        The nature of molecular mechanisms governing embryo development is largely unknown, but recent reports have demonstrated that proper execution of programmed cell death is crucial for this process. The main objective of this study is to examine the mode of programmed cell death during nuclear transfer embryos development in porcine. In particular, the relative employment of two major pathways in programmed cell death; e.g. apoptosis (type I) and autophagy (type II) was compared. Oocytes use in the study was matured in vitro in the presence of 10% FBS maturation medium. After nuclear transfer embryos were cultured for each programmed cell death control factor [Cysteamine(Cyst : 0.4mM), 3-methyladenine(3MA : 2.5mM) and Rapamycin(RP : 100nM)] in TCM-199 medium supplemented with 0.1% BSA. In this study results of among the blastocysts development in 3MA; PCNA, MAP1LC3A and ATG5 RNA gene expression level increased in the order of IVF<Cyst < 3MA < RP. However Casp-3 and TNF-r RNA gene expression level decreased in the order of IVF < 3MA and RP< Cyst. The expression of mTOR showed a pattern opposite to that of MAP1LC3A. That is, its expression was the lowest in Cyst group. And next experiments analysis of MMP expression patterns. Analysed this MMPs enzyme activation to evaluate the effectiveness of high quality brastocyst culture in porcine. In this results of the enzymatic activity of MMP-2 and MMP-9 was assessed in culture, the level of active MMP-9 was higher expression in the medium of each 3MA and RP treatment group, with the level of another treatment group being relatively higher. These results suggest that the autophagy activation culture medium is more effective for stable and innovative nuclear transfer embryos development.

        Disheveled is a major regulator of WNT signalling pathway. It has been shown that WNT signaling is important for regulating synaptic plasticity. However, it is not still clear how Dsh is regulating synaptic plasticity. In this study, we used various methods to investigate how Dsh regulates synaptic plasticity. Our further studies will reveal unknown molecular and cellular mechanisms underlying WNT signaling dependent synaptic plasticity.

        독점규제법상 지주회사 규제는 경제력집중 억제를 위한 규제의 하나로 1986년 동법의 개정 시에 도입되었다. 특히 1999년 동법 개정에 의하 여 지주회사 규제는 원칙적 설립 금지에서 설립 허용과 일정한 제한을 가하는 방식으로 변경되었다. 이와 같은 제도 변화에는 경제력집중의 수 단이 될 수 있는 등의 지주회사가 갖는 부정적 측면이 완화되었고, 반면 효율적인 조직 운영과 같은 긍정적 측면이 활용될 필요가 있다는 인식이 영향을 미쳤다. 이러한 이유로 지주회사의 설립‧전환을 원칙적으로 허용 하면서 대신 일정한 제한을 부과하는 방식으로 법 개정이 이루어졌다. 이후 공정거래위원회는 재벌로 대표되는 대규모기업집단의 집단 구조가 계열사의 순환출자 방식에 기반함으로써 매우 복잡하고 불투명한 구조를 취하고 있고, 이를 개선하는 방안으로 지주회사 체제를 적극적으로 권장 하는 정책을 추진하여 왔다. 이 과정에서 1999년 법 개정에 의하여 도 입된 지주회사에 대한 여러 제한은 처음 입법 당시에 비하여 상당히 완 화된 내용을 갖게 되었다. 이러한 정책의 추진에 의하여 지주회사의 수 는 증가하였지만, 반면 지주회사의 부정적 측면을 최소화하기 위하여 도 입된 규제의 의의를 퇴색시키는 결과를 낳았다. 이러한 상황에서 지주회 사 규제의 의의를 다시 확인하고, 현재의 규제 내용이 이에 상응하는 것 인지를 살펴볼 필요가 있다. 또한 금산분리 원칙의 완화를 의도하는 공 정거래위원회의 개정안도 이러한 맥락에서 검토되어야 할 것이다.

        Pro-inflammatory cytokines, interleukin-1β (IL1B), IL6, and tumor necrosis factor-alpha (TNF), are known to play important roles in regulating the endometrial function in the uterus during the estrous cycle and pregnancy in several species. However, the expression and function of these cytokines and their receptors in the uterine endometrium during the estrous cycle have not been studied in pigs. Thus, this study determined the expression and regulation of IL1B, IL6, TNF and their respective receptors, IL1R1, IL1RAP, IL6R, GP130, TNFRSF1A, and TNFRSF1B during the estrous cycle in pigs. To analyze levels of each gene expression in the uterine endometrium we obtained from endometrial tissues on Days 0, 3, 6, 9, 12, 15, and 18 of the estrous cycle. Real-time RT-PCR analysis showed that levels of IL1B, IL1RAP, IL6R, GP130, TNF, TNFRSF1A, and TNFRSF1B mRNAs were highest on Day 15 or 18 of the estrous cycle, which corresponds to the proestrus period. Levels of IL1R1 were highest on Day 0, while levels of IL6 were biphasic with high levels on Day 6 and Day 15. The abundance of IL1B, IL6, IL6R, and TNF mRNAs was decreased by progesterone, while levels of GP130 were increased by progesterone in endometrial tissue explants. These results showed that expression of pro-inflammatory cytokines and their receptors changed stage-specifically during the estrous cycle and regulated by progesterone in the uterine endometrium in pigs, suggesting that these pro-inflammatory cytokines may be involved in the regulation endometrial function during the estrous cycle in pigs.

        최근 중국에서는 많은 환경문제가 제기되어 왔으며 정부의 관심도 증가하여 특히, 난파물 제거작업에 대한 전반적인 접근방식에 큰 영향을 미쳐 왔다. 따라 서 대부분의 난파물 제거작업에 대해 환경적인 요소를 우선적으로 고려하며 진 행하게 되었다. 한편, 최근 시장경제 논리 경향에 따라 난파물 제거 작업에 대 한 정부당직의 단계별 비규제화를 보이는 부분도 있어 왔다. 이러한 상황에서 이 논문은 강제적인 난파물 제거와 관련된 전형적인 행정상 및 민사상 문제들을 명 확히 하고, 중국의 현행규정 상 논란이 되고 있는 규정에 대한 살펴보고자 한다. 아 울러 난파물 제거 비용에 대한 부분도 고찰해 보고자 한다.

        Ribosomal protein L21 (RPL21) plays an important role in ribosome assembly. It is considered to be a major cause for the occurrence of the hypotrichosis simplex (HTS), a type of sustained hair loss from early childhood to adulthood. In this study, the full-length sequence of pig RPL21 gene (GenBank accession number: KU891824) was cloned and identified for the first time. We found it contains a 483-bp open reading frame (ORF) encoding 160 amino acids. It is located in the plus strand of chromosome 11, which spans 2,167 bp from 4,199,792 to 4,201,958. We found RPL21 expression level is closely related to cell proliferation and cell cycle arrest. In the knockdown group, the cell proliferation activity was significantly decreased (P<0.01) and an obvious accumulation of cells at the G2/M phase with a simultaneous up-regulation of p53 and p21 was observed. This likely due to knockdown of RPL21 triggered ribosomal stress, which affected the normal ribosome assembly and caused defective ribosome biogenesis. The unassembled RPs were released consequently from the nucleolus to the nucleoplasm where they can activate p53-dependent cell-cycle responsive factors and led to a G2/M arrest. We expect these results may provide valid information for further study on the pig RPL21 gene and the cause of hypo trichosis simplex.

        Fatty acid synthesis (FASN) is an enzyme responsible for the de novo synthesis of long-chain fatty acids. During oncogenesis, FASN plays a role in growth and survival rather than acting within the energy storage pathways. Here, the function of FASN during early embryonic development was studied using its specific inhibitor C75. We found that the presence of the inhibitor reduced blastocyst hatching. FASN inhibition decreased Cpt1 expression, leading to a reduction in mitochondrial copy numbers and ATP content. This inhibition of FASN results in the down-regulation of the AKT pathway, thereby triggering apoptosis through the activation of the p53 pathway. Activation of the apoptotic pathways also leads to increased accumulation reactive oxygen species and autophagy. In addition, the FASN inhibitor can impair cell proliferation, a parameter of blastocyst quality for outgrowth. The level of OCT4, an important factor in embryonic development, decreased after treatment with the FASN inhibitor. These results show that FASN exerts an effect on the early embryonic development by regulation of both fatty acid oxidation and the AKT pathway in pigs.

        PP2A-B55α, a regulatory subunit of PP2A plays an important roles in regulating cell proliferation and survival. However, the functions for PP2A-B55α in mouse early embryo development is not clear. The objective of present were to investigate the expression patterns and to explore its biological function during mouse early development. Thetranscripts of PP2A-B55α were detected at all developmental stages in mouse embryo and decreased during embryo development. Immunostaining revealed that PP2A-B55α was present in both the nucleus and cytoplasm in early cleavage stage embryos. In the late embryonic development, PP2A-B55α was predominantly located in the cytoplasm. Knockdown (KD) of PP2A-B55α using double strand RNA not affect the proportion of cleaved embryos, but resulted in significantly decreased development to blastocyst stage and reduced total cell number in blastocyst. KD PP2A-B55α is able to induce sustained DNA damage and reduced the transcripts of non-homologous end joining (NHEJ) or homologous recombination (HR) pathways relative genes in mouse early embryo. KD PP2A-B55αcaused apoptosis and increase the transcripts of pro-apoptotic genes in blastocyst. Furthermore, The KDPP2A-B55α showed significantly lower cell proliferating rates (from 5-Bromo-deoxyuridineassayresults) in blastocysts and to talareas of out growth potential was decreased. These observation provide novel in sight into PP2A-B55α expression patterns in mouse early embryos and down-regulation of PP2A-B55α negatively impacted blastocyst development, total cell number, DNA damage, apoptosis, and proliferation and post-hatchingevents.

        Zinc (Zn2+) is one of essential factors during mammalian oocyte maturation and fertilization. Previous studies showed that depletion of cellular Zn by metalion chelator impair asymmetric division of oocyte. But the detailed mechanism of these phenomena is unclear. We found that depletions of zinc by cell-permeable heavy metal chelator N,N,N',N'-tetrakis(2-pyridylmethyl)ethane-1,2-diamine (TPEN) caused the decrease of cytoplasmic actin mesh level. Spire2-GFP is co-localized with zinc at the cortex and intracellular vesicle. By the treatment of TPEN, number of Spire2-GFP decorated vesicle is drastically decreased, indicating that Zn2+is essential for the localization of the spire in mouse oocyte. Two putative zinc-binding regions were located in the C-terminal part of Spire2. Mutations of zinc binding site on spire abolish its localization at the intracellular vesicle. Over expression of C-terminal region containing zinc binding site of spire impair oocyte maturations and decrease cytoplasmic actin mesh. Taken together, these results suggest that intracellular zinc is crucial for the proper localizations of spire in the mouse oocyte, and unraveling the novel regulatory mode of actin nucleator spire by Zn2+.

        Melatonin has an important role as anti-oxidative effect and reducing of endoplasmic reticulum(ER)-stress on oocyte maturation and embryo development. Under ER-stress condition, unfolding protein response (UPR) is a defence mechanism in mammalian cells. Recently, regulation of UPR signaling genes are involved in oocyte maturation, embryo development and female reproduction. However, there is no report on the role of melatonin for UPR signaling and ER-stress mediated apoptosis during pig oocyte maturation progression. Moreover, the changes of UPR genes expression according to the porcine oocyte maturation is not yet fully understood. Here, we investigated the changes of UPR signal (BIP/GRP78, ATF4, p90/p50ATF6, and XBP1) and ER-stress apoptotic factor CHOP genes expressions in porcine oocyte maturation by Western blot and RT-PCR analysis. During oocyte maturation, UPR marker and CHOP genes expressions were significantly increased in matured oocytes or cumulus-oocyte complexes (COCs). UPR markers expressions were significantly increased by ER-stress inducer, tunicamycin (Tm), treated (1, 5, 10 μg/ml) groups in a dose-dependent manner compared with control group. To confirm the reducing of ER-stress by melatonin (0.1 μM), we were compared to the effects of ER-stress inhibitor, TUDCA (200 μM), after pre-treated Tm (5 μg/ml) for 22 h maturation. Expressions of UPR markers and meiotic maturation were recovered by melatonin (0.1 μM) in COCs. And, we observed the role of Grp78/Bip as UPR signaling beginning marker using siRNA. In result, reduction of Grp78/Bip gene expression by siRNA was induced the inhibition of oocyte maturation (32.5±10.1 vs control; 77.8±5.3), and p50ATF6 protein level was significantly decreased (p<0.001) in cultured COCs for 44 h. In addition, these results were recovered through the addition of melatonin (0.1 μM) or TUDCA (200 μM) in maturation medium. These results demonstrated that the regulation of UPR signaling via Grp78/Bip gene induction plays a critical role in porcine oocyte maturation in vitro. Furthermore, this present study first confirmed a functional link between inhibition effect of ER-stress by melatonin and regulating of UPR signaling in porcine oocyte maturation. In conclusion, melatonin improves the oocyte maturation and cumulus cells expansion of COCs through the regulation of UPR signal pathway by BIP/GRP78 against the ER-stress during porcine oocyte maturation periods.

        The migration, adhesion, and proliferation of conceptuses during pregnancy are tightly controlled processes that are mediated by various factors including cytokines, growth factors, and hormones. Among many factors, chemokines play key roles in lymphocyte trafficking, cellular proliferation, vascularization, and embryogenesis in many mammalian species. Especially, it has been shown that C-X-C chemokine ligand 12 (CXCL12) plays an important role in early pregnancy by promoting trophoblast invasion, proliferation, and differentiation through its receptor, C-X-C chemokine receptor 4 (CXCR4) in humans. However, expression and function of CXCL12 in the uterine endometrium during pregnancy have not been well studied in pigs. Thus, we determined expression of CXCL12 and its receptor, CXCR4, in the uterine endometrium during the estrous cycle and pregnancy in pigs. We obtained endometrial tissues from gilts on day (D) 12 and D15 of the estrous cycle and D12, D15, D30, D60, D90, and D114 of pregnancy, conceptus tissues from D12 and D15 of pregnancy, and chorioallantoic tissues from D30, D60, D90, and D114 of pregnancy. Real-time RT-PCR analysis showed that levels of CXCL12 and CXCR4 mRNAs changed in the uterine endometrium during pregnancy. Levels of CXCL12 and CXCR4 mRNAs on D15 of pregnancy were higher than those on D15 of the estrous cycle. After D15 of pregnancy levels of CXCL12 and CXCR4 mRNAs gradually decreased toward term of pregnancy, and CXCL12 and CXCR4 were expressed in the chorioallantoic tissues during the mid- to late pregnancy. CXCL12 and CXCR4 mRNAs were expressed in chorioallantoic tissues during mid- to late pregnancy, and RT-PCR analysis showed that CXCL12 and CXCR4 mRNAs were detectable in conceptus on D12 and D15 of pregnancy. Immunohistochemistry showed that CXCL12 proteins were localized to endometrial luminal and glandular epithelial cells during the estrous cycle and pregnancy, and to chorionic epithelial cells during mid- to late pregnancy. Abundance of CXCL12 mRNAs, but not CXCR4, in the uterine endometrium was increased by the treatment of IFNG. These results showed that CXCL12 and CXCR4 were expressed in the uterine endometrium, conceptus, and chorioallantoic tissues and IFNG increased endometrial CXCL12 expression in pigs, suggesting that CXCL12 and its receptor may play a key role in regulation of the establishment and maintenance of pregnancy by affecting the conceptus development in pigs.         S100As are calcium-binding proteins with two EF-hand calcium-binding motifs. In several studies, S100A proteins are described to play important roles in pro-inflammatory responses including damage-associated molecular pattern (DAMP) signaling and in the establishment of pregnancy. However, the role of S100As have not been determined in the uterine endometrium during the estrous cycle in pigs. Thus, this study was performed to investigate expression and regulation of S100A8, S100A9, and S100A12 in the uterine endometrial tissues during the estrous cycle in pigs. Real-time RT-PCR analysis showed that S100A8, S100A9, and S100A12 mRNAs were expressed in the uterine endometrium during the estrous cycle with higher levels on days 15 and 18 of the estrous cycle than other days of cycle. To investigate the effects of steroid hormones, estradiol (E2) and progesterone (P4), on expression of S100A8, S100A9, and S100A12 mRNAs, endometrial tissue explants from immature pigs were treated with steroid hormones. Levels of S100A8, S100A9, and S100A12 were increased by the treatment of P4, and the increased levels of S100A8, S100A9, and S100A12 by P4 were not inhibited by the treatment of progesterone receptor antagonist, RU486. However, levels of S100A8, S100A9, and S100A12 were decreased by treatment of MEK inhibitor, U0126. These results exhibited that S100As were expressed in the uterine endometrium during the estrous cycle in a cyclic stage-specific manner, and their expression was affected by P4. These suggest that S100As may play an important role in endometrial function during the proestrous period of the estrous cycle in pigs.         Interferon tau (IFNT), produced by the mononuclear trophectoderm, signals the process of maternal recognition of pregnancy in ruminants, but its expression in vivo is not well characterized. Objectives of this study were to determine IFNT gene isoforms expressed in the bovine uterus, and to identify differences in promoter sequences of IFNT genes that differ in their expression. Through the RNA-seq analysis of bovine conceptuses on days 17, 20 and 22 (day 0 = day of estrus), the expression of only two IFNT transcripts, IFNT1 and IFNTc1, were found, which were indeed classified into the IFNT gene clade. IFNT mRNAs were highest on day 17, and then decreased on days 20, and 22, which were also supported by the results of quantitative RT-PCR. Bovine ear-derived fibroblast (EF) cells were then cotransfected with luciferase reporter constructs carrying 5‘-upstream (positions -1000 to +51) regions of IFNT1 or IFNTc1 and various transcription factor expression plasmids. CDX2, either alone or with other Ap-1, ETS2 and/or CREBBP transcription factors, was found to increase luciferase activity approximately 10 and 18 fold more than twice of those cotransfected with bIFNT1, c1-Luc construct. Furthermore, The degree of transcriptional activation by a combination of the AP1, ETS2, CREBBP and/or CDX2 expression vectors was similar to that of CDX2 along plasmid. However, expression patterns of these Luc activity differented. Whereas bIFNTc1-Luc showed lowest antivity had than bIFNT1-Luc reports. Although, lowest antivity had of the bIFNTc1 –Luc report, cotransfected with the bIFNTc1-Luc construct and AP1(JUN) or/and ETS2 expression plasmid, Luc activity was enhanced approximately 2 and 4-fold more than the bIFNT1-Luc. Furthermore, along CDX2 expression factor had high effect on activity of bIFNT1-Luc reporter than the c1 gene in EF cells. These results suggest that two forms of IFNT genes are expressed in utero and their transcriptional regulations differ.

        The regulation for industrial accident rate level assessment of construction companies was introduced to the construction industry in the Republic of Korea since 1993 and has brought positive outcome on industrial accidents reduction at construction work sites. There were considerable decrease of industrial accident ratio and enforcing of contrators' safety organizations from the beginning of the regulation for industrial accident rate level assessment. In spite of these positive outcomes, there were some negative effects such as contractors' shrinking accident reports to keep good accident ratios since these figures had a great impact on pre-qualification stage of bidding when general contractors were competing for new construction projects. In addition, Comprehensive evaluation bid system, which replaces the lowest price bid system is applied to government-ordered construction projects since 2016. Comprehensive evaluation bid system includes construction company's accident rate as one of the evaluation items and carries out with the industrial accident rate level assessment of construction companies at the same time. The regulations of two systems have been called for improvement to unify these different procedures and standards which have led business stakeholders to confusion for several years. This study aims to devote on lessening shrinking accident reports and to reduce the waste of business stakeholders through changing the regulation for industrial accident rate level assessment.

        Recent decades have seen a number of policies have been launched to promote information disclosure on food products marketed in developed countries. There has also been increasing efforts to promote healthy eating in developing economies (Cowburn & Stockley 2005). A good example appears in China, where, in an effort to improve health education, nutrition labels became mandatory under the Nutrition Labeling Act (NLA) since 2013. Additionally, food labeling regulations were issued by India, Mercosur members and South Africa. While nutritional labeling has received a lot of attention both in academia and from the press, there are some key gaps in the nutrition labeling literature. One issue of how standardized information requirements affect firm marketing responses (e.g., advertising levels. R&D investment, and corporate social responsibility programs) and performance has been understudied. Another clear gap in the literature is how nutritional labeling requirements affect developing economies.
