인삼근에 있어서 신아의 휴면타파 및 맹아과과에 대한 생리기구 해명을 위한 기초자료를 얻고자 일년생근의 저온처리 및 맹아과정중 ABA 함량 및 GA활성의 변화를 처리단계별로 조사하여 휴면 및 맹아와의 관계를 실험하였다. 1. 4℃ 처리후 15℃ 처리에서의 맹아률은 30일 처리에서 35%, 60일 이상 처리에서 100%이었으나 4℃ 나 15℃ 에서 연속처리한 경우에는 전혀 맹아하지 않았다. 맹아시 및 50% 맹아일수는 저온처리기간이 길어 질수록 저온처리후부터의 일수가 짧아졌으며, 매아시부터 50% 맹아까지의 일수도 단축되었다. 2. 두부를 포함한 근상부에서의 ABA함량은 4℃ 나 15℃ 에서 모두 처리기간이 경과함에 따라 점차 증가하였으며 그 증가정도는 15℃ 에서 높았다. 근하부에서의 ABA함량은 처리기간중 15℃ 에서 미미한 증가를 보였으나 4℃ 에서는 거의 변화가 없었다. 또, 4℃ 에서 60일처리후 15℃ 의 처리기간중 ABA함량은 근상부에서는 크게 증가하였으나 근하부에서는 오히려 다소 감소하였다. 3. 근상부에서의 GA활성은 4℃ 나 15℃ 에서 모두 점차 계속 감소하였는데, 그 감소정도는 15℃ 에서 컸다. 근하부에서의 GA활성은 근상부 및 근하부에서 모두 크게 증가하였는데, 특히, 15℃ 처리기간중에는 근의 부위에 관계없이 Rf치가 낮은 쪽에서는 증가하였으나 높은 쪽에서는 상대적으로 감소하였다. 4. 이상의 결과를 종합하여 보면 인삼근에 있어서 신아의 휴면타파 및 맹아는 ABA와는 별로 관계가 없고 온도숙건에 지배되어 나타나는 GA의 생합성 능력과 그 생합성에 의한 활성증가와 밀접한 관계가 있는 것으로 추정된다.
The present study was carried out to get the basic information for clarifying physiological mechanism of breaking dormancy and sprouting in Panax ginseng roots. Changes in Abscisic acid (ABA) content and Gibberellin (GA) activity were investigated in one-year-old root during stratification at 4~circC . 15~circC . and 15~circC after 60day-treatment at 4~circC . Sprouting rate at 15~circC was 35% in 30days storage at 4~circC and 100% in longer than 60days, but there was no sprout in both the constant treatment at 4~circC or 15~circC regardless of the treatment period. The longer the period of low temperature treatment. number of days to the first and 50% sprouting was shortened, and number of days to 50% from first sprouting was also shortened. ABA content in the upper part of root(contained bud) was gradually increased at both 4~circC and 15~circC as the treatment period was extended. and the degree of increase was higher at 15~circC . In the lower part. it showed a slight increase at 15~circC . while showed little change at 4~circC throughout the treatment period. In the 15~circC treatment after 60days at 4~circC , it was greatly increased in the upper part. while rather slightly decreased in the lower part of root. GA activity in the upper part was gradually decreased at both 4~circC and 15~circC , and the degree of decrease was higher at 15~circC . In the lower part. it was similar tendency to those in the upper part. In the 15~circC treatment after 60days at 4~circC . it was remarkably increased in both the upper and lower part. The increase was great in the low Rf region, while the decrease appeared relatively in the high Rf region compared to those of 60day-treatment at 4~circC . The above results indicated that the breaking dormancy and sprouting of bud were closely associated with the degree of GA activities in response to temperature condition .during stratification rather than the direct effect associated with the changes in ABA content.
