본 연구는 t-butyl hydroperoxide(t-BHP)의 산화적 손상에 대한 원지(Polygalae Radix, PR)추출물의 항산화 효과를 알아보기 위하여 배양 C6 glioma 세포를 재료로 세포생존율을 비롯하여 superoxide dismutase(SOD) 유사 활성을 비롯하여 lactate dehydrogenase(LDH) 활성을 조사하였다. 본 실험에서 t-BHP는 농도 의존적으로 배양 C6 glioma 세포의 생존율을 유의하게 감소시켰다. 한편, t-BHP의 세포독성에 대한 원지 추출물의 항산화 효과의 정량분석에 있어서 원지 추출물은 t-BHP의 산화적 손상으로 감소된 세포생존율의 유의한 증가를 비롯하여 LDH 활성감소 및 SOD 유사 활성을 보임으로서 항산화 효과를 나타냈다.
The purpose of this study is to elucidate the antioxidant effect of Polygalae Radix (PR) extract on the cultured glioma cells damaged by reactive oxygen species(ROS). For this study, the C6 glioma cells were exposed to various concentrations of t-butyl hydroperoxide (t-BHP). And also, the antioxidant effect of PR extract on the t-BHP-mediated cytotoxicity was measured by the cell viability, lactate dehydrogenase (LDH) activity and superoxide dismutase (SOD)-like activity. In this study, the cell viability of C6 glioma cells was decreased according to the treated dosage of t-BHP, and XTT90 and XTT50 values were determined at 25 and 60 µM of PR extract, respectively. And also, t-BHP was highly toxic in this culture for XTT50 value was lower than 100 µM by the toxic criteria. In the antioxidant effect of PR extract against the the cytotoxicity induced by t-BHP, PR extract significantly increased the cell viability compared with t-BHP-treated group. PR extract also showed not only the decrease of LDH activity but also the increse of SOD-like activity. From these results, it is suggested that t-BHP-like ROS was highly toxic on cultured C6 glioma cells, and also, PR extract showed antioxidant effect on t-BHP-induced oxidative damage by the decrease of LDH activity and the increase of SOD-like activity.
