기주목(상수리나무, 밤나무, 자작나무, 버드나무)별 겨우살이 ethanol추출물과 5가지 분획물 (hexane fr., chloroform fr., ethyl acetate fr., butanol fr., aqueous fr.)을 이용하여 in vitro에서 DPPH를 이용한 수소전자공여능, DNA에 대한 산화적 손상 및 손상억제 효과 등에 미치는 영향을 연구하기 위하여 comet assay를 실시하였다. 4종의 한국산 겨우살이의 ethanol추출물과 분획물 중 ethyl acetate fr.들이 높은 수소전자공여능을 보였다. Comet assay분석에서 head DNA의 형태를 유지하거나 또는 tail DNA의 생성을 억제하는 겨우살이 fr.들이 DNA에 대하여 항산화적 기능이 있음을 보여 주었다. 또는 comet assay 유용한 parameter인 tail length(μm)는 positiye control인 H2O2 (10-3M)로 40min동안 세포에 처리하여 산화적 손상을 준 결과, 149μm의 tail length를 보였으며 , 상수리나무 겨우살이의 ethyl acetate fr.(0.1mg/ml)을 H2O2 (10-3M)와 동시 처리에는 110μm, 자작나무 겨우살이 ethyl acetate fr.은 104μm, 상수리나무 겨우살이 butanol fr.은 123μm, 자작나무 겨우살이 butanol fr.은 118μm로 tail length가 감소하는 것으로 보아 겨우살이가 산화적 DNA손상을 방어한다는 것을 알 수 있었다. 이상의 결과로 볼 때 한국산 겨우살이는 산화적 stress를 일으키는 활성 산소 자유라디칼의 생성에 대해 보호활성을 나타내어 유용한 chemopreventive agent의 기능이 기대된다.
Korean mistletoes extracts were investigated for in vitro antioxidation activity, with 1,1-diphenyl-2-picrylhydrazine(DPPH), and an inhibitory effect on oxidative DNA damage by using comet assay. The Korean mistletoes were 4 different kinds classified by their host plants (Korean Viscum sp. in Quercus acutissima Carr., Korean Viscum sp. in Castanea crenata, Korean Viscum sp. in Betula platyphylla, and Korean Viscum sp. in Salix koreensis). The samples were extracted with ethanol, and fractonationed with n-butanol, ethyl acetate, chloroform, n-hexane, and second distilled water. Among them, ethyl acetate fraction from Korean Viscum sp. in Betula platyphylla showed the strongest activities to electron donating ability on 1,1-diphenyl-2-picrylhydrazyl(DPPH) and the protective effect on oxidative DNA damage.
