In spite of medicinal importance, Astragalus membranaceus, a perennial herbal plant, has been little studied for its molecular breeding. In this study, in planta genetic transformation using its seedling was performed by means of agroinfiltration. In the analysis of fluorometric and histochemical GUS assay, it was determined that A. membranaceus seemed not very susceptible to Agrobacterium infection. However, a successful transformation could be achieved when 2 to 3 days-old seedlings, prior to agroinfiltration, were moderately wounded by NaOH and hydrogen peroxide treatment. The results can suggest an efficient way for developing a more desirable A. membranaceus cultivars in light of cultivation as well as functional properties.
