        The objective of this study was to investigate the effect of bacterial contamination on elapsed time after preservation on boar semen. Known numbers of Escherichia coli (E. coli) were inoculated to freshly ejaculated semen and sperm parameters such as viability, motility, agglutination, acrosome integrity and hypo-osmotic swelling test were perform-ed during 7 days of liquid preservation. Semen samples were prepared using antibiotic free BTS extender and 4 di-fferent levels of E. coli were treated to semen with following concentrations; 3,000, 5,000, 7,000, 10,000 CFU/ml of sperms. Semen samples were preserved at 17℃ for 7 days in semen storage until analyzed. Aliquots were subjected to measure the sperm viability, motility and agglutination using computer assisted sperm analysis (CASA) system, acrosome integrity was performed using chlortetracycline (CTC) staining method and hypo-osmotic swelling test was performed using hypotonic solution from day 1 (day of semen collection) to 7. Detrimental effects on sperm motility and viability were observed 3 days after preservation at the level of 5,000 CFU/ml (p<0.05). Percentage of sperm abnor-mality was higher (p<0.05) in over 5,000 CFU/ml groups. Sperm agglutination rate was also significantly higher (p< 0.05) in groups of 5,000 and 7,000 CFU/ml. The rate of acrosome reacted sperm was higher as preservation time goes in all the samples but the pattern was clearly higher among E. coli contaminated groups (p<0.05). The sperm mem-brane integrity in terms of hypo-osmotic test, E. coli affects little compared to other sperm parameters. The deleterious effects observed due to the bacterial contamination in semen suggest that importance of hygiene protocol to minimize the bacterial contamination during semen collection and processing.

        우리 헌법상 기본권의 하나로 규정되고 있는 환경권은 개별 법령을 통하여 구체적 권리로 인정될 수 있다. 개별 법령에서 환경의 개념을 설정할 때, 지나치게 포괄적인 환경개념을 추구하는 것은 자칫 환경의 보전을 위한 노력을 공허하게 만들 우려가 있다. 이는 해양환경의 개념 설정에도 동일하게 적용될 수 있다. 다만, 해양환경은 육상 환경과 구별되는 특성이 두드러지는 바, 해양환경의 보전을 위한 해양환경정책은 기존 전통적인 환경정책과는 달리, 해양환경의 보전과 친환경적인 개발 그리고 참여와 협력의 거버넌스를 포함하는 통합적인 접근을 추구해야 한다. 따라서 해양환경의 독자성에 대한 이해를 기반으로 한 해양환경정책의 수립은 향후 육상과 해양을 아우를 단일 법전의 통합 환경법 제정에 있어 해양환경의 보전을 위한 실효적 법체계 구축의 초석이 될 것으로 기대한다.

        Maintaining and preserving mushroom mycelial cultures for ensuring the genetic, morphological, physiological traits and viability over time are essential elements in mushroom production. A number of different mushroom species can be preserved by many means, such as cryopreservation, freeze-drying, and placement in preservation solutions like mineral oil, distilled water, or glycerol. This study tested simple and low-cost preservation methods of the mushroom strains from various species, namely Pleurotus ostreatus, Agaricus bisporus, Cordyceps militaris. Each mycelium was cultured and preserved in different storage temperatures and methods. Temperature were differed at 4, 14, 24, 34°C at agar slants. Different storage methods were cryobial tube with sterile water, cryobial tube + 10% glycerol, agar slant of test tube, and agar slant + mineral oil at 4°C for 2 years. The mushroom cultures were better in the viability and recovery at 4°C than those of preservation at higher temperatures. Result showed that all the mushroom strains tested remained viable without any contamination under all different storage methods over two year duration of the experiment. However, all the strains studied could be best stored in sterile water with 10% glycerol for the viability.

        This study was carried out to investigate the general characteristics of semen such as semen volume, pH, sperm motility and sperm concentration of the semen collected from Shih Tzu dogs (age of 24 to 48 months, weight of 4 to 8 kg) by using the method of digital manipulation of the penis. The effect of preservation temperature and time on motility of fresh semen was also investigated in the present study. Semen was collected for 16 times from 4 male Shih Tzu dogs by multiple ejaculations (four times ejaculation per dog). The average of semen volume, semen pH, sperm motility and sperm concentration of the second fraction containing small volume of the initial third fraction per ejaculation were 2.11 ± 0.31 ml, 6.25 ± 0.07, 97.59 ± 1.03% and 2.05 ± 0.14 × 108 cells/ml, respectively. Average semen volume per ejaculate, semen pH, sperm motility and sperm concentration of the first fraction from the ejaculation were 1.12 ± 0.15 ml, 5.99 ± 0.14, 16.09 ± 6.18% and 5.16 ± 2.03 × 105 cells/ml, respectively. Those of second fraction were 2.07 ± 0.29 ml, 6.36 ± 0.13, 97.31 ± 1.36% and 2.15 ± 0.30×108 cells/ml, respectively. Those of third fraction were 2.60 ± 0.29 ml, 6.63 ± 0.08, 95.72 ± 1.61% and 6.03 ± 1.83 × 107 cells/ml, respectively. Sperm motility was significantly higher at 17℃ preservation temperature than at 5℃ or 36℃ during preservation period except 1 h preservation (P<0.05). When preservation temperature was 17℃, sperm motility was 96.69 ± 1.49% at 1 h, 91.38 ± 1.90% at 6 h, 88.38 ± 2.34% at 12 h, 78.13 ± 4.58% at 18 h, 58.44 ± 8.57% at 24 h and 29.56 ± 5.06% at 30 h, respectively.

        The study focuses on the quality assessment of Black Bengal buck semen preserved at chilled condition. In this in vitro trial, collected semen from Black Bengal bucks was preserved at chilling temperature (4▲5줛) in tris-glucosecitrate yolk medium of 1:5 ratios for four days. Artificial Vagina (AV) method was utilized to collect semen from buck. General evaluation of semen includes the color, mass activity and density were measured by direct visual examination. However, computer-assisted sperm analysis (CASA) and phase contrast microscopy were used to figure out the motility (%), hyper-activated (HYP) motility (%) and number of abnormal spermatozoa (%) initially, and at every 24 h intervals. The result revealed that spermatozoa preserved at chilling temperature showed significantly (P<0.05) lower motility and HYP motility with the progression of preservation. The number of phenotypically abnormal spermatozoa significantly (P<0.05) increased following preservation. Although significant positive correlation (r=0.945; P<0.05) was existed between % motile and % HYP motile spermatozoa however, the % of morphologically abnormal spermatozoa was negatively correlated with % motile (r=긏0.997; P<0.05) and % HYP motile spermatozoa (r=긏0.946; P<0.01). Therefore, we concluded that the quality of chilled semen progressively losses its viability and doesn…t remain useable after certain period of preservation with respect to its motility and morphology.

        The efficiency of artificial insemination (AI) for horses remains unsatisfactory. It is mainly because each process of AI causes a detrimental effect on semen quality. To sustain quality of semen properly, several factors including libido of stallions and sperm damage during sperm processing and preservation should be considered. Stallions with decent libido produce a high ratio of sperm to seminal plasma in their ejaculates, which is the ideal semen composition for maintaining sperm quality. Thus, to maximize the fertility rate upon AI, stallions should be appropriately managed to enhance their libido. Seminal plasma should have a positive effect on horse fertility in the case of natural breeding, whereas the effects of seminal plasma on both sperm viability and quality in the context of AI remain controversial. Centrifugation of semen is performed during semen processing to remove seminal plasma and to isolate fine quality sperm from semen. However, the centrifugation process can also result in sperm loss and damage. To solve this problem, several different centrifugation techniques such as Cushion Fluid along with dual and single Androcoll-ETM were developed to minimize loss of sperm and to damage at the bottom of the pellet. Most recently, a new technique without centrifugation was developed with the purpose of separating sperm from semen. AI techniques have been advanced to deliver sperm to optimal region of female reproductive tract at perfect timing. Recombinant equine luteinizing hormone (reLH) and low dose insemination techniques have been developed to maximize both fertility rate and the efficiency of AI. Horse breeders should consider that the entire AI procedure should be optimized for each stallion due to variation in individual horses for a uniformed AI protocol.

        Alpha-tocopherol as an antioxidant acts in preservation of chilled semen by preserving cell membrane damage from lipid peroxidation. Optimum concentrations of α-tocopherol in egg yolk-citrate (EYC) extender need to be studied in crossbred bull’s semen. Different concentrations of α-tocopherol viz. 0, 1, 2, 4 and 6mg per ml of extender were used. Semen was collected once a week from four bulls used to regular collection, aged 4 to 7 years, weighing 320 to 450 kg, and with body condition score 4 to 4.5 and scrotal circumference 23 to 32 cm. Semen was evaluated routinely and sperm morphology was viewed under light microscope at ×1,000 magnification after fixing with buffered formal saline. Over 90% had normal head, acrosome, mid-piece and tail. Semen was diluted with egg-yolk-citrate extender to produce 15×106 spermatozoa/ml and 0, 1, 2, 4 and 6 mg/ml α-tocopherol were added. The semens amples were kept at 8℃. Sperm motility and viability were examined daily up to 5 days under light microscopy at ×200 magnification. Sperm viability was acceptable (≥40%) up to the 4th day with all concentrations of α-tocopherol and up to the 5th day with 2 mg/ml α-tocopherol. Sperm motility was acceptable (≥40%) up to the 3rd day irrespective of α-tocopherol concentration, and up to the 4th day with 2 mg/ml α-tocopherol. It is suggested that the lifespan of chilled semen may be extended up to 4 days by adding 2mg/ml α-tocopherol.

        This research was carried out to find herbal preservatives for Makgeolli, as Makgeolli loses its commercial value due to overproduced acidic materials. When Makgeolli was kept at 25℃ to find the changes in acidity, total microbial cell number, yeast cell number, and bacterial species variety, a sudden increase of acidity as well as the disappearance of yeast cells occurred at day 6, and Makgeolli was changed to complete off-flavor. Acetobacter pasteurianus is the main acidifier in Makgeolli and shows a synergy effect in acid formation when cultured in combination with Lactobacillus casei. Among 12 herbs, the ethanol extract of Sutellaria baicalensis showed antimicrobial activity against A. pasteurianus, whereas the ethanol extract of Coptidis rhizoma showed antimicrobial activity against L. casei. Makgeolli added with Sutellaria baicalensis extracts demonstrated a lower acidity than that with Coptidis rhizoma extracts, which indicates that the inhibition of an acetic acid former is more important than that of a lactic acid former in Makgeolli preservation. Sutellaria baicalensis extracts prolonged the shelf life of Makgeolli by 1~2 weeks at a minimal inhibitory concentration (0.63 ㎎/㎖) during storage at 10℃.

        이 연구의 목적은 버려진 굴 패각을 재활용하여 감소시키는 것으로 굴 패각과 해조펄프를 이용하여 식품의 보존에 대한 항산화비드를 활용하는데 있다. 이 연구는 두 부분으로 나누었다. 첫 번째는 파일럿 규격조건으로 설계하고 제조하였다. 둘째는 [A],[B],[C-a],[C-b]화합물에 대한 최적제조 조건을 다루어 확립하고 이들 생성물을 분석하였다. 파일럿 규격 제조는 해조펄프의 분자수식합성과/Cl-전분 그리고 그의 다양한 비드 형태들이 극성용매에서 다양한 중량비율로 액상 혼합하여 제조되었다. 부가적으로, 혼합과정에서 해조펄프의 비율이 증가함으로서, 해조펄프/Ag-굴 패각을 섞인 것의 항균성은 감소되었으나 항산화와 비드의 견고성 성질은 증가되었다. 파일럿 규격의 생성라인은 해조 펄프와 Cl-전분을 분자수식으로 합성하였다. 그리고 극성용매를 사용하여 여러 가지 중량비율로 다양한 형태의 비드를 제조하였다.

        Historic buildings are history of human kind and a crystal of culture. Therefore, we have a responsibility and obligation to succeed to descendants. Once such heritage is damaged, it is hard to restore its original form. Thus, various attempts to preserve its value and authenticity have been made internationally. Among these, the international preservation tendency of historic building in terms of materials has been given to the preservation of materials as well as the preservation of appearance. In accordance with such trend, Korea also presented an equivalent general principle, and has managed historic buildings with the separate related instruction. The most general method to preserve historic buildings in Korea is a repair work. Since the foundation of Korea, the repair work tendency has been carried out in the principle of phenomenon change and original form maintenance. But, such tendency focuses on the appearance such as style, structure and trimming method, and there is insufficient consideration on the authentic preservation of materials. Accordingly, this study attempts to examine preservation principle and methods in the repair works, and also to investigate the afforestation condition for the restoration work. And, by suggesting preservation method of authentic materials, it aims to prepare the reference for the future repair work.

        Bacterial contamination reduces the semen quality, semen preservation, and cause of disease spread as well. Sperm fertility is essential factor of reproductive performance in swine. Sperm fertility is affected by semen quality such as sperm motility, abnormality, morphology, and rate of bacterial contamination. This study was conducted to determine the relationship between elapsed time after semen preservation on the changes of bacteria and semen quality. Semen was diluted with BTS extender without antibiotic for 7 days and sperm parameter and fertility were measured. Sperm motility was measured by CASA and total bacteria number was counted after 22 24 hr incubation from counting agar plate in which sperm dilute to 10 106 in 0.9% saline solution and inoculate to agar. Acrosomal integrity was measured by Chlortetracycline (CTC) staining. CTC patterns were uniform fluorescence over the whole head (pattern A), characteristic of uncapacitated acrosome-intact spermatozoa; fluorescence-free band in the post-acrosomal region (pattern B), characteristic of capacitated acrosome-intact spermatozoa; and almost no fluorescence over the whole head except for a thin band in the equatorial segment (pattern C), characteristic of acrosome reacted spermatozoa. Total number of bacteria was significantly increased (p<0.0001) 3 days after preservation. Sperm motility, viability, and morphological abnormality on elapsed time after preservation were lower from 5 (77.24±6.47, p<0.001) and 7 days (77.24±6.47, p< 0.001) after preservation compared to 1 (15.71±7.18) and 3 days(18.39±7.22) after preservation, respectively. Sperm viability was significantly lower (53.25±35.03, p<0.0001) at 7 days after preservation. Mohological abnormality of sperm was lower (p<0.001) at 1 (15.71±7.18) and 3 (18.39±7.22) days compared to (5 21.84±7.91) and 7 (22.59± 9.93) days after preservation. Acrosomal integrity and capacitation rate (pattern A) were significantly lower (p<0.001) from 5 days after preservation.

        France is known to be making particular efforts to maintain its traditional urban architectural culture by diverse measures including the implementation of urban policies. However, France is facing up to the need to pursue modern urbanization in keeping with the requirements of the current times. Thus, this paper examines with what priority France is attempting to reflect in its current urban policies its determination to retain its urban architectural tradition and to recreate its capital city of Paris as a future European hub city. To that end, the paper first seeks to analyze Paris' policies for urban architecture from diachronic perspectives in a bid to determine Paris' urban architectural culture. Second, the study attempts to examine within the purview of the paradigm of contemporary urban architectural designs how Paris is pursuing the two conflicting purposes of the preservation of tradition and modern urbanization through the Paris Local Urbanization Plan (or Plan Local d'Urbanisme [PLU]). First, the findings indicate that Paris is applying the principle of a sustainable development plan in all fields of environment, economy and society. In terms of environmental sustainability, Paris is trying to improve the life quality of its citizens through the establishment of efficient mass transportation systems and the expansion of its green belt areas. In terms of social sustainability, Paris is implementing policies to ensure social diversity through housing policies. Also, in terms of economic sustainability, Paris is trying to expand employment and bolster its urban functions by conserving commercial activities and developing peripheral urban areas. Second, the findings indicate that Paris' policy of recovering its traditions takes priority over that of creating a sustainable city.

        The standard does much to improve merchantable quality, distribution efficiency and fair dealings by shipping of the standard agricultural products. Mushrooms notified as the standard are five items; Pleurotus ostreatus, Pleurotus eryngii, Flammulina velutipes, Agaricus bisporus and Ganoderma lucidum. But many farmers are suffering from a strict standards. So these standard is required modification to fit farmhouse situations. This study was carried out to investigate mushroom quality of farm shipping and quality change during preservation at various treatment. Flammulina velutipes and Agaricus bisporus preserved at different temperature( 4℃, 10℃, 20℃) to investigate quality changes. Pileus diameters of Flammulina velutipes was 67% of first grade and 33% of second grade at the early stage. After two weeks, pileus diameters was not signigicant changes; 50% of first grade, 50% of second grade at 4℃ and 50% of second grade, 50% of third grade at 10℃. Although there were no significant changes of diameter at 20℃, most of the fruit bodies were decayed. In case of Agaricus bisporus, pileus diameter was 15% of first grade, 81% of second grade and 4% of third grade at the early stage. The more storage period longed, the more the color of the fruit body was browned. But pileus diameters was not signigicant changes. Hardness and chromaticity of these mushroom was declined as time passed. Now we are carrying out more research on some mushroom’s standards for improve consumer confidence.

        Geomorphological landscape of the border region in Gangwon province can be divided by coastal area in the east, mountainous area in the middle and east, and plain area in the west along DMZ (Demilitarized Zone). According to this landscape, various geomorphological units such as mountain landform, river landform, coastal landform and volcanic landform are surveyed. Because there are unique cultural, historical, and ecological heritages, the region has the potential for a global attraction. Therefore, the geopark as one of institutions of natural environment preservation in UNESCO is alternatives to the activated program for DMZ where has undergone a conflict of value between preservation and development, as well as to the preservation of geomorphological landscape in the border region.

        본 연구는 자연형 도시공원 생태자원의 효율적인 보전관리를 위한 기초자료를 제공하기 위하여 식생의 자연성 평가를 실시하였다.도시식생의 자연성 평가를 위하여 전형적인 중ㆍ소도시인 익산시의 5개 자연형 도시공원을 연구 대상지로 선정하였으며,자연성 평가를 실시한 결과는 다음과 같다.1. 익산시 5개 도시자연공원의 식물상은 84과 208속 261종 1아종 3품종 34변종의 총 299종류로 조사되었다.2. 익산시 5개 도시자연공원에 분포하는 식물들의 생활형의 특징은 H-D4-R5의 유형으로 나타났고 생육형에서는 직립형이우세하였다.3. 생활형 스펙트럼을 분석한 결과 M(소형지상식물): 20.0%±1.0%, N(미소지상식물): 10.2%±1.5%, E(착생식물): 2.5%±1.0%, Ch(지표식물): 5.2%±0.8%, H(반지중식물): 22.3%±2.5%, G(지중식물): 9.6%±1.1%, HH(수생식물): 4.5%±0.8%, Th(1년생식물):27.3%±3.7%로 나타났다.4. 식생 자연성 평가에서 III등급인 배산을 제외한 4개 도시자연공원은 IV등급이다. 전체적으로 익산시 주요 도시자연공원의자연성은 높지 않은 것으로 나타났다.
