        작물 재배 시 주요 해충 발생에 대해 한두 달 이상 앞선 계절전망이 가능하다면 농가의 해충관리 의사결정이 보다 효율적으로 이루어질 수 있을 것이다. 본 연구에서는 국내 해충 발생과 통계적으로 유의미한 원격상관관계에 있는 기후현상을 찾기 위해 Moving Window Regression (MWR) 기법을 활용하였다. 벼멸구의 발생과 비래는 장기간에 걸쳐 여러 지역에서 연속적으로 일어나는 사건이기 때문에 비슷한 시공간적 규모 를 갖는 기후현상과 통계적인 연관성을 가질 가능성이 높아 본 연구의 대상 해충으로 선택하였다. MWR 통계 분석의 반응변수로써 1983년부터 2014년까지 국내 벼멸구 발생면적 자료를 사용하였고, 10개의 기후모형에서 생산되는 10개의 기후변수를 예보 선행시간별로 추출하여 설명변 수로 사용하였다. 최종적으로 선정된 각 MWR 모형의 특정 시기와 지역의 기후변수는 연간 벼멸구 발생면적 자료와 통계적으로 유의한 상관관 계를 보였다. 결론적으로, 본 연구에서 개발한 MWR 통계 모형을 통해 국내 벼멸구 발생 위험도에 따른 선제적 대응을 위한 벼멸구 계절전망이 가능할 것으로 보인다.

        A seasonal outlook for crop insect pests is most valuable when it provides accurate information for timely management decisions. In this study, we investigated the probable relationship between climatic phenomena and pest infestations in Korea using two statistical methods. Brown planthopper was selected because of its migration characteristics, which fits well with the concept of our statistical modelling – utilizing a long-term, multi-regional influence of selected climatic phenomena to predict a dominant biological event at certain time and place. The moving window regression (MWR) model showed high correlation between the national infestation trends of brown planthopper and some tempo-spatial climatic variables near its sequential migration path, while the climate index regression (CIR) model resulted in a relatively low correlation compared to the MWR model. Overall, the statistical models developed in this study showed a promising predictability for rice brown planthopper infestation in Korea, although the dynamical relationships between the infestation and selected climatic phenomena need to be further elucidated.

        애멸구(Laodelphax striatellus)는 rice stripe virus (RSV)의 매개충으로 벼에 큰 피해를 주는 해충이다. 본 연구에서는 애멸구의 장·단시형, 암·수, 약·성충에 대한 RSV 보독률과 이병률을 비교하였다. 애멸구의 장·단시형의 RSV 보독률은 각각 60.7%, 63.1%로 크게 차이는 없었다. 암·수에 대한 RSV 보독률은 각각 61.9%, 52.2%로 암컷의 보독률이 더 높았으나 유의성은 없었다. 약·성충의 보독률을 비교한 결과 각각 51.2%, 58.7%로 역시 크게 차이가 나지는 않았다. RSV에 감염된 애멸구에 노출된 건전한 벼의 이병률은 장시형은 53.3%, 단시형은 48.2%를 보였으며, 약·성충의 이병률은 각각 38.2%, 42.6%를 보여 유의성은 없었다. 반면 암컷은 50.5%의 이병률을 보이고 수컷은 22.3%의 이병률을 보여 암컷이 수컷에 비해 22.3% 이병률이 높아 유의성이 있었다. 또한 벼와 애멸구의 RSV 감염여부에 의한 애멸구의 발육기간은 건전한 벼에 RSV 감염 애멸구를 접종 했을 때 가장 긴 것으로 나타났으며, 건전한 벼에 건전한 애멸구를 접종 했을 때 발육기간이 가장 짧은 것으로 나타났다.

        The small brown planthopper (SBPH), Laodelphax striatellus Fallén (Hemiptera: Delphacidae) is one of the major insect pest against rice, Oryza sativa L. in Korea. High density of SBPH could cause severe damage on rice plant by directly sucking and indirectly transmitting viral pathogens, Rice stripe virus and Rice streaked dwarf virus. As a preliminary study for de novo whole-genome sequencing of SBPH, we investigated 6 transcriptomes isolated from different developmental stages, sex, and tissue (egg, 1st ~ 3rd nymphs, 4th ~ 5th nymphs, female and male adults, salivary gland). Clean-sequence data of 19.3 Gb were obtained from total 47.8 Gb raw data after adaptor and quality trimming (Q30) and overlapped reads joining. As a suitable assembler, Bridger was selected based on the results of reference mapping (93.45%) and CEGMA completeness (95.97%). Finally, we obtained 158,207 reads (size range: 201 ~ 22,162 bp; Mean size: 1,048.04 bp; N50: 2,417 bp) after clustering the assembly results by CD-HIT-EST (similarity threshold: 99%). Based on these results, we are conducting further studies such as transcript expression pattern among different developmental stages and gene annotation.

        The small brown planthopper (SBPH), Laodelphax striatellus Fallen, is an important pest that causes severe yield losses by transmitting plant viruses to rice. For the efficient control of SBPH in Korea, the respective resistance levels in the immigrant and indigenous populations need to be discerned. The resistance levels to 10 insecticides (three carbamates, an organophosphate, four neonicotinoids, and a phenylpyrazole) were evaluated in 21 field populations collected from either SBPH-immigrating or indigenous regions during two different seasons (early spring vs. late summer). Imidacloprid resistance was most widely observed in many regional populations, followed by thiamethoxam resistance. Interestingly, the resistance level to imidacloprid was significantly higher in both immigrant and late-summer-collected populations than in indigenous and early spring-collected populations, respectively [3.3- (p = 0.018) and 2.6-fold (p = 0.026)]. Moreover, the late summer immigrant population exhibited higher imidacloprid resistance (2.4-fold) than the early spring-collected population from the same region, suggesting that the migratory SBPH that immigrated into Korea already exhibited imidacloprid resistance traits and were further selected after inhabitation. All field populations showed little resistance to fipronil (0.1- to 0.7-fold), suggesting that it is the most effective among the tested insecticides to control field populations of SBPH. The coefficient of variation of the resistance ratio (RR) among different regional populations and the correlation coefficient of RR among different insecticides have been suggested as supplementary parameters when determining appropriate insecticides as respective indicators for the dispersion status of resistance among SBPH populations and the possibility of cross resistance among tested insecticides.

        애멸구(Laodelphax striatellus (Fallén))는 벼줄무늬잎마름병을 매개하여 심각한 피해를 야기하는 벼 해충으로 국내에서 월동하는 토착종 이며 중국에서 국내 서해안지역으로 비래하기도 한다. 본 연구는 애멸구의 분산과 개체군 동태의 기초자료로 활용하기 위해 국내 16지역에서 2014년 4월과 7월에 애멸구를 채집하여 발생시기에 따른 날개형태의 비율과 홍집게벌 기생율 현황을 파악하였다. 수컷은 4월보다 7월에 단시형 의 비율이 유의미하게 낮았고, 암컷 단시형 비율은 차이가 없었다. 회귀분석 결과 4월의 단시형 비율이 위도가 증가함에 따라 증가하는 경향을 보 였다. 약충 및 성충의 기생율은 7월보다 4월에 높았다. 또한 서해안지역의 약충 기생율이 대체적으로 높은 경향을 보였다.

        Rice stripe virus (RSV) is one of the serious plant pathogenic viruses for rice and mediated by small brown planthopper, Laodalphax striatellus. So far, the studies have been mainly focused on the interaction between the host plant and the virus. In this study, for better comprehension of the interactions among Rice stripe virus, rice and small brown planthopper, transcriptomes of the RSV-viruliferous (RVLS) and non-viruliferous L. striatellus (NVLS) were comparatively analysed. For this, non-viruliferous L. striatellus were collected from non-infected rice field and fed RSV-infected rice for 5 days. With the RNAs prepared from the RSV-viruliferous and the non-viruliferous small brown planthoppers, we conducted Illumina RNA sequencing (Hiseq 2000) and then two transcriptome databases were generated from RVLS and NVLS, respectively. The transcriptome of RVLS and NVLS were campared to figure out how the gene expression of the insects affected by Rice Stripe Virus. RSV-dependently regulated genes analysed from this study may have important functions in the transmission and replication of RSV.

        The brown planthopper (BPH), Nilaparvata lugens (Stål), is an important pest in rice paddies. Because BPH cannot survive under low temperature conditions (<12℃), its outbreak in temperature areas depends on migration from overwintering areas of BPH. Geospatial distribution of BPH overwinterng area would be changed under future climate. The objective of the present study was to assess potential areas for BPH overwintering in Asia under current and future climate conditions. To reduce model uncertainty, we explored an ensemble modeling approach using five species distribution models (SDMs) and climate data from five general circulation models (GCMs). For future projections, outputs from an SDM with five sets of GCM data were averaged into a single map. Then, five maps from individual SDMs were summarized to assess the potential overwintering areas of BPH. Overall, the BPH overwintering areas expanded northward under future climate conditions. The degree of expansion in overwintering areas differed by region. For example, the uppermost boundary of BPH overwintering areas shifted north about 150 Km in southern China by 2050s. In Japan, small areas in the southern Kyushu were projected as potential areas of BPH overwintering. These results indicated that migration of BPH into East Asia including northern China, Korea, as well as Japan would occur early in the growing season, which could result in considerable damages in rice production.

        최근 WMO는 온실가스 배출량 시나리오(SRES)를 대신하여 대표농도경로(RCP)를 바탕으로 새로운 기후변화 시나리오를 생산하였으며 기상연구소는 RCP 시나리오를 바탕으로 한반도의 새로운 기후변화 시나리오를 생산하였다. 본 연구에서는 과거 관측값을 바탕으로 평년(1981-2010)의 애멸구의 우화시기와 세대수를 추정하였으며, RCP 8.5 시나리오를 바탕으로 2020년대(2015-2024), 2050년대(2045-2054)와 2090년대(2085-2094) 애멸구의 우화시기와 세대수를 예측하였다. 평년 애멸구 월동 1세대수의 우화일인 176.0±0.97일과 비교하여 2050년대에서는 13.2±0.18일(162.8±0.91일), 2090년대에는 32.1±0.61일(143.9±1.08일) 앞당겨질 것을 예측되었다. 그리고 애멸구의 연간 세대수는 2050년대에서는 현재보다 2.0±0.02세대, 2090년대에는 5.2±0.06세대 증가할 것으로 예측되었다.

        벼멸구에 감수성이며 고품질인 ‘황금누리’와 벼멸구 저항성 유전자 Bph18을 가지고 있는 SR30071-3-7-23-6-1-1-1계통을 교배하여 전통육종법과 분자육종법을 활용하여 새로운 벼멸구 저항성 계통을 선발하여 병해충 저항성과 농업형질을 조사 수행하였다. 1. 20개체의 F1을 양성하여 F2 1,200개체를 포장에 전개하 였다. 포장에서 선발된 개체에 대해 실내미질선발을 수행하여 23개체의 F3를 공시하였으며 벼멸구 유묘검정, 도열병 밭못자 리검정, 흰잎마름병 K1 균계 접종검정을 실시한 후 F4 세대 를 거쳐 100계통을 F5 세대로 공시하였다. 2. F5 세대에서 DNA 마커 검정으로 100계통 중 Bph18은 92계통, Xa3는 96계통, Stv-bi는 98계통에서 저항성 유전자가 확인되었다. 생물검정에서는 100계통 중 벼멸구 유묘검정은 94계통이 저항성을, 흰잎마름병 K3 균계검정은 27계통 및 73 계통이 각각 저항성 및 중도저항성 반응을 보였다. 3. F5 세대 100계통 중 주요 농업적 형질이 우수한 11계통 을 생산력검정시험에 공시하였고, 보통기 보비재배에서 농업 형질이 양호하고, 수량이 높으며 병해충 저항성을 보이는 우 량한 4계통을 선발하여 이삭모양과 초형이 우수하며 출수기는 중만생종이며, 간장이 작고 수량이 높은 HR28011-1-2-3을 ‘익 산562호’로 계통명을 부여하였다. 본 실험에서 육성한 ‘익산562호’는 지역적응성 평가를 통한 품종화를 추진 중이며 벼멸구 및 기타 주요 병해충에 복합저 항성 벼품종육성을 위한 유전자원으로 활용되기를 기대한다.

        Rice stripe virus disease (RSVD), one of the most serious disease of rice is mediated through the sucking by small brown planthopper, Laodalphax striatellus. So far, the studies have been mainly focused on the interaction between the host plant and the virus. In this study, for better comprehension of the interactions among the host plant, vector insect and plant-pathogenic virus, we investigated transcriptome of the vector insect and the differences between viruliferous and naïve L.striatellus. For this, naïve L. striatellus were collected from non-infected rice field and 50 L.striatellus of them were fed RSV-infected rice for 5 days. With the RSV-viruliferous and the naïve insects, we conducted Illumina RNA sequencing (Hiseq 2000) and obtained 175,243,488 and 146,031,348 reads from viruliferous and naïve L.striatellus, respectively. These reads were assembled into contigs and two transcriptome databases were generated. The transcriptome of naïve and RSV-viruliferous L. striatellus were campared to figure out up-regulated or down-regulated genes. These RSV-dependently regulated genes may have important function in the behavior of planthoppers or the transmission of RSV.

        Brown planthopper, Nilaparvata lugens is giving enormous damage to rice production. In this work, virulence of four Beauveria bassiana isolates against brown planthoppers was investigated by applying fungal granules on the water of pots, and we further examined the growth of hypha on the rice plants from the water surcae to explain the insecticidal mode of action. We used Beauveria bassiana (Bb) ERL 836, 1170, 1575 and 1578 isolates, which produced ~2×108 conidia/g of millet grain in a solid culture. Rice seeding was grown in breeding boxes at 28±2℃ for 5 days. Mycotized millet grains were treated on the water in a box at 1 g/box and the rice seeding was infested with 18~25 brown planthopper adults per box. A chemical pesticide and water treatment served as controls. Among the treatments, Bb ERL 836-treated plants had the lowest damage, rather than the other fungal treatments in laboratory assays. Hyphal growth on the stem of rice plants was observed in Petridish conditions under a fluorescent microscopy. This work suggests a possible control of brown planthoppers using entomopathogenic fungi.

        Among hemipteran insects which is the most important insect vector of plant viruses, small brown planthopper, Laodelphax striatellus, transmits the rice stripe virus (RSV) causing rice stripe disease. For effective control of RSV, it is important to understand interaction between RSV and L. striatellus. Therefore, in this study, expressed sequence tag (EST) databases were generated based on 454 GS-FLX pyrosequencing for comparative transcriptome analysis between nonviruliferous and RSV-viruliferous L. striatellus. By comparing the two EST libraries, we showed that 108 host genes were significantly up-regulated and 28 host genes were significantly down-regulated in viruliferous insects. Interestingly, genes encoding ribosomal proteins were mainly up-regulated in viruliferous L. striatellus, whereas genes related to translation were concentrated in the downregulated cohort. These RSV-dependently regulated genes may have important function in the behavior of planthopper or the transmission of RSV.

         ,  , The developmental period of Laodelphax striatellus Fallen, a vector of rice stripe virus (RSV), was investigated at ten constant temperatures from 12.5 to 35±1℃ at 30 to 40% RH, and a photoperiod of 14:10 (L:D) h. Eggs developed successfully at each temperature tested and their developmental time decreased as temperature increased. Egg development was fasted at 35℃(5.8 days), and slowest at 12.5℃ (44.5 days). Nymphs could not develop to the adult stage at 32.5 or 35℃. The mean total developmental time of nymphal stages at 12.5, 15, 17.5, 20, 22.5, 25, 27.5 and 30℃ were 132.7, 55.9, 37.7, 26.9, 20.2, 15.8, 14.9 and 17.4 days, respectively. One linear model and four nonlinear models (Briere 1, Lactin 2, Logan 6 and Poikilotherm rate) were used to determine the response of developmental rate to temperature. The lower threshold temperatures of egg and total nymphal stage of L. striatellus were 10.2℃ and 10.7℃, respectively. The thermal constants (degree-days) for eggs and nymphs were 122.0 and 238.1DD, respectively. Among the four nonlinear models, the Poikilotherm rate model had the best fit for all developmental stages (r<, SUP>, 2<, /SUP>, =0.98∼0.99). The distribution of completion of each development stage was well described by the two-parameter Weibull function (r<, SUP>, 2<, /SUP>, =0.84∼0.94). The emergence rate of L. striatellus adults using DYMEX<, SUP>, ®<, /SUP>, was predicted under the assumption that the physiological age of over-wintered nymphs was 0.2 and that the Poikilotherm rate model was applied to describe temperature-dependent development. The result presented higher predictability than other conditions.

        Rice stripe virus (RSV), the type member of the genus Tenuivirus, causes rice stripe disease and the viral transmission is mediated through the sucking by small brown planthopper, Laodelphax striatellus. Considerations have been mainly focused on the protection of rice from RSV and/or the planthopper, rather than the interaction between RSV and the insect. To clarify the interaction, in this work, mRNA was extracted from RSV-viruliferous planthopper with non-viruliferous control, and expressed sequence tag (EST) databases were generated based on 454 GS-FLX pyrosequencing technology for comparative analysis. RSV-viruliferous planthopper had ca. 2500 isotigs, which included genes on biological process (19%), cellular component (13%), molecular function (22%) and no hits (46%) from gene ontology (GO) analysis; this structure was similar to the control. However, in the viruliferous planthopper, 109 isotigs were up-regulated and 660 isotigs were down-regulated, compared to the non-viruliferous control. These RSV-dependently regulated genes may have important function in the behavior of planthopper or the transmission of RSV.

        In the previous study, the effect of orange-eyed color on mating of BPH was not clearly occurred, and the mating preference and multiple mating of female were cautiously suggested. To understand about the BPH female mate selection in the male-male competition-free conditions and the point of time for multiple mating, we designed four different combinations of the mate choice test with one virgin orange-eyed female (org/org) and two male BPH adults, orange-eyed male (org/org) and homozygous normal-eyed male (+/+). We gave the four different mating chances to female by the order of incubation (the first male and the following second male) and the incubation time (one day and days on end) of the second male. The distribution of the eye-color phenotype in F1 progeny and their hatching-order were observed in each mating combination. In the results, fourteen females out of twenty in four combinations produced progenies in accordance with mating chance priority of the first male, while three females of one combination selected the second male as a gamete. Interestingly, in three females of one combination with the continuous incubation of the second male, after eggs fertilized by the first male were continuously produced and then egg-laying was finished, progenies of the second male were started to be produced. From these results, it was suggested that mating order determines egg-laying order of female and the second mating of female would be occurred when sperm of the first male was almost exhausted in the reproductive organ.

        Role of flavonoids in plant against herbivores is well known. Many studies showed that flavonoids are deterrent or have anti-feeding effects to herbivores. In this study phenolic compounds were analyzed as defense materials to brown planthopper (BPH) feeding in four rice varieties using HPLC. Two of them, Chunambyeo and Nackdongbyeo, are BPH-susceptible, and the other two, Chungmo-1006 and Hanambyeo, are BPH-resistant. Changes in flavonoids were investigated by the function of damage period by BPH, Twelve flavonoids were characterized in the four varieties damaged by BPH. Among those flavonoids, apigenin and qurecetin were significantly higher in Chungmo-1006 damaged by BPH. Thus, resistance of Chungmo-1006 to BPH may be attributed to the increase of the two flavonoids contents.

        In the previous study, we found an orange-eyed mutant of the brown planthopper (BPH). And we confirmed that it’s orange phenotype is controlled by single recessive allele in the autosome. To understand the effect of the orange eye color on mating of BPH, we designed two different combinations of the mate choice test with three virgin female and male BPH adults. The one consisted of orange-eyed female (org/org), orange-eyed male (org/org), and homozygous normal-eyed male (+/+) (female mate choice). The other was composed of orange-eyed male (org/org), orange-eyed female (org/org), and homozygous normal-eyed female (+/+) (male mate choice). In female mate choice test, four mating types could be distinguished according to the distribution of the eye-color phenotype in F1 progeny and their hatching-order in each mating pair. Two mating types showed only one eye-color phenotype, normal and orange, respectively, and the other two produced both eye-color phenotypes in a different hatching-order. In male mate choice test, both phenotypes of offsprings were also produced in most mating pairs. From these results, the effect of eye color on mating of BPH was not clearly found, but the multiple mating in both sexes and the mating preference by female are cautiously suggested.

        An expression of adult wing form in reaction to rearing density during nymphal stage was investigated in the brown planthopper, Nilaparvata lugens. Under mass rearing condition, the S-BPH and the 2007-BPH population predominantly showed a short-winged, brachypterous, form and a long-winged, macropterous, form, respectively. At rearing density of less than 5 nymphs in the 2007-BPH, 90% of females showed brachypterous form, but all males became macropterous form. The ratio of macropterous form in the 2007-BPH males decreased by 75% in 10 to 15 rearing density, but increased by more than 95% again at 20 to 30 rearing density. In the case of the 2007-BPH females, the ratio of macropterous form gradually grew from 31% at 10 nymphal density to 92% at 20 nymphal density. All females originated from the S-BPH showed brachypterous form, regardless of nymphal density. The ratio of macropterous males in the S-BPH rapidly went down from 74% at 1 nymphal density to 10% at 10 nymphal density. At 20 nymphal rearing density, all males of the S-BPH showed brachypterous form. On the other hand, other brachypterous (OJ67-BPH) and macropterous (2006-BPH) population produced similar results with above the two populations at 1 nymphal rearing density. In summary, these results demonstrate that wing form dimorphism in N. lugens is largely influenced by nymphal density and the wing form at a specific density (low or high) can be different by sex or N. lugens populations.

        The small brown planthopper (SBPH), Laodelphax striatelleus (Fallén), is a insect vector of Rice stripe virus (RSV) in temperate countries such as Korea, Japan, China, and Taiwan. As SBPH is able to overwinter successfully in these areas, RSV disease in subsequent rice fields has been believed to be endemic. In Korea, however, the RSV disease outbreaks have been observed mainly but not continuously at some western regions since 2001, caused a severe damage to the rice production. Although many efforts are underway to explain the outbreak phenomenon, the exact related factors are not known yet. In the meantime of the study on SBPH population dynamics in 2009, we catched unusually large numbers of SBPH adults by aerial net traps, maximally over 900, in early June at western coastal counties such as Taean, Seocheon, Buan, Sinan, and Jindo in Korea. Age distribution changes of SBPH in winter and post-winter seasons at some selected fields shows that the adults might be not related to overwintering population. The adults of overwintering population emerged from early April. Newly hatched nymphs of first generation were found from mid-May. In late May, just before the unusual catch of adults, the developmental stages of SBPH were mostly below 5th instars. This means that the big adult populations would be results of mass migration of SBPH abroad. We present also spacial distribution and host relationship of overwintering population as well as viruliferous rate changes of immigratory population.
