Twenty two common millet (Panicum miliaceum L.) varieties collected from Korea, China and Russia were investigated for their phylogenetic relationship using 5S ribosomal DNA sequences with a hope to provide the basic information on their exact origin. Sequences of 5S rDNA were isolated by PCR. The primers, 5s-rRNA1 and 5s-rRNA2, were designed to isolate the complete NTS. Genomic DNA amplification produced two fragments with different length, 900 bp and 400 bp fragments, confirming the presence of two types of 5S rDNA repeats that differed from each other in the length of the NTS region. Amplified DNAs of 400 bp fragment were subcloned and used for further investigation. The obtained NTS sequences ranged from 200 to 300 bp and homology of sequences among plant materials was much higher than long repeat. CLUSTALW multiple aligment of 5S rDNA sequences from 22 different common millets revealed the clear difference by their origin. And critically different areas with insert or deletion were also confirmed. Those sequence difference seems to be used for discrimination of cultivars from different origin and use as molecular markers for origin identification. In phylogenic tree construction, the clear classification was shown where the genotypes from China and Russia is positioned together and stay away from domestic genotypes.
The cereal grain crops have strong flexibility against adverse environment and they have various functional compounds. The objective of the present study was to screen phenolic compounds in sorghum [Sorghum bicolor (L.) Moench], foxtail millet (Setaria italica), common millet (Panicum miliaceum L.) by high performance liquid chromatography (HPLC) with photodiode array (PDA) detector. Sorghum contained the highest amount of phenolic compounds among three different crops (sorghum, foxtail millet, common millet). Especially Moktaksusu showed the highest amount of phenolic compounds concentrations and biggest regional differences. The comparison of average phenolic compounds in sorghums by regions showed order to Milyang (963.3~;~mug~cdotg-1 ), Yeongyang (923.1~;~mug~cdotg-1 ), Gijang (831.3~;~mug~cdotg-1 ) and Bonghwa (735.6~;~mug~cdotg-1 ). Among the sorghum cultivars, Moktaksusu (1407.9~;~mug~cdotg-1 ) had the highest concentration of phenolic compounds. The average phenolic compounds of foxtail millets showed similar amount among Milyang (319.0~;~mug~cdotg-1 ), Gijang (288.1~;~mug~cdotg-1 ) and Bonghwa (281.9~;~mug~cdotg-1 ) areas. The phenolic compounds of Yeongyang (246.6~;~mug~cdotg-1 ) slightly low and that showed similar concentrations among three different regions. The concentration of phenolic compounds in foxtail millets, Chungchajo (335.6~;~mug~cdotg-1 ) showed the highest concentrations. The average phenolic compounds of common millets showed the highest concentrations in Milyang (305.5~;~mug~cdotg-1 ), Bonghwa (262.0~;~mug~cdotg-1 ), Gijang (195.1~;~mug~cdotg-1 ), Yeongyang (237.2~;~mug~cdotg-1 ) in decreasing order. The concentration of phenolic compounds of common millets was the highest in the Norangchalgijang (337.0~;~mug~cdotg-1 ), Hwanggumgijang (250.0~;~mug~cdotg-1 ) was also relatively higher than others. The results of this study will provide basic information for breeding sorghums, foxtail millets and common millets with higher phenolic compound concentrations.
경사도가 높거나 농지의 지형적 토성적 이유로 농기계의 작업효율이 낮은 한계경지는 산간지역에 많이 분포하고 있어 이와 같은 불량환경에 적응성이 큰 조, 기장, 수수의 생육 및 수량특성을 검정하여 한계경지 적응 적정 작목 및 품종을 선발코자 실시한 연구결과는 다음과 같다. 1. 토양 pH는 평탄지인 대조구가 7.85로 약 알카리성을 나타내었으며, 경사지 및 척박지는 각각 6.3, 6.2로 약산성을 나타내었으나, 자갈밭은 5.1로 강한 산성을 나타내었다. EC는 대조구에 비해 한계농경지(경사지, 척박지, 자갈밭)가 낮았으며, 특히 척박지는 0.05 dS m1로 가장 낮았다. 2. 조의 한계경지별 출수기 및 성숙기는 큰 차이가 없었다. 간장은 경사지에서 169.5 cm로 가장 컸으며, 자갈밭이 143.7 cm로 가장 작았다. 1수립수는 대조구를 제외하면 경사지가 4913.9립으로 많았으며, 3.3 m2 당 이삭수는 자갈밭이 85.3이삭으로 대조구에 비해 25% 적었다. 품종별 생육기간은 3품종 모두 112~113일로 차이가 없었다. 3. 기장의 파종기로부터 출수소요일은 한계경지간 큰 차이가 없었으며, 1수립수는 경사지가 787.1립으로 대조구 다음으로 가장 많았다. 품종간 1수립수는 벼룩기장이 827.2개로 가장 많았으나, 1000립중은 5.5g으로 황기장 6.2g에 비해 작았다. 4. 한계경지별 수수의 출수기 및 성숙기는 큰 차이가 없었다. 1수립수는 대조구와 경사지가 각각 2598.1, 2563.8개로 가장 많았으며, 이삭수도 대조구와 경시지가 각각 26.7, 26.0이삭/3.3 m2 으로 가장 많았다. 품종별 생육기간은 3품종 모두 122~123이로 큰 차이가 없었으며, 1수립수는 목탁수수가 2357.6립으로 가장 많았으며, 메수수가 2071.8개로 가장 적었다. 1000립중은 장목수수가 23.8g으로 가장 무거웠다. 5. 조의 수량은 대조구가 295 kg/10a으로 가장 많았으며, 다음으로는 경사지가 282.0 kg/10a으로 많았다. 품종별로는 몽당조가 252.3 kg/10a으로 수량이 가장 많았다. 기장의 수량은 대조구가 217.0 kg/10a으로 가장 많았으며, 다음으로는 경사지가 196.0 kg/10a으로 많았다. 품종별로는 벼룩기장이 173.8 kg/10a으로 가장 많았으며, 흰기장이 122.2 kg/10a으로 수량이 가장 낮았다. 수수의 수량은 대조구가 313.0 kg/10a으로 가장 많았으며, 다음은 경사지가 301.7 kg이었으며, 품종별로 는 목탁수수가 236.5 kg/10a로 가장 수량이 많았다.
수년 동안 국내 전역에서 수집된 기장 137 유전자원을 공시하여 농업적 특성조사를 통한 품종육성의 기초자료를 제공코자 수행하였던 바 그 결과를 요약하면 다음과 같다. 1. 공시된 137자원의 수형은 편수형, 산수형, 편밀수형(편수형과 밀수형의 중간형태) 및 산편수형(산수형 과 편수형의 중간 형태)의 4가지 수형으로 구분되었다. 수형별 빈도를 보면 편수형이 91자원으로 66.4%를 차지하였다. 2. 종피색은 황백색, 황갈색, 흑갈색, 황색 및 갈색 5가지로 분류되었으며, 종피색별 빈도를 보면 황백색 이 54자원 39.4%로 가장 많았으며, 종실형태는 방추형이 62.8%, 단방 추형이 37.2%이었다. 3. 출수소요일은 68~85 일의 범위를 나타내어 변이폭이 컷으며, 평균은 73.7일이었다. 빈도별 분포는 70~75 일이 46.7%로 가장 많았다. 생육기간은 109~124 일의 범위를 나타내었고, 평균 112일이었다. 생 육기간의 빈도를 보면 111~115 일이 83자원으로 60.6%로 가장 많았다. 4. 간장의 분포는 181~190cm 가 65자원으로 47.4%로 가장 많았으며, 160 cm 이하가 2.9%로 가장 적었다. 수장은 156~201cm 의 범위를 보였고, 평균 181.4 cm이었으며, 빈도별 분포는 31~35cm 가 74자원으로 54.0%로 가장 많았다. 5. 1수립수의 범위는 432~2,665 개로 변이폭이 상당히 컷으며, 평균은 1,429.5개이었다. 1수립수 분포는 1,200~1,400 개가 20.4%로 가장 많았으며, 1,000개 이하 및 2,000개 이상이 각각 13.1%, 8.6%를 차지 하였다. 6. 등숙률은 10% 이하에서 90% 이상까지 변이의 폭이 매우 컷다. 1000립중은 범위가 2.2~6.0g 으로 변 이폭이 상당히 컷으며, 평균은 4.69 g이었다. 7. 간장은 수장 및 출수소요일과 유의한 정의 상관관계를 나타내었으며, 생육기간은 1000립중과 유의한 정의 상관관계를 나타내었다. 등숙률도 1000립중과 유의한 정의 상관관계를 보여 등숙률이 높은 자 원일수록 1000립중이 높았다.
Crop characteristics of 137 common millet(Panicum miliaceum L.) resources collected in Korea were investigated in order to establish basic data for the improvement of crop breeding. Spike types of 137 plant resources were classified as 4 types of oblate, spreading, oblate-compact(intermediary type between oblate and compact) and oblate-spreading(intermediary type between oblate and compact) type, of which oblate spike type was the highest percentage of 66.4%(91 plant resources) of 137 germplasm. Seed color was classified as 5 colors of dark brown, yellow, brown, yellowish brown, and yellowish white, of which yellowish white was the highest percentage of 66.4%(54 plant resources) among them. Seed type was classified as 2 types of pyramid shape and short pyramid shape, which showed 62.8% and 37.2% respectively. Days from seeding to heading date showed the range from 68 to 85 days with 73.7 days in the average and the highest frequency proportion of it was the group from 70 to 75 days, which occupied 46.7%(64 plant resources) of 137 germplasm. Days from seeding to physiological maturity showed the range from 109 to 124 days with 112.0 days in the average and the highest frequency proportion(60.6%) of it was the group from 111 to 115 days, while there were also occupied with 6.6% below 110 days and 5.1% over 120 days among 137 germplasm. Group from 181cm to 190cm of culm length showed the highest frequency proportion of 47.4%, whereas below 160cm was the lowest occupancy of 2.9%. Spike length showed the range from 181 to 190cm with 181.4cm in the average and the highest frequency distribution of it was the group from 31 to 35cm with the proportion of 54.0%. Number of grains per spike showed the range from 432 to 2,665 grains with 1,429.5 grains in the average and group from 1,200 to 1,400 grains per spike was the highest frequency distribution with the proportion of 20.4%. There were also occupied with 13.1% below 1,000 grains and 8.6% over 2,000 grains per spike among 137 germplasm. Culm length and spike length were positively significant correlation coefficient with days from seeding to heading date. Days from seeding to physiological maturity and percentage of ripened grains were highly significant positive correlation coefficient with 1000 grain weight, showing that the longer and higher the days from seeding to physiological maturity and percentage of ripened grains, the higher 1000 grain weight.
Amino acid composition of proteins in Italian millet, Common millet and sorghum were invstigated by HCI hydrolysis method. The optimum condition was obtained by hydrolysis at 110 for 24hr. As major amino acids from protein hydrolyzate, the content of tyosine, arginine and phebylalanine were 7.06%, 6.79% and 6.44%, respectively. The content of glutamic acid in Common millet, Italian millet and Sorghum were 5.73%, 5.64% and 5.46%, respectively. Glycine content was about 2.93% in three samples. Contents of crude protein and pure protein in Italian millet, Common millet and sorghum were determined by micro-kjeldahl method. Crude protein contents were slightly higher than that of pure protein. Protein content of sorghum was higher than those of Italian millet and Common millet. For SDS-PAGE analysis, Italian millet showed more soluble proteins including 50kDa, 30kDa and smaller proteins than other cereals. In particular, Common millet and Sorghum only solubilized proteins less than 15kDa.
