid content (TFC) were found in water extract, and TPC of 80% methanol extract was higher than that of 70% ethanol extract and TFC of 70% ethanol extract was higher than that of 80% methanol extract. Water extract exhibited the strongest DPPH, ABTS radicals, and nitrite scavenging activities, Fe2+ chelating ability, and FRAP among the three extracts. In addition, antioxidant activity of 80% methanol extract was higher than that of 70% ethanol extract in most of the experiments. As a result, antioxidant activity of Lentinula edodes GNA01 showed a difference according to extraction solvent and concentration; nevertheless, water extract exhibiting high polarity had the strongest antioxidant effect. Consequently, water extract from Lentinula edodes GNA01 is anticipated to be useful for the development of a high value-added functional product

        In 2010, a new variety for sawdust cultivation,was produced by monokaryotic-monokaryotic crossing between Sanjo 701ho and FMRI0995. The optimum temperature for Sanjo 708ho mycelial growth was 25oC; mycelial growth on potato dextrose agar was 51.6 mm over 7 days. The fruiting pattern was sporadic, and the optimal temperature range for fruiting was 5~20oC. Regarding fruiting body characteristics, the pileus was 65.8 mm in size, 16.1 mm in thickness, and hemispherical in shape. The stipe was 42.6 mm in and 19.6 mm in thickness. Mproductivity was good during thelow-temperature period from autumn to spring, with an average productivity of 310.7 g/bag.

        This study was performed to determine the optimal condition for Lentinula edodes JMI-10079 mycelium cultivation on the root of Polygonum multiflorum. We also analyzed the proximate composition, total amino acids, and minerals in the root of P. multiflorum cultivated with L. edodes JMI-10079 mycelia. The optimal temperature and pH for L. edodes JMI-10079 mycelium cultivation on the P. multiflorum root were 25oC and pH 5–6 respectively, whereas the optimal carbon and nitrogen sources were glucose and maltose, respectively. The content of crude protein, crude fat, and ash in the P. multiflorum root cultivated with L. edodes JMI-10079 mycelia was higher than that in the uncultivated P. multiflorum root. The content of crude fiber was the highest in the control. Total amino acid analysis revealed that the contents of total amino acids and total essential amino acids were increased by higher root of P. multiflorum concentration.

        Lentinula edodes, the popular shiitake mushroom, is one of the most important cultivated edible mushrooms. It is used as a food and for medicinal purposes. Here, we present the 46.1 Mb draft genome of L. edodes, comprising 13,028 predicted gene models. The genome assembly consists of 31 scaffolds. Gene annotation provides key information about various signaling pathways and secondary metabolites. This genomic information should help establish the molecular genetic markers for MAS/MAB and increase our understanding of the genome structure and function.
