치자에서 분리한 황색색소에 대한 랫드 및 토끼에 있어서의 단회투여 경구 독성시험을 수행하였다. 황색색소는 랫드의 경우 5, 000, 2, 500, 1, 250, 625, 312.5 및 0㎎/㎏의 용량으로 경구로, 토끼에는 5, 000, 2, 500 및 1, 250㎎/㎏의 용량으로 경구로 암·수 각각에 1회 투여한 후 14일 관찰하였다. 시험결과에 있어 랫드 및 토끼의 모든 황색색소 투여군에서 사망예, 임상증상, 체중변화, 육안적 소견 및 병리조직학적 소견에서 특기할만한 이상은 관찰되지 않았으며 랫드와 토기에서 황색색소의 LD_50치를 산출할 수 없었다. 따라서 LD_50치는 랫드 암·수 모두에서 체중 ㎏당 5, 000㎎ 이상인 것으로 판단되었으며, 토끼 암·수 모두에서 체중 ㎏당 5, 000㎎ 이상인 것으로 판단되었다. 이상의 결과로부터 치자로부터 분리한 황색색소는 랫드 및 토끼에서 단회투여 경구 독성시험에서 어떠한 독성 및 부작용을 유발하지 않는 안전한 제제로 사료된다.
This study was performed to evaluate single-dose toxicity of the gardenia yellow pigment in Sprague-Dawley rats and New-Zealand White rabbits via oral routes. The yellow pigment was administered in rats at does levels of 5, 000, 2, 500, 1, 250, 625, 312.5㎎/㎏ and 0㎎/㎏. And also yellow pigment was administered in rabbits at does levels of 5, 000, 2, 500, 1, 250㎎/㎏ and 0 unit/㎏. The rats and rabbits of both sexes were observed daily for 14 days after single oral administration. Yellow pigment treated rats and rabbits did not induce any mortalities and abnormal signs in clinical findings, body weights, gross findings and histopathological finding. Based on these results, it is impossible to estimate LD_50 values in rats and rabbits. Therefore, it was concluded that gardenia yellow pigment have no effect on acute toxicity and side effect in rats and rabbits.
